    Adsorption of C2H2-C2O3 on Si(001)

    The adsorption of maleic anhydride on the Si(001) surface was investigated using the first-principles pseudopotential formalism. Our results indicate that C2H2-C2O3 adsorbs through a [2+2] cycloaddition of the C-C bond to the dangling bonds of Si-Si dimers, as observed for other small hydrocarbons. According to our calculations, the adsorption of a second maleic anhydride molecule will preferentially occur on first-neighbours Si-Si dimer sites in either the dimer chain our dimer row, i.e. considering a (4Ă—2) reconstruction, C2H2-C2O3 can either form a linear chain or a zig-zag chain, depending on the growth conditions. Although both structures are found to be stable from the energetic point of view, only our STM theoretical images for the linear chain are comparable to available experimental data

    Dietary fatty acids on subcutaneous adipose tissue modulation in transition dairy goats

    The goal of the present study was to evaluate the metabolic and immune response of peripartal dairy goats to dietary supplementation with fish oil or stearic acid. 15 multiparous alpine dairy goats were involved in the trial. Starting from the last week of gestation until 3 weeks after kidding date the experimental diets, based on alfalfa and mix hays and a concentrate mix, were added either with protected fish oil (FO) or with stearic acid (ST). Feed intake, body weight, energy balance, milk production and composition were measured weekly. Adipose tissue biopsies were performed on day -7, 7 and 21 relative to kidding date and samples were immediately fixed in formalin, paraffin embedded and Hematoxilin Eosin stained. The results discussed in the present work are relative to a subsample of 8 goats, representative of the two experimental groups. Hematological and histological data were analyzed by a Generalized Estimating Equation (GEE) in IBM SPSS 19.0 was used. Production parameters were analyzed by a MIXED repeated model in SAS 9.2. No differences were observed between FO and ST in milk production, BCS, weight, dry matter intake and milk components except for a higher milk protein percentage in the 7 to 21 d period for ST. BHB serum content was higher in ST overall the experiment, whereas NEFA and ALAT serum content were higher at day 7 in FO compared to ST (P < 0.08). ALAT was higher also at day 21 in FO. Treatment had no effect on blood cellular component except for WBC in FO group, where a significant decrease at 7 d was observed. WBC and HCM parameters were in the physiological range for dairy goats during transition period. Histologic adipose tissue analysis revealed a significant decreased adipocytes surface between -7 and 21 d in ST, whereas in FO the adipocyte surface reduction was related to the -7 to 7 d interval reaching a plateau until day 21. The EB pattern and the NEFA serum content at 7d in particular for FO are well correlated with histologic observations indicating goats were using fat depots to cope their negative energy balance. NEFA levels did not confirm the histological evidence at day 21 for ST suggesting a possible different action on subcutaneous adipose tissue during time. Results suggest a modulation in lipid storage management during peripartal negative energy balance by saturated vs. unsaturated dietary fatty acid supplementation that did not affect production levels of goats

    New insights on Flavescence dor&#233;e phytoplasma ecology in the vineyard agro-ecosystem in southern Switzerland

    Phytoplasmas associated with Flavescence dor\ue9e (FDp) grapevine disease are quarantine pathogens controlled through mandatory measures including the prompt eradication and destruction of diseased plants, and the insecticide treatments against the insect vector, the ampelophagous leafhopper Scaphoideus titanus. In the present study, a multidisciplinary approach has been applied to investigate the FDp ecological cycle in a test vineyard agro-ecosystem in Canton Ticino, south Switzerland. Despite the scarce population density of S. titanus, a regular trend of new infections (3.4% of the total vines) through the years was observed. The leafhopper Orientus ishidae was found as the most abundant among the captured insect species known as phytoplasma vectors (245 out of 315 specimens). The population of O. ishidae was evidenced prevalently (167 specimens) in the south-western side of the vineyard and within the neighbouring forest constituted mainly by hazel (Corylus avellana) and willow (Salix spp.). These plant species were found infected by FDp related strains (30% of analysed trees) for the first time in this study. Interestingly, O. ishidae was found to harbour FDp related strains in high percentage (26% of the analysed pools). In addition, 16SrV phytoplasma group was detected for the first time in the insect Hyalesthes obsoletus and a FDp related strain in Thamnotettix dilutior, present in low populations within the test vineyard. Molecular characterisation and phylogenetic analyses of methionine aminopeptidase (map) gene sequences of FDp and related strains, here identified, revealed the great prevalence of the map-type FD2 in grapevines (97%) and in O. ishidae pools (72%). Such a map-type was found also in hazel and in T. dilutior, but not in S. titanus. Moreover, map-types FD1 and FD3 were identified for the first time in Switzerland in several host plants and phytoplasma vectors, including grapevine (FD1), S. titanus (FD1) and O. ishidae (FD1 and FD3). Based on the data obtained in this study, it is reasonable to hypothesise that the ecological cycle of FDp could be related not exclusively to the grapevine-specific feeding diet of S. titanus, but it could include other insect vector(s) and/or plant host(s). Further studies will be needed to prove the role of O. ishidae as vector able to transmit FDp from wild plants (e.g. hazel) to grapevine

