    Transcranial magnetic stimulation effects support an oscillatory model of ERP genesis

    Whether prestimulus oscillatory brain activity contributes to the generation of post-stimulus-evoked neural responses has long been debated, but findings remain inconclusive. We first investigated the hypothesized relationship via EEG recordings during a perceptual task with this correlational evidence causally probed subsequently by means of online rhythmic transcranial magnetic stimulation. Both approaches revealed a close link between prestimulus individual alpha frequency (IAF) and P1 latency, with faster IAF being related to shorter latencies, best explained via phase-reset mechanisms. Moreover, prestimulus alpha amplitude predicted P3 size, best explained via additive (correlational and causal evidence) and baseline shift mechanisms (correlational evidence), each with distinct prestimulus alpha contributors. Finally, in terms of performance, faster prestimulus IAF and shorter P1 latencies were both associated with higher task accuracy, while lower prestimulus alpha amplitudes and higher P3 amplitudes were associated with higher confidence ratings. Our results are in favor of the oscillatory model of ERP genesis and modulation, shedding new light on the mechanistic relationship between prestimulus oscillations and functionally relevant evoked components

    Hierarchical psychophysiological pathways subtend perceptual asymmetries in Neglect

    Stroke patients with left Hemispatial Neglect (LHN) show deficits in perceiving left contralesional stimuli with biased visuospatial perception towards the right hemifield. However, very little is known about the functional organization of the visuospatial perceptual neural network and how this can account for the profound reorganization of space representation in LHN. In the present work, we aimed at (1) identifying EEG measures that discriminate LHN patients against controls and (2) devise a causative neurophysiological model between the discriminative EEG measures. To these aims, EEG was recorded during exposure to lateralized visual stimuli which allowed for pre-and post-stimulus activity investigation across three groups: LHN patients, lesioned controls, and healthy individuals. Moreover, all participants performed a standard behavioral test assessing the perceptual asymmetry index in detecting lateralized stimuli. The between-groups discriminative EEG patterns were entered into a Structural Equation Model for the identification of causative hierarchical associations (i.e., pathways) between EEG measures and the perceptual asymmetry index. The model identified two pathways. A first pathway showed that the combined contribution of pre-stimulus frontoparietal connectivity and individual-alpha-frequency predicts post-stimulus processing, as measured by visual-evoked N100, which, in turn, predicts the perceptual asymmetry index. A second pathway directly links the inter-hemispheric distribution of alpha-amplitude with the perceptual asymmetry index. The two pathways can collectively explain 83.1% of the variance in the perceptual asymmetry index. Using causative modeling, the present study identified how psychophysiological correlates of visuospatial perception are organized and predict the degree of behavioral asymmetry in LHN patients and controls

    Morphological and quantitative analysis of BCL6 expression in human colorectal carcinogenesis.

    The aim of the present study was to determine whether BCL6 is expressed during malignant transformation of the large bowel and to assess whether, and to what extent, immunoreactivity is related to the different stages of neoplastic progression. Samples of normal colorectal mucosa (n=22), microadenomas (n=22) and colorectal cancer (n=22), were analyzed by immunohistochemistry, immunofluorescence coupled with confocal microscopy and western blotting. Our results clearly outlined the marked increase occurring in both intensity and density of BCL6 protein expression in the normal mucosa-microadenoma-carcinoma sequence. Immunohistochemistry and immunofluorescence analyses showed that BCL6 is expressed at low levels in normal mucosa and increases in microadenoma and in cancer with statistical significance. These results were confirmed by western blotting data. The increasing expression of BCL6 in human colorectal cancer development suggests the involvement of BCL6 in tumor progression, from the earliest stages of carcinogenesis with significant increase in cancer. The enhanced understanding of the biological role of BCL6, previously shown to exert a key role in lymphomagenesis, may lead to a re-evaluation of this protein and may highlight the importance of performing further studies in order to identify novel therapeutic targets for colorectal cancer

    Impact of a Shorter Brine Soaking Time on Nutrient Bioaccessibility and Peptide Formation in 30-Months-Ripened Parmigiano Reggiano Cheese

    Reducing the salt content in food is an important nutritional strategy for decreasing the risk of diet-related diseases. This strategy is particularly effective when applied to highly appreciated food having good nutritional characteristics, if it does not impact either upon sensory or nutritional properties of the final product. This work aimed at evaluating if the reduction of salt content by decreasing the brine soaking time modifies fatty acid and protein bioaccessibility and bioactive peptide formation in a 30-month-ripened Parmigiano Reggiano cheese (PRC). Hence, conventional and hyposodic PRC underwent in vitro static gastrointestinal digestion, and fatty acid and protein bioaccessibility were assessed. The release of peptide sequences during digestion was followed by LC–HRMS, and bioactive peptides were identified using a bioinformatic approach. At the end of digestion, fatty acid and protein bioaccessibility were similar in conventional and hyposodic PRC, but most of the bioactive peptides, mainly the ACE-inhibitors, were present in higher concentrations in the low-salt cheese. Considering that the sensory profiles were already evaluated as remarkably similar in conventional and hyposodic PRC, our results confirmed that shortening brine soaking time represents a promising strategy to reduce salt content in PRC

