    A novel framework to harmonise satellite data series for climate applications

    Fundamental and thematic climate data records derived from satellite observations provide unique information for climate monitoring and research. Since any satellite operates over a limited period of time only, creating a climate data record requires the combination of space-borne measurements from a series of several (often similar) satellite sensors. A simple combination of calibrated measurements from several sensors, however, can produce an inconsistent climate data record. This is particularly true of older, historic sensors whose behavior in space was often different from their behavior during pre-launch calibration in the laboratory. More scientific value can be derived from considering the series of historical and present satellites as a whole. Here we consider harmonisation as a process that obtains new calibration coefficients for revised sensor calibration models by comparing calibrated measurements over appropriate satellite-to-satellite match-ups, such as simultaneous nadir overpasses. When we perform a comparison of two sensors, however, we must consider that those sensors are not observing exactly the same Earth radiance. This is in part due to differences in exact location and time tolerated by the match-up process itself, but also due to differences in the spectral response functions of the two instruments, even when nominally observing the same spectral band. To derive a harmonised data set we do not aim to correct for spectral response function differences, but to reconcile the calibration of different sensors given their estimated spectral response function differences. Here we present the concept of a framework that establishes calibration coefficients and their uncertainty and error covariance for an arbitrary number of sensors in a metrologically-rigorous manner. We describe harmonisation and its mathematical formulation as an inverse problem. Solving this problem is challenging when some hundreds of millions of match-ups are involved and the errors of fundamental sensor measurements are correlated. We solve the harmonisation problem as marginalised errors in variables regression. The algorithm involves computation of first and second order partial derivatives, for which the corresponding computer source code is generated by Automatic Differentiation. Finally we present re-calibrated AVHRR radiances from a series of 10 sensors. It is shown that the new time series have much less match-up differences while being consistent with uncertainty statistics

    Alternative particle formation pathways in the eastern tropical North Pacific's biological carbon pump

    A fraction of organic carbon produced in the oceans by phytoplankton sinks storing 5‐15 gigatonnes of carbon annually in the ocean interior. The accepted paradigm is that rapid aggregation of phytoplankton cells occurs forming large, fresh particles which sink quickly; this concept is incorporated into ecosystem models used to predict the future climate. Here we demonstrate a slower, less efficient export pathway in the Eastern Tropical North Pacific. Lipid biomarkers suggest the large, fast‐sinking particles found beneath the mixed layer are compositionally distinct from those found in the mixed layer and thus not directly and efficiently formed from phytoplankton cells. We postulate they are formed from the in situ aggregation of smaller, slow‐sinking particles over time in the mixed layer itself. This export pathway is likely widespread where smaller phytoplankton species dominate. Its lack of representation in biogeochemical models suggests they may be currently over‐estimating the ability of the oceans to store carbon if large, fast‐sinking, labile particles dominate simulated particle export. Plain Language Summary The oceans are one of the largest sinks of atmospheric carbon dioxide on our planet. One method by which this occurs is through the production of organic material (phytoplankton ‐ plant‐like cells) in the surface ocean, which capture atmospheric carbon dioxide during photosynthesis. Eventually, the phytoplankton die and sink out of the surface ocean, transporting huge amounts of carbon to the deep ocean where it is stored for centuries or even millennia. Our current understanding is that generally, most organic material sinks quickly as large, fast‐sinking (100s of metres per day) particles (clumps of dead phytoplankton cells). However in our study in the Equatorial Pacific Ocean we were able to show that a different and much slower process occurs where phytoplankton first aggregate to smaller, slower sinking detrital particles and eventually form, very degraded larger particles that sink to the deep. This has consequences for estimating ocean carbon storage as smaller particles are respired much quicker than larger particles. Thus where they are an important part of this carbon sink, such as in the Equatorial Pacific, the proportion phytoplankton‐captured atmospheric carbon dioxide being stored in the deep ocean is likely reduced

    Testing variational estimation of process parameters and initial conditions of an earth system model

    We present a variational assimilation system around a coarse resolution Earth System Model (ESM) and apply it for estimating initial conditions and parameters of the model. The system is based on derivative information that is efficiently provided by the ESM's adjoint, which has been generated through automatic differentiation of the model's source code. In our variational approach, the length of the feasible assimilation window is limited by the size of the domain in control space over which the approximation by the derivative is valid. This validity domain is reduced by non-smooth process representations. We show that in this respect the ocean component is less critical than the atmospheric component. We demonstrate how the feasible assimilation window can be extended to several weeks by modifying the implementation of specific process representations and by switching off processes such as precipitation

    Coccolithophore calcification fails to deter microzooplankton grazers.

