    NMR quantification of 16-O-methylcafestol and kahweol in Coffea canephora var. robusta beans from different geographical origins

    Diterpenes have recently received a great deal of interest as tools to investigate the botanical origin of coffee. Specifically, kahweol has been proposed as a marker of Coffea arabica while 16-O-methylcafestol (16-OMC) is a Coffea canephora specific marker and its detection and quantification allow the authenticity of pure C. arabica roasted coffee blends to be assessed. In this study, we evaluated the possibility of the industrial use of the quantification of these diterpenes to assess the relative amounts of the two coffee species in blends. The content of 16-OMC and kahweol was determined in 78 samples (i.e., 39 green and the corresponding 39 roasted beans) of C. canephora from different geographical origins using a recently published NMR approach. Our results show a small natural variability in 16-OMC content for the Asian samples (average content = 1837 \ub1 113 mg/kg) while a much larger spread was found for the African samples (average content = 1744 \ub1 322 mg/kg). This large variability prevents the use of 16-OMC to quantify C. canephora in unknown roasted coffee blends. We also show that kahweol cannot be considered a specific C. arabica marker since it was detected almost all coffees and quantified in about 30% of the C. canephora samples

    Mean Field Theory of the Localization Transition

    A mean field theory of the localization transition for bosonic systems is developed. Localization is shown to be sensitive to the distribution of the random site energies. It occurs in the presence of a triangular distribution, but not a uniform one. The inverse participation ratio, the single site Green's function, the superfluid order parameter and the corresponding susceptibility are calculated, and the appropriate exponents determined.     Dimensional crossover of a boson gas in multilayers

    We obtain the thermodynamic properties for a non-interacting Bose gas constrained on multilayers modeled by a periodic Kronig-Penney delta potential in one direction and allowed to be free in the other two directions. We report Bose-Einstein condensation (BEC) critical temperatures, chemical potential, internal energy, specific heat, and entropy for different values of a dimensionless impenetrability P0P\geqslant 0 between layers. The BEC critical temperature TcT_{c} coincides with the ideal gas BEC critical temperature T0T_{0} when P=0P=0 and rapidly goes to zero as PP increases to infinity for any finite interlayer separation. The specific heat CVC_{V} \textit{vs} TT for finite PP and plane separation aa exhibits one minimum and one or two maxima in addition to the BEC, for temperatures larger than TcT_{c} which highlights the effects due to particle confinement. Then we discuss a distinctive dimensional crossover of the system through the specific heat behavior driven by the magnitude of PP. For T<TcT<T_{c} the crossover is revealed by the change in the slope of logCV(T)\log C_{V}(T) and when T>TcT>T_{c}, it is evidenced by a broad minimum in CV(T)C_{V}(T).Comment: Ten pages, nine figure

    Outcome comparison between radiation therapy and surgery as primary treatment for dogs with periarticular histiocytic sarcoma: An Italian Society of Veterinary Oncology study

    Localized histiocytic sarcoma may occur as a primary lesion in periarticular tissues of large appendicular joints. Treatment options for the primary lesion include radical surgical excision, radiation therapy (RT), or both, in combination with chemotherapy for potential systemic metastases. In an effort to better characterize the time to progression (TTP) following surgical vs non-surgical approaches for periarticular histiocytic sarcoma (PAHS), a contemporary European population of affected dogs was retrospectively surveyed. Medical records were queried for newly-diagnosed PAHS cases undergoing surgery (predominantly limb amputation) or RT followed by systemic chemotherapy. Of 49 dogs, 34 underwent RT and 15 underwent surgery. All dogs received adjuvant chemotherapy. There was no statistically significant difference in TTP or overall survival between groups. The median TTP was 336 days for the operated dogs and 217 days for the irradiated dogs (P =.117). The median overall survival time was 398 days for the operated dogs and 240 days for the irradiated dogs (P =.142). On multi-variable analysis, the variables significantly associated with an increased risk of both tumour progression and tumour-related death were regional lymph node and distant metastasis at admission. Survival and local control rates following RT may be comparable to radical resection. These data may better inform shared decision-making processes between multi-disciplinary care providers and owners

    Interaction effects in non-Hermitian models of vortex physics

    Vortex lines in superconductors in an external magnetic field slightly tilted from randomly-distributed parallel columnar defects can be modeled by a system of interacting bosons in a non-Hermitian vector potential and a random scalar potential. We develop a theory of the strongly-disordered non-Hermitian boson Hubbard model using the Hartree-Bogoliubov approximation and apply it to calculate the complex energy spectra, the vortex tilt angle and the tilt modulus of (1+1)-dimensional directed flux line systems. We construct the phase diagram associated with the flux-liquid to Bose-glass transition and find that, close to the phase boundary, the tilted flux liquid phase is characterized by a band of localized excitations, with two mobility edges in its low-energy spectrum.Comment: 19 pages, 19 figures, To appear in Phys.     Feline large granular lymphocyte lymphoma: An Italian Society of Veterinary Oncology (SIONCOV) retrospective study

