    Force Spectroscopy And Two Photon Excited Luminescence In An Optical Tweezers System

    Up to now optical spectroscopies have analyzed the scattered light or the heat generated by absorption as a function of the wavelength to get information about the samples. Among the light matter interaction phenomena one that has almost never been used for spectroscopy is the direct photon momenta transfer. Probably because the forces involved are very small, varying from hundreds of femto to tens of pico Newtons. However, the nowadays very popular Optical Tweezers can easily accomplish the task to measure the photon momenta transfer and may be the basis for the Optical Force Spectroscopy. We demonstrate its potential as such a tool by observing more than eight Mie resonance peaks of a single polystyrene microsphere, and showed the capability to selective couple the light to either the TE, TM or both microsphere modes depending of the beam size, the light polarization and the beam positioning. The Mie resonances can change the optical force values by 30-50%. Our results also clearly show how the beam polarization breaks the usually assumed azimuthal symmetry by Optical Tweezers theories. We also obtained the spectrum from the two photon excited luminescence using the Optical Tweezers to hold a single bead suspended and a femtosecond Ti:sapphire laser for the non-linear excitation. This spectrum shows the pair of peaks due to both TE and TM spherical cavity modes. We have been able to observe more than 14 Mie resonance peaks in the TPE luminescence.     Impact of dietary Chlorella vulgaris and carbohydrate-active enzymes incorporation on plasma metabolites and liver lipid composition of broilers

    Research Areas: Veterinary SciencesBackground: Chlorella vulgaris has been proposed as a sustainable green feedstock in poultry nutrition due to its ease of cultivation, minimal environmental impact and balanced nutritional composition. However, the majority of studies documents the use of C. vulgaris as a dietary supplement in broilers instead of a feed ingredient. To the best of our knowledge, no report has shown the effect of a high-level incorporation (>2 % in the diet) of C. vulgaris on plasma metabolites and hepatic lipid composition of broilers. One hundred and twenty Ross 308 male birds were housed in 40 wired-floor cages and randomly distributed by the following experimental diets at 22 days of age (n = 10) during 15 days: (1) a corn-soybean meal based diet (control); (2) based diet with 10% of C. vulgaris; (3) diet 2 supplemented with 0.005% Rovabio® Excel AP; and (4) diet 2 supplemented with 0.01% of a pre-selected four-CAZyme mixture. Results: The inclusion of C. vulgaris at 10% in the diet, regardless of the presence of exogenous CAZymes, changed plasma metabolites but did not compromise broilers growth. Plasma total lipids increased in broilers fed C. vulgaris combined with the two feed CAZymes (p < 0.001) compared with the control diet. Moreover, the supplementation with Rovabio® increased total cholesterol and LDL-cholesterol, while the addition of the four-CAZyme mixture increased triacylglycerols, VLDL-cholesterol and ALP activity. In opposition, HDL-cholesterol levels decreased in broilers fed microalga alone (p = 0.002). Regarding hepatic composition, the inclusion of C. vulgaris in broiler diets, individually or combined with exogenous CAZymes, had a minor effect on fatty acids but improved the n-6/n-3 ratio and total carotenoids. Conclusions: In summary, the inclusion of a high level (10%) of C. vulgaris in broiler´s diet, regardless of the presence of exogenous CAZymes, improved hepatic antioxidant composition and did not impair broiler’s performance. In addition, the feed supplementation with CAZymes increased broilers lipemia.     An individual alginate lyase is effective in the disruption of Laminaria digitata recalcitrant cell wall

