ONLINE HIERARCHICAL MODELS FOR SURFACE RECONSTRUCTION
Applications based on three-dimensional object models are today very common, and can be found in many fields as design, archeology, medicine, and entertainment. A digital 3D model can be obtained by means of physical object measurements performed by using a 3D scanner. In this approach, an important step of the 3D model building process consists of creating the object's surface representation from a cloud of noisy points sampled on the object itself. This process can be viewed as the estimation of a function from a finite subset of its points. Both in statistics and machine learning this is known as a regression problem.
Machine learning views the function estimation as a learning problem to be addressed by using computational intelligence techniques: the points represent a set of examples and the surface to be reconstructed represents the law that has generated them. On the other hand, in many applications the cloud of sampled points may become available only progressively during system operation. The conventional approaches to regression are therefore not suited to deal efficiently with this operating condition.
The aim of the thesis is to introduce innovative approaches to the regression problem suited for achieving high reconstruction accuracy, while limiting the computational complexity, and appropriate for online operation. Two classical computational intelligence paradigms have been considered as basic tools to address the regression problem: namely the Radial Basis Functions and the Support Vector Machines. The original and innovative aspect introduced by this thesis is the extension of these tools toward a multi-scale incremental structure, based on hierarchical schemes and suited for online operation. This allows for obtaining modular, scalable, accurate and efficient modeling procedures with training algorithms appropriate for dealing with online learning. Radial Basis Function Networks have a fast configuration procedure that, operating locally, does not require iterative algorithms. On the other side, the computational complexity of the configuration procedure of Support Vector Machines is independent from the number of input variables. These two approaches have been considered in order to analyze advantages and limits of each of them due to the differences in their intrinsic nature
Glycosaminoglycan profile in macrophages exposed to Candida albicans and interleukins
Glycosaminoglycans (GAG), are extracellular matrix macromolecules that affect the phagocytic properties of macrophages. In order to assess whether the interaction between macrophages and Candida albicans (iCa) provokes changes in the phenotype, we analyzed the GAG profiles in two macrophage lines, ANA-1 (from murine bone-marrow) and BV-2 (from murine brain). We also investigated GAG modulation by interleukin-1alpha (IL-1alpha) and interleukin-6 (IL-6). During iCa treatment and even after the addition of ILs, ANA-1 accumulated less total GAG compared to controls. IL-1 treatment, combined with iCa exposure, induced a decrease in heparan sulfate and chondroitin sulfate chains, and an increase in the hyaluronic acid percentage. IL-6 treatment, with or without iCa, decreased the hyaluronic acid/sulfated GAG ratio. The GAG pattern in BV-2 appears to be different to ANA-1 and iCa exposure does not induce any difference in total GAG. The inhibitory effect induced by ILs on GAG synthesis is less than that observed in ANA-1 and the GAG elution profile is modulated to a lesser extent by treatment with ILs and/or iCa compared to the ANA-1. We suggest that the observed changes in the expression of the individual GAG classes may be responsible for the macrophage functional heterogeneity
Polymorphisms in toll-like receptor genes and susceptibility to pulmonary aspergillosis
Toll-like receptors (TLRs) are important components of innate immunity. We investigated the association between polymorphisms in the TLR2, TLR4, and TLR9 genes and susceptibility to noninvasive forms of pulmonary aspergillosis. A significant association was observed between allele G on Asp299Gly (TLR4) and chronic cavitary pulmonary aspergillosis (odds ratio [OR], 3.46; P =.003). Susceptibility to allergic bronchopulmonary aspergillosis was associated with allele C on T-1237C (TLR9) (OR, 2.49; P =. 043). No particular polymorphism was associated with severe asthma with fungal sensitization. Geraniol Treatment for Irritable Bowel Syndrome: A Double-Blind Randomized Clinical Trial
Geraniol is an acyclic monoterpene alcohol with well-known anti-inflammatory and antimicrobial properties which has shown eubiotic activity towards gut microbiota (GM) in patients with irritable bowel syndrome (IBS). Methods: Fifty-six IBS patients diagnosed according to Rome III criteria were enrolled in an interventional, prospective, multicentric, randomized, double-blinded, placebo-controlled trial. In the treatment arm, patients received a low-absorbable geraniol food supplement (LAGS) once daily for four weeks. Results: Patients treated with LAGS showed a significant reduction in their IBS symptoms severity score (IBS-SSS) compared to the placebo (195 vs. 265, p = 0.001). The rate of responders according to IBS-SSS (reduction ≥ 50 points) was significantly higher in the geraniol vs placebo group (52.0% vs. 16.7%, p = 0.009) mainly due to the IBS mixed subtype. There were notable differences in the microbiota composition after geraniol administration, particularly a significant decrease in a genus of Ruminococcaceae, Oscillospira (p = 0.01), a decreasing trend for the Erysipelotrichaceae and Clostridiaceae families (p = 0.1), and an increasing trend for other Ruminococcaceae taxa, specifically Faecalibacterium (p = 0.09). The main circulating proinflammatory cytokines showed no differences between placebo and geraniol arms. Conclusion: LAGS was effective in treating overall IBS symptoms, together with an improvement in the gut microbiota profile, especially for the IBS mixed subtype
