    Exopolysaccharides produced by Lactobacillus and Bifidobacterium strains abrogate in vitro the cytotoxic effect of bacterial toxins on eukaryotic cells

    Aims: To evaluate the capability of the exopolysaccharides (EPS) produced by lactobacilli and bifidobacteria from human and dairy origin to antagonize the cytotoxic effect of bacterial toxins. Methods and Results: The cytotoxicity of Bacillus cereus extracellular factors on Caco-2 colonocytes in the presence/absence of the EPS was determined by measuring the integrity of the tissue monolayer and the damage to the cell membrane (extracellular lactate dehydrogenase activity). Additionally, the protective effect of EPS against the haemolytic activity of the streptolysin-O was evaluated on rabbit erythrocytes. The EPS produced by Bifidobacterium animalis ssp. lactis A1 and IPLA-R1, Bifidobacterium longum NB667 and Lactobacillus rhamnosus GG were able to counteract the toxic effect of bacterial toxins on the eukaryotic cells at 1 mg ml-1 EPS concentration. The EPS A1 was the most effective in counteracting the effect of B. cereus toxins on colonocytes, even at lower doses (0.5 mg ml-1), whereas EPS NB667 elicited the highest haemolysis reduction on erythrocytes. Conclusions: The production of EPS by lactobacilli and bifidobacteria could antagonize the toxicity of bacterial pathogens, this effect being EPS and biological marker dependent.     Bifidobacterium adolescentis as a key member of the human gut microbiota in the production of GABA

    Gamma aminobutyric acid (GABA) is the principal inhibitory neurotransmitter playing a key role in anxiety and depression disorders in mammals. Recent studies revealed that members of the gut microbiota are able to produce GABA modulating the gut–brain axis response. Among members of the human gut microbiota, bifidobacteria are well known to establish many metabolic and physiologic interactions with the host. In this study, we performed genome analyses of more than 1,000 bifidobacterial strains publicly available revealing that Bifidobacterium adolescentis taxon might represent a model GABA producer in human gastrointestinal tract. Moreover, the in silico screening of human/animal metagenomic datasets showed an intriguing association/correlation between B. adolescentis load and mental disorders such as depression and anxiety. Interestingly, in vitro screening of 82 B. adolescentis strains allowed identifying two high GABA producers, i.e. B. adolescentis PRL2019 and B. adolescentis HD17T2H, which were employed in an in vivo trial in rats. Feeding Groningen rats with a supplementation of B. adolescentis strains, confirmed the ability of these microorganisms to stimulate the in vivo production of GABA highlighting their potential implication in gut–brain axis interactions

    The 2011 October Draconids outburst-I. Orbital elements, meteoroid fluxes and 21P/Giacobini-Zinner delivered mass to Earth

    On 2011 October 8, the Earth crossed the dust trails left by comet 21P/Giacobini-Zinner during its 19th and 20th century perihelion approaches with the comet being close to perihelion. The geometric circumstances of that encounter were thus favourable to produce a meteor storm, but the trails were much older than in the 1933 and 1946 historical encounters. As a consequence the 2011 October Draconid display exhibited several activity peaks with Zenithal Hourly Rates of about 400 meteors h-1. In fact, if the display had not been forecasted, it could have passed almost unnoticed as was strongly attenuated for visual observers due to the Moon. This suggests that most meteor storms of a similar nature could have passed historically unnoticed under unfavourable weather and Moon observing conditions. The possibility of obtaining information on the physical properties of cometary meteoroids penetrating the atmosphere under low geocentric velocity encounter circumstances motivated us to set up a special observing campaign. Added to the Spanish Fireball Network wide-field all-sky and CCD video monitoring, other high-sensitivity 1/2 arcsec black and white CCD videocameras were attached to the modified medium-field lenses for obtaining high-resolution orbital information. The trajectory, radiant and orbital data of October 16 Draconid meteors observed at multiple stations are presented. The results show that the meteors appeared from a geocentric radiant located at α = 263.0 ± 0°.4 and δ =+55.3 ± 0°.3 that is in close agreement with the radiant predicted for the 1873-1894 and the 1900 dust trails. The estimated mass of material from 21P/Giacobini-Zinner delivered to Earth during the 6 h outburst was around950 ±150 kg

    Multi-instrumental observations of the 2014 Ursid meteor outburst

    The Ursid meteor shower is an annual shower that usually shows little activity. However, its Zenith hourly rate sometimes increases, usually either when its parent comet, 8P/Tuttle, is close to its perihelion or its aphelion. Outbursts when the comet is away from perihelion are not common and outbursts when the comet is close to aphelion are extremely rare. The most likely explanation offered to date is based on the orbital mean motion resonances. The study of the aphelion outburst of 2000 December provided a means of testing that hypothesis. A new aphelion outburst was predicted for 2014 December. The SPanish Meteor Network, in collaboration with the French Fireball Recovery and InterPlanetary Observation Network, set up a campaign to monitor this outburst and eventually retrieve orbital data that expand and confirm previous preliminary results and predictions. Despite unfavourable weather conditions over the south of Europe over the relevant time period, precise trajectories from multistation meteor data recorded over Spain were obtained, as well as orbital and radiant information for four Ursid meteors. The membership of these four meteors to the expected dust trails that were to provoke the outburst is discussed, and we characterize the origin of the outburst in the dust trail produced by the comet in the year AD 1392.Peer reviewe

