    NFV Orchestration over Disaggregated Metro Optical Networks with End-to-End Multi-Layer Slicing enabling Crowdsourced Live Video Streaming

    Network infrastructure must support emerging applications, fulfill 5G requirements, and respond to the sudden increase of societal need for remote communications. Remarkably, crowdsourced live video streaming (CLVS) challenges operators' infrastructure with tides of users attending major sport or public events that demand high bandwidth and low latency jointly with computing capabilities at the networks' edge. The Metro-Haul project entered the scene proposing a cost-effective, agile, and disaggregated infrastructure for the metro segment encompassing optical and packet resources jointly with computing capabilities. Recently, a major Metro-Haul outcome took the form of a field trial of network function virtualization (NFV) orchestration over the multi-layer packet and disaggregated optical network testbed that demonstrated a CLVS use case. We showcased the average service creation time below 5 min, which met the key performance indicator as defined by the 5G infrastructure public private partnership. In this paper, we expand our field trial demonstration with a detailed view of the Metro-Haul testbed for the CLVS use case, the employed components, and their performance. The throughput of the service is increased from approximately 9.6 Gbps up to 35 Gbps per virtual local area network with high-performance VNFs based on single-root input/output virtualization technology

    Coarse graining ππ scattering

    We carry out an analysis of ππ scattering in the I J = 00, 11 and 20 channels in configuration space up to a maximal center-of-mass energy √ s = 1.4 GeV. We separate the interaction into two regions marked by an elementarity radius of the system; namely, a long distance region above which pions can be assumed to interact as elementary particles and a short distance region where many physical effects cannot be disentangled. The long distance interaction is described by chiral dynamics, where a two-pionexchange potential is identified, computed and compared to lattice calculations. The short distance piece corresponds to a coarse grained description exemplified by a superposition of delta-shell potentials sampling the interaction with the minimal wavelength. We show how the so constructed nonperturbative scattering amplitude complies with the proper analytic structure, allowing for an explicit N/D type decomposition in terms of the corresponding Jost functions and fulfilling dispersion relations without subtractions. We also address renormalization issues in coordinate space and investigate the role of crossing when fitting the scattering amplitudes above and below threshold to Roy-equation results. At higher energies, we show how inelasticities can be described by one single complex and energy dependent parameter.     Current and Future Drug Targets in Weight Management

    Obesity will continue to be one of the leading causes of chronic disease unless the ongoing rise in the prevalence of this condition is reversed. Accumulating morbidity figures and a shortage of effective drugs have generated substantial research activity with several molecular targets being investigated. However, pharmacological modulation of body weight is extremely complex, since it is essentially a battle against one of the strongest human instincts and highly efficient mechanisms of energy uptake and storage. This review provides an overview of the different molecular strategies intended to lower body weight or adipose tissue mass. Weight-loss drugs in development include molecules intended to reduce the absorption of lipids from the GI tract, various ways to limit food intake, and compounds that increase energy expenditure or reduce adipose tissue size. A number of new preparations, including combinations of the existing drugs topiramate plus phentermine, bupropion plus naltrexone, and the selective 5-HT2C agonist lorcaserin have recently been filed for approval. Behind these leading candidates are several other potentially promising compounds and combinations currently undergoing phase II and III testing. Some interesting targets further on the horizon are also discussed

    Canagliflozin and Renal Outcomes in Type 2 Diabetes and Nephropathy

    BACKGROUND Type 2 diabetes mellitus is the leading cause of kidney failure worldwide, but few effective long-term treatments are available. In cardiovascular trials of inhibitors of sodium–glucose cotransporter 2 (SGLT2), exploratory results have suggested that such drugs may improve renal outcomes in patients with type 2 diabetes. METHODS In this double-blind, randomized trial, we assigned patients with type 2 diabetes and albuminuric chronic kidney disease to receive canagliflozin, an oral SGLT2 inhibitor, at a dose of 100 mg daily or placebo. All the patients had an estimated glomerular filtration rate (GFR) of 30 to 300 to 5000) and were treated with renin–angiotensin system blockade. The primary outcome was a composite of end-stage kidney disease (dialysis, transplantation, or a sustained estimated GFR of <15 ml per minute per 1.73 m 2), a doubling of the serum creatinine level, or death from renal or cardiovascular causes. Prespecified secondary outcomes were tested hierarchically. RESULTS The trial was stopped early after a planned interim analysis on the recommendation of the data and safety monitoring committee. At that time, 4401 patients had undergone randomization, with a median follow-up of 2.62 years. The relative risk of the primary outcome was 30% lower in the canagliflozin group than in the placebo group, with event rates of 43.2 and 61.2 per 1000 patient-years, respectively (hazard ratio, 0.70; 95% confidence interval [CI], 0.59 to 0.82; P=0.00001). The relative risk of the renal-specific composite of end-stage kidney disease, a doubling of the creatinine level, or death from renal causes was lower by 34% (hazard ratio, 0.66; 95% CI, 0.53 to 0.81; P<0.001), and the relative risk of end-stage kidney disease was lower by 32% (hazard ratio, 0.68; 95% CI, 0.54 to 0.86; P=0.002). The canagliflozin group also had a lower risk of cardiovascular death, myocardial infarction, or stroke (hazard ratio, 0.80; 95% CI, 0.67 to 0.95; P=0.01) and hospitalization for heart failure (hazard ratio, 0.61; 95% CI, 0.47 to 0.80; P<0.001). There were no significant differences in rates of amputation or fracture. CONCLUSIONS In patients with type 2 diabetes and kidney disease, the risk of kidney failure and cardiovascular events was lower in the canagliflozin group than in the placebo group at a median follow-up of 2.62 years

