    Why Young patients with cancer requiere different coping tools?

    The medical field has made great advancements in recent decades, but even one in every two men and one in three women will suffer from cancer throughout their lives [1]. Furthermore, cancer was the leading cause of death due to illness in the adolescent and young adult (18-39 year old) population [2]. Despite these figures, no one thinks he is close to suffering from cancer. We prefer to think that it is a disease related to some other collective that we perceive as distant. We are not prepared to deal with the processes involved in the diagnosis and treatment of cancer. Therefore, it is a disease that poses challenges beyond the medicine itself and that forces to take action in relation to the personal and social environment of the patient, measures that will probably transform their present and future lifestyle. This becomes especially relevant if we focus on cancer in young patients [3]

    Toxicity of cadmium and zinc on two microalgae, Scenedesmus obliquus and Desmodesmus pleiomorphus, from Northern Portugal

    Aquatic environments often contain toxic heavy metals that may enter the food web via uptake by microalgae and eventually cause severe poisoning problems at higher trophic levels. The effects of Cd and Zn cations upon growth of two native green microalgal species, Scenedesmus obliquus and Desmodesmus pleiomorphus (previously isolated from a polluted site in Northern Portugal), were accordingly evaluated. Growth inhibition of the microalgal cells was determined following exposure for 96 h to several initial concentrations of aqueous solutions of either of those two metals. At the higher end of Cd and Zn experimental concentration ranges, a significant reduction in cell density was observed in the cultures; EC50 values, calculated after fitting a Weibull model to the experimental data, were 0.058 and 1.92 mg L−1 for Cd and 16.99 and 4.87 mg L−1 for Zn in the case of S. obliquus and D. pleiomorphus, respectively. One observed that S. obliquus can tolerate higher Zn concentrations than D. pleiomorphus, but the reverse holds regarding exposure to Cd

    A Comparison of Two Methods for Enhancing the Recovery of Seagrasses into Propellor Scars: Mechanical Injection of a Nutrient and Growth Hormone Solution vs. Defecation by Roosting Seabirds: Final Report.

    Based on the recovery rates for Thalassia testudinum measured in this study for scars of these excavation depths and assuming a linear recovery horizon, we estimate that it would take ~ 6.9 years (95% CI. = 5.4 to 9.6 years) for T. testudinum to return to the same density as recorded for the adjacent undisturbed population. The application of water soluble fertilizers and plant growth hormones by mechanical injection into the sediments adjacent to ten propellor scars at Lignumvitae State Botanical Site did not significantly increase the recovery rate of Thalassia testudinum or Halodule wrightii. An alternative method of fertilization and restoration of propellor scars was also tested by a using a method of “compressed succession” where Halodule wrightii is substituted for T. testudinum in the initial stages of restoration. Bird roosting stakes were placed among H.wrightii bare root plantings in prop scars to facilitate the defecation of nitrogen and phosphorus enriched feces. In contrast to the fertilizer injection method, the bird stakes produced extremely high recovery rates of transplanted H. wrightii. We conclude that use of a fertilizer/hormone injection machine in the manner described here is not a feasible means of enhancing T. testudinum recovery in propellor scars on soft bottom carbonate sediments. Existing techniques such as the bird stake approach provide a reliable, and inexpensive alternative method that should be considered for application to restoration of seagrasses in these environments.     Durability of carbon fibre reinforced polymer laminates

    Due to their excellent mechanical properties and easy application Carbon Fibre Reinforced Polymers (CFRP) are being successfully used to repair and renew old and damaged civil engineering infrastructures. In particular, the method of strengthening concrete members with externally bonding of CFRP laminates has become increasingly popular. In fact, extensive research has shown that this technique can lead to a substantial improvement in the fl exural and shear strength capacities of the member repaired. In such applications, materials involved will be exposed to outdoor environmental conditions, including humidity, water from rain, saline-water, ground water and high alkalinity solutions due to chemical composition of the concrete itself, as well as solar ultraviolet (UV) radiation. Thus, it is of great relevancy to study the durability of the materials involved, namely epoxy adhesives and CFRP laminates, as well as, the integrity of the entire system, when subjected to those environmental factors. This work is part of a much larger research study to predict the long-term behaviour of several commercial CFRP laminate/adhesive systems used in the rehabilitation of concrete structures. In the whole project it is recognised that the durability of these solutions depends not only on the durability of the composite materials but also on the performance of the systems used to bond them to the concrete. This present document provides details about the durability of CFRP laminates and the study carried out to investigate the environmental and accelerated ageing effects on three types of commercially available CFRP laminates. The changes in CFRP laminates properties after exposure up to 18 months under different environmental conditions – immersion in demineralised water, saline and alkaline solutions at room temperature, 40 ° C and 60 °C, as well as under constant humidity condensation atmosphere at 40 ° C – are reported and discussed. The results of tests performed in accelerated weathering chambers – with UV radiation – are also included. Results obtained offer valuable information about the progression of material degradation under different ageing conditions (some of them accelerated), which can be used, not only to characterise the behaviour of CFRP laminates, but also for a better understanding of the degradation mechanisms involved, which is a base for the design of service life-prediction models

