Coincidence of Small-scale Spatial Discontinuities in Leaf Morphology and Nuclear Microsatellite Variation of Quercus petraea and Q. robur in a Mixed Forest
Background and Aims The taxon complex comprising Quercus petraea and Q. robur shows distinct morphologies and ecological preferences, but mostly low differentiation in various types of molecular markers at a broad spatial range. Local, spatially explicit analyses may reveal patterns induced by microevolutionary processes operating mainly over short distances. However, no attempts have been made to date to explore the potential of spatial analyses combining morphological and genetic data of these oaks. Methods A mixed oak stand was studied to elucidate the small-scale population genetic structure. All adult individuals were classified and putative hybrids were identified using multivariate discrimination analysis of leaf morphological characters. Likewise, all trees were genotyped with five nuclear microsatellites, and a Bayesian assignment method was applied based on maximum likelihood of multilocus genotypes for taxon and putative hybrid classification. Key Results Multivariate analyses of leaf morphological data recognized two groups with few individuals as putative hybrids. These groups were significantly differentiated at the five microsatellites, and genetic taxon assignment coincided well with morphological classification. Furthermore, most putative hybrids were assigned to the taxon found in their spatial neighbourhood. When grouping trees into clusters according to their spatial positions, these clusters were clearly dominated by one taxon. Discontinuities in morphological and genetic distance matrices among these clusters showed high congruence. Conclusions The spatial-genetic analyses and the available literature led to the assumption that reproductive barriers, assortative mating, limited seed dispersal and microsite-induced selection in favour of the locally adapted taxon at the juvenile stage may reinforce taxon-specific spatial aggregation that fosters species separation. Thus, the results tend to support the hypothesis that Q. petraea and Q. robur are distinct taxa which share a recent common ancestry. Chloroplast DNA analysis in oak stands (Quercus robur L.) in North Rhine-Westphalia with presumably Slavonian origin: Is there an association between geographic origin and bud phenology?
Slavonian oaks (Quercus robur subsp. slavonica) have been introduced into Germany in the second half of the 19th century from the lowlands of the rivers Save and Drava in today’s Croatia. If compared to indigenous oak stands, they are characterized by good growth, comparatively low seed production and a late bud burst. Based on the information of European-wide variation patterns at chloroplast DNA markers in oaks we adapted chloroplast microsatellites for the analysis of all oak stands of presumably Slavonian origin in the Münsterland and lower Rhine regions. We were able to distinguish between Slavonian haplotypes with no natural occurrence in the study area and indigenous types that do not occur in the Balkan region. A generally high differentiation among stands was observed at chloroplast markers (GST = 0.674). Based on the haplotype information and historic records we found that stands with Slavonian material have been established between the years 1878 and 1903. In a total of 910 analysed trees the Slavonian haplotypes 5, 2 or 17 were the most frequent ones but a considerable amount of samples with indigenous haplotype 1 or haplotype10 with presumed origin in Southwestern Europe was also present. A clear association between haplotype 2 and late bud burst was detected in adult stands and in a field trial established with seeds from Slavonian and indigenous oak stands. The information about the haplotype composition in all Slavonian stands can be used as reference for the certification of reproductive material. The analysis of cpDNA haploytpes in old oak stands that had been established before the introduction of foreign seed material can give valuable information for the identification of indigenous oak stands
Application of genetic markers to the discrimination of European Black Poplar ( Populus nigra ) from American Black Poplar ( P. deltoides ) and Hybrid Poplars ( P. x canadensis ) in Switzerland
