    IL-12 and IL-4 activate a CD39-dependent intrinsic peripheral tolerance mechanism in CD8+ T cells

    Immune responses to protein antigens involve CD4+ and CD8+ T cells, which follow distinct programs of differentiation. Naïve CD8 T cells rapidly develop cytotoxic T-cell (CTL) activity after T-cell receptor stimulation, and we have previously shown that this is accompanied by suppressive activity in the presence of specific cytokines, i.e. IL-12 and IL-4. Cytokine-induced CD8+ regulatory T (Treg) cells are one of several Treg-cell phenotypes and are Foxp3− IL-10+ with contact-dependent suppressive capacity. Here, we show they also express high level CD39, an ecto-nucleotidase that degrades extracellular ATP, and this contributes to their suppressive activity. CD39 expression was found to be upregulated on CD8+ T cells during peripheral tolerance induction in vivo, accompanied by release of IL-12 and IL-10. CD39 was also upregulated during respiratory tolerance induction to inhaled allergen and on tumor-infiltrating CD8+ T cells. Production of IL-10 and expression of CD39 by CD8+ T cells was independently regulated, being respectively blocked by extracellular ATP and enhanced by an A2A adenosine receptor agonist. Our results suggest that any CTL can develop suppressive activity when exposed to specific cytokines in the absence of alarmins. Thus negative feedback controls CTL expansion under regulation from both nucleotide and cytokine environment within tissues

    Misconceptions and alternative conceptions refer to students\u27 conceptions that are different from those held by the scientific community (Sungur, Tekkaya, & Geban, 2001, p. 91). All students have misconceptions due to prior experiences they have had. Students\u27 alternative conceptions are problems for educators because they can block new learning (Sewell, 2002). Therefore, teachers play a crucial role in helping students construct new knowledge upon whatever foundation that already exists. This review of literature examines the findings of research regarding misconceptions, common misconceptions that students have in life science, and how teachers can teach difficult concepts so students are more likely to truly understand what they have learned.     A pregnancy test: women workers and the hybrid American welfare state, 1940-1993

    This dissertation shows that the US developed the outlines of a maternity policy at least by WWII. Rarely were the needs of pregnant workers or new mothers at the top of social policy initiatives. However, when European countries were developing their plans, reformers and bureaucrats sought to establish similar plans in the United States and, for a while, seemed like they might. Politics intervened in the form of the Cold War. With a few state level exceptions, the experiences of WWII were largely dismantled in the wake of political changes, business and medical opposition and the Red Scare. Subsequent policies that emerged grew largely in the private sector where women’s disadvantages in the workforce constrained maternity in the blossoming system of employee fringe benefits. Where they could, unions defended women’s access to contractual benefits, but this effort was hampered by the marginalization of maternity in the private system. Finally, with the emergence of a rights framework in the 1970s, feminist lawyers forced the inclusion of pregnancy into the central operating welfare state of private workforce relationships and benefits, leading to the current national maternity policy. However, conservatism and globalization limited this approach and indicate the necessity of public social supports for maternity, or family, benefits

    Reliability of perceptions of voice quality: evidence from a problem asthma clinic population

    <p>Introduction: Methods of perceptual voice evaluation have yet to achieve satisfactory consistency; complete acceptance of a recognised clinical protocol is still some way off.</p> <p>Materials and methods: Three speech and language therapists rated the voices of 43 patients attending the problem asthma clinic of a teaching hospital, according to the grade-roughness-breathiness-asthenicity-strain (GRBAS) scale and other perceptual categories.</p> <p>Results and analysis: Use of the GRBAS scale achieved only a 64.7 per cent inter-rater reliability and a 69.6 per cent intra-rater reliability for the grade component. One rater achieved a higher degree of consistency. Improved concordance on the GRBAS scale was observed for subjects with laryngeal abnormalities. Raters failed to reach any useful level of agreement in the other categories employed, except for perceived gender.</p> <p>Discussion: These results should sound a note of caution regarding routine adoption of the GRBAS scale for characterising voice quality for clinical purposes. The importance of training and the use of perceptual anchors for reliable perceptual rating need to be further investigated.</p&gt

    Mapping change in medical student attitudes regarding lesbian, gay and bisexual healthcare

    Despite significant social advances among lesbian, gay, bisexual, and transgender (LGBT) individuals, healthcare disparities remain prevalent. Medical education represents a prime opportunity for addressing this need. Thus, we undertook this study to explore second year medical students’ attitudes regarding equal access to healthcare services including erectile dysfunction medication, family planning medications and technology, and partner health insurance coverage. Nearly 1500 allopathic medical students were surveyed from 2006 to 2014 during their second pre-clinical training year. While attitudes regarding care between LGB patients and heterosexual patients were often similar, there was a significant change in favor of providing insurance coverage for in-vitro and assisted reproduction services for lesbian women (increasing from 70% of students in favor in 2008 to 87% of students in 2013). Similarly, the proportion of students in favor of providing health insurance coverage for prescription drugs to same sex partners significantly increased from 2009 to 2013. Additionally, students noted healthcare providers should be required to provide equal access to care for all patients regardless of marital status or sexual practices (96% agreement in 2006 and 98.6% agreement in 2014). Even with this significant increase in medical student approval of certain healthcare services for LGB patients, continuing to integrate LGBT healthcare topics into medical education curriculum remains critically important

