    A Web-based spatial decision supporting system for land management and soil conservation

    Abstract. Today it is evident that there are many contrasting demands on our landscape (e.g. food security, more sustainable agriculture, higher income in rural areas, etc.) as well as many land degradation problems. It has been proved that providing operational answers to these demands and problems is extremely difficult. Here we aim to demonstrate that a spatial decision support system based on geospatial cyberinfrastructure (GCI) can address all of the above, so producing a smart system for supporting decision making for agriculture, forestry, and urban planning with respect to the landscape. In this paper, we discuss methods and results of a special kind of GCI architecture, one that is highly focused on land management and soil conservation. The system allows us to obtain dynamic, multidisciplinary, multiscale, and multifunctional answers to agriculture, forestry, and urban planning issues through the Web. The system has been applied to and tested in an area of about 20 000 ha in the south of Italy, within the framework of a European LIFE+ project (SOILCONSWEB). The paper reports – as a case study – results from two different applications dealing with agriculture (olive growth tool) and environmental protection (soil capability to protect groundwater). Developed with the help of end users, the system is starting to be adopted by local communities. The system indirectly explores a change of paradigm for soil and landscape scientists. Indeed, the potential benefit is shown of overcoming current disciplinary fragmentation over landscape issues by offering – through a smart Web-based system – truly integrated geospatial knowledge that may be directly and freely used by any end user (www.landconsultingweb.eu). This may help bridge the last very important divide between scientists working on the landscape and end users

    Design, structural and functional characterization of a Temporin-1b analog active against Gram-negative bacteria

    Background Temporins are small antimicrobial peptides secreted by the Rana temporaria showing mainly activity against Gram-positive bacteria. However, different members of the temporin family, such as Temporin B, act in synergy also against Gram-negative bacteria. With the aim to develop a peptide with a wide spectrum of antimicrobial activity we designed and analyzed a series of Temporin B analogs. Methods Peptides were initially obtained by Ala scanning on Temporin B sequence; antimicrobial activity tests allowed to identify the TB-G6A sequence, which was further optimized by increasing the peptide positive charge (TB-KKG6A). Interactions of this active peptide with the LPS of E. coli were investigated by CD, fluorescence and NMR. Results TB-KKG6A is active against Gram-positive and Gram-negative bacteria at low concentrations. The peptide strongly interacts with the LPS of Gram-negative bacteria and folds upon interaction into a kinked helix. Conclusion Our results show that it is possible to widen the activity spectrum of an antimicrobial peptide by subtle changes of the primary structure. TB-KKG6A, having a simple composition, a broad spectrum of antimicrobial activity and a very low hemolytic activity, is a promising candidate for the design of novel antimicrobial peptides. General significance The activity of antimicrobial peptides is strongly related to the ability of the peptide to interact and break the bacterial membrane. Our studies on TB-KKG6A indicate that efficient interactions with LPS can be achieved when the peptide is not perfectly amphipathic, since this feature seems to help the toroidal pore formation process

    Crowd guilds: Worker-led reputation and feedback on crowdsourcing platforms

    Crowd workers are distributed and decentralized. While decentralization is designed to utilize independent judgment to promote high-quality results, it paradoxically undercuts behaviors and institutions that are critical to high-quality work. Reputation is one central example: crowdsourcing systems depend on reputation scores from decentralized workers and requesters, but these scores are notoriously inflated and uninformative. In this paper, we draw inspiration from historical worker guilds (e.g., in the silk trade) to design and implement crowd guilds: centralized groups of crowd workers who collectively certify each other’s quality through double-blind peer assessment. A two-week field experiment compared crowd guilds to a traditional decentralized crowd work model. Crowd guilds produced reputation signals more strongly correlated with ground-truth worker quality than signals available on current crowd working platforms, and more accurate than in the traditional model

    The Italian multiple sclerosis register

    The past decade has seen extraordinary increase in worldwide availability of and access to several large multiple sclerosis (MS) databases and registries. MS registries represent powerful tools to provide meaningful information on the burden, natural history, and long-term safety and effectiveness of treatments. Moreover, patients, physicians, industry, and policy makers have an active interest in real-world observational studies based on register data, as they have the potential to answer the questions that are most relevant to daily treatment decision-making. In 2014, the Italian MS Foundation, in collaboration with the Italian MS clinical centers, promoted and funded the creation of the Italian MS Register, a project in continuity with the existing Italian MS Database Network set up from 2001. Main objective of the Italian MS Register is to create an organized multicenter structure to collect data of all MS patients for better defining the disease epidemiology, improving quality of care, and promoting research projects in high-priority areas. The aim of this article is to present the current framework and network of the Italian MS register, including the methodology used to improve the quality of data collection and to facilitate the exchange of data and the collaboration among national and international groups

    Talents from Abroad. Foreign Managers and Productivity in the United Kingdom.

    In this paper, we test the contribution of foreign management on firms’ competitiveness. We use a novel dataset on the careers of 165,084 managers employed by 13,106 companies in the United Kingdom in the period 2009-2017. We find that a domestic manufacturing firm becomes on average between 9% and 12% more productive after hiring at least one foreign manager. Interestingly, productivity gains by domestic firms after recruiting foreign managers are similar in magnitude to gains after foreign acquisitions as from previous literature. Eventually, we do not find significant gains by foreign-owned firms hiring foreign managers. Our identification strategy combines difference-in-difference and matching techniques to challenge reverse causality. We proxy firms’ competitiveness either by total factor productivity or by technical efficiency derived from stochastic frontier analyses. Eventually, we argue that limits to the circulation of talents, as for example in case of a Brexit event, may hamper the allocation of labor productive resources

    Mirna profile is altered in a modified eae mouse model of multiple sclerosis featuring cortical lesions

    Cortical lesions represent a hallmark of multiple sclerosis and are proposed as a predictor of disease severity. microRNAs are suggested to be important players in the disease pathogenesis and the experimental autoimmune encephalomyelitis animal model. We implemented a mouse model recapitulating more closely the human pathology as it is characterized by both an autoimmune heterogeneity and the presence of cortical lesions, two parameters missing in experimental autoimmune encephalomyelitis. In our model, mice clustered in two groups displaying high or low clinical scores. Upon cortical cytokine injection, lesions appeared with a specific topography while cortical miRNA profiles were altered. These two features differed according to disease severity. We evidenced changes in miRNA regulators and targets suggesting that miRNA alteration had functional repercussions that could explain the differences in cortical lesions. This model represents a crucial tool for the study of both miRNA involvement and cortical lesion formation in disease pathogenesis