    Phytoplankton play a central role in the regulation of global carbon and nutrient cycles, forming the basis of the marine food webs. A group of biogeochemically important phytoplankton, the coccolithophores, produce calcium carbonate scales that have been hypothesized to deter or reduce grazing by microzooplankton. Here, a meta-analysis of mesocosm-based experiments demonstrates that calcification of the cosmopolitan coccolithophore, Emiliania huxleyi, fails to deter microzooplankton grazing. The median grazing to growth ratio for E. huxleyi (0.56 � 0.40) was not significantly different among non-calcified nano- or picoeukaryotes (0.71 � 0.31 and 0.55 � 0.34, respectively). Additionally, the environmental concentration of E. huxleyi did not drive preferential grazing of non-calcified groups. These results strongly suggest that the possession of coccoliths does not provide E. huxleyi effective protection from microzooplankton grazing. Such indiscriminate consumption has implications for the dissolution and fate of CaCO3 in the ocean, and the evolution of coccoliths

    Esta investigação inscreve-se dominantemente no âmbito dos estudos da criança e ancora a sua área específica nas questões linguísticas, assumindo os pressupostos teóricos e metodológicos da análise do discurso (de tradição francófona), considerando também contributos de áreas conexas. Analisa textos de manuais/livros escolares usados no sistema de ensino português, pretendendo tornar visíveis as imagens que estes constroem das crianças e da infância, bem como os recursos linguísticos mais relevantes associados a tal construção. Procura desvendar a representatividade das tipologias textuais nos manuais e de que forma estas contribuem para a construção das referidas imagens. Conclui que as crianças dos textos são seres sem voz e sem poder, consumidoras, mas não coprodutoras de cultura, com os textos inculcando valores conformadores da manutenção da ordem social estabelecida, sendo a forma como as tipologias textuais estão organizadas contribuidora para se alcançar esse propósito.This research forms primarily part of the child studies and is based on its specific field of linguistic issues, assuming the theoretical and methodological assumptions of the speech analysis (of French-speaking tradition), having also regard the inputs of related fields. It analyses texts from handbooks/textbooks used on the Portuguese educational system, intending to reveal pictures that these books construct about children and childhood, as well as the most relevant linguistic resources associated with such construction. This paper aims at unravelling the representation of the text typologies in the handbooks and how these can contribute for building of such pictures. From this, one can conclude that children from the texts are voiceless and have no power, they are consumers but not co-producers of culture, with these texts instilling conservative values for maintaining the established social order, but also that the way texts typologies are organized to contribute for reaching that propose.Esta investigación se ensere en el ámbito de los estudios del niño y más precisamente en la área de las cuestiones lingüísticas, asumiendo los presupuestos teóricos e metodológicos de la análisis del discurso (de tradición francófona), considerando también aportes de las áreas conexas.     Geographical, seasonal, and depth variation in sinking particle speeds in the North Atlantic

    Particle sinking velocity is considered to be a controlling factor for carbon transport to the deep sea and thus carbon sequestration in the oceans. The velocities of the material exported to depth are considered to be high in high-latitude productive systems and low in oligotrophic distributions. We use a recently developed method based on the measurement of the radioactive pair 210Po-210Pb to calculate particle sinking velocities in the temperate and oligotrophic North Atlantic during different bloom stages. Our estimates of average sinking velocities (ASVs) show that slowly sinking particles (<100?m?d?1) contribute significantly to carbon flux at all the locations except in the temperate regions during the bloom. ASVs appear to vary strongly with season, which we propose is caused by changes in the epipelagic community structure. Our results are the first field data to confirm the long-standing theory that particle sinking velocities increase with depth, with increases of up to 90% between 50 and 150?m depth

    AAV-mediated in vivo knockdown of luciferase using combinatorial RNAi and U1i

    RNA interference (RNAi) has been successfully employed for specific inhibition of gene expression; however, safety and delivery of RNAi remain critical issues. We investigated the combinatorial use of RNAi and U1 interference (U1i). U1i is a gene-silencing technique that acts on the pre-mRNA by preventing polyadenylation. RNAi and U1i have distinct mechanisms of action in different cellular compartments and their combined effect allows usage of minimal doses, thereby avoiding toxicity while retaining high target inhibition. As a proof of concept, we investigated knockdown of the firefly luciferase reporter gene by combinatorial use of RNAi and U1i, and evaluated their inhibitory potential both in vitro and in vivo. Co-transfection of RNAi and U1i constructs showed additive reduction of luciferase expression up to 95% in vitro. We attained similar knockdown when RNAi and U1i constructs were hydrodynamically transfected into murine liver, demonstrating for the first time successful in vivo application of U1i. Moreover, we demonstrated long-term gene silencing by AAV-mediated transduction of murine muscle with RNAi/U1i constructs targeting firefly luciferase. In conclusion, these results provide a proof of principle for the combinatorial use of RNAi and U1i to enhance target gene knockdown in vivo