European Black Poplar (Populus nigra) is considered a rare and endangered tree species because of severe reduction of its natural riverine habitat and potential hybridisation with the related non-indigenous taxa P. deltoides and P. x canadensis. As it is difficult to distinguish these taxa solely based on their morphology, we applied a PCR-based assay with an easy-to-use and robust molecular marker set (cpDNA trnL-trnF/RsaI RFLP, nDNA win3 and nDNA POPX/MspI RFLP) in order to identify pure P. nigra. Different plant tissues could be used for fast and standardised DNA extraction. The application of the three marker types was tested on a number of different Populus taxa, and they were also used for the verification of pure P. nigra in a sample of 304 putative P. nigra individuals from Switzerland. Cross-checking of the DNA data with those using a traditional allozyme approach resulted in complete agreement. The availability of molecular identification methods is an important prerequisite for the conservation of European Black Poplar, because pure, non-introgressed plant material can then be used in restoration projects of European floodplain
Narrow genetic base in forest restoration with holm oak (Quercus ilex L.) in Sicily
In order to empirically assess the effect of actual seed sampling strategy on
genetic diversity of holm oak (Quercus ilex) forestations in Sicily, we have
analysed the genetic composition of two seedling lots (nursery stock and
plantation) and their known natural seed origin stand by means of six nuclear
microsatellite loci. Significant reduction in genetic diversity and significant
difference in genetic composition of the seedling lots compared to the seed
origin stand were detected. The female and the total effective number of
parents were quantified by means of maternity assignment of seedlings and
temporal changes in allele frequencies. Extremely low effective maternity
numbers were estimated (Nfe 2-4) and estimates accounting for both
seed and pollen donors gave also low values (Ne 35-50). These values
can be explained by an inappropriate forestry seed harvest strategy limited to
a small number of spatially close trees

Leaf morphological differentiation between Quercus robur and Quercus petraea is stable across western European mixed oak stands
© INRA, EDP Sciences 2002Leaf morphology was assessed in nine mixed oak stands (Quercus petraea and Q. robur ) located in eight European countries. Exhaustive sampling was used in an area of each stand where the two species coexisted in approximately equal proportions (about 170 trees/species/stand). Fourteen leaf characters were assessed on each of 5 to10 leaves collected from the upper part of each tree. Three multivariate statistical techniques (CDA, canonical discriminant analysis; PCA, principal component analysis; MCA, multiple correspondence analysis) were used in two different ways: first on the total set of leaves over all stands (global analysis) and second, separately within each stand (local analysis). There was a general agreement of the results among the statistical methods used and between the analyses conducted (global and local). The first synthetic variable derived by each multivariate analysis exhibited a clear and sharp bimodal distribution, with overlapping in the central part. The two modes were interpreted as the two species, and the overlapping region was interpreted as an area where the within-species variations were superimposed. There was no discontinuity in the distribution or no visible evidence of a third mode which would have indicated the existence of a third population composed of trees with intermediate morphologies. Based on petiole length and number of intercalary veins, an "easy to use" discriminant function applicable to a major part of the natural distribution of the species was constructed. Validation on an independent set of trees provided a 98% rate of correct identification. The results were interpreted in the light of earlier reports about extensive hybridization occurring in mixed oak stands. Maternal effects on morphological characters, as well as a lower frequency or fitness of hybrids in comparison with parent species could explain the maintenance of two modes, which might be composed of either pure species or pure species and introgressed forms.Antoine Kremer, Jean Luc Dupouey, J. Electromagnetic Field Effect or Simply Stress? Effects of UMTS Exposure on Hippocampal Longterm Plasticity in the Context of Procedure Related Hormone Release