    2700 years of Mediterranean environmental change in central Italy : a synthesis of sedimentary and cultural records to interpret past impacts of climate on society

    Abrupt climate change in the past is thought to have disrupted societies by accelerating environmental degradation, potentially leading to cultural collapse. Linking climate change directly to societal disruption is challenging because socioeconomic factors also play a large role, with climate being secondary or sometimes inconsequential. Combining paleolimnologic, historical, and archaeological methods provides for a more secure basis for interpreting the past impacts of climate on society. We present pollen, non-pollen palynomorph, geochemical, paleomagnetic and sedimentary data from a high-resolution 2700 yr lake sediment core from central Italy and compare these data with local historical documents and archeological surveys to reconstruct a record of environmental change in relation to socioeconomic history and climatic fluctuations. Here we document cases in which environmental change is strongly linked to changes in local land management practices in the absence of clear climatic change, as well as examples when climate change appears to have been a strong catalyst that resulted in significant environmental change that impacted local communities. During the Imperial Roman period, despite a long period of stable, mild climate, and a large urban population in nearby Rome, our site shows only limited evidence for environmental degradation. Warm and mild climate during the Medieval Warm period, on the other hand, led to widespread deforestation and erosion. The ability of the Romans to utilize imported resources through an extensive trade network may have allowed for preservation of the environment near the Roman capital, whereas during medieval time, the need to rely on local resources led to environmental degradation. Cool wet climate during the Little Ice Age led to a breakdown in local land use practices, widespread land abandonment and rapid reforestation. Our results present a high-resolution regional case study that explores the effect of climate change on society for an under-documented region of Europe. (C) 2015 Elsevier Ltd. All rights reserved

    Oleoylethanolamide enhances beta-adrenergic-mediated thermogenesis and white-to-brown adipocyte phenotype in epididymal white adipose tissue in rat

    β-adrenergic receptor activation promotes brown adipose tissue (BAT) β-oxidation and thermogenesis by burning fatty acids during uncoupling respiration. Oleoylethanolamide (OEA) can inhibit feeding and stimulate lipolysis by activating peroxisome proliferator-activating receptor-α (PPARα) in white adipose tissue (WAT). Here we explore whether PPARα activation potentiates the effect of β3-adrenergic stimulation on energy balance mediated by the respective agonists OEA and CL316243. The effect of this pharmacological association on feeding, thermogenesis, β-oxidation, and lipid and cholesterol metabolism in epididymal (e)WAT was monitored. CL316243 (1 mg/kg) and OEA (5 mg/kg) co-administration over 6 days enhanced the reduction of both food intake and body weight gain, increased the energy expenditure and reduced the respiratory quotient (VCO2/VO2). This negative energy balance agreed with decreased fat mass and increased BAT weight and temperature, as well as with lowered plasma levels of triglycerides, cholesterol, nonessential fatty acids (NEFAs), and the adipokines leptin and TNF-α. Regarding eWAT, CL316243 and OEA treatment elevated levels of the thermogenic factors PPARα and UCP1, reduced p38-MAPK phosphorylation, and promoted brown-like features in the white adipocytes: the mitochondrial (Cox4i1, Cox4i2) and BAT (Fgf21, Prdm16) genes were overexpressed in eWAT. The enhancement of the fatty-acid β-oxidation factors Cpt1b and Acox1 in eWAT was accompanied by an upregulation of de novo lipogenesis and reduced expression of the unsaturated-fatty-acid-synthesis enzyme gene, Scd1. We propose that the combination of β-adrenergic and PPARα receptor agonists promotes therapeutic adipocyte remodelling in eWAT, and therefore has a potential clinical utility in the treatment of obesity
