Lack of mutations of exon 2 of the MEN1 gene in endocrine and nonendocrine sporadic tumors
In addition to the mutations that underlie most cases of the multiple endocrine neoplasia type 1 (MEN1) syndrome, somatic mutations of the MEN1 gene have also been described in sporadic tumors like gastrinomas, insulinomas and bronchial carcinoid neoplasm. We examined exon 2 of this gene, where most of the mutations have been described, in 148 endocrine and nonendocrine sporadic tumors. DNA was obtained by phenol/chloroform extraction and ethanol precipitation from 92 formalin-fixed, paraffin-embedded samples, and from 40 fresh tumor tissue samples. We used 5 pairs of primers to encompass the complete coding sequence of exon 2 of the MEN1 gene that was screened by the polymerase chain reaction-single-stranded conformation polymorphism (PCR-SSCP) technique in 78 sporadic thyroid cancers: 28 follicular adenomas, 35 papillary carcinomas, 14 follicular carcinomas, and 1 anaplastic thyroid carcinoma. We also examined 46 adrenal lesions (3 hyperplasias, 3 adenomas and 35 adrenocortical carcinomas, 2 pheochromocytomas, 2 ganglioneuroblastomas, and 1 lymphoma) and 24 breast cancers (6 noninvasive, 16 infiltrating ductal, and 2 invasive lobular tumors). The PCR product of 5 tumors suspected to present band shifts by SSCP was cloned. Direct sense and antisense sequencing did not identify mutations. These results suggest that the MEN1 gene is not important in breast, thyroid or adrenal sporadic tumorigenesis. Because the frequency of mutations varies significantly among tumor subgroups and allelic deletions are frequently observed at 11q13 in thyroid and adrenal cancers, another tumor suppressor gene residing in this region is likely to be involved in the tumorigenesis of these neoplasms.86186
Nanoparticles In Treatment Of Thermal Injured Rats: Is It Safe?
The aim of this study was to assess whether thermal trauma induced oxidative stress altered the balance between oxidant and antioxidant systems in the blood of burn wound rats in the absence and presence of silver nanoparticles and S-nitrosoglutathione, GSNO. Free silver nanoparticles, free GSNO and silver nanoparticles + GSNO had no cytotoxic effects. Under anesthesia, the shaved dorsum of the rats was exposed to 90°C (burn group) water bath. Studied compounds were administered topically immediately and at 28 days after the burn injury, four times a day. Silver nanoparticles and silver nanoparticles + GSNO were no toxic in vitro and in vivo. There were no significant differences in the levels of urea, creatinine, aminotransferases and hematological parameters, in control-burn groups (free silver nanoparticles) and treated-burn groups (free GSNO or silver nanoparticles + GSNO). There were no differences in lipid peroxidation and in the levels of protein carbonyls and glutathione, used as oxidative stress markers. A little inflammatory cell response, papillary dermis vascularization, fibroblasts differentiated into contractile myofibroblasts and the presence of a large amount of extracellular matrix were evidenced in treated groups following skin injury. A multi-resolution, non-parametric, Bayesian framework for identification of spatially-varying model parameters
This paper proposes a hierarchical, multi-resolution framework for the
identification of model parameters and their spatially variability from noisy
measurements of the response or output. Such parameters are frequently
encountered in PDE-based models and correspond to quantities such as density or
pressure fields, elasto-plastic moduli and internal variables in solid
mechanics, conductivity fields in heat diffusion problems, permeability fields
in fluid flow through porous media etc. The proposed model has all the
advantages of traditional Bayesian formulations such as the ability to produce
measures of confidence for the inferences made and providing not only
predictive estimates but also quantitative measures of the predictive
uncertainty. In contrast to existing approaches it utilizes a parsimonious,
non-parametric formulation that favors sparse representations and whose
complexity can be determined from the data. The proposed framework in
non-intrusive and makes use of a sequence of forward solvers operating at
various resolutions. As a result, inexpensive, coarse solvers are used to
identify the most salient features of the unknown field(s) which are
subsequently enriched by invoking solvers operating at finer resolutions. This
leads to significant computational savings particularly in problems involving
computationally demanding forward models but also improvements in accuracy. It
is based on a novel, adaptive scheme based on Sequential Monte Carlo sampling
which is embarrassingly parallelizable and circumvents issues with slow mixing
encountered in Markov Chain Monte Carlo schemes
Undamning the Douro river catchment: a stepwise approach for prioritizing dam removal
Dams provide water supply, flood protection, and hydropower generation benefits, but
also harm native species by altering the natural flow regime, and degrading the aquatic and riparian
habitats. In the present study, which comprised the Douro River basin located in the North of
Portugal, the cost-benefit assessment of dams was based upon a balance between the touristic
benefits of a dammed Douro, and the ecological benefits of less fragmented Douro sub-catchments.