    Changes in Older and Younger Woods in West-Central Ohio

    Author Institution: Dept. of Biological Sciences, Wright State University, OHThis study examines changes in two forest stands in the Quercus-Acer saccharum forest region of west central Ohio: an old-growth stand changing from Quercus-dominated to Acer saccharum-dominated and a stand established following agricultural abandonment about 1950. Both stands are in the Wright State University woods. Permanent plots were sampled in 1980 (younger stand only), 1982 (older stand only), 1993, and 2000. The older stand had more small, fewer intermediate, and more large stems than the younger stand. The plot in the new stand showed a bell-shaped distribution with most stems established shortly after land abandonment. Mortality decreased and growth increased with stem size for both stands. Acer saccharum in all sizes and large Quercus dominated the older stand. The younger stand was dominated by Robinia pseudo-acacia with Acer saccharum also important. In the older plots small stems generally were clustered, intermediate-sized stems randomly distributed, and the largest stems regularly distributed. In the younger plot small stems were aggregated while larger ones were randomly distributed. Quercus regenerated well until the late 1800s, singly or in small groups, but few stems have become established since 1900. Quercus may need fires or grazing to regenerate successfully. Both stands are changing to increased dominance by Acer saccharum and other shade-tolerant species as they lose species (Robinia pseudo-acacia in the younger stand, Quercus in the older stand) more successful under past than present conditions

    Tissue engineering for total meniscal substitution : Animal study in sheep model

    Objective: The aim of the study was to investigate the use of a novel hyaluronic acid/polycaprolactone material for meniscal tissue engineering and to evaluate the tissue regeneration after the augmentation of the implant with expanded autologous chondrocytes. Two different surgical implantation techniques in a sheep model were evaluated. Methods: Twenty-four skeletally mature sheep were treated with total medial meniscus replacements, while two meniscectomies served as empty controls. The animals were divided into two groups: cell-free scaffold and scaffold seeded with autologous chondrocytes. Two different surgical techniques were compared: in 12 animals, the implant was sutured to the capsule and to the meniscal ligament; in the other 12 animals, also a transtibial fixation of the horns was used. The animals were euthanized after 4 months. The specimens were assessed by gross inspection and histology. Results: All implants showed excellent capsular ingrowth at the periphery. Macroscopically, no difference was observed between cell-seeded and cell-free groups. Better implant appearance and integrity was observed in the group without transosseous horns fixation. Using the latter implantation technique, lower joint degeneration was observed in the cell-seeded group with respect to cell-free implants. The histological analysis indicated cellular infiltration and vascularization throughout the implanted constructs. Cartilaginous tissue formation was significantly more frequent in the cell-seeded constructs. Conclusion: The current study supports the potential of a novel HYAFF/polycaprolactone scaffold for total meniscal substitution. Seeding of the scaffolds with autologous chondrocytes provides some benefit in the extent of fibrocartilaginous tissue repair

    Minute aphids belonging to the species Myzus fataunae Shinji (Hemiptera: Aphididae) were found at a nursery in Seminole County, Florida. Morphological and molecular data support this determination. The Florida population only colonized species of Pilea Lindl. in our host range experiments. It did not colonize Fatoua villosa. Nakai. Likewise, it did not colonize tested common Florida species of Urticaceae other than Pilea spp. Myzus fataunae is adventive, and it appears to be established in the United States

    Fishing For Food: Values and Benefits Associated With Coastal Infrastructure

    While there is substantial literature about the socio-cultural characteristics and values associated with recreational and commercial fisheries in the U.S., studies directed at those who ‘fish for food’—those who depend on consuming their catch to various degrees—are relatively sparse. Using qualitative data collected through 80 semi-structured interviews with fishers in the summer and fall of 2018 in Carteret County, North Carolina, this study aims to better understand the group of recreational fishers who consume their catch by describing social and cultural dimensions and values associated with fishing for food, examining the role of infrastructure in facilitating access to benefits associated with this activity, and considering how knowledge of existing licensing regulations surrounding subsistence license waivers affect this fishing community. Interviews conducted at free public fishing structures in the region revealed that fishers derive a variety of values and benefits from fishing at these sites, including access to recreation, nutrition, a social community, and mental health benefits, which were found to be negatively impacted by Hurricane Florence in September 2018. We also found an informal economy of sharing catch on- and off-site that extends the reach and benefits facilitated by public infrastructure to people beyond those using it directly. Overall, we call for conceptualizations of ‘fishing for food’ that include aspects that go beyond traditional definitions of ‘subsistence’ or ‘recreational’ fishing such as food security, access, and less obvious social and cultural motivations behind the activity. These findings are a compelling rationalization for the creation and maintenance of formal and informal fishing places locally and, by extension, in other coastal areas, given the array of benefits provided by access to these types of locations