Harmful effects of electromagnetic fields (EMF) on cognitive and behavioural features of humans and rodents have been controversially discussed and raised persistent concern about adverse effects of EMF on general brain functions. In the present study we applied radio-frequency (RF) signals of the Universal Mobile Telecommunications System (UMTS) to full brain exposed male Wistar rats in order to elaborate putative influences on stress hormone release (corticosteron; CORT and adrenocorticotropic hormone; ACTH) and on hippocampal derived synaptic long-term plasticity (LTP) and depression (LTD) as electrophysiological hallmarks for memory storage and memory consolidation. Exposure was computer controlled providing blind conditions. Nominal brain-averaged specific absorption rates (SAR) as a measure of applied mass-related dissipated RF power were 0, 2, and 10 W/kg over a period of 120 min. Comparison of cage exposed animals revealed, regardless of EMF exposure, significantly increased CORT and ACTH levels which corresponded with generally decreased field potential slopes and amplitudes in hippocampal LTP and LTD. Animals following SAR exposure of 2 W/kg (averaged over the whole brain of 2.3 g tissue mass) did not differ from the sham-exposed group in LTP and LTD experiments. In contrast, a significant reduction in LTP and LTD was observed at the high power rate of SAR (10 W/kg). The results demonstrate that a rate of 2 W/kg displays no adverse impact on LTP and LTD, while 10 W/kg leads to significant effects on the electrophysiological parameters, which can be clearly distinguished from the stress derived background. Our findings suggest that UMTS exposure with SAR in the range of 2 W/kg is not harmful to critical markers for memory storage and memory consolidation, however, an influence of UMTS at high energy absorption rates (10 W/kg) cannot be excluded
Padrão geográfico de diversidade genética em populações naturais de Pau-rosa (Aniba rosaeodora), na Amazônia Central
Rosewood (Aniba rosaeodora Ducke, Lauraceae) is an Amazonian evergreen tree and a source of the purest linalool, the main component of its essential oil, which is very valuable in the international perfumery market. After decades of over-exploitation it is currently considered as threatened. We evaluated the genetic diversity and its distribution in four populations in Central Amazonia. Thirty-five reliable RAPD markers were generated, of which 32 were polymorphic (91.4%). Variation was higher within the populations (76.5%; p < 0.0001) and geographic distribution contributed to population differentiation (23.4%; p < 0.0001). The Amazon River had a small influence on gene flow (3.3%; p < 0.0001), but we identified evidence of gene flow across the river. There were significant differences in marker frequencies (p < 0.05), in agreement with the low gene flow (Nm = 2.02). The correlation between genetic distance and gene flow was - 0.95 (p = 0.06) and between geographic distance and gene flow was -0.78 (p = 0.12). There was a geographic cline of variability across an East-West axis, influenced as well by the Amazon River, suggesting the river could be a barrier to gene flow. Although threatened, these Rosewood populations retain high diversity, with the highest levels in the Manaus population, which has been protected for over 42 years in a Reserve.O Pau-rosa (Aniba rosaeodora Ducke, Lauraceae) é uma árvore amazônica fonte do mais puro linalol, o qual é o principal componente do seu óleo essencial e muito valioso no mercado internacional de perfumaria. Após várias décadas de intensa exploração, a espécie foi levada à categoria de ameaçada de extinção. Quatro populações naturais distribuídas na bacia Amazônia Central foram avaliadas quanto ao nível e a distribuição da diversidade genética. Trinta e cinco marcadores RAPD reprodutíveis foram gerados, dos quais 32 foram polimórficos (91,4%). A diversidade foi maior dentro das populações (76,5%; p < 0,0001) e a distribuição geográfica contribuiu para a diferenciação entre as populações (23,4%; p < 0,0001). A AMOVA indicou que pode haver uma influência parcial do Rio Amazonas no fluxo gênico (3,3%; p < 0,0001), mas foram identificadas evidências de fluxo gênico atravessando o rio. Houve diferenças significativas nas freqüências dos marcadores (p < 0,05) e o fluxo gênico estimado foi relativamente baixo (Nm = 2,02). A correlação entre a distância genética e o fluxo gênico foi de - 0,95 (p = 0,06) e para a distância geográfica e o fluxo gênico foi de - 0,78 (p = 0,12). Houve um padrão geográfico de variabilidade ao longo do eixo Leste - Oeste, influenciado também pelo Rio Amazonas, o que sugere que o rio poderia funcionar como uma barreira para o fluxo gênico. Apesar de ameaçadas, estas populações de Pau-rosa possuem alta diversidade, com o maior valor na população de Manaus, que vem sendo protegida por 42 anos em uma reserva