    Feline large granular lymphocyte (LGL) lymphoma is an uncommon subtype of lymphoma characterized by a grave prognosis and scarce response to chemotherapy. There are limited reports on clinico-pathological and prognostic factors. One-hundred and 9 cats with newly diagnosed LGL lymphoma that underwent initial staging (including hematology, serum biochemistry, thoracic radiographs and abdominal ultrasound), and followed-up were retrospectively evaluated. LGL lymphoma was localized within the gastrointestinal tract with or without extra-intestinal involvement in 91.7% of the cases, and at extra-gastrointestinal sites in 8.3%. Symptoms were frequent. Anemia (31.2%) and neutrophilia (26.6%) were commonly observed, and 14 (12.8%) cats had neoplastic circulating cells. Frequent biochemistry abnormalities included elevated ALT (39.4%) and hypoalbuminemia (28.4%). Twenty (54.1%) of 37 cats had elevated serum LDH. Treatment varied among cats, and included surgery (11%), chemotherapy (23%), corticosteroids (38.5%) and no treatment (27.5%). Median time to progression (MTTP) was 5 days, and median survival time (MST) 21 days. MST was significantly shorter in the case of substage b, circulating neoplastic cells, lack of chemotherapy administration, and lack of treatment response. A small subset of cats (7.3%) survived more than 6 months, suggesting that a more favorable clinical course can be found among LGL lymphoma patients

    Breed-associated risks for developing canine lymphoma differ among countries: an European canine lymphoma network study

    Canine breeds may be considered good animal models for the study of genetic predisposition to cancer, as they represent genetic clusters. From epidemiologic and case collection studies it emerges that some breeds are more likely to develop lymphoma or specific subtypes of lymphoma but available data are variable and geographically inconsistent. This study was born in the context of the European Canine Lymphoma Network with the aim of investigating the breed prevalence of canine lymphoma in different European countries and of investigating possible breed risk of lymphoma overall and/or different lymphoma subtypes

    NKG2A is a late immune checkpoint on CD8 T cells and marks repeated stimulation and cell division

    The surface inhibitory receptor NKG2A forms heterodimers with the invariant CD94 chain and is expressed on a subset of activated CD8 T cells. As antibodies to block NKG2A are currently tested in several efficacy trials for different tumor indications, it is important to characterize the NKG2A(+) CD8 T cell population in the context of other inhibitory receptors. Here we used a well-controlled culture system to study the kinetics of inhibitory receptor expression. Naive mouse CD8 T cells were synchronously and repeatedly activated by artificial antigen presenting cells in the presence of the homeostatic cytokine IL-7. The results revealed NKG2A as a late inhibitory receptor, expressed after repeated cognate antigen stimulations. In contrast, the expression of PD-1, TIGIT and LAG-3 was rapidly induced, hours after first contact and subsequently down regulated during each resting phase. This late, but stable expression kinetics of NKG2A was most similar to that of TIM-3 and CD39. Importantly, single-cell transcriptomics of human tumor-infiltrating lymphocytes (TILs) showed indeed that these receptors were often coexpressed by the same CD8 T cell cluster. Furthermore, NKG2A expression was associated with cell division and was promoted by TGF-beta in vitro, although TGF-beta signaling was not necessary in a mouse tumor model in vivo. In summary, our data show that PD-1 reflects recent TCR triggering, but that NKG2A is induced after repeated antigen stimulations and represents a late inhibitory receptor. Together with TIM-3 and CD39, NKG2A might thus mark actively dividing tumor-specific TILs.Experimental cancer immunology and therap

    Benchmarking of cell type deconvolution pipelines for transcriptomics data

    Many computational methods have been developed to infer cell type proportions from bulk transcriptomics data. However, an evaluation of the impact of data transformation, pre-processing, marker selection, cell type composition and choice of methodology on the deconvolution results is still lacking. Using five single-cell RNA-sequencing (scRNA-seq) datasets, we generate pseudo-bulk mixtures to evaluate the combined impact of these factors. Both bulk deconvolution methodologies and those that use scRNA-seq data as reference perform best when applied to data in linear scale and the choice of normalization has a dramatic impact on some, but not all methods. Overall, methods that use scRNA-seq data have comparable performance to the best performing bulk methods whereas semi-supervised approaches show higher error values. Moreover, failure to include cell types in the reference that are present in a mixture leads to substantially worse results, regardless of the previous choices. Altogether, we evaluate the combined impact of factors affecting the deconvolution task across different datasets and propose general guidelines to maximize its performance. Inferring cell type proportions from transcriptomics data is affected by data transformation, normalization, choice of method and the markers used.