    Research Areas: Science & Technology - Other TopicsIn the present study, 199 pre-selected Carbohydrate-Active enZymes (CAZymes) and sulfatases were assessed, either alone or in combination, to evaluate their capacity to disrupt Laminaria digitata cell wall, with the consequent release of interesting nutritional compounds. A previously characterized individual alginate lyase, belonging to the family 7 of polysaccharide lyases (PL7) and produced by Saccharophagus degradans, was shown to be the most efcient in the in vitro degradation of L. digitata cell wall. The alginate lyase treatment, compared to the control, released up to 7.11 g/L of reducing sugars (p< 0.001) and 8.59 mmol/100 g dried alga of monosaccharides (p< 0.001), and reduced cell wall fuorescence intensity by 39.1% after staining with Calcofuor White (p= 0.001). The hydrolysis of gel-forming polymer alginate by the alginate lyase treatment could prevent the trapping of fatty acids and release benefcial monounsaturated fatty acids, particularly 18:1c9 (p < 0.001), to the extracellular medium. However, no liberation of proteins (p > 0.170) or pigments (p > 0.070) was observed. Overall, these results show the ability of an individual alginate lyase, from PL7 family, to partially degrade L. digitata cell wall under physiological conditions.     Effect of Dietary Laminaria digitata with Carbohydrases on Broiler Production Performance and Meat Quality, Lipid Profile, and Mineral Composition

A total of 120 22-day old broilers were fed the following dietary treatments (n = 10) for 14 days: cereal-based diet (control); control diet with 15% L. digitata (LA); LA diet with 0.005% Rovabio® Excel AP (LAR); LA diet with 0.01% alginate lyase (LAE). Final body weight was lower and feed conversion ratio higher with LA diet than with the control. The ileal viscosity increased with LA and LAR diets relative to control but without differences between LAE and control. The pH of thigh meat was higher, and the redness value of breast was lower with LA diet than with control. Meat overall acceptability was positively scored for all treatments. The γ-tocopherol decreased, whereas total chlorophylls and carotenoids increased in meat with alga diets relative to control. The percentage of n-3 polyunsaturated fatty acids (PUFA) and accumulation of bromine and iodine in meat increased with alga diets compared with control.     Molecular determinants of ligand specificity in family 11 carbohydrate binding modules - An NMR, X-ray crystallography and computational chemistry approach

    12 pags, 6 figs, 1 tabThe direct conversion of plant cell wall polysaccharides into soluble sugars is one of the most important reactions on earth, and is performed by certain microorganisms such as Clostridium thermocellum (Ct). These organisms produce extracellular multi-subunit complexes (i.e. cellulosomes) comprising a consortium of enzymes, which contain noncatalytic carbohydrate-binding modules (CBM) that increase the activity of the catalytic module. In the present study, we describe a combined approach by X-ray crystallography, NMR and computational chemistry that aimed to gain further insight into the binding mode of different carbohydrates (cellobiose, cellotetraose and cellohexaose) to the binding pocket of the family 11 CBM. The crystal structure of C. thermocellum CBM11 has been resolved to 1.98 Å in the apo form. Since the structure with a bound substrate could not be obtained, computational studies with cellobiose, cellotetraose and cellohexaose were carried out to determine the molecular recognition of glucose polymers by CtCBM11. These studies revealed a specificity area at the CtCBM11 binding cleft, which is lined with several aspartate residues. In addition, a cluster of aromatic residues was found to be important for guiding and packing of the polysaccharide. The binding cleft of CtCBM11 interacts more strongly with the central glucose units of cellotetraose and cellohexaose, mainly through interactions with the sugar units at positions 2 and 6.     Mesenchymal stem cells from patients with chronic myeloid leukemia do not express BCR-ABL and have absence of chimerism after allogeneic bone marrow transplant

    Bone marrow is a heterogeneous cell population which includes hematopoietic and mesenchymal progenitor cells. Dysregulated hematopoiesis occurs in chronic myelogenous leukemia (CML), being caused at least in part by abnormalities in the hematopoietic progenitors. However, the role of mesenchymal stem cells (MSCs) in CML has not been well characterized. The objectives of the present study were to observe the biological characteristics of MSCs from CML patients and to determine if MSCs originate in part from donors in CML patients after bone marrow transplantation (BMT). We analyzed MSCs from 5 untreated patients and from 3 CML patients after sex-mismatched allogeneic BMT. Flow cytometry analysis revealed the typical MSC phenotype and in vitro assays showed ability to differentiate into adipocytes and osteoblasts. Moreover, although some RT-PCR data were contradictory, combined fluorescence in situ hybridization analysis showed that MSCs from CML patients do not express the bcr-abl gene. Regarding MSCs of donor origin, although it is possible to detect Y target sequence by nested PCR, the low frequency (0.14 and 0.34%) of XY cells in 2 MSC CML patients by fluorescence in situ hybridization analysis suggests the presence of contaminant hematopoietic cells and the absence of host-derived MSCs in CML patients. Therefore, we conclude that MSCs from CML patients express the typical MSC phenotype, can differentiate into osteogenic and adipogenic lineages and do not express the bcr-abl gene. MSCs cannot be found in recipients 12 to 20 months after BMT. The influence of MSCs on the dysregulation of hematopoiesis in CML patients deserves further investigation

