Correlazione genotipo-fenotipo in pazienti con malformazioni della corteccia cerebrale
Lo studio delle malformazioni corticali ne ha evidenziato, soprattutto negli
ultimi anni, l'alta frequenza in soggetti con ritardo mentale, ritardo dello sviluppo ed
epilessia, specialmente in etĂ pediatrica. In particolare la diagnostica per immagini
costituisce il principale mezzo per l'identificazione e la classificazione delle diverse
sindromi malformative, permettendo un rapido inquadramento diagnostico. La genetica
molecolare e l'analisi di linkage rappresentano gli strumenti piĂą importanti per
identificare alterazioni in geni noti e per indirizzare lo studio su nuovi loci, al fine di
delineare i fattori genetici di rischio per le sindromi identificate. Negli ultimi dieci anni
l’avvento della diagnostica mediante neuro-immagini ad alta risoluzione ha consentito
l’individuazione o l’ulteriore caratterizzazione di vari tipi di malformazioni corticali.
Le variazioni nella distribuzione e profonditĂ dei solchi corticali, nello spessore della
corteccia cerebrale, nei confini fra sostanza bianca e sostanza grigia e nell’intensità di
segnale permettono il riconoscimento di differenti pattern malformativi. Uno sviluppo
anomalo corticale rappresenta una maggiore causa di epilessia, che costituisce
generalmente il fenotipo clinico caratteristico di questi pazienti. Le piĂą gravi
malformazioni possono tuttavia manifestarsi con quadri di grave ritardo dello sviluppo e
precoce insorgenza delle crisi, con conseguente svantaggio riproduttivo. Recentemente
è stato dimostrato che diversi geni sono coinvolti nel processo di migrazione neuronale,
attraverso cui si formano i sei strati definitivi della corteccia cerebrale. In particolare è
stato visto che i geni implicati in questo processo sono FLNA, richiesto per
l'allontanamento dei neuroni dalla zona ventricolare, LIS1 e DCX, necessari per la
successiva migrazione di questi neuroni verso la corteccia, e i geni RELN, DAB1,
VLDLR, APOER2, CDK5 e P35 che permettono il passaggio dei neuroni attraverso la
piastra corticale. Lo studio dei geni che guidano la migrazione neuronale è di
fondamentale importanza per la comprensione delle anomalie dello sviluppo corticale
che spesso sono associate a gravi forme di epilessia e di ritardo mentale.
In questo studio è stato effettuata un'analisi clinica, neuroradiologica e
molecolare di pazienti con malformazioni della corteccia cerebrale determinate da
disturbi della migrazione neuronale (lissencefalie, eterotopia nodulare periventricolare)
e da anomalie dell’organizzazione corticale (polimicrogirie). E' stata eseguita una

analisi di mutazione mediante DHPLC e sequenziamento diretto dei geni che sono
responsabili di tali malformazioni: LIS1, DCX, FLNA, GPR56. Inoltre, l'elevata
sensibilitĂ e la natura quantitativa della metodica dHPLC ci hanno consentito di
caratterizzare due casi di mosaicismo somatico del gene FLNA. La DHPLC rappresenta
quindi una valida metodica da utilizzare quando si deve effettuare, in un numero elevato
di individui, un’analisi di mutazione rapida e sensibile di un gene di grandi dimensioni
come FLNA. I risultati da noi ottenuti evidenziano inoltre come tale metodica sia di
grande utilitĂ per l'identificazione e la caratterizzazione di mutazioni presenti sotto
forma di mosaico somatico. Infine l'utilizzo di una nuova metodica definita MLPA ci ha
permesso di identificare delezioni e duplicazioni genomiche in pazienti con lissencefalia
e eterotopia a banda sottocorticale che erano risultati negativi all'analisi di mutazione
dei geni LIS1 e DCX effettuata con le metodiche standard.
I risultati ottenuti sono stati utilizzati per effettuare correlazioni genotipofenotipo
all’interno della casistica di pazienti con malformazioni della corteccia
cerebrale pervenuti al nostro centro. In particolare, la tipologia di mutazioni da noi
individuate (mutazioni missenso, mutazioni troncanti, delezioni o duplicazioni) e la loro
localizzazione all’interno della proteina (ad esempio mutazioni in regioni conservate o
in domini coinvolti in meccanismi funzionali importanti) sono stati correlati con i dati
neuroradiologici, il livello cognitivo e la presenza di epilessia.