High flow nasal oxygen vs. conventional oxygen therapy over respiratory oxygenation index after esophagectomy: an observational study
Background: Postoperative pulmonary complications after esophagectomy still represent a matter of concern. High flow nasal cannula (HFNC) early after major abdominal and thoracic surgery has demonstrated some advantages over conventional oxygen therapy. Data about respiratory effect of HFNC after esophagectomy is scarce. The primary aim of this study is to investigate if the early use of HFNC after esophagectomy could enhance patients' postoperative respiratory oxygenation (ROX) index and, ultimately, reduce postoperative pneumonia. Methods: In this single center retrospective study all patients undergoing to esophagectomy for cancer from May 2020 to November 2022 were evaluated. Historical cohort (HC) received postoperative oxygen supplementation with Venturi mask or nasal goggles, and a cohort was put under HFNC (HFNC cohort). ROX index, blood gas analysis, radiological atelectasis score (RAS), post-operative complications' data and information on hospital stay have been collected and analyzed. Results: Seventy-one patients were included for the final statistical analysis, 31 in the HFNC and 40 in the HC cohort. Mean age was 64±10 years and body mass index (BMI) was 26 [24-29] kg/m2. ROX index was higher in the HFNC patients than in the HC, 20.8 [16.7-25.9] vs. 14.9 [10.8-18.2] (P<0.0001). In the HFNC cohort patients, pH was higher, 7.42 [7.40-7.44] vs. 7.39 [7.37-7.43] than HC, while PaCO2 was lower in HFNC cohort compared with HC, 39 [36-41] vs. 42 [39-45] mmHg, respectively (P=0.01). RAS was similar between the two cohorts of patients, 1.5±0.98 vs. 1.4±1.04 in the HFNC and the HC cohort, respectively (P=0.611). Lower acute respiratory failure (ARF) rate was recorded among HFNC than HC cohort, 0% vs. 13% respectively, P=0.06. No difference in pneumonia frequency between two cohorts was shown. Conclusions: HFNC improved the ROX index after esophagectomy through significant respiratory rate reduction. This tool should be considered for early respiratory support after extubation in this category of patients, not only as a rescue therapy for ARF, but also to optimize early postoperative respiratory function. Whether this will improve patients' outcomes requires further large randomized controlled trials
Visual Localization in the Presence of Appearance Changes Using the Partial Order Kernel
Visual localization across seasons and under varying weather and lighting conditions is a challenging task in robotics. In this paper, we present a new sequence-based approach to visual localization using the Partial Order Kernel (POKer), a convolution kernel for string comparison, that is able to handle appearance changes and is robust to speed variations. We use multiple sequence alignment to construct directed acyclic graph representations of the database image sequences, where sequences of images of the same place acquired at different times are represented as alternative paths in a graph. We then use the POKer to compute the pairwise similarities between these graphs and the query image sequences obtained in a subsequent traversal of the environment, and match the corresponding locations. We evaluated our approach on a dataset which features extreme appearance variations due to seasonal changes. The results demonstrate the effectiveness of our approach, where it achieves
higher precision and recall than two state-of-the-art baseline method
The endocannabinoid system controls food intake via olfactory processes
Comment in Sensory systems: the hungry sense. [Nat Rev Neurosci. 2014] Inhaling: endocannabinoids and food intake. [Nat Neurosci. 2014]; International audience; Hunger arouses sensory perception, eventually leading to an increase in food intake, but the underlying mechanisms remain poorly understood. We found that cannabinoid type-1 (CB1) receptors promote food intake in fasted mice by increasing odor detection. CB1 receptors were abundantly expressed on axon terminals of centrifugal cortical glutamatergic neurons that project to inhibitory granule cells of the main olfactory bulb (MOB). Local pharmacological and genetic manipulations revealed that endocannabinoids and exogenous cannabinoids increased odor detection and food intake in fasted mice by decreasing excitatory drive from olfactory cortex areas to the MOB. Consistently, cannabinoid agonists dampened in vivo optogenetically stimulated excitatory transmission in the same circuit. Our data indicate that cortical feedback projections to the MOB crucially regulate food intake via CB1 receptor signaling, linking the feeling of hunger to stronger odor processing. Thus, CB1 receptor-dependent control of cortical feedback projections in olfactory circuits couples internal states to perception and behavior