Focused on four sub-catchments (Sabor, Tâmega, Côa and Corgo), a probabilistic stream connectivity
model was developed and implemented to recommend priorities for dam removal, where this action
could significantly improve the movement of potadromous fish species along the local streams.
The proposed model accounts for fish movement across the dam or weir (permeability), which
is a novel issue in connectivity models. However, before any final recommendation on the fate
of a dam or weir, the connectivity results will be balanced with other important socio-economic
interests. While implementing the connectivity model, an inventory of barriers (dams and weirs)
was accomplished through an observation of satellite images. Besides identification and location of
any obstacles, the inventory comprised the compilation of data on surrounding land use, reservoir
water use, characteristics of the riparian gallery, and permeability conditions for fish, among others.
All this information was stored in a geospatial dataset that also included geographical information
on the sub-catchment drainage network. The linear (drainage network) and point (barriers) source
data were processed in a computer program that provided or returned numbers for inter-barrier
stream lengths (habitat), and the barrier permeability. These numbers were finally used in the
same computer program to calculate a habitat connector index, and a link improvement index,
used to prioritize dam removal based upon structural connectivity criteria. The results showed
that habitat patch connectivity in the Sabor, Tâmega and Côa sub-catchments is not dramatically
affected by the installed obstacles, because most link improvement values were generally low. For the
opposite reason, in the Corgo sub-catchment, obstacles may constitute a relatively higher limitation
to connectivity, and in this case the removal of eight obstacles could significantly improve this
connectivity. Using the probabilistic model of structural connectivity, it was possible to elaborate
a preliminary selection of dams/weirs that critically limit stream connectivity, and that will be the
focus of field hydraulic characterization to precisely determine fish movement along the associated
river stretches. Future work will also include the implementation of a multi-criteria decision support
system for dam removal or mitigation of the critical structures, as well to define exclusion areas for

Inhomogeneous cosmologies with Q-matter and varying
Starting from the inhomogeneous shear--free Nariai metric we show, by solving
the Einstein--Klein--Gordon field equations, how a self--interacting scalar
field plus a material fluid, a variable cosmological term and a heat flux can
drive the universe to its currently observed state of homogeneous accelerated
expansion. A quintessence scenario where power-law inflation takes place for a
string-motivated potential in the late--time dominated field regime is

Cosmic Microwave Background Anisotropy with Cosine-Type Quintessence
We study the Cosmic Microwave Background (CMB) anisotropies produced by
cosine-type quintessence models. In our analysis, effects of the adiabatic and
isocurvature fluctuations are both taken into account. For purely adiabatic
fluctuations with scale invariant spectrum, we obtain a stringent constraint on
the model parameters using the CMB data from COBE, BOOMERanG and MAXIMA.
Furthermore, it is shown that isocurvature fluctuations have significant
effects on the CMB angular power spectrum at low multipoles in some parameter
space, which may be detectable in future satellite experiments. Such a signal
may be used to test the cosine-type quintessence models.Comment: 21 pages, 9 figure