I risultati di questo studio sono importanti per formulare linee guida cliniche e
genetiche per il trattamento dei pazienti con malformazioni corticali e per offrire una
adeguata consulenza genetica alle famiglie dei pazienti affetti

A partire dal commento approntato dall'autrice per l'edizione anastatica a cura di Arnaldo Bruni dell'Outline Engravings and Descriptions of the Woburn Abbey Marbles (Firenze, Polistampa, 2012), il catalogo della collezione di sculture del duca di Bedford che ospita la Dissertation on an Ancient Hymn to the Graces di Foscolo, il saggio discute i rapporti di quest'ultima opera con l'altro inserto foscoliano interpolato nella descrizione del gruppo canoviano delle Grazie di Philip Hunt. L'attenzione si concentra sui modelli che Foscolo poté aver presenti per la selezione delle fonti all'interno della vasta produzione settecentesca di repertori iconografici e mitografici. Si discute poi dell'impiego nella Dissertation del topos del manoscritto ritrovato
Foscolo e l’estetica di Lessing
Il saggio ripercorre il diagramma della riflessione di Foscolo attorno a uno degli snodi cruciali della cultura europea tra Sette e Ottocento, quello del rapporto fra poesia e arti figurative. In particolare, si intende fare il punto su una questione già da altri dibattuta, ossia sul ruolo che nello sviluppo delle idee di Foscolo sull’argomento ebbe l’incontro con un testo capitale dell’estetica neoclassica, il trattato Laocoonte (1766) di Gotthold Ephraim Lessing. Secondo l’ipotesi che qui si sostiene, la piena assimilazione delle idee di Lessing da parte di Foscolo e la loro applicazione in una personale rielaborazione è testimoniata soltanto nei materiali relativi al secondo carme e quindi, all’incirca, a partire dal 1813, raggiungendo il culmine nella Dissertation on an Ancient Hymn to the Graces del 1822.L’essai retrace l’histoire de la réflexion de Foscolo sur l’une des questions cruciales de la culture européenne entre xviiie et xixe siècle, celle de la relation entre la poésie et les arts figuratifs. On voudrait faire le point ici sur un problème que la critique foscolienne a souvent évoqué, à savoir le rôle joué dans l’élaboration de la pensée de Foscolo par sa rencontre avec un texte d’une importance capitale dans l’esthétique néoclassique, le Laocoon (1766) de Gotthold Ephraïm Lessing. Nous avançons l’hypothèse que des traces de l’assimilation des idées de Lessing et de leur réélaboration personnelle de la part de Foscolo apparaissent pour la première fois dans les matériaux théoriques liés aux Grâces, à partir de 1813. Cette appropriation atteint ensuite son sommet dans la Dissertation on an Ancient Hymn to the Graces (1822).The essay retraces the development of Foscolo’s views on one of the most disputed points in European culture between the eighteenth and the nineteenth century: the relationship between poetry and the fine arts. It deals with an issue that Foscolo’s scholars have already addressed, namely the role that a work of paramount importance in neoclassical aesthetics, Lessing’s Laocoon (1766), played in the development of his ideas on the subject. The essay will argue that Lessing’s ideas were fully appropriated and re-elaborated only in writings relating to the Grazie, which he began in 1813. Migrating focal seizures and myoclonic status in ARV1-related encephalopathy
Objective: To report longitudinal clinical, EEG, and MRI findings in 2 sisters carrying compound heterozygous ARV1 mutations and exhibiting a peculiar form of developmental and epileptic encephalopathy (DEE). Neuropathologic features are also described in one of the sisters. Methods: Clinical course description, video-EEG polygraphic recordings, brain MRI, skin and muscle biopsies, whole-exome sequencing (WES), and brain neuropathology. Results: Since their first months of life, both girls exhibited severe axial hypotonia, visual inattention, dyskinetic movements, severe developmental delay, and slow background EEG activity. Intractable nonmotor seizures started in both at the eighth month of life, exhibiting the electroclinical characteristics of epilepsy of infancy with migrating focal seizures (EIMFS). In the second year of life, continuous epileptiform EEG activity of extremely high amplitude appeared in association with myoclonic status, leading to severely impaired alertness and responsiveness. Repeated brain MRI revealed progressive atrophic changes and severe hypomyelination. WES identified a compound heterozygous in the ARV1 gene [(p.Ser122Glnfs*7) and (p.Trp163*)] in one patient and was subsequently confirmed in the other. Both sisters died prematurely during respiratory infections. Postmortem neuropathologic examination of the brain, performed in one, revealed atrophic brain changes, mainly involving the cerebellum. Conclusions: This report confirms that biallelic ARV1 mutations cause a severe form of DEE and adds epilepsy with migrating focal seizures and myoclonic status to the spectrum of epilepsy phenotypes. Considering the potential role of human ARV1 in glycosylphosphatidylinositol (GPI) anchor biosynthesis, this severe syndrome can be assigned to the group of inherited GPI deficiency disorders, with which it shares remarkably similar clinical and neuroimaging features. CDKL5 deficiency disorder: progressive brain atrophy may be part of the syndrome
