Unveiling interactions between DNA and cytotoxic 2-arylpiperidinyl-1,4-naphthoquinone derivatives: A combined electrochemical and computational study
Indexación: Scopus.Three 2-arylpiperidinyl-1,4-naphthoquinone derivatives were synthesized and evaluated in vitro to determine their cytotoxicity on cancer and normal cell lines. In order to establish their possible action mechanism, the electrochemical behaviour of these quinones was examined using cyclic voltammetry (CV) as technique by using a three-electrode setup: a glassy carbon, Ag/AgCl (in 3 M KCl), and platinum wire as working, reference, and counter electrodes, respectively. Kinetic studies were done to determine the control of the reduction reaction and the number of transferred electrons in the process. Furthermore, the addition of dsDNA to the quinone solutions allowed for the observation of an interaction between each quinone and dsDNA as the current-peaks became lower in presence of dsDNA. Otherwise, motivated to support the aforementioned results, electronic structure calculations at the TPSS-D3/6-31+G(d,p) level of theory were carried out in order to find the most favourable noncovalently bonded complexes between quinones and DNA. Noncovalent complexes formed between DNA and 2-arylpiperidinyl-1,4-naphthoquinones and stabilized by π-stacking interactions along with the well-known hydrogen-bonded complexes were found, with the former being more stable than the latter. Forest landscape restoration in the drylands of Latin America
Forest Landscape Restoration (FLR) involves the ecological restoration of degraded forest landscapes, with the aim of benefiting both biodiversity and human well-being. We first identify four fundamental principles of FLR, based on previous definitions. We then critically evaluate the application of these principles in practice, based on the experience gained during an international, collaborative research project conducted in six dry forest landscapes of Latin America. Research highlighted the potential for FLR; tree species of high socioeconomic value were identified in all study areas, and strong dependence of local communities on forest resources was widely encountered, particularly for fuelwood. We demonstrated that FLR can be achieved through both passive and active restoration approaches, and can be cost-effective if the increased provision of ecosystem services is taken into account. These results therefore highlight the potential for FLR, and the positive contribution that it could make to sustainable development. However, we also encountered a number of challenges to FLR implementation, including the difficulty of achieving strong engagement in FLR activities among local stakeholders, lack of capacity for community-led initiatives, and the lack of an appropriate institutional and regulatory environment to support restoration activities. Successful implementation of FLR will require new collaborative alliances among stakeholders, empowerment and capacity building of local communities to enable them to fully engage with restoration activities, and an enabling public policy context to enable local people to be active participants in the decision making process. © 2012 by the author(s). A DNA Microarray-Based Assay to Detect Dual Infection with Two Dengue Virus Serotypes
Here; we have described and tested a microarray based-method for the screening of dengue virus (DENV) serotypes. This DNA microarray assay is specific and sensitive and can detect dual infections with two dengue virus serotypes and single-serotype infections. Other methodologies may underestimate samples containing more than one serotype. This technology can be used to discriminate between the four DENV serotypes. Single-stranded DNA targets were covalently attached to glass slides and hybridised with specific labelled probes. DENV isolates and dengue samples were used to evaluate microarray performance. Our results demonstrate that the probes hybridized specifically to DENV serotypes; with no detection of unspecific signals. This finding provides evidence that specific probes can effectively identify single and double infections in DENV samples
Determining the neutrino mass ordering and oscillation parameters with KM3NeT/ORCA
[EN] The next generation of water Cherenkov neu-trino telescopes in the Mediterranean Sea are under con- struction offshore France (KM3NeT/ORCA) and Sicily (KM3NeT/ARCA). The KM3NeT/ORCA detector features an energy detection threshold which allows to collect atmo- spheric neutrinos to study flavour oscillation. This paper reports the KM3NeT/ORCA sensitivity to this phenomenon. The event reconstruction, selection and classification are described. The sensitivity to determine the neutrino mass ordering was evaluated and found to be 4.4¿ if the true order- ing is normal and 2.3¿ if inverted, after 3 years of data taking.
The precision to measure ¿m² and ¿¿¿ were also estimated and found to be 85.10¿¿ eV² and (¿¹.¿ -3.1)¿ for normal neu- trino mass ordering and, 75.10¿¿ eV² and (¿².¿ -7.0)¿ for inverted ordering. Finally, a unitarity test of the leptonic mixing matrix by measuring the rate of tau neutrinos is described. Sensitivity to light sterile neutrino mixing parameters with KM3NeT/ORCA
[EN] KM3NeT/ORCA is a next-generation neutrino telescope optimised for atmospheric neutrino oscillations studies. In this paper, the sensitivity of ORCA to the presence of a light sterile neutrino in a 3+1 model is presented. After three years of data taking, ORCA will be able to probe the active-sterile mixing angles ¿¿¿, ¿¿¿, ¿¿¿ and the effective angle ¿µ¿, over a broad range of mass squared difference ¿m² ~ [10¿5, 10] eV2, allowing to test the eV-mass sterile neutrino hypothesis as the origin of short baseline anomalies, as well as probing the hypothesis of a very light sterile neutrino, not yet constrained by cosmology. Calidad de la carne en corderos suplementados con tres concentraciones de zinc en una dieta energética
Objective: To evaluate the effect of three doses of Zn supplemented in the diet on the quality and concentration of Zn in meat.
