    Intersection types and (positive) almost-sure termination

    Randomized higher-order computation can be seen as being captured by a λ-calculus endowed with a single algebraic operation, namely a construct for binary probabilistic choice. What matters about such computations is the probability of obtaining any given result, rather than the possibility or the necessity of obtaining it, like in (non)deterministic computation. Termination, arguably the simplest kind of reachability problem, can be spelled out in at least two ways, depending on whether it talks about the probability of convergence or about the expected evaluation time, the second one providing a stronger guarantee. In this paper, we show that intersection types are capable of precisely characterizing both notions of termination inside a single system of types: the probability of convergence of any λ-term can be underapproximated by its type, while the underlying derivation's weight gives a lower bound to the term's expected number of steps to normal form. Noticeably, both approximations are tight-not only soundness but also completeness holds. The crucial ingredient is non-idempotency, without which it would be impossible to reason on the expected number of reduction steps which are necessary to completely evaluate any term. Besides, the kind of approximation we obtain is proved to be optimal recursion theoretically: no recursively enumerable formal system can do better than that

    Phytochemical investigations on Artemisia alba Turra growing in the North-East of Italy

    Artemisia alba Turra (Asteraceae) is an Euro-Mediterranean plant used in Veneto (North-East of Italy) as traditional medicine for the treatment of various diseases. A. alba is a taxonomically problematic species, characterized by common polymorphism leading to a quite high variability in secondary metabolites content. Nonetheless, the phytochemical knowledge on its phytoconstituents, especially non-volatile components, is limited. In the present paper, the phytochemical composition of a tincture obtained from the aerial parts of A. alba growing in Veneto is presented. Extensive chromatographic separations led to the isolation of three new sesquiterpene derivatives, whose structures were elucidated by 1D and 2D NMR experiments and mass spectrometry.     Optimal fishery with coastal catch

    In many spatial resource models, it is assumed that an agent is able to harvest the resource over the complete spatial domain. However, agents frequently only have access to a resource at particular locations at which a moving biomass, such as fish or game, may be caught or hunted. Here, we analyze an infinite time‐horizon optimal control problem with boundary harvesting and (systems of) parabolic partial differential equations as state dynamics. We formally derive the associated canonical system, consisting of a forward–backward diffusion system with boundary controls, and numerically compute the canonical steady states and the optimal time‐dependent paths, and their dependence on parameters. We start with some one‐species fishing models, and then extend the analysis to a predator–prey model of the Lotka–Volterra type. The models are rather generic, and our methods are quite general, and thus should be applicable to large classes of structurally similar bioeconomic problems with boundary controls. Recommedations for Resource Managers Just like ordinary differential equation‐constrained (optimal) control problems and distributed partial differential equation (PDE) constrained control problems, boundary control problems with PDE state dynamics may be formally treated by the Pontryagin's maximum principle or canonical system formalism (state and adjoint PDEs). These problems may have multiple (locally) optimal solutions; a first overview of suitable choices can be obtained by identifying canonical steady states. The computation of canonical paths toward some optimal steady state yields temporal information about the optimal harvesting, possibly including waiting time behavior for the stock to recover from a low‐stock initial state, and nonmonotonic (in time) harvesting efforts. Multispecies fishery models may lead to asymmetric effects; for instance, it may be optimal to capture a predator species to protect the prey, even for high costs and low market values of the predators

    The effects of iodine supplementation in pregnancy on iodine status, thyroglobulin levels and thyroid function parameters: Results from a randomized controlled clinical trial in a mild-to-moderate iodine deficiency area

    Background: Iodine supplementation during pregnancy in areas with mild-to-moderate iodine deficiency is still debated. Methods: A single-center, randomized, single-blind and placebo-controlled (3:2) trial was conducted. We enrolled 90 women before 12 weeks of gestation. From enrollment up until 8 weeks after delivery, 52 women were given an iodine supplement (225 ug/day, potassium iodide tablets) and 38 were given placebo. At recruitment (T0), in the second (T1) and third trimesters (T2), and 8 weeks after delivery (T3), we measured participants\u2019 urinary iodine-to-creatinine ratio (UI/Creat), thyroid function parameters (thyroglobulin (Tg), TSH, FT3, and FT4), and thyroid volume (TV). The newborns\u2019 urinary iodine concentrations were evaluated in 16 cases. Results: Median UI/Creat at recruitment was 53.3 ug/g. UI/Creat was significantly higher in supplemented women at T1 and T2. Tg levels were lower at T1 and T2 in women with UI/Creat 65 150 ug/g, and in the Iodine group at T2 (p = 0.02). There was a negative correlation between Tg and UI/Creat throughout the study (p = 0.03, r = 120.1268). A lower TSH level was found in the Iodine group at T3 (p = 0.001). TV increased by + 067.43% in the Iodine group, and by + 0611.17% in the Placebo group. No differences were found between the newborns\u2019 TSH levels on screening the two groups. Conclusion: Tg proved a good parameter for measuring iodine intake in our placebo-controlled series. Iodine supplementation did not prove harmful to pregnancy in areas of mild-to-moderate iodine deficiency, with no appreciable harmful effect on thyroid function

