    II. Apples to apples A2A^2: cluster selection functions for next-generation surveys

    We present the cluster selection function for three of the largest next-generation stage-IV surveys in the optical and infrared: Euclid-Optimistic, Euclid-Pessimistic and the Large Synoptic Survey Telescope (LSST). To simulate these surveys, we use the realistic mock catalogues introduced in the first paper of this series. We detected galaxy clusters using the Bayesian Cluster Finder (BCF) in the mock catalogues. We then modeled and calibrated the total cluster stellar mass observable-theoretical mass (MCLMhM^*_{\rm CL}-M_{\rm h}) relation using a power law model, including a possible redshift evolution term. We find a moderate scatter of σMCLMh\sigma_{M^*_{\rm CL} | M_{\rm h}} of 0.124, 0.135 and 0.136 dex\rm dex for Euclid-Optimistic, Euclid-Pessimistic and LSST, respectively, comparable to other work over more limited ranges of redshift. Moreover, the three datasets are consistent with negligible evolution with redshift, in agreement with observational and simulation results in the literature. We find that Euclid-Optimistic will be able to detect clusters with >80%>80\% completeness and purity down to 8×1013h1M8\times10^{13} h^{-1} M_{\odot} up to z<1z<1. At higher redshifts, the same completeness and purity are obtained with the larger mass threshold of 2×1014h1M2\times10^{14} h^{-1} M_{\odot} up to z=2z=2. The Euclid-Pessimistic selection function has a similar shape with 10%\sim10\% higher mass limit. LSST shows 5%\sim 5\% higher mass limit than Euclid-Optimistic up to z<0.7z<0.7 and increases afterwards, reaching values of 2×1014h1M2\times10^{14} h^{-1} M_{\odot} at z=1.4z=1.4. Similar selection functions with only 80%80\% completeness threshold have been also computed.     Studying the role of the strawberry Fra protein family in the flavonoid metabolism during fruit ripening

    Strawberry fruits are highly appreciated worldwide due to their pleasant flavor and aroma and to the health benefits associated to their consumption. An important part of these properties is due to their content in secondary metabolites, especially phenolic compounds, of which flavonoids are the most abundant in the strawberry fruit. Although the flavonoid biosynthesis pathway is uncovered, little is known about its regulation. The strawberry Fra a (Fra) genes constitute a large family of homologs of the major birch pollen allergen Bet v 1 and for which no equivalents exist in Arabidopsis. Our group has shown that Fra proteins are involved in the formation of colored compounds in strawberries (Muñoz et al., 2010), which mainly depends on the production of certain flavonoids; that they are structurally homologs to the PYR/PYL/RCAR Arabidopsis ABA receptor, and that they are able to bind flavonoids (Casañal et al., 2013). With these previous results, our working hypothesis is that the Fra proteins are involved in the regulation of the flavonoids pathway. They would mechanistically act as the ABA receptor, binding a protein interactor and a ligand to regulate a signaling cascade and/or act as molecular carriers. The main objective of this research is to characterize the Fra family in strawberry and gain insight into their role in the flavonoid metabolism. By RNAseq expression analysis in ripening fruits we have identified transcripts for 10 members of the Fra family. Although expressed in all tissues analyzed, each family member presents a unique pattern of expression, which suggests functional specialization for each Fra protein. Then, our next approach was to identify the proteins that interact with Fras and their ligands to gain knowledge on the role that these proteins play in the flavonoids pathway. To identify the interacting partners of Fras we have performed a yeast two hybrid (Y2H) screening against cDNA libraries of strawberry fruits at the green and red stages. A protein that shares a 95% homology to the Heat stress transcription factor A-4-C like of Fragaria vesca (HSA4C) interacts specifically with Fra1 and not with other family members, which suggests functional diversification of Fra proteins in specific signaling pathways. The Y2H screening is not yet saturated, so characterization of other interacting proteins with other members of the Fra family will shed light on the functional diversity within this gene family. This research will contribute to gain knowledge on how the flavonoid pathway, and hence, the fruit ripening, is regulated in strawberry; an economically important crop but for which basic research is still very limited.     The Añarbe reservoir is located in the Iberic-Macaronesian region and is a water body provisionally identified as heavily modified, comparable to 'lake', that according to the "A" classification system of the WFD is "siliceous, lowlands, big and very deep", and according to the "B" system is ¨lowlands, northern, warm monomictic and of acidic waters". Two annual cycles of studies have been completed (from May of 2004 to January of 2006) looking to the main biological, physicochemical and hydromorphological parameters. The trophic state is oligotrophic for most of the parameters, except for the total phosphorus, that classifies it as mesotrophic. It does not present algal blooms and the average oxygen concentration in the hypolimnion in the zone of the dam during the stratification is 4.5 mg O2/l. The percentage of variation of the water volume of the reservoir respective to the maximum volume varies between 40 and 33% for the years 2004 and 2005, respectively. It shows a good ecological potential and could be considered as a reference within its group.El embalse del Añarbe se encuentra en la Región ibérico-macaronésica y es una masa de agua identificada provisionalmente como muy modificada, asimilable a 'lago', que según el Sistema A de clasificación de la DMA es del tipo 'silíceo, tierras bajas, grande y muy profundo' y según el Sistema B; 'tierras bajas, septentrional, monomíctico cálido y de aguas 'acidas'.     Consumer visual appraisal and shelf life of leg chops from suckling kids raised with natural milk or milk replacer