    Impact of Extreme Obesity and Diet-Induced Weight Loss on the Fecal Metabolome and Gut Microbiota

    Scope: A limited number of human studies have characterized fecal microbiota and metabolome in extreme obesity and after diet-induced weight loss. Methods and results: Fecal samples from normal-weight and extremely obese adults and from obese participants before and after moderate diet-induced weight loss are evaluated for their interaction with the intestinal adenocarcinoma cell line HT29 using an impedance-based in vitro model, which reveals variations in the interaction between the gut microbiota and host linked to obesity status. Microbiota composition, short chain fatty acids, and other intestinal metabolites are further analyzed to assess the interplay among diet, gut microbiota, and host in extreme obesity. Microbiota profiles are distinct between normal-weight and obese participants and are accompanied by fecal signatures in the metabolism of biliary compounds and catecholamines. Moderate diet-induced weight loss promotes shifts in the gut microbiota, and the primary fecal metabolomics features are associated with diet and the gut–liver and gut–brain axes. Conclusions: Analyses of the fecal microbiota and metabolome enable assessment of the impact of diet on gut microbiota composition and activity, supporting the potential use of certain fecal metabolites or members of the gut microbiota as biomarkers for the efficacy of weight loss in extreme obesity

    Orbits and emission spectra from the 2014 Camelopardalids

    We have analyzed the meteor activity associated with meteoroids of fresh dust trails of Comet 209P/LINEAR, which produced an outburst of the Camelopardalid meteor shower (IAU code #451, CAM) in May 2014. With this aim, we have employed an array of high-sensitivity CCD video devices and spectrographs deployed at 10 meteor observing stations in Spain in the framework of the Spanish Meteor Network (SPMN). Additional meteoroid flux data were obtained by means of two forward-scatter radio systems. The observed peak zenithal hourly rate (ZHR) was much lower than expected, of around 20 meteors h-1. Despite of the small meteor flux in the optical range, we have obtained precise atmospheric trajectory, radiant and orbital information for 11 meteor and fireball events associated with this stream. The ablation behaviour and low tensile strength calculated for these particles reveal that Camelopardalid meteoroids are very fragile, mostly pristine aggregates with strength similar to that of the Orionids and the Leonids. The mineral grains seem to be glued together by a volatile phase. We also present and discuss two unique emission spectra produced by two Camelopardalid bright meteors. These suggest a non-chondritic nature for these particles, which exhibit Fe depletion in their composition.Comment: Accepted for publication in Monthly Notices of the Royal Astronomical Society on Sept. 22, 201

    In vitro evaluation of different prebiotics on the modulation of gut microbiota composition and function in morbid obese and normal-weight subjects

    The gut microbiota remains relatively stable during adulthood; however, certain intrinsic and environmental factors can lead to microbiota dysbiosis. Its restoration towards a healthy condition using best-suited prebiotics requires previous development of in vitro models for evaluating their functionality. Herein, we carried out fecal cultures with microbiota from healthy normal-weight and morbid obese adults. Cultures were supplemented with different inulin-type fructans (1-kestose, Actilight, P95, Synergy1 and Inulin) and a galactooligosaccharide. Their impact on the gut microbiota was assessed by monitoring gas production and evaluating changes in the microbiota composition (qPCR and 16S rRNA gene profiling) and metabolic activity (gas chromatography). Additionally, the effect on the bifidobacterial species was assessed (ITS-sequencing). Moreover, the functionality of the microbiota before and after prebiotic-modulation was determined in an in vitro model of interaction with an intestinal cell line. In general, 1-kestose was the compound showing the largest effects. The modulation with prebiotics led to significant increases in the Bacteroides group and Faecalibacterium in obese subjects, whereas in normal-weight individuals, substantial rises in Bifidobacterium and Faecalibacterium were appreciated. Notably, the results obtained showed differences in the responses among the tested compounds but also among the studied human populations, indicating the need for developing population-specific products

    Se presenta una investigación acerca del uso y la efectividad de los mapas conceptuales en la enseñanza. La experiencia se realizó en tres universidades cubanas y las materias en las que se emplearon los medios de enseñanza son tan disímiles como: Botánica, Programación Lógica y Estructuras de Datos. Los mapas fueron confeccionados por profesores de vasta experiencia en cada una de las materias y su aplicación en el aula se ha sometido a validaciones estadísticas con resultados satisfactorios. Como parte de la investigación se asocian nuevos recursos a los mapas, entre los que se destacan los agentes inteligentes, que guían el proceso de enseñanza, así como herramientas para la visualización de programas que permiten apreciar el funcionamiento de los algoritmos que se implementen.The present research is about the use and effectiveness of concept maps in teaching. The experiment was conducted in three Cuban universities and educational facilities created were used in subjects as diverse as: Botany, Logic Programming and Data Structures. The systems were developed by teachers with extensive experience and its application in the classroom has undergone successful statistical validations. As part of the research, new resources are associated with the maps, especially the intelligent agents that guide the teaching process, also highlights program visualization tools that allow us to appreciate the performance of the algorithms are implemented.Facultad de Informátic
