    Pregnancy in the female athlete - Part 1: antenatal

    As female participation in sport and exercise continues to increase, there is a strong likelihood that a strength and conditioning (S&C) coach will work with a pregnant athlete. High profile athletes such as Serena Williams and Dame Jessica Ennis-Hill have shown recently that it is not uncommon for female athletes to become mothers during their careers, rather than after they have retired. However, historically pregnancy was perceived as time spent away from sport and being active was wrongly associated with a high, unreasonable risk to the unborn foetus. The challenge for the S&C coach working with pregnant athletes is navigating the wide range of global recommendations that are often aimed at active or inactive individuals, but not at elite athletes. It is fundamental that any coach working with pregnant athletes is knowledgeable about the physiological and anatomical changes that occur throughout the trimesters, the nutritional demands of pregnancy, and its effect on weight management in order to appropriately facilitate athlete and foetal health during the antenatal period. Therefore, the aim of this initial review is to summarise current literature and provide key insights that promote best practice for practitioners working with pregnant athletes. The use of ‘women’ throughout this paper will refer to the general population and ‘sportswomen’ to the athletic population. ‘Antenatal’ refers to a pregnant woman before childbirth and the term ‘postnatal’ means after childbirth, something which will be covered in part 2 of this review series

    The Werner's Syndrome protein collaborates with REV1 to promote replication fork progression on damaged DNA

    AbstractDNA damage tolerance pathways facilitate the bypass of DNA lesions encountered during replication. These pathways can be mechanistically divided into recombinational damage avoidance and translesion synthesis, in which the lesion is directly bypassed by specialised DNA polymerases. We have recently shown distinct genetic dependencies for lesion bypass at and behind the replication fork in the avian cell line DT40, bypass at the fork requiring REV1 and bypass at post-replicative gaps requiring PCNA ubiquitination by RAD18. The WRN helicase/exonuclease, which is mutated in the progeroid and cancer predisposition disorder Werner's Syndrome, has previously been implicated in a RAD18-dependent DNA damage tolerance pathway. However, WRN has also been shown to be required to maintain normal replication fork progression on a damaged DNA template, a defect reminiscent of REV1-deficient cells. Here we use the avian cell line DT40 to demonstrate that WRN assists REV1-dependent translesion synthesis at the replication fork and that PCNA ubiquitination-dependent post-replicative lesion bypass provides an important backup mechanism for damage tolerance in the absence of WRN protein

    Elite male Flat jockeys display lower bone density and lower resting metabolic rate than their female counterparts: implications for athlete welfare

    To test the hypothesis that daily weight-making is more problematic to health in male compared with female jockeys, we compared the bone-density and resting metabolic rate (RMR) in weight-matched male and female Flat-jockeys. RMR (kcal.kg-1 lean mass) was lower in males compared with females as well as lower bone-density Z-scores at the hip and lumbar spine. Data suggest the lifestyle of male jockeys’ compromise health more severely than females, possibly due to making-weight more frequently

    Calibrated and completeness-corrected optical stellar density maps of the Northern Galactic Plane

This is an Open Access article distributed under the terms of the Creative Commons Attribution License (http://creativecommons.org/licenses/by/4.0/), which permits unrestricted reuse, distribution, and reproduction in any medium, provided the original work is properly cited.Following on from the second release of calibrated photometry from IPHAS, the INT/WFC Photometric Hα Survey of the Northern Galactic Plane, we present incompleteness-corrected stellar density maps in the r and i photometric bands. These have been computed to a range of limiting magnitudes reaching to 20th magnitude in r and 19th in i (Vega system), and with different angular resolutions – the highest resolution available being 1 arcmin2. The maps obtained cover 94 per cent of the 1800 square degree IPHAS footprint, spanning the Galactic latitude range, −5◦ < b < +5◦, north of the celestial equator. The corrections for incompleteness, due to confusion and sensitivity loss at the faint limit, have been deduced by the method of artificial source injection. The presentation of this method is preceded by a discussion of other more approximate methods of determining completeness. Our method takes full account of position-dependent seeing and source ellipticity in the survey data base. The application of the star counts to testing reddened Galactic disc models is previewed by a comparison with predicted counts along three constant-longitude cuts at 30◦, 90◦ and 175◦: some overprediction of the most heavily reddened 30◦ counts is found, alongside good agreement at 90◦ and 175◦.     A 3D extinction map of the northern Galactic plane based on IPHAS photometry

All rights reserved.We present a 3D map of extinction in the northern Galactic plane derived using photometry from the INT/WFC Photometric Hα Survey of the northern Galactic plane. The map has fine angular (~10 arcmin) and distance (100 pc) sampling allied to a significant depth (≳5 kpc). We construct the map using a method based on a hierarchical Bayesian model described in a previous article by Sale. In addition to mean extinction, we also measure differential extinction, which arises from the fractal nature of the interstellar medium, and show that it will be the dominant source of uncertainty in estimates of extinction to some arbitrary position. The method applied also furnishes us with photometric estimates of the distance, extinction, effective temperature, surface gravity, and mass for ~38 million stars.     Application of a single-objective, hybrid genetic algorithm approach to pharmacokinetic model building.