    BACKGROUND: The use of milk replacers to feed suckling kids could affect the shelf life and appearance of the meat. Leg chops were evaluated by consumers and the instrumental color was measured. A machine learning algorithm was used to relate them. The aim of this experiment was to study the shelf life of the meat of kids reared with dam's milk or milk replacers and to ascertain which illuminant and instrumental color variables are used by consumers as criteria to evaluate that visual appraisal. RESULTS: Meat from kids reared with milk replacers was more valuable and had a longer shelf life than meat from kids reared with natural milk. Consumers used the color of the whole surface of the leg chop to assess the appearance of meat. Lightness and hue angle were the prime cues used to evaluate the appearance of meat. CONCLUSION: Illuminant D65 was more useful for relating the visual appraisal with the instrumental color using a machine learning algorithm. The machine learning algorithms showed that the underlying rules used by consumers to evaluate the appearance of suckling kid meat are not at all linear and can be computationally schematized into a simple algorithm

    Active Rho is localized to podosomes induced by oncogenic Src and is required for their assembly and function

    Transformation of fibroblasts by oncogenic Src causes disruption of actin stress fibers and formation of invasive adhesions called podosomes. Because the small GTPase Rho stimulates stress fiber formation, Rho inactivation by Src has been thought to be necessary for stress fiber disruption. However, we show here that Rho[GTP] levels do not decrease after transformation by activated Src. Inactivation of Rho in Src-transformed fibroblasts by dominant negative RhoA or the Rho-specific inhibitor C3 exoenzyme disrupted podosome structure as judged by localization of podosome components F-actin, cortactin, and Fish. Inhibition of Rho strongly inhibited Src-induced proteolytic degradation of the extracellular matrix. Furthermore, development of an in situ Rho[GTP] affinity assay allowed us to detect endogenous Rho[GTP] at podosomes, where it colocalized with F-actin, cortactin, and Fish. Therefore, Rho is not globally inactivated in Src-transformed fibroblasts, but is necessary for the assembly and function of structures implicated in tumor cell invasion

    Prolonged oral cannabinoid administration prevents neuroinflammation, lowers β-amyloid levels and improves cognitive performance in Tg APP 2576 mice

    This is an Open Access article distributed under the terms of the Creative Commons Attribution License.-- et al.[Background]: Alzheimer's disease (AD) brain shows an ongoing inflammatory condition and non-steroidal anti-inflammatories diminish the risk of suffering the neurologic disease. Cannabinoids are neuroprotective and anti-inflammatory agents with therapeutic potential. [Methods]: We have studied the effects of prolonged oral administration of transgenic amyloid precursor protein (APP) mice with two pharmacologically different cannabinoids (WIN 55,212-2 and JWH-133, 0.2 mg/kg/day in the drinking water during 4 months) on inflammatory and cognitive parameters, and on 18F-fluoro-deoxyglucose (18FDG) uptake by positron emission tomography (PET). [Results]: Novel object recognition was significantly reduced in 11 month old Tg APP mice and 4 month administration of JWH was able to normalize this cognitive deficit, although WIN was ineffective. Wild type mice cognitive performance was unaltered by cannabinoid administration. Tg APP mice showed decreased 18FDG uptake in hippocampus and cortical regions, which was counteracted by oral JWH treatment. Hippocampal GFAP immunoreactivity and cortical protein expression was unaffected by genotype or treatment. In contrast, the density of Iba1 positive microglia was increased in Tg APP mice, and normalized following JWH chronic treatment. Both cannabinoids were effective at reducing the enhancement of COX-2 protein levels and TNF-α mRNA expression found in the AD model. Increased cortical β-amyloid (Aβ) levels were significantly reduced in the mouse model by both cannabinoids. Noteworthy both cannabinoids enhanced Aβ transport across choroid plexus cells in vitro. [Conclusions]: In summary we have shown that chronically administered cannabinoid showed marked beneficial effects concomitant with inflammation reduction and increased Aβ clearance.This work was supported by the Spanish Ministry of Science and Technology (SAF 2005-02845 to M.L.C).     A numerical approach to analyze the performance of a PEF-Ohmic heating system in microbial inactivation of solid food

    Pulsed Electric Fields (PEF) technology has been recently proposed as a new ohmic-heating system for the heat treatment of solid products in short periods (less than 1 min). However, similar to traditional ohmic heating, non-homogeneous distribution of temperature has been observed and cold points appeared in the interphase between the solid treated product and the electrodes, which can limit the technology for assuring food safety for treated solid products. In this investigation, a computational axisymmetric model of a lab-scale PEF system for a solid product (agar cylinder) was developed. This model was used to predict the temperature and the electric field distribution, treatment time, and the microbial inactivation (Salmonella Typhimurium 878) in the solid product after a PEF-ohmic treatment. Using a factorial analysis, a total of 8 process conditions with different settings of applied field strength levels (2.5–3.75 kV/cm), frequencies (100–200 Hz), and initial agar and electrode temperature (40–50°C) were simulated for the agar cylinder in order to identify the effect and optimal values of these parameters, which offer the most temperature homogeneity. The results showed that the initial temperature of the agar and the electrodes was of great importance in achieving the best temperature uniformity, limiting the occurrence of cold points, and therefore, improving the homogeneity in the level of inactivation of Salmonella Typhimurium 878 all over the agar cylinder. A treatment of 2.3 s would be enough at 3.75 kV/cm, 200 Hz with an initial temperature of 50°C of the agar and the electrodes, for a 5-Log10 reduction of Salmonella Typhimurium 878 in the whole product with a deviation of 9°C between the coldest and hottest point of the solid