The clinical phenotype of Cyclin-Dependent Kinase-Like 5 (CDKL5) deficiency disorder (CDD) has been delineated but neuroimaging features have not been systematically analyzed. We studied brain magnetic resonance imaging (MRI) scans in a cohort of CDD patients and reviewed age at seizure onset, seizure semiology, head circumference. Thirty-five brain MRI from 22 unrelated patients were included. The median age at study entry was 13.4 years. In 14/22 patients (85.7%), MRI in the first year of life was unremarkable in all but two. In 11/22, we performed MRI after 24 months of age (range 2.5-23 years). In 8 out of 11 (72.7%), MRI showed supratentorial atrophy and in six cerebellar atrophy. Quantitative analysis detected volumetric reduction of the whole brain (-17.7%, P-value = 0.014), including both white matter (-25.7%, P-value = 0.005) and cortical gray matter (-9.1%, P-value = 0.098), with a reduction of surface area (-18.0%, P-value = 0.032), mainly involving the temporal regions, correlated with the head circumference (& rho; = 0.79, P-value = 0.109). Both the qualitative structural assessment and the quantitative analysis detected brain volume reduction involving the gray and white matter. These neuroimaging findings may be related to either progressive changes due to CDD pathogenesis, or to the extreme severity of epilepsy, or both. Larger prospective studies are needed to clarify the bases for the structural changes we observed
Prediction of fatty acid composition in intact and minced fat of European autochthonous pigs breeds by near infrared spectroscopy
The fatty acids profile has been playing a decisive role in recent years, thanks to technological, sensory and health demands from producers and consumers. The application of NIRS technique on fat tissues, could lead to more efficient, practical, and economical in the quality control. The study aim was to assess the accuracy of Fourier Transformed Near Infrared Spectroscopy technique to determine fatty acids composition in fat of 12 European local pig breeds. A total of 439 spectra of backfat were collected both in intact and minced tissue and then were analyzed using gas chromatographic analysis. Predictive equations were developed using the 80% of samples for the calibration, followed by full cross validation, and the remaining 20% for the external validation test. NIRS analysis of minced samples allowed a better response for fatty acid families, n6 PUFA, it is promising both for n3 PUFA quantification and for the screening (high, low value) of the major fatty acids. Intact fat prediction, although with a lower predictive ability, seems suitable for PUFA and n6 PUFA while for other families allows only a discrimination between high and low values.info:eu-repo/semantics/publishedVersio
Mutations in TUBG1, DYNC1H1, KIF5C and KIF2A cause malformations of cortical development and microcephaly.
International audienceThe genetic causes of malformations of cortical development (MCD) remain largely unknown. Here we report the discovery of multiple pathogenic missense mutations in TUBG1, DYNC1H1 and KIF2A, as well as a single germline mosaic mutation in KIF5C, in subjects with MCD. We found a frequent recurrence of mutations in DYNC1H1, implying that this gene is a major locus for unexplained MCD. We further show that the mutations in KIF5C, KIF2A and DYNC1H1 affect ATP hydrolysis, productive protein folding and microtubule binding, respectively. In addition, we show that suppression of mouse Tubg1 expression in vivo interferes with proper neuronal migration, whereas expression of altered Îł-tubulin proteins in Saccharomyces cerevisiae disrupts normal microtubule behavior. Our data reinforce the importance of centrosomal and microtubule-related proteins in cortical development and strongly suggest that microtubule-dependent mitotic and postmitotic processes are major contributors to the pathogenesis of MCD

Individual common variants exert weak effects on the risk for autism spectrum disorders.
While it is apparent that rare variation can play an important role in the genetic architecture of autism spectrum disorders (ASDs), the contribution of common variation to the risk of developing ASD is less clear. To produce a more comprehensive picture, we report Stage 2 of the Autism Genome Project genome-wide association study, adding 1301 ASD families and bringing the total to 2705 families analysed (Stages 1 and 2). In addition to evaluating the association of individual single nucleotide polymorphisms (SNPs), we also sought evidence that common variants, en masse, might affect the risk. Despite genotyping over a million SNPs covering the genome, no single SNP shows significant association with ASD or selected phenotypes at a genome-wide level. The SNP that achieves the smallest P-value from secondary analyses is rs1718101. It falls in CNTNAP2, a gene previously implicated in susceptibility for ASD. This SNP also shows modest association with age of word/phrase acquisition in ASD subjects, of interest because features of language development are also associated with other variation in CNTNAP2. In contrast, allele scores derived from the transmission of common alleles to Stage 1 cases significantly predict case status in the independent Stage 2 sample. Despite being significant, the variance explained by these allele scores was small (Vm< 1%). Based on results from individual SNPs and their en masse effect on risk, as inferred from the allele score results, it is reasonable to conclude that common variants affect the risk for ASD but their individual effects are modest
