Jane Austen's Pride and Prejudicewas the novel of Victorian period. This Victorian era was the timeof traditions when many aspectshas emerged in the field of literature. As a writer of Victorian period Jane Austenwas very much successfulwith the work of romantic fictions.Pride and Prejudicehas been considered herfirst novel and the story of it was located in the country of Hertfordshire. The prominent family of this novel was known as the Bennet family. They used to live in a peaceful place called Longbourn. The main attraction of the Bennet family was centered around their daughters' marriage issue. In this novel the two main male characters were Mr. Bingley and his friend Darcy, and other characters did playsomevital role regarding the concepts of love and marriage. Surrounded by the wealthy handsome men, Mrs. Bennet was finding the possible match among those opulent persons for her daughters. At first glance Mr. Bingley has found his love towards Jane, while on the other hands Mr. Darcy proved to be the perfect match for Elizabeth. With these two wealthy and decent man, there was the presence of a villain like person named Mr. Wickham, who has settled himself finally with Lydia, the youngest girl of the Bennet family. The main female protagonist of this novel was Elizabeth. She was not a complicated character rather than funny and witty in nature. Her perfect match Mr. Darcy was different than her. He was snobbish, proud and reservedtype of character. They both were always sunk in teasing and quarrelling but somehow they were essentially attracted with each other. Elizabeth has confessed that she hate him and also said it frankly after knowing that Mr. Darcy will forgive her. With all those small and big incidents mixed with sadness and happiness, they both eventually got married and lived happily ever after. This paper will actually present the importance of love and marriage in human's life and how Jane Austen beautifully portrayed these themes of love and marriage which are not only the embellishment of life but also urged for the economical needs for the people through her famous novel Pride and Prejudice

Citrate Binding to the Membrane Protein Proteorhodopsin

Proteorhodopsin (pR) is an intrinsic membrane protein with an important role in solar-energy storage of the biosphere. Earlier work in our lab has shown that polyhistidine-tagged pR can be purified by means of selective precipitation with citrate under specific conditions, as can a number of mutants based on this His6-tagged pR. Purification of a heterologously-expressed trans-membrane protein by a simple salt such as citrate is novel. However, such a phenomenon leads to several questions: How does citrate cause pR precipitation? Does the polyhistidine-tag assist in such a precipitation? Is this precipitation pH-specific? Does citrate affect the function of pR? Does citrate-induced pR precipitation have any biological significance? Are there other ions that could cause pR precipitation? This dissertation focuses on understanding the nature of the interaction of pR with citrate and other anions, and in particular on trying to take advantage of this interaction in order to develop a novel membrane protein purification method. The end goal that branches out of these two aims is to utilize the compact citrate interaction site identified in pR, by incorporating it into other membrane proteins and using it to permit their purification by similar simple procedures. In Chapter 1, I briefly provide some background information on the wide variety of structurally-similar proteins as rhodopsins that include pR. I also describe the general importance of developing purification methods for 7-helix membrane proteins, including pR. Chapter 2 focuses on the investigation of the nature of citrate-binding site of pR. To address the main question of how citrate aids in pR purification, site-directed mutagenesis technique was applied to generate several single, double, triple or quadruple mutants of pR in a histidine-tag free background, which were then tested for their reactivity to citrate. Several different anions were tested to examine if precipitation of pR was specific to citrate or whether the precipitation is susceptible to other negatively charged salts. Photocycle of pR progresses through several intermediates, each with a distinct absorption maximum (described in subsection 1.3.1). M-intermediate is detected at pH ≥ 8 with λmax = 410 nm. Flash spectroscopy involves excitation of pR at a particular wavelength that leads to transient absorption, thus, signaling the formation of the corresponding intermediate. Flash-induced transient visible absorption measurements were used to assay the effect of exposing pR to citrate on its physiological function. Chapter 3 describes the development of a method of purification of pR using simple salts, citrate and phosphate. Chapter 4 begins an exploration of a future direction. The ultimate objective is to apply the above techniques for the general purpose of 7-helix membrane protein purification, especially for the important class of pharmacological receptors known as GPCRs. An attempt at heterologous expression in E. coli, and purification, of a mammalian GPCR, is described therein. Such a method would be desirable for obtaining proteins for structural, functional and pharmacological studies

Estimation study about the relationship between exchange rate flexibility and current account adjustment has been through three stages, the first stage was analysis of correlation among exchange rates variability (proxied by REER and NEER) and exchange rate regimes classification. The second step was estimating the relationship that the former was mentioned with VAR as benchmark model. The third step was applying the nonlinear estimation with Threshold VAR. The results of analysis showed that exchange rate regime classification may not capture actual exchange rate variability and flexibility exchange rate can accelerate current account adjustment in Indonesia if the changes of Indonesia exchange rate less than 27.7059 (low regime) whereas in high regime exchange rate is persistent increasing so that the system between exchange rate and current account become unstable. Bank Indonesia as monetary authorities must keep the changes of exchange rate less than 27.7059, due to exchange rate can affect current account adjustment, so can anticipate if there is current account deficit in Indonesia economy

Modeling And Simulation Of Wind Turbine For Partial Load Operation

    Get PDF
The purpose of this research is mainly to model a wind turbine with doubly fed induction generator (DFIG) and to investigate its behavior in partial load operation using computer simulation. System behavior analysis and simulation are two particular approaches applied in this research. The wind turbine model is developed whereby the behavior of its main components is described by mathematical model and transformed in simulation model in MATLAB/Simulink. The simulation model is carried out in partial load operation at a wind speed of 9 m/s. The physical quantities including generator speed, torque and electrical power output are measured and evaluated. By using variable speed generator, a wind turbine model with optimum power generation at wind speed between 4 m/s to 13 m/s is created. This research shows that if mathematical models represent the wind turbine accurately, then the proposed model can be used to observe the dynamic behavior of wind turbine precisely, efficiently and inexpensively. The simulation result is expected to be a reference for extending the knowledge of dynamic behavior of wind turbines and optimize the performance of future large-scale wind turbine systems

Cost-effectiveness of household contact investigation for detection of tuberculosis in Pakistan

Objectives Despite WHO guidelines recommending household contact investigation, and studies showing the impact of active screening, most tuberculosis (TB) programmes in resource-limited settings only carry out passive contact investigation. The cost of such strategies is often cited as barriers to their implementation. However, little data are available for the additional costs required to implement this strategy. We aimed to estimate the cost and cost-effectiveness of active contact investigation as compared with passive contact investigation in urban Pakistan. Methods We estimated the cost-effectiveness of ‘enhanced’ (passive with follow-up) and ‘active’ (household visit) contact investigations compared with standard ‘passive’ contact investigation from providers and the programme’s perspective using a simple decision tree. Costs were collected in Pakistan from a TB clinic performing passive contact investigation and from studies of active contact tracing interventions conducted. The effectiveness was based on the number of patients with TB identified among household contacts screened. Results The addition of enhanced contact investigation to the existing passive mode detected 3.8 times more cases of TB per index patient compared with passive contact investigation alone. The incremental cost was US30perindexpatient,whichyieldedanincrementalcostofUS30 per index patient, which yielded an incremental cost of US120 per incremental patient identified with TB. The active contact investigation was 1.5 times more effective than enhanced contact investigation with an incremental cost of US$238 per incremental patient with TB identified. Conclusion Our results show that enhanced and active approaches to contact investigation effectively identify additional patients with TB among household contacts at a relatively modest cost. These strategies can be added to the passive contact investigation in a high burden setting to find the people with TB who are missed and meet the End TB strategy goals.publishedVersio