    Direct evidence for microdomain-specific localization and remodeling of functional L-type calcium channels in rat and human atrial myocytes

    Background—Distinct subpopulations of L-type calcium channels (LTCCs) with different functional properties exist in cardiomyocytes. Disruption of cellular structure may affect LTCC in a microdomain-specific manner and contribute to the pathophysiology of cardiac diseases, especially in cells lacking organized transverse tubules (T-tubules) such as atrial myocytes (AMs). Methods and Results—Isolated rat and human AMs were characterized by scanning ion conductance, confocal, and electron microscopy. Half of AMs possessed T-tubules and structured topography, proportional to cell width. A bigger proportion of myocytes in the left atrium had organized T-tubules and topography than in the right atrium. Super-resolution scanning patch clamp showed that LTCCs distribute equally in T-tubules and crest areas of the sarcolemma, whereas, in ventricular myocytes, LTCCs primarily cluster in T-tubules. Rat, but not human, T-tubule LTCCs had open probability similar to crest LTCCs, but exhibited ≈40% greater current. Optical mapping of Ca2+ transients revealed that rat AMs presented ≈3-fold as many spontaneous Ca2+ release events as ventricular myocytes. Occurrence of crest LTCCs and spontaneous Ca2+ transients were eliminated by either a caveolae-targeted LTCC antagonist or disrupting caveolae with methyl-ÎČ-cyclodextrin, with an associated ≈30% whole-cell ICa,L reduction. Heart failure (16 weeks post–myocardial infarction) in rats resulted in a T-tubule degradation (by ≈40%) and significant elevation of spontaneous Ca2+ release events. Although heart failure did not affect LTCC occurrence, it led to ≈25% decrease in T-tubule LTCC amplitude. Conclusions—We provide the first direct evidence for the existence of 2 distinct subpopulations of functional LTCCs in rat and human AMs, with their biophysical properties modulated in heart failure in a microdomain-specific manner

    Nanoscale Study of Calcium Handling Remodeling in Right Ventricular Cardiomyocytes Following Pulmonary Hypertension

    Pulmonary hypertension is a complex disorder characterized by pulmonary vascular remodeling and right ventricular hypertrophy, leading to right heart failure. The mechanisms underlying this process are not well understood. We hypothesize that the structural remodeling occurring in the cardiomyocytes of the right ventricle affects the cytosolic Ca2+ handling leading to arrhythmias. After 12 days of monocrotaline-induced pulmonary hypertension in rats, epicardial mapping showed electrical remodeling in both ventricles. In myocytes isolated from the hypertensive rats, a combination of high-speed camera and confocal line-scan documented a prolongation of Ca2+ transients along with a higher local Ca2+-release activity. These Ca2+ transients were less synchronous than in controls, likely due to disorganized transverse-axial tubular system. In fact, following pulmonary hypertension, hypertrophied right ventricular myocytes showed significantly reduced number of transverse tubules and increased number of axial tubules; however, Stimulation Emission Depletion microscopy demonstrated that the colocalization of L-type Ca2+ channels and RyR2 (ryanodine receptor 2) remained unchanged. Finally, Stimulation Emission Depletion microscopy and super-resolution scanning patch-clamp analysis uncovered a decrease in the density of active L-type Ca2+ channels in right ventricular myocytes with an elevated open probability of the T-tubule anchored channels. This may represent a general mechanism of how nanoscale structural changes at the early stage of pulmonary hypertension impact on the development of the end stage failing phenotype in the right ventricle

    Surgical management and outcomes in patients with acute type A aortic dissection and cerebral malperfusion

    Objective: The study objective was to evaluate the surgical results in patients with acute type A aortic dissection and cerebral malperfusion. Methods: From 2000 to 2019, 234 patients with type A aortic dissection and cerebral malperfusion were stratified into 3 groups: 50 (21%) with syncope (group 1), 152 (65%) with persistent loss of focal neurological function (group 2), and 32 (14%) with coma (group 3). Results were evaluated and compared by univariable and multivariable analyses. Results: Median age was higher in group 1, and incidence of cardiogenic shock was higher in group 3. The femoral artery was the most common cannulation site, whereas the axillary artery was used in 18% of group 1, 30% of group 2, and 25% of group 3 patients (P = .337). Antegrade cerebral perfusion was performed in more than 80% of patients, and ascending aorta/arch replacement was performed in 40% of group 1, 27% of group 2, and 31% of group 3 (P = .21). In-hospital mortality was 18% in group 1, 27% in group 2, and 56% in group 3 (P = .001). Survival at 5 years is 57.0% in group 1, 57.7% in group 2, and 38.7% in group 3 (P = .0005). On multivariable analysis, age, cardiopulmonary bypass time, and group 3 versus group 2 were independent risk factors for mortality, whereas axillary cannulation was a protective factor. Conclusions: Patients with aortic dissection and cerebral malperfusion without preoperative coma showed acceptable mortality, and those with coma had a high in-hospital mortality regardless of the type of brain protection. Overall axillary artery cannulation appeared to be a protective factor