Coexpression of vesicular glutamate transporters 1 and 2, glutamic acid decarboxylase and calretinin in rat entorhinal cortex
We studied the distribution and coexpression of vesicular glutamate transporters (VGluT1, VGluT2), glutamic acid decarboxylase
(GAD) and calretinin (CR, calcium-binding protein) in rat entorhinal cortex, using immunofluorescence staining and multichannel
confocal laser scanning microscopy. Images were computer processed and subjected to automated 3D object recognition, colocalization
analysis and 3D reconstruction. Since the VGluTs (in contrast to CR and GAD) occurred in fibers and axon terminals only, we
focused our attention on these neuronal processes. An intense, punctate VGluT1-staining occurred everywhere in the entorhinal
cortex. Our computer program resolved these punctae as small 3D objects. Also VGluT2 showed a punctate immunostaining pattern,
yet with half the number of 3D objects per tissue volume compared with VGluT1, and with statistically significantly larger
3D objects. Both VGluTs were distributed homogeneously across cortical layers, with in MEA VGluT1 slightly more densely distributed
than in LEA. The distribution pattern and the size distribution of GAD 3D objects resembled that of VGluT2. CR-immunopositive
fibers were abundant in all cortical layers. In double-stained sections we noted ample colocalization of CR and VGluT2, whereas
coexpression of CR and VGluT1 was nearly absent. Also in triple-staining experiments (VGluT2, GAD and CR combined) we noted
coexpression of VGluT2 and CR and, in addition, frequent coexpression of GAD and CR. Modest colocalization occurred of VGluT2
and GAD, and incidental colocalization of all three markers. We conclude that the CR-containing axon terminals in the entorhinal
cortex belong to at least two subpopulations of CR-neurons: a glutamatergic excitatory and a GABAergic inhibitory
FimH Adhesin of Type 1 Fimbriae Is a Potent Inducer of Innate Antimicrobial Responses Which Requires TLR4 and Type 1 Interferon Signalling
Components of bacteria have been shown to induce innate antiviral immunity via Toll-like receptors (TLRs). We have recently shown that FimH, the adhesin portion of type 1 fimbria, can induce the innate immune system via TLR4. Here we report that FimH induces potent in vitro and in vivo innate antimicrobial responses. FimH induced an innate antiviral state in murine macrophage and primary MEFs which was correlated with IFN-β production. Moreover, FimH induced the innate antiviral responses in cells from wild type, but not from MyD88−/−, Trif−/−, IFN−α/βR−/− or IRF3−/− mice. Vaginal delivery of FimH, but not LPS, completely protected wild type, but not MyD88−/−, IFN-α/βR−/−, IRF3−/− or TLR4−/− mice from subsequent genital HSV-2 challenge. The FimH-induced innate antiviral immunity correlated with the production of IFN-β, but not IFN-α or IFN-γ. To examine whether FimH plays a role in innate immune induction in the context of a natural infection, the innate immune responses to wild type uropathogenic E. coli (UPEC) and a FimH null mutant were examined in the urinary tract of C57Bl/6 (B6) mice and TLR4-deficient mice. While UPEC expressing FimH induced a robust polymorphonuclear response in B6, but not TLR4−/− mice, mutant bacteria lacking FimH did not. In addition, the presence of TLR4 was essential for innate control of and protection against UPEC. Our results demonstrate that FimH is a potent inducer of innate antimicrobial responses and signals differently, from that of LPS, via TLR4 at mucosal surfaces. Our studies suggest that FimH can potentially be used as an innate microbicide against mucosal pathogens
Research Priorities for Neglected Infectious Diseases in Latin America and the Caribbean Region

infectious diseases (NIDs) can be assessed
in different ways, but it is important to
realize that regional priorities may significantly
differ one from another. The region
of Latin America and the Caribbean (LAC)
is—along with Africa and Asia—more
affected by NIDs than other regions of the
world. Some of the Latin American NIDs
are common to other continents, while
others are very specific or disproportionately
affect the Latin American region [1–
3] (Table 1). Because of its huge ecological
diversity, ongoing environmental changes,
and massive migrations, LAC is also a
catalyst for the (re-)emergence and spreading
of NIDs, both inside and outside the
subcontinent. Following a colloquium on
NIDs in LAC held in Lima, Peru, between
12 and 14 November 2009, a thematic
workshop was organized with the support
of the European Commission (EC). It
involved 29 scientists (16 from the Americas,
two from the Democratic Republic of
Congo and India, respectively, and nine
from Europe) working on different NIDs
and representing several research areas
from basic to applied. This report summarizes
the consensus comments of the expert
group after oral and written consultation. It
is envisaged that this document should
stimulate a debate within the scientific
community and serve as a recommendation
for future actions by international or
regional funding agencies in the area of
