    Vers un monitoring électrique des garnitures mécaniques d'étanchéité

    Cet article s'intéresse à l'évolution en temps réel du contact d'une garniture mécanique d'étanchéité en fonctionnement. Afin de simuler ce contact frottant, un banc spécifique de mesures a été développé, mettant en jeux des bagues identiques au dispositif industriel et présentant des surfaces avec des finitions industrielles. Ce banc permet d'obtenir des informations sur l'état mécanique du contact en le faisant traverser par un courant électrique sonde. Deux paramètres importants ont été explorés : la lubrification et la charge. L'étude présentée montre des signatures électriques spécifiques qui font de la mesure électrique un outil adapté pour suivre en temps réel l'évolution de l'état du contact

We model uncertainty on task durations by intervals. With this problem formulation, our goal is to assert possible and necessary criticality of the different tasks and to compute their possible earliest starting dates, latest starting dates, and floats. This paper combines various results and provides a complete solution to the problem. We present the complexity results of all considered subproblems and efficient algorithms to solve them

    Mule Regulates the Intestinal Stem Cell Niche via the Wnt Pathway and Targets EphB3 for Proteasomal and Lysosomal Degradation

    The E3 ubiquitin ligase Mule is often overexpressed in human colorectal cancers, but its role in gut tumorigenesis is unknown. Here, we show in vivo that Mule controls murine intestinal stem and progenitor cell proliferation by modulating Wnt signaling via c-Myc. Mule also regulates protein levels of the receptor tyrosine kinase EphB3 by targeting it for proteasomal and lysosomal degradation. In the intestine, EphB/ephrinB interactions position cells along the crypt-villus axis and compartmentalize incipient colorectal tumors. Our study thus unveils an important new avenue by which Mule acts as an intestinal tumor suppressor by regulation of the intestinal stem cell niche

    TRAF6 and IRF7 Control HIV Replication in Macrophages

    The innate immune system recognizes virus infection and evokes antiviral responses which include producing type I interferons (IFNs). The induction of IFN provides a crucial mechanism of antiviral defense by upregulating interferon-stimulated genes (ISGs) that restrict viral replication. ISGs inhibit the replication of many viruses by acting at different steps of their viral cycle. Specifically, IFN treatment prior to in vitro human immunodeficiency virus (HIV) infection stops or significantly delays HIV-1 production indicating that potent inhibitory factors are generated. We report that HIV-1 infection of primary human macrophages decreases tumor necrosis factor receptor-associated factor 6 (TRAF6) and virus-induced signaling adaptor (VISA) expression, which are both components of the IFN signaling pathway controlling viral replication. Knocking down the expression of TRAF6 in macrophages increased HIV-1 replication and augmented the expression of IRF7 but not IRF3. Suppressing VISA had no impact on viral replication. Overexpression of IRF7 resulted in enhanced viral replication while knocking down IRF7 expression in macrophages significantly reduced viral output. These findings are the first demonstration that TRAF6 can regulate HIV-1 production and furthermore that expression of IRF7 promotes HIV-1 replication

    Induction of Selective Blood-Tumor Barrier Permeability and Macromolecular Transport by a Biostable Kinin B1 Receptor Agonist in a Glioma Rat Model

    Treatment of malignant glioma with chemotherapy is limited mostly because of delivery impediment related to the blood-brain tumor barrier (BTB). B1 receptors (B1R), inducible prototypical G-protein coupled receptors (GPCR) can regulate permeability of vessels including possibly that of brain tumors. Here, we determine the extent of BTB permeability induced by the natural and synthetic peptide B1R agonists, LysdesArg9BK (LDBK) and SarLys[dPhe8]desArg9BK (NG29), in syngeneic F98 glioma-implanted Fischer rats. Ten days after tumor inoculation, we detected the presence of B1R on tumor cells and associated vasculature. NG29 infusion increased brain distribution volume and uptake profiles of paramagnetic probes (Magnevist and Gadomer) at tumoral sites (T1-weighted imaging). These effects were blocked by B1R antagonist and non-selective cyclooxygenase inhibitors, but not by B2R antagonist and non-selective nitric oxide synthase inhibitors. Consistent with MRI data, systemic co-administration of NG29 improved brain tumor delivery of Carboplatin chemotherapy (ICP-Mass spectrometry). We also detected elevated B1R expression in clinical samples of high-grade glioma. Our results documented a novel GPCR-signaling mechanism for promoting transient BTB disruption, involving activation of B1R and ensuing production of COX metabolites. They also underlined the potential value of synthetic biostable B1R agonists as selective BTB modulators for local delivery of different sized-therapeutics at (peri)tumoral sites

    Amplification des effets des explosions sous l'effet du rayonnement thermique

    National audienceLes explosions de poussières affectent tout le tissu industriel avec souvent, des conséquences désastreuses. De nombreux progrès ont été accomplis lors de ces dernières décennies dans la compréhension et la modélisation de ce type d'explosion en démontrant par exemple les similitudes qu'il peut y avoir avec les explosions de gaz. Mais on s'interroge sur le comportement des fines poussières métalliques capables d'engendrer des explosions extrêmement puissantes. L'objectif du projet RADIAN, soutenu par la région Picardie, est de découvrir la raison de la violence de ces explosions. Dans cette communication, on propose d'identifier à l'aide de quelques observations expérimentales l'importance potentielle du rayonnement thermique dans les processus de propagation des flammes dans des nuages d'air et de particules d'aluminium et d'imaginer, à l'aide de modèles théoriques quel pourrait être le comportement d'une flamme dominée par le rayonnement thermique. L'incidence de la taille des particules est discutée. On montre en particulier que des régimes d'explosion atypiques sont possibles contre lesquels les moyens de protection habituels pourraient bien être inopérants

    Influence of a Laser Irradiation and Laser Scribing on Magnetic Properties of GO Silicon Steels Sheets Using a Nanosecond Fiber Laser

    International audienceImproving the performance of electrical steels within the magnetic circuits is essential to save energy. The domain refinement through local surface treatment by laser is an effective technique to reduce the iron losses in grain-oriented iron silicon steels. To interpret the mechanism of this technique, we have quantitatively studied the impact of nanosecond pulse laser treatment on the magnetic properties of grain-oriented Fe(3%wt)Si sheets. We measured the total power loss and apparent permeability of the samples using a Single-Sheet Tester (SST). The laser treatment resulted in a loss reduction of up to 24% compared to the average power loss of standard samples at 50 Hz. At mid-induction levels, the reduction was also accompanied by an improvement in apparent permeability. A dynamic magnetic behavior law was used to identify a dynamic property Λ including information on density, surface area and wall mobility and another internal permeability property µ representative of static field and magnetization characteristics. Lastly, we presented the behavior of these properties under different laser treatment