    Genetic Deletion of the Nociceptin/Orphanin FQ Receptor in the Rat Confers Resilience to the Development of Drug Addiction

    The nociceptin (NOP) receptor is a G-protein-coupled receptor whose natural ligand is the nociceptin/orphanin FQ (N/OFQ) peptide. Evidence from pharmacological studies suggests that the N/OFQ system is implicated in the regulation of several addiction-related phenomena, such as drug intake, withdrawal and relapse. Here, to further explore the role of NOP system in addiction, we used NOP (-/-) rats to study the motivation for cocaine, heroin and alcohol self-administration in the absence of N/OFQ function. Conditioned place preference (CPP) and saccharin (0.2% w/v) self-administration were also investigated. Results showed that NOP (-/-) rats self-administer less cocaine (0.25, 0.125 or 0.5 mg/infusion) both under a Fixed Ratio 1 and a Progressive Ratio schedule of reinforcement compared to wild type (Wt) controls. Consistently, cocaine (10 mg/kg, i.p.) was able to induce CPP in Wt but not in NOP (-/-). When NOP (-/-) rats were tested for heroin (20 μg/infusion) and ethanol (10% v/v) self-administration, they showeda significantly lower drug intake compared to Wt. Conversely, saccharin self-administration was not affected by NOP deletion, excluding the possibility of nonspecific learning deficits or generalized disruption of reward mechanisms in NOP (-/-) rats. These findings were confirmed with pharmacological experiments using two selective NOP antagonists, SB-612111 and LY2817412. Both drugs attenuated alcohol self-administration in Wt rats but not in NOP (-/-) rats. In conclusion, our results demonstrate that genetic deletion of NOP receptors confers resilience to drug abuse and support a role for NOP receptor antagonism as a potential treatment option for drug addiction.Neuropsychopharmacology accepted article preview online, 26 August 2016. doi:10.1038/npp.2016.171

    The Gene Ontology (GO) knowledgebase (http://geneontology.org) is a comprehensive resource concerning the functions of genes and gene products (proteins and noncoding RNAs). GO annotations cover genes from organisms across the tree of life as well as viruses, though most gene function knowledge currently derives from experiments carried out in a relatively small number of model organisms. Here, we provide an updated overview of the GO knowledgebase, as well as the efforts of the broad, international consortium of scientists that develops, maintains, and updates the GO knowledgebase. The GO knowledgebase consists of three components: (1) the GO-a computational knowledge structure describing the functional characteristics of genes; (2) GO annotations-evidence-supported statements asserting that a specific gene product has a particular functional characteristic; and (3) GO Causal Activity Models (GO-CAMs)-mechanistic models of molecular "pathways" (GO biological processes) created by linking multiple GO annotations using defined relations. Each of these components is continually expanded, revised, and updated in response to newly published discoveries and receives extensive QA checks, reviews, and user feedback. For each of these components, we provide a description of the current contents, recent developments to keep the knowledgebase up to date with new discoveries, and guidance on how users can best make use of the data that we provide. We conclude with future directions for the project

    The genetic epidemiology of joint shape and the development of osteoarthritis

    Congruent, low-friction relative movement between the articulating elements of a synovial joint is an essential pre-requisite for sustained, efficient, function. Where disorders of joint formation or maintenance exist, mechanical overloading and osteoarthritis (OA) follow. The heritable component of OA accounts for ~ 50% of susceptible risk. Although almost 100 genetic risk loci for OA have now been identified, and the epidemiological relationship between joint development, joint shape and osteoarthritis is well established, we still have only a limited understanding of the contribution that genetic variation makes to joint shape and how this modulates OA risk. In this article, a brief overview of synovial joint development and its genetic regulation is followed by a review of current knowledge on the genetic epidemiology of established joint shape disorders and common shape variation. A summary of current genetic epidemiology of OA is also given, together with current evidence on the genetic overlap between shape variation and OA. Finally, the established genetic risk loci for both joint shape and osteoarthritis are discussed