    Determination of the uptake and translocation of nitrogen applied at different growth stages of a melon crop (Cucumis melo L.) using 15N isotope.

    In order to establish a rational nitrogen (N) fertilisation and reduce groundwater contamination, a clearer understanding of the N distribution through the growing season and its dynamics inside the plant is crucial. In two successive years, a melon crop (Cucumis melo L. cv. Sancho) was grown under field conditions to determine the uptake of N fertiliser, applied by means of fertigation at different stages of plant growth, and to follow the translocation of N in the plant using 15N-labelled N. In 2006, two experiments were carried out. In the first experiment, labelled 15N fertiliser was supplied at the female-bloom stage and in the second, at the end of fruit ripening. Labelled 15N fertiliser was made from 15NH415NO3 (10 at.% 15N) and 9.6 kg N ha−1 were applied in each experiment over 6 days (1.6 kg N ha−1 d−1). In 2007, the 15N treatment consisted of applying 20.4 kg N ha−1 as 15NH415NO3 (10 at.% 15N) in the middle of fruit growth, over 6 days (3.4 kg N ha−1 d−1). In addition, 93 and 95 kg N ha−1 were supplied daily by fertigation as ammonium nitrate in 2006 and 2007, respectively. The results obtained in 2006 suggest that the uptake of N derived from labelled fertiliser by the above-ground parts of the plants was not affected by the time of fertiliser application. At the female-flowering and fruit-ripening stages, the N content derived from 15N-labelled fertiliser was close to 0.435 g m−2 (about 45% of the N applied), while in the middle of fruit growth it was 1.45 g m−2 (71% of the N applied). The N application time affected the amount of N derived from labelled fertiliser that was translocated to the fruits. When the N was supplied later, the N translocation was lower, ranging between 54% at female flowering and 32% at the end of fruit ripening. Approximately 85% of the N translocated came from the leaf when the N was applied at female flowering or in the middle of fruit growth. This value decreased to 72% when the 15N application was at the end of fruit ripening. The ammonium nitrate became available to the plant between 2 and 2.5 weeks after its application. Although the leaf N uptake varied during the crop cycle, the N absorption rate in the whole plant was linear, suggesting that the melon crop could be fertilised with constant daily N amounts until 2–3 weeks before the last harvest

    La rabia es una zoonosis ampliamente generalizada, y poco notificada, así, por falta de tratamiento a tiempo resulta fatal en 100% de los casos, ya sea en el hombre como en animal. Se considera al perro el principal reservorio de transmisión en el ciclo urbano, se hace necesario investigar los reservorios naturales del virus, para garantizar el control de la enfermedad. En este trabajo, se reporta por primera vez en la provinciade Huambo un caso de rabia en una Civeta Africana (Civettictis civetta). El caso fue detectado en el barrio Raimundo de la administración de barrio Comandante Xavier Samacau del municipio Huambo sede. El animal presentó un comportamiento agresivo mordiendo a personas y objetos inanimados que encontraba en su camino, resultando lesionadas una mujer y una niña de 3 años. El mismo fue capturado, sacrificado y enviado al Instituto de Servicio Veterinario, donde el encéfalo fue sometido a investigación por Inmunofluorescencia directa con resultados positivos a Lyssavirus. El comportamiento clínico de este animal, caracterizado por elevada agresividad con ataque a objetos inanimados y al hombre así como el diagnóstico positivo por inmunofluorescencia permitieron establecer el diagnóstico de rabi
