    Options of public income support for the unemployed in the Philippines and social protection

    How can the income support for the unemployed Filipino workers be improved? The study analyzes both arguments in favor of, and against each of the following programs: unemployment insurance, unemployment assistance, severance pay, unemployment insurance savings accounts, public works, and self-employment programs. In doing so, it addresses the following questions: How does a candidate program interact with other labor market institutions? Does the program respond to a country's income shocks such as recessions, and natural disasters? Does the country have sufficient administrative capacity to carry out a program? Does the program fit into existing formal, and informal mechanisms of social risk management? Is there a risk of disrupting, or displacing existing mechanisms, such as transfers between family members? Is the program attuned to the prevailing norms, and culture? The study concludes that all programs have certain advantages, and disadvantages - an argument for the multiplicity of programs and flexibility of their use. Nonetheless, to enhance income protection, the paper proposes a two-prong approach: a) the expansion of public works, and b) the introduction of a new program - individual savings accounts, i.e., either as unemployment insurance savings accounts, or comprehensive savings accounts covering contingencies such as education, health, housing, and old age.Environmental Economics&Policies,Services&Transfers to Poor,Safety Nets and Transfers,Rural Poverty Reduction,Health Economics&Finance

    Exact moments in a continuous time random walk with complete memory of its history

    We present a continuous time generalization of a random walk with complete memory of its history [Phys. Rev. E 70, 045101(R) (2004)] and derive exact expressions for the first four moments of the distribution of displacement when the number of steps is Poisson distributed. We analyze the asymptotic behavior of the normalized third and fourth cumulants and identify new transitions in a parameter regime where the random walk exhibits superdiffusion. These transitions, which are also present in the discrete time case, arise from the memory of the process and are not reproduced by Fokker-Planck approximations to the evolution equation of this random walk.Comment: Revtex4, 10 pages, 2 figures. v2: applications discussed, clarity improved, corrected scaling of third momen

    Trajectory design and optimization of a solar sail sun probe

    There is a desire to observe the sun's poles to further deepen our understanding of solar activity. However, because of the large speeds needed to perform out-of-ecliptic plane maneuvers, chemical and electric rocket propulsion mechanisms have been proven to be costly and impractical, leaving alternative space technology systems like solar sails to be considered for these applications. In this paper, we study the possibility of using a solar sail as a probe observing the sun. We design and optimize the trajectories of the solar sail probe through the surface constraint approach, with the assumption that the sail moves on a displaced spherical surface. We first review the surface constraint approach, focusing on its important assumptions and limitations. Then, we solve and obtain a family of radial and azimuthal trajectory equations by choosing the correct constraint equation. We characterize the trajectories based on the functional dependence of the sail's orientation with the polar angle. Finally, we determine the trajectories of the probe that will give us the minimum flight time. Results show that increasing the number of mission stages decreases the total flight time, with minimal changes in the sail's radial and polar velocities. Furthermore, changing the functional dependence of the clock angle resets the azimuthal velocity, making the sail not reverse its direction and directly approach the sun along the spherical surface.Comment: 25 pages, 8 figure

    Reduced insulin secretion correlates with decreased expression of exocytotic genes in pancreatic islets from patients with type 2 diabetes.

    Reduced insulin release has been linked to defect exocytosis in β-cells. However, whether expression of genes suggested to be involved in the exocytotic process (exocytotic genes) is altered in pancreatic islets from patients with type 2 diabetes (T2D), and correlate to insulin secretion, needs to be further investigated. Analysing expression levels of 23 exocytotic genes using microarray revealed reduced expression of five genes in human T2D islets (χ(2)=13.25; p<0.001). Gene expression of STX1A, SYT4, SYT7, SYT11, SYT13, SNAP25 and STXBP1 correlated negatively to in vivo measurements of HbA1c levels and positively to glucose stimulated insulin secretion (GSIS) in vitro in human islets. STX1A, SYT4 and SYT11 protein levels correspondingly decreased in human T2D islets. Moreover, silencing of SYT4 and SYT13 reduced GSIS in INS1-832/13 cells. Our data support that reduced expression of exocytotic genes contributes to impaired insulin secretion, and suggest decreased expression of these genes as part of T2D pathogenesis

    Discovery of (R)-N-benzyl-2-(2,5-dioxopyrrolidin-1-yl)propanamide [(R)-AS-1], a novel orally bioavailable EAAT2 modulator with drug-like properties and potent antiseizure activity in vivo

    [Image: see text] (R)-7 [(R)-AS-1] showed broad-spectrum antiseizure activity across in vivo mouse seizure models: maximal electroshock (MES), 6 Hz (32/44 mA), acute pentylenetetrazol (PTZ), and PTZ-kindling. A remarkable separation between antiseizure activity and CNS-related adverse effects was also observed. In vitro studies with primary glia cultures and COS-7 cells expressing the glutamate transporter EAAT2 showed enhancement of glutamate uptake, revealing a stereoselective positive allosteric modulator (PAM) effect, further supported by molecular docking simulations. (R)-7 [(R)-AS-1] was not active in EAAT1 and EAAT3 assays and did not show significant off-target activity, including interactions with targets reported for marketed antiseizure drugs, indicative of a novel and unprecedented mechanism of action. Both in vivo pharmacokinetic and in vitro absorption, distribution, metabolism, excretion, toxicity (ADME-Tox) profiles confirmed the favorable drug-like potential of the compound. Thus, (R)-7 [(R)-AS-1] may be considered as the first-in-class small-molecule PAM of EAAT2 with potential for further preclinical and clinical development in epilepsy and possibly other CNS disorders

    TIGER : The gene expression regulatory variation landscape of human pancreatic islets

    Genome-wide association studies (GWASs) identified hundreds of signals associated with type 2 diabetes (T2D). To gain insight into their underlying molecular mechanisms, we have created the translational human pancreatic islet genotype tissue-expression resource (TIGER), aggregating >500 human islet genomic datasets from five cohorts in the Horizon 2020 consortium T2DSystems. We impute genotypes using four reference panels and meta-analyze cohorts to improve the coverage of expression quantitative trait loci (eQTL) and develop a method to combine allele-specific expression across samples (cASE). We identify >1 million islet eQTLs, 53 of which colocalize with T2D signals. Among them, a low-frequency allele that reduces T2D risk by half increases CCND2 expression. We identify eight cASE colocalizations, among which we found a T2D-associated SLC30A8 variant. We make all data available through the TIGER portal (http://tiger.bsc.es), which represents a comprehensive human islet genomic data resource to elucidate how genetic variation affects islet function and translates into therapeutic insight and precision medicine for T2D.Peer reviewe
