Op weg naar nieuwe koppelingen : de rol van natuur- en milieuorganisaties in gebiedsgericht beleid
Op weg naar nieuwe koppelingen belicht de positie en rol van natuur- en milieuorganisaties in gebiedgericht beleid aan de hand van het revitaliseringsproces in de Wijde Biesbosch. Aanleiding is de onvrede die er onder dergelijke groepen en de koepelorganisatie Stichting Natuur en Milieu bestaat over hun invloed in gebiedsgerichte processen. Op basis van een uitgebreide procesanalyse komen de onderzoekers tot aanbevelingen voor invloedsvergroting. Suggesties op gebiedsniveau liggen op het vlak van gezamenlijke visie-ontwikkeling en prioriteitsstelling, coalitievorming, mobilisering van de lokale achterban en beïnvloeding van de provinciale regie en inrichting van het proces
Erasmus Computing Grid: Het bouwen van een 20 Tera-FLOPS Virtuele Supercomputer.
Het Erasmus Medisch Centrum en de Hogeschool
Rotterdam zijn in 2005 een samenwerking begonnen
teneinde de ongeveer 95% onbenutte rekencapaciteit
van hun computers beschikbaar te maken voor onderzoek
en onderwijs. Deze samenwerking heeft geleid tot
het Erasmus Computing GRID (ECG), een virtuele
supercomputer met na voltooiing een rekencapaciteit
van 20 Teraflops. Dit artikel schetst enige achtergronden
van grid computing, beschrijft een aantal toepassingen
die mogelijk zijn met een grid infrastructuur en geeft
de wijze weer waarop het ECG wordt vormgegeven. In
het verlengde hiervan bevat het een pleidooi om grid
computing binnen het onderwijs een betere basis te
geven om op die manier vanuit het onderwijs een
substantiële bijdrage te leveren aan het versterken van
(rekenintensief) onderzoek
Profiling of a high mannose-type N-glycosylated lipase using hydrophilic interaction chromatography-mass spectrometry
Many industrial enzymes exhibit macro- and micro-heterogeneity due to co-occurring post-translational modifications. The resulting proteoforms may have different activity and stability and, therefore, the characterization of their distributions is of interest in the development and monitoring of enzyme products. Protein glycosylation may play a critical role as it can influence the expression, physical and biochemical properties of an enzyme. We report the use of hydrophilic interaction liquid chromatography-mass spectrometry (HILIC-MS) to profile intact glycoform distributions of high mannose-type N-glycosylated proteins, using an industrially produced fungal lipase for the food industry as an example. We compared these results with conventional reversed phase LC-MS (RPLC-MS) and sodium dodecyl sulfate-polyacrylamide gel-electrophoresis (SDS-PAGE). HILIC appeared superior in resolving lipase heterogeneity, facilitating mass assignment of N-glycoforms and sequence variants. In order to understand the glycoform selectivity provided by HILIC, fractions from the four main HILIC elution bands for lipase were taken and subjected to SDS-PAGE and bottom-up proteomic analysis. These analyses enabled the identification of the most abundant glycosylation sites present in each fraction and corroborated the capacity of HILIC to separate protein glycoforms based on the number of glycosylation sites occupied. Compared to RPLC-MS, HILIC-MS reducted the sample complexity delivered to the mass spectrometer, facilitating the assignment of the masses of glycoforms and sequence variants as well as increasing the number of glycoforms detected (69 more proteoforms, 177% increase). The HILIC-MS method required relatively short analysis time (<30 min), in which over 100 glycoforms were distinguished. We suggest that HILIC(-MS) can be a valuable tool in characterizing bioengineering processes aimed at steering protein glycoform expression as well as to check the consistency of product batche
Human plasma phospholipid transfer protein increases the antiatherogenic potential of high density lipoproteins in transgenic mice
Plasma phospholipid transfer protein (PLTP) transfers phospholipids
between lipoprotein particles and alters high density lipoprotein (HDL)
subfraction patterns in vitro, but its physiological function is poorly
understood. Transgenic mice that overexpress human PLTP were generated.
Compared with wild-type mice, these mice show a 2.5- to 4.5-fold increase
in PLTP activity in plasma. This results in a 30% to 40% decrease of
plasma levels of HDL cholesterol. Incubation of plasma from transgenic
animals at 37 degrees C reveals a 2- to 3-fold increase in the formation
of pre-beta-HDL compared with plasma from wild-type mice. Although
pre-beta-HDL is normally a minor subfraction of HDL, it is known to be a
very efficient acceptor of peripheral cell cholesterol and a key mediator
in reverse cholesterol transport. Further experiments show that plasma
from transgenic animals is much more efficient in preventing the
accumulation of intracellular cholesterol in macrophages than plasma from
wild-type mice, despite lower total HDL concentrations. It is concluded
that PLTP can act as an antiatherogenic factor preventing cellular
cholesterol overload by generation of pre-beta-HDL
Intravascular ultrasound predictors of outcome after peripheral balloon angioplasty
Objective:This study investigates the potential role of intravascular ultrasound (IVUS) in the outcome in patients undergoing percutaneous transluminal angioplasty (PTA) of the superficial femoral artery.Materials:Angiographic and the qualitative and quantitative IVUS data obtained at the narrowest site derived from 39 patients before and after PTA were analysed.Results:Angiographically the diameter of the remaining stenosis seen after PTA was classified as < 50% in 31 patients (success); in eight patients a failure was encountered. Evaluating at 6 months the functional and anatomic results of the PTA in 31 patients, the intervention was a success in 14 patients (Group I) and a failure in 17 patients (Group II). The remaining eight patients defined as angiographic failure following PTA comprised Group III. Neither qualitative nor quantitative IVUS data obtained before PTA could predict outcome. Conversely, after PTA, the extent of dissection was significantly more severe in Groups II and III than in Group I. Similarly, significant differences were found between Groups I and II for mean free lumen area (13.2 vs. 9.7 mm2, respectively) and mean free lumen diameter (4.1 vs. 3.5 mm, respectively). Quantitative data obtained in Group II were similar to those in Group III.Conclusion:This preliminary study demonstrates that following PTA the extent of dissection, free lumen area and diameter seen with IVUS are predictive factors of patency. Future studies with more patients are mandatory to further highlight the sensitivity of these observations
Evidence for a mixed mass composition at the `ankle' in the cosmic-ray spectrum
We report a first measurement for ultra-high energy cosmic rays of the
correlation between the depth of shower maximum and the signal in the water
Cherenkov stations of air-showers registered simultaneously by the fluorescence
and the surface detectors of the Pierre Auger Observatory. Such a correlation
measurement is a unique feature of a hybrid air-shower observatory with
sensitivity to both the electromagnetic and muonic components. It allows an
accurate determination of the spread of primary masses in the cosmic-ray flux.
Up till now, constraints on the spread of primary masses have been dominated by
systematic uncertainties. The present correlation measurement is not affected
by systematics in the measurement of the depth of shower maximum or the signal
in the water Cherenkov stations. The analysis relies on general characteristics
of air showers and is thus robust also with respect to uncertainties in
hadronic event generators. The observed correlation in the energy range around
the `ankle' at differs significantly from
expectations for pure primary cosmic-ray compositions. A light composition made
up of proton and helium only is equally inconsistent with observations. The
data are explained well by a mixed composition including nuclei with mass . Scenarios such as the proton dip model, with almost pure compositions, are
thus disfavoured as the sole explanation of the ultrahigh-energy cosmic-ray
