    Repostaje en tierra de un avión mediante algoritmos genéticos

    [Resumen] En este trabajo se presenta una solución alternativa, basada en técnicas de la inteligencia artificial, al problema de la gestión de la distribución de combustible dentro de una aeronave comercial. En concreto se ha abordado el repostaje de la aeronave antes de un vuelo. El repostaje en tierra consiste en decidir la cantidad de combustible que se dispondrá en cada depósito atendiendo a criterios de seguridad, estabilidad del avión y eficiencia energética. Para ello se define un problema de optimización cuya función de coste nace del conocimiento específico de los factores que influyen en una aeronave que tiene relación con el posicionamiento del combustible. Después su resolución se realiza mediante algoritmos genéticos. Los resultados obtenidos en simulación mejoran la solución manual estándar que se viene aplicando.Ministerio de Economía y Competitividad; DPI2015-71320-RED

    Paecilomyces fumosoroseus blastospore production using liquid culture in a bioreactor

    There are many advantages to using liquid cultures for the production of blastospores. These include mainly the processes of scale up which are relatively easy, as well as the control of parameters such astemperature, aeration and pH. In this work, we evaluated the production of Paecilomyces fumosoroseus blastospores using a low-cost liquid culture medium in a fermenter in comparison to a mediumcommonly used for this purpose, with regard to yield and viability of blastospores. The two media contained the same concentration of glucose but differed in N source (M1 containing casamino acidsand M2 provided with collagen peptone and yeast extract). Starting with an inoculum of 1x106 blastospores/ml, M2 medium produced 2x1010 blastospores/ml after incubation for 72 h at 520 rev/minagitation and 1 v/v/m (volume air/volume liquid.min) aeration, while only 2.4 x 108/ml were produced with M1. In addition, the microorganisms in medium M1 grew more slowly during log phase and reached an earlier plateau at 36 h fermentation. The medium containing collagen peptone and yeast extract is an excellent alternative for the production of P. fumosoroseus blastospores, providing lower cost, higher yield and shorter propagation time, but formulation does need to be improved

    CorE from Myxococcus xanthus Is a Copper-Dependent RNA Polymerase Sigma Factor

    The dual toxicity/essentiality of copper forces cells to maintain a tightly regulated homeostasis for this metal in all living organisms, from bacteria to humans. Consequently, many genes have previously been reported to participate in copper detoxification in bacteria. Myxococcus xanthus, a prokaryote, encodes many proteins involved in copper homeostasis that are differentially regulated by this metal. A σ factor of the ECF (extracytoplasmic function) family, CorE, has been found to regulate the expression of the multicopper oxidase cuoB, the P1B-type ATPases copA and copB, and a gene encoding a protein with a heavy-metal-associated domain. Characterization of CorE has revealed that it requires copper to bind DNA in vitro. Genes regulated by CorE exhibit a characteristic expression profile, with a peak at 2 h after copper addition. Expression rapidly decreases thereafter to basal levels, although the metal is still present in the medium, indicating that the activity of CorE is modulated by a process of activation and inactivation. The use of monovalent and divalent metals to mimic Cu(I) and Cu(II), respectively, and of additives that favor the formation of the two redox states of this metal, has revealed that CorE is activated by Cu(II) and inactivated by Cu(I). The activation/inactivation properties of CorE reside in a Cys-rich domain located at the C terminus of the protein. Point mutations at these residues have allowed the identification of several Cys involved in the activation and inactivation of CorE. Based on these data, along with comparative genomic studies, a new group of ECF σ factors is proposed, which not only clearly differs mechanistically from the other σ factors so far characterized, but also from other metal regulators

    Las encuestas nacionales sobre salud bucodental tienen como función básica proporcionar una idea de conjunto sobre salud y necesidades de tratamiento poblacional con el fin de vigilar la evolución de las tasas de morbilidad. Nos permiten conocer: - La medida en que los servicios odontológicos existentes responden a las necesidades de la población. - La naturaleza y cuantía de los servicios de prevención y restauración necesarios. - Los recursos necesarios para implantar, mantener, aumentar o reducir los programas de salud bucodental, estimando las necesidades cuantitativas y el tipo de personal requerido. En 1993, el Consejo General de Colegios de Odontólogos y Estomatólogos de España (actualmente denominado Consejo General de Colegios de Dentistas de España) encargó la realización de una encuesta epidemiológica bucodental, siguiendo los criterios establecidos por la Organización Mundial de la Salud (OMS) para la ejecución de estudios transversales tipo Pathfinder. Ese estudio, publicado en 19951, se realizó una década después del anterior, desarrollado en 1984 bajo supervisión de la OMS. En los años 2000, 2005 y 20105 se realizaron las correspondientes encuestas, siempre financiadas el Consejo General de Dentistas. Transcurridos 5 años desde entonces, la necesidad de monitorizar la situación oral de nuestra población es la principal justificación de este proyecto

    Proteomic Analysis Reveals That Iron Availability Alters the Metabolic Status of the Pathogenic Fungus Paracoccidioides brasiliensis