Design / methodology / approach: 3 doses of Zn were supplemented: a) 21 mg Zn / Kg of dry matter (DM) from diet only, b) Zn80 (diet + ZnSO4) and c) Zn400 (diet + ZnSO4). Twelve Katahdin lambs randomly distributed in three groups, weighing 29.72 ± 2.16 Kg and age 9 ± 1 months. The consumed DM was restricted for all lambs to 718.62 ± 10.84 g.
Findings / Conclusion: The luminosity of the psoas major muscle was Zn21 = 36.50, Zn80 = 38.14 and Zn400 = 35.35 (quadratic effect, P <0.01). The redness of the psoas major muscle was between 18.81 to 19.72 and the intensity of the yellow color was from 3.63 to 3.82. The pH of the gracilis and psoas major muscles were 6.21 and 6.91 and there were no differences in pH, lightness and hardness. The water retention capacity of the gracilis muscle was Zn21 = 63.68%, Zn80 = 65.32% and Zn400 = 83.80% (linear effect, P <0.05). The 80 mg / kg dose of Zn improved the Zn content in meat.Objetivo: Evaluar el efecto de tres dosis de Zn suplementado en la dieta sobre la calidad y concentración de Zn en la carne.
Diseño/metodología/aproximación: Se suplementaron 3 dosis de Zn: a) 21 mg Zn/Kg de materia seca (MS) proveniente de la dieta solamente, b) Zn80 (dieta + ZnSO4) y c) Zn400 (dieta + ZnSO4). Doce corderos de raza Katahdin distribuidos aleatoriamente en tres grupos, con peso de 29.72 ± 2.16 Kg y edad de 9 ± 1 meses. La MS consumida fue restringida para todos los corderos a 718.62 ± 10.84 g.
Hallazgos/conclusión: La luminosidad del músculo psoas major fue Zn21 = 36.50, Zn80 = 38.14 y Zn400 = 35.35 (efecto cuadrático, P < 0.01). El enrojecimiento del músculo psoas major fue entre 18.81 a 19.72 y la intensidad del color amarillo fue de 3.63 a 3.82. El pH de los músculos gracilis y psoas major fueron de 6.21 y 6.91 y no hubo diferencias en el pH, luminosidad y dureza. La capacidad de retención de agua del músculo gracilis fue Zn21 = 63.68 %, Zn80 = 65.32 % y Zn400 = 83.80 % (efecto lineal, P < 0.05). La dosis de 80 mg/kg de Zn mejoró el contenido de Zn en la carne
Implementation and first results of the KM3NeT real-time core-collapse supernova neutrino search
[EN] The KM3NeT research infrastructure is uncon-
struction in the Mediterranean Sea. KM3NeT will study atmospheric and astrophysical neutrinos with two multi- purpose neutrino detectors, ARCA and ORCA, primar-ily aimed at GeV¿PeV neutrinos.
Thanks to the multi-photomultiplier tube design of the digital optical modules, KM3NeT is capable of detecting the neutrino burst from a Galactic or near-Galactic core-collapse supernova. This potential is already exploitable with the first detection units deployed in the sea. This paper describes the real-time imple- mentation of the supernova neutrino search, operating on the two KM3NeT detectors since the first months of 2019. A quasi-online astronomy analysis is introduced to study the time profile of the detected neutrinos for especially signifi- cant events. The mechanism of generation and distribution of alerts, as well as the integration into the SNEWS and SNEWS
2.0 global alert systems, are described. The approach for the follow-up of external alerts with a search for a neutrino excess in the archival data is defined. Finally, an overview of the cur-rent detector capabilities and a report after the first two years

Nanobeacon: A time calibration device for the KM3NeT neutrino telescope
[EN] The KM3NeT Collaboration is currently constructing a multi-site high-energy neutrino telescope in the Mediterranean Sea consisting of matrices of pressure-resistant glass spheres, each holding a set of 31 small-area photomultipliers. The main goals of the telescope are the observation of neutrino sources in the Universe and the measurement of the neutrino oscillation parameters with atmospheric neutrinos. A relative time synchronisation between photomultipliers of the nanosecond arder needed to guarantee the required angular resolution of the detector. Due to the large detector volumes to be instrumented by KM3NeT, a cost reduction of the different systems is a priority. To this end, the inexpensive Nanobeacon has been designed and developed by the KM3NeT Collaboration to be used for detector time-calibration studies. At present, more than 600 Nanobeacons have been already produced. The characterisation of the optical pulse and the wavelength emission profile of the devices is critica! for the time calibration. The optical pulse rise time has been quantified as less than 3 ns, while the Full Width Half Maximum is less than 6 ns. The wavelength drift, due to a variation of the supply voltage, has also been qualified as lower than 10 nm for the full range of the Nanobeacon.