    New integrative computational approaches unveil the Saccharomyces cerevisiae pheno-metabolomic fermentative profile and allow strain selection for winemaking

    During must fermentation by Saccharomyces cerevisiae strains thousands of volatile aroma compounds are formed. The objective of the present work was to adapt computational approaches to analyze pheno-metabolomic diversity of a S. cerevisiae strain collection with different origins. Phenotypic and genetic characterization together with individual must fermentations were performed, and metabolites relevant to aromatic profiles were determined. Experimental results were projected onto a common coordinates system, revealing 17 statistical-relevant multi-dimensional modules, combining sets of most-correlated features of noteworthy biological importance. The present method allowed, as a breakthrough, to combine genetic, phenotypic and metabolomic data, which has not been possible so far due to difficulties in comparing different types of data. Therefore, the proposed computational approach revealed as successful to shed light into the holistic characterization of S. cerevisiae pheno-metabolome in must fermentative conditions.     Modulatory effects of cAMP and PKC activation on gap junctional intercellular communication among thymic epithelial cells

    <p>Abstract</p> <p>Background</p> <p>We investigated the effects of the signaling molecules, cyclic AMP (cAMP) and protein-kinase C (PKC), on gap junctional intercellular communication (GJIC) between thymic epithelial cells (TEC).</p> <p>Results</p> <p>Treatment with 8-Br-cAMP, a cAMP analog; or forskolin, which stimulates cAMP production, resulted in an increase in dye transfer between adjacent TEC, inducing a three-fold enhancement in the mean fluorescence of coupled cells, ascertained by flow cytometry after calcein transfer. These treatments also increased Cx43 mRNA expression, and stimulated Cx43 protein accumulation in regions of intercellular contacts. VIP, adenosine, and epinephrine which may also signal through cyclic nucleotides were tested. The first two molecules did not mimic the effects of 8-Br-cAMP, however epinephrine was able to increase GJIC suggesting that this molecule functions as an endogenous inter-TEC GJIC modulators. Stimulation of PKC by phorbol-myristate-acetate inhibited inter-TEC GJIC. Importantly, both the enhancing and the decreasing effects, respectively induced by cAMP and PKC, were observed in both mouse and human TEC preparations. Lastly, experiments using mouse thymocyte/TEC heterocellular co-cultures suggested that the presence of thymocytes does not affect the degree of inter-TEC GJIC.</p> <p>Conclusions</p> <p>Overall, our data indicate that cAMP and PKC intracellular pathways are involved in the homeostatic control of the gap junction-mediated communication in the thymic epithelium, exerting respectively a positive and negative role upon cell coupling. This control is phylogenetically conserved in the thymus, since it was seen in both mouse and human TEC preparations. Lastly, our work provides new clues for a better understanding of how the thymic epithelial network can work as a physiological syncytium.</p

    Complete Inactivation of Sebum-Producing Genes Parallels the Loss of Sebaceous Glands in Cetacea

    Publisher's version (Ăștgefin grein)Genomes are dynamic biological units, with processes of gene duplication and loss triggering evolutionary novelty. The mammalian skin provides a remarkable case study on the occurrence of adaptive morphological innovations. Skin sebaceous glands (SGs), for instance, emerged in the ancestor of mammals serving pivotal roles, such as lubrication, waterproofing, immunity, and thermoregulation, through the secretion of sebum, a complex mixture of various neutral lipids such as triacylglycerol, free fatty acids, wax esters, cholesterol, and squalene. Remarkably, SGs are absent in a few mammalian lineages, including the iconic Cetacea. We investigated the evolution of the key molecular components responsible for skin sebum production: Dgat2l6, Awat1, Awat2, Elovl3, Mogat3, and Fabp9. We show that all analyzed genes have been rendered nonfunctional in Cetacea species (toothed and baleen whales). Transcriptomic analysis, including a novel skin transcriptome from blue whale, supports gene inactivation. The conserved mutational pattern found in most analyzed genes, indicates that pseudogenization events took place prior to the diversification of modern Cetacea lineages. Genome and skin transcriptome analysis of the common hippopotamus highlighted the convergent loss of a subset of sebum-producing genes, notably Awat1 and Mogat3. Partial loss profiles were also detected in non-Cetacea aquatic mammals, such as the Florida manatee, and in terrestrial mammals displaying specialized skin phenotypes such as the African elephant, white rhinoceros and pig. Our findings reveal a unique landscape of “gene vestiges” in the Cetacea sebum-producing compartment, with limited gene loss observed in other mammalian lineages: suggestive of specific adaptations or specializations of skin lipids.This work was supported by Project No. 031342 cofinanced by COMPETE 2020, Portugal 2020 and the European Union through the ERDF, and by Fundacža~o para a Cie^ncia e a Tecnologia through national funds.     Health-related quality of life in well-differentiated metastatic gastroenteropancreatic neuroendocrine tumors

