    Gold oxide films grown in the confined aqueous layer between gold and organic solvents

    The properties of anodic passive films potentiostatically formed on polycrystalline gold in aqueous phosphate solutions were studied using voltammetry, electrochemical impedance spectroscopy, and contact angle measurements. The nature of the gold oxide layer was analyzed as a function of a potential holding in the aqueous double layer charge region at the interface between gold and the aqueous layer confined by insoluble organic solvents (hexane, chloroform, anisole, butyl acetate, xylene, and isopropyl ether). Different growth conditions change the homogeneity of the oxide layer leading to different passive properties. The nature of the incipient oxide layer was analyzed as a function of the cycling time in the double layer-oxide monolayer potential region. The replacement of the electrolyte by hexane allows the increase of the optical signal.     This paper includes commentaries on outcomes of esophageal surgery, including the mechanisms by which fundoduplication improves lower esophageal sphincter (LES) pressure; the efficacy of the Linx\u2122 management system in improving LES function; the utility of radiologic characterization of antireflux valves following surgery; the correlation between endoscopic findings and reported symptoms following antireflux surgery; the links between laparoscopic sleeve gastrectomy and decreased LES pressure, endoscopic esophagitis, and gastroesophageal reflux disease (GERD); the less favorable outcomes following fundoduplication among obese patients; the application of bioprosthetic meshes to reinforce hiatal repair and decrease the incidence of paraesophageal hernia; the efficacy of endoluminal antireflux procedures, and the limited efficacy of revisional antireflux operations, underscoring the importance of good primary surgery and diligent work-up to prevent the necessity of revisional procedures

    Elucidation of the tumoritropic principle of hypericin

    Hypericin is a potent agent in the photodynamic therapy of cancers. To better understand its tumoritropic behaviour, we evaluated the major determinants of the accumulation and dispersion of hypericin in subcutaneously growing mouse tumours. A rapid exponential decay in tumour accumulation of hypericin as a function of tumour weight was observed for each of the six tumour models investigated, and a similar relationship was found between tumour blood flow and tumour weight. Moreover, there was a close correlation between the higher hypericin uptake in RIF-1 tumours compared to R1 tumours and tumour vessel permeability. To define the role of lipoproteins in the transport of hypericin through the interstitial space, we performed a visual and quantitative analysis of the colocalisation of hypericin and DiOC18-labelled lipoproteins in microscopic fluorescent overlay images. A coupled dynamic behaviour was found early after injection (normalised fluorescence intensity differences were on the whole less than 10%), while a shifted pattern in localisation of hypericin and DiOC18 was seen after 24 h, suggesting that during its migration through the tumour mass, hypericin is released from the lipoprotein complex. In conclusion, we were able to show that the tumour accumulation of hypericin is critically determined by a combination of biological (blood flow, vessel permeability) and physicochemical elements (affinity for interstitial constituents)

    Perceived stress, exercise habits, and exercise addiction in Israeli army reserves: a pilot study

    Being an army reservist involves stress and uncertainty. While some individuals use exercise to cope with stress, no previous research has ever studied the relationship between army reservists exercise habits and perceived stress. The current study examined the hypothesis that Israeli army reserves in combat roles would report greater perceived stress and (to cope with the stress) exhibit more intensive exercise habits, as well as higher risk for exercise addiction, than the army reserves who are in a non-combat (i.e., office job) role and controls who are not in the army reserve. Participants (n = 277) completed questionnaires assessing their regular exercises habits, perceived stress, and risk for exercise addiction. The results showed that combat reserves scored higher on all dependent measures than non-combat reserves and controls. Perceived stress accounted for 38.8% to 56.6% of the variance in the risk of exercise addiction. Findings suggest that Israeli army reserves in combat role exercise more, for longer episodes, with greater reported intensity, and are at a greater risk for exercise addiction than the reserves in non-combat roles and controls. The higher risk for exercise addiction in the combat reserves, accompanied by a lower predictive power of the perceived stress, reflects a weaker association between these two variables in this group, most likely because combat reserves feel obliged to be physically fit in case of active deployment. This is the first study to show that differences in reservists’ roles is associated with different levels of risk for exercise addiction

    Wettability Modification of Nanomaterials by Low-Energy Electron Flux

    Controllable modification of surface free energy and related properties (wettability, hygroscopicity, agglomeration, etc.) of powders allows both understanding of fine physical mechanism acting on nanoparticle surfaces and improvement of their key characteristics in a number of nanotechnology applications. In this work, we report on the method we developed for electron-induced surface energy and modification of basic, related properties of powders of quite different physical origins such as diamond and ZnO. The applied technique has afforded gradual tuning of the surface free energy, resulting in a wide range of wettability modulation. In ZnO nanomaterial, the wettability has been strongly modified, while for the diamond particles identical electron treatment leads to a weak variation of the same property. Detailed investigation into electron-modified wettability properties has been performed by the use of capillary rise method using a few probing liquids. Basic thermodynamic approaches have been applied to calculations of components of solid–liquid interaction energy. We show that defect-free, low-energy electron treatment technique strongly varies elementary interface interactions and may be used for the development of new technology in the field of nanomaterials