    A limitation in traditional stepwise population pharmacokinetic model building is the difficulty in handling interactions between model components. To address this issue, a method was previously introduced which couples NONMEM parameter estimation and model fitness evaluation to a single-objective, hybrid genetic algorithm for global optimization of the model structure. In this study, the generalizability of this approach for pharmacokinetic model building is evaluated by comparing (1) correct and spurious covariate relationships in a simulated dataset resulting from automated stepwise covariate modeling, Lasso methods, and single-objective hybrid genetic algorithm approaches to covariate identification and (2) information criteria values, model structures, convergence, and model parameter values resulting from manual stepwise versus single-objective, hybrid genetic algorithm approaches to model building for seven compounds. Both manual stepwise and single-objective, hybrid genetic algorithm approaches to model building were applied, blinded to the results of the other approach, for selection of the compartment structure as well as inclusion and model form of inter-individual and inter-occasion variability, residual error, and covariates from a common set of model options. For the simulated dataset, stepwise covariate modeling identified three of four true covariates and two spurious covariates; Lasso identified two of four true and 0 spurious covariates; and the single-objective, hybrid genetic algorithm identified three of four true covariates and one spurious covariate. For the clinical datasets, the Akaike information criterion was a median of 22.3 points lower (range of 470.5 point decrease to 0.1 point decrease) for the best single-objective hybrid genetic-algorithm candidate model versus the final manual stepwise model: the Akaike information criterion was lower by greater than 10 points for four compounds and differed by less than 10 points for three compounds. The root mean squared error and absolute mean prediction error of the best single-objective hybrid genetic algorithm candidates were a median of 0.2 points higher (range of 38.9 point decrease to 27.3 point increase) and 0.02 points lower (range of 0.98 point decrease to 0.74 point increase), respectively, than that of the final stepwise models. In addition, the best single-objective, hybrid genetic algorithm candidate models had successful convergence and covariance steps for each compound, used the same compartment structure as the manual stepwise approach for 6 of 7 (86 %) compounds, and identified 54 % (7 of 13) of covariates included by the manual stepwise approach and 16 covariate relationships not included by manual stepwise models. The model parameter values between the final manual stepwise and best single-objective, hybrid genetic algorithm models differed by a median of 26.7 % (q₁ = 4.9 % and q₃ = 57.1 %). Finally, the single-objective, hybrid genetic algorithm approach was able to identify models capable of estimating absorption rate parameters for four compounds that the manual stepwise approach did not identify. The single-objective, hybrid genetic algorithm represents a general pharmacokinetic model building methodology whose ability to rapidly search the feasible solution space leads to nearly equivalent or superior model fits to pharmacokinetic data

    The influence of acute exercise on bone biomarkers: protocol for a systematic review with meta-analysis. [Protocol]

    Background: Bone is a plastic tissue that is responsive to its physical environment. As a result, exercise interventions represent a potential means to influence bone. However, little is currently known about how various exercise and participant characteristics interact to influence bone metabolism. Acute, controlled, interventions provide an in-vivo model through which the acute bone response to exercise can be investigated, typically by monitoring circulating bone biomarkers. Currently, substantial heterogeneity in factors such as study design, quality, exercise and participant characteristics render it difficult to synthesize and evaluate the available evidence. Using a systematic review and meta-analytic approach, the aim of this investigation is to quantify the effect of an acute exercise bout on circulating bone biomarkers as well as examine potential factors that may moderate this response e.g., variation in participant, exercise and sampling characteristics. Methods: This protocol was designed in accordance with PRISMA-P guidelines. Seven databases (Medline, Embase, Sport Discus, Cochrane CENTRAL, PEDro, LILACS and Ibec) will be systematically searched and supplemented by secondary screening of the reference lists of all included articles. The PICOS (Population, Intervention, Comparator, Outcomes and Study Design) approach was used to guide the determination of eligibility criteria. Participants of any age, sex, training or health status will be considered for inclusion. We will select studies that have measured the bone biomarker response before and after an acute exercise session. All biomarkers considered to represent bone metabolism will be considered for inclusion and sensitivity analyses will be conducted using reference biomarkers for the measurement of bone resorption and formation (namely β-CTX-1 and P1NP). Multi-level, meta-regression models within a Bayesian framework will be used to explore the main effect of acute exercise on bone biomarkers as well as potential moderating factors. Risk of bias for each individual study will be evaluated using a modified version of the Downs and Black checklist while certainty in resultant outcomes will be assessed using the Grading of Recommendations Assessment, Development and Evaluation (GRADE) approach. Discussion: A better understanding of the bone metabolic response to an acute bout of exercise has the potential to advance our understanding of the mechanisms through which this stimulus impacts bone metabolism, including factors that may moderate this response. Additionally, we will identify current gaps in the evidence base and provide recommendations to inform future research.     New young planetary nebulae in IPHAS

We search for very small-diameter galactic planetary nebulae (PNe) representing the earliest phases of PN evolution. The IPHAS catalogue of H-emitting stars provides a useful basis for this study since all sources present in this catalogue must be of small angular diameter. Methods. The PN candidates are selected based on their location in two colour-colour diagrams: IPHAS (r' - H) vs. (r' - i'), and 2MASS (J - H) vs. (H - ). Spectroscopic follow-up was carried out on a sample of candidates to confirm their nature. Results. We present a total of 83 PN candidates. We were able to obtain spectra or find the classification from the literature for 35 candidates. Five of these objects are likely to be new PNe, including one large bipolar PN discovered serendipitously close to an emission-line star. PN distances deduced from extinction-distance relations based on IPHAS field-star photometry are presented for the first time. These yield distance estimates for our objects in the range 2 kpc and 6 kpc. From the data in hand, we conclude that four of the discovered objects are probably young PNe.Peer reviewe