    The Initial Conditions of Clustered Star Formation I: NH3 Observations of Dense Cores in Ophiuchus

    We present combined interferometer and single dish telescope data of NH3 (J,K) = (1,1) and (2,2) emission towards the clustered star forming Ophiuchus B, C and F Cores at high spatial resolution (~1200 AU) using the Australia Telescope Compact Array, the Very Large Array, and the Green Bank Telescope. While the large scale features of the NH3 (1,1) integrated intensity appear similar to 850 micron continuum emission maps of the Cores, on 15" (1800 AU) scales we find significant discrepancies between the dense gas tracers in Oph B, but good correspondence in Oph C and F. Using the Clumpfind structure identifying algorithm, we identify 15 NH3 clumps in Oph B, and 3 each in Oph C and F. Only five of the Oph B NH3 clumps are coincident within 30" (3600 AU) of a submillimeter clump. We find v_LSR varies little across any of the Cores, and additionally varies by only ~1.5 km/s between them. The observed NH3 line widths within the Oph B and F Cores are generally large and often mildly supersonic, while Oph C is characterized by narrow line widths which decrease to nearly thermal values. We find several regions of localized narrow line emission (\Delta v < 0.4 km/s), some of which are associated with NH3 clumps. We derive the kinetic temperatures of the gas, and find they are remarkably constant across Oph B and F, with a warmer mean value (T_K = 15 K) than typically found in isolated regions and consistent with previous results in clustered regions. Oph C, however, has a mean T_K = 12 K, decreasing to a minimum T_K = 9.4 K towards the submillimeter continuum peak, similar to previous studies of isolated starless cores.     Comparison of the biological effects of gadodiamide (Omniscan) and gadoteridol ({ProHance}) by means of multi-organ and plasma metabolomics

    Gadolinium-based contrast agents (GBCAs) are massively employed in radiology to increase the diagnostic power of MRI. However, investigations aiming at detecting possible metabolic perturbations or adverse health effects due to gadolinium deposition are still lacking. In this work, aqueous organs extract and plasma samples were analyzed by GC-MS and H-1-NMR, respectively, to investigate the effects of multiple administrations of one linear (Omniscan) and one macrocyclic (ProHance) GBCA, on the main metabolic pathways in healthy mice. Multivariate analysis revealed that plasma metabolome was not differently perturbed by the two GBCAs, while, the multiorgan analysis displayed a clear separation of the Omniscan-treated from the control and the ProHance-treated groups. Interestingly, the most affected organs were the brain, cerebellum and liver. Thus, this work paves the way to both the safest use of the commercially available GBCAs and the development of new GBCAs characterized by lower general toxicity

    Recalcitrant compounds removal in raw leachate and synthetic effluents using the white-rot fungus Bjerkandera adusta

    Recalcitrant compounds limit the efficiency of conventional biological processes for wastewater treatment, representing one of the major issues in the field. This study focused on the treatment of three effluents with White-Rot-Fungus (WRF) Bjerkandera adusta MUT 2295 in batch tests, with biomass cultivated in attached form on polyurethane foam cubes (PUFs) to test its efficiency in the removal of the target effluents' recalcitrant fraction. Treatment efficiency of B. adusta was evaluated on landfill leachate (Canada) and two solutions containing synthetic recalcitrant compounds, which were prepared with tannic and humic acid. Chemical Oxygen Demand (COD) and color removal, the production of manganese peroxidases, and the consumption of a co-substrate (glucose) were monitored during the experiment. Biological Oxygen Demand (BOD5) and fungal dry weight were measured at the beginning and at the end of the experiment. After co-substrate addition, effluent COD was 2300 ± 85, 2545 ± 84, and 2580 ± 95 (mg/L) in raw leachate and tannic and humic acids, respectively. COD removal of 48%, 61%, and 48% was obtained in raw leachate and in the synthetic effluents containing tannic and humic acids, respectively. Color removal of 49%, 25%, and 42% was detected in raw leachate and in tannic and humic acid solutions, respectively. COD and color removals were associated with the increase of fungal dry weight, which was observed in all the trials. These results encourage the use of the selected fungal strain to remove tannic acid, while further investigations are required to optimize leachate and humic acid bioremediation