    Photochemical internalisation of chemotherapy potentiates killing of multidrug-resistant breast and bladder cancer cells

    Multidrug resistance (MDR) is the major confounding factor in adjuvant solid tumour chemotherapy. Increasing intracellular amounts of chemotherapeutics to circumvent MDR may be achieved by a novel delivery method, photochemical internalisation (PCI). PCI consists of the co-administration of drug and photosensitiser; upon light activation the latter induces intracellular release of organelle-bound drug. We investigated whether co-administration of hypericin (photosensitiser) with mitoxantrone (MTZ, chemotherapeutic) plus illumination potentiates cytotoxicity in MDR cancer cells. We mapped the extent of intracellular co-localisation of drug/photosensitiser. We determined whether PCI altered drug-excreting efflux pump P-glycoprotein (Pgp) expression or function in MDR cells. Bladder and breast cancer cells and their Pgp-overexpressing MDR subclones (MGHU1, MGHU1/R, MCF-7, MCF-7/R) were given hypericin/MTZ combinations, with/without blue-light illumination. Pilot experiments determined appropriate sublethal doses for each. Viability was determined by the 3-[4,5-dimethylthiazolyl]-2,5-diphenyltetrazolium bromide assay. Intracellular localisation was mapped by confocal microscopy. Pgp expression was detected by immunofluorescence and Pgp function investigated by Rhodamine123 efflux on confocal microscopy. MTZ alone (0.1–0.2 μg ml−1) killed up to 89% of drug-sensitive cells; MDR cells exhibited less cytotoxicity (6–28%). Hypericin (0.1–0.2 μM) effects were similar for all cells; light illumination caused none or minimal toxicity. In combination, MTZ /hypericin plus illumination, potentiated MDR cell killing, vs hypericin or MTZ alone. (MGHU1/R: 38.65 and 36.63% increase, P<0.05; MCF-7/R: 80.2 and 46.1% increase, P<0.001). Illumination of combined MTZ/hypericin increased killing by 28.15% (P<0.05 MGHU1/R) compared to dark controls. Intracytoplasmic vesicular co-localisation of MTZ/hypericin was evident before illumination and at serial times post-illumination. MTZ was always found in sensitive cell nuclei, but not in dark resistant cell nuclei. In illuminated resistant cells there was some mobilisation of MTZ into the nucleus. Pgp expression remained unchanged, regardless of drug exposure. Pgp efflux was blocked by the Pgp inhibitor verapamil (positive control) but not impeded by hypericin. The increased killing of MDR cancer cells demonstrated is consistent with PCI. PCI is a promising technique for enhancing treatment efficacy

    Development of Bacterial Biofilms on Artificial Corals in Comparison to Surface-Associated Microbes of Hard Corals

    Numerous studies have demonstrated the differences in bacterial communities associated with corals versus those in their surrounding environment. However, these environmental samples often represent vastly different microbial micro-environments with few studies having looked at the settlement and growth of bacteria on surfaces similar to corals. As a result, it is difficult to determine which bacteria are associated specifically with coral tissue surfaces. In this study, early stages of passive settlement from the water column to artificial coral surfaces (formation of a biofilm) were assessed. Changes in bacterial diversity (16S rRNA gene), were studied on artificially created resin nubbins that were modelled from the skeleton of the reef building coral Acropora muricata. These models were dip-coated in sterile agar, mounted in situ on the reef and followed over time to monitor bacterial community succession. The bacterial community forming the biofilms remained significantly different (R = 0.864 p<0.05) from that of the water column and from the surface mucus layer (SML) of the coral at all times from 30 min to 96 h. The water column was dominated by members of the α-proteobacteria, the developed community on the biofilms dominated by γ-proteobacteria, whereas that within the SML was composed of a more diverse array of groups. Bacterial communities present within the SML do not appear to arise from passive settlement from the water column, but instead appear to have become established through a selection process. This selection process was shown to be dependent on some aspects of the physico-chemical structure of the settlement surface, since agar-coated slides showed distinct communities to coral-shaped surfaces. However, no significant differences were found between different surface coatings, including plain agar and agar enhanced with coral mucus exudates. Therefore future work should consider physico-chemical surface properties as factors governing change in microbial diversity