Gastroenteropancreatic neuroendocrine tumors (GEP-NETs) are rare neoplasms capable of producing hormones. The development of new treatments has improved progression-free survival, albeit with increased toxicity. Health-related quality of life (HRQoL) has become an important endpoint in clinical research to evaluate patients’ well-being in such a contradictory scenario. In this review, we examine key reported outcomes across clinical studies exploring HRQoL in patients with GEP-NETs. We have conducted a review of the literature using PubMed, The Cochrane Library, EMBASE, and Google Scholar. Selection criteria for articles were (1) publication in English between 1995 and 2014, (2) patients with GEP-NET, and (3) analysis of HRQoL, including mental health and psychological symptoms. Forty-nine studies met the inclusion criteria (31 clinical trials, 14 observational studies, and 4 developments of NET-specific HRQoL instruments). The scope and nature of the literature was diverse with 27 instruments used to measure aspects of HRQoL. EORTC QLQ-C30 was the most frequently used, in 38 of the 49 studies. Standardized measures revealed that in spite of generally good HRQoL, GEP-NET patients have specific psychological and physical complaints. The clinical benefit of somatostatin analogs and sunitinib has been clearly supported by HRQoL assessment. Improvement in HRQoL scores or symptom relief over time was also reported in 14 trials of peptide receptor radionuclide therapy, however the absence of randomized studies obviate definitive conclusions. We have also identified several unanswered questions that should be addressed in further research concerning chemotherapy, everolimus, surgery, local ablative therapies, and chemoembolization. Future research should incorporate GEP-NET-specific HRQoL instruments into phase III trials.     Oral implant osseointegration model in C57Bl/6 mice: microtomographic, histological, histomorphometric and molecular characterization

    Despite the successful clinical application of titanium (Ti) as a biomaterial, the exact cellular and molecular mechanisms responsible for Ti osseointegration remains unclear, especially because of the limited methodological tools available in this field. Objective: In this study, we present a microscopic and molecular characterization of an oral implant osseointegration model using C57Bl/6 mice. Material and Methods: Forty-eight male wild-type mice received a Ti implant on the edentulous alveolar crest and the peri-implant sites were evaluated through microscopic (ÎŒCT, histological and birefringence) and molecular (RealTimePCRarray) analysis in different points in time after surgery (3, 7, 14 and 21 days). Results: The early stages of osseointegration were marked by an increased expression of growth factors and MSC markers. Subsequently, a provisional granulation tissue was formed, with high expression of VEGFb and earlier osteogenic markers (BMPs, ALP and Runx2). The immune/inflammatory phase was evidenced by an increased density of inflammatory cells, and high expression of cytokines (TNF, IL6, IL1) chemokines (CXCL3, CCL2, CCL5 and CXC3CL1) and chemokine receptors (CCR2 and CCR5). Also, iNOS expression remained low, while ARG1 was upregulated, indicating predominance of a M2-type response. At later points in time, the bone matrix density and volume were increased, in agreement with a high expression of Col1a1 and Col21a2. The remodelling process was marked by peaks of MMPs, RANKL and OPG expression at 14 days, and an increased density of osteoclasts. At 21 days, intimate Ti/bone contact was observed, with expression of final osteoblast differentiation markers (PHEX, SOST), as well as red spectrum collagen fibers. Conclusions: This study demonstrated a unique molecular view of oral osseointegration kinetics in C57Bl/6 mice, evidencing potential elements responsible for orchestrating cell migration, proliferation, ECM deposition and maturation, angiogenesis, bone formation and remodeling at the bone-implant interface in parallel with a novel microscopic analysis