    Paracoccidioides brasiliensis is a thermodimorphic fungus and the causative agent of paracoccidioidomycosis (PCM). The ability of P. brasiliensis to uptake nutrients is fundamental for growth, but a reduction in the availability of iron and other nutrients is a host defense mechanism many pathogenic fungi must overcome. Thus, fungal mechanisms that scavenge iron from host may contribute to P. brasiliensis virulence. In order to better understand how P. brasiliensis adapts to iron starvation in the host we compared the two-dimensional (2D) gel protein profile of yeast cells during iron starvation to that of iron rich condition. Protein spots were selected for comparative analysis based on the protein staining intensity as determined by image analysis. A total of 1752 protein spots were selected for comparison, and a total of 274 out of the 1752 protein spots were determined to have changed significantly in abundance due to iron depletion. Ninety six of the 274 proteins were grouped into the following functional categories; energy, metabolism, cell rescue, virulence, cell cycle, protein synthesis, protein fate, transcription, cellular communication, and cell fate. A correlation between protein and transcript levels was also discovered using quantitative RT-PCR analysis from RNA obtained from P. brasiliensis under iron restricting conditions and from yeast cells isolated from infected mouse spleens. In addition, western blot analysis and enzyme activity assays validated the differential regulation of proteins identified by 2-D gel analysis. We observed an increase in glycolytic pathway protein regulation while tricarboxylic acid cycle, glyoxylate and methylcitrate cycles, and electron transport chain proteins decreased in abundance under iron limiting conditions. These data suggest a remodeling of P. brasiliensis metabolism by prioritizing iron independent pathways

    Epidemiological trends of HIV/HCV coinfection in Spain, 2015-2019

    Altres ajuts: Spanish AIDS Research Network; European Funding for Regional Development (FEDER).Objectives: We assessed the prevalence of anti-hepatitis C virus (HCV) antibodies and active HCV infection (HCV-RNA-positive) in people living with HIV (PLWH) in Spain in 2019 and compared the results with those of four similar studies performed during 2015-2018. Methods: The study was performed in 41 centres. Sample size was estimated for an accuracy of 1%. Patients were selected by random sampling with proportional allocation. Results: The reference population comprised 41 973 PLWH, and the sample size was 1325. HCV serostatus was known in 1316 PLWH (99.3%), of whom 376 (28.6%) were HCV antibody (Ab)-positive (78.7% were prior injection drug users); 29 were HCV-RNA-positive (2.2%). Of the 29 HCV-RNA-positive PLWH, infection was chronic in 24, it was acute/recent in one, and it was of unknown duration in four. Cirrhosis was present in 71 (5.4%) PLWH overall, three (10.3%) HCV-RNA-positive patients and 68 (23.4%) of those who cleared HCV after anti-HCV therapy (p = 0.04). The prevalence of anti-HCV antibodies decreased steadily from 37.7% in 2015 to 28.6% in 2019 (p < 0.001); the prevalence of active HCV infection decreased from 22.1% in 2015 to 2.2% in 2019 (p < 0.001). Uptake of anti-HCV treatment increased from 53.9% in 2015 to 95.0% in 2019 (p < 0.001). Conclusions: In Spain, the prevalence of active HCV infection among PLWH at the end of 2019 was 2.2%, i.e. 90.0% lower than in 2015. Increased exposure to DAAs was probably the main reason for this sharp reduction. Despite the high coverage of treatment with direct-acting antiviral agents, HCV-related cirrhosis remains significant in this population

    Conservação de café torrado e moído durante o armazenamento. Parte 2: Massa específica e porosidade

    The determination of physical properties is an important factor in the design of machinery and the scaling of post-harvest operations. The present study evaluates the influence of the level of roasting and the size of grinding on the physical properties of coffee during storage. The following physical properties were evaluated: true and bulk density, and intergranular porosity. Raw coffee beans (Coffea canephora and Coffea arabica), hulled and dried, were roasted to two different levels: medium light (SCAA#65) and moderately dark (SCAA#45). The beans were then grinded into three different sizes: fine (0.59 mm), medium (0.84 mm) and coarse (1.19 mm). An additional coffee lot was kept whole. Following grinding, samples were stored at two different temperatures (10 and 30 ºC) and analyzed after five different storage durations (0, 30, 60, 120 and 180 days). The medium light roast had higher values for each of the measured physical properties. Finely ground samples had higher true and bulk densities, and porosities. It is concluded that the size of grinding, level of roasting and duration of storage significantly affect the physical properties of coffee.A determinação das propriedades físicas é fator importante na formulação de projetos de maquinários e dimensionamento de operações pós-colheita. De forma a permitir o dimensionamento correto e o uso desses maquinários, objetivou-se avaliar e determinar, durante o armazenamento, as propriedades físicas: massa específica unitária e aparente e porosidade intergranular, além de avaliar a influência dos níveis de torrefação e moagem sobre essas propriedades. Grãos de café cru (Coffea canephora e Coffea arabica) foram utilizados, descascados e secados e só então torrados em dois níveis: média clara (SCAA#65) e moderadamente escura (SCAA#45). Os grãos foram moídos em três granulometrias: fina (0,59 mm), média (0,84 mm) e grossa (1,19 mm), além do lote de café inteiro. Realizada a moagem as amostras foram armazenadas em duas temperaturas (10 e 30 ºC) e analisadas em cinco tempos distintos de armazenamento (0, 30, 60, 120 e 180 dias). A torra média clara permitiu maiores valores das propriedades físicas; já as amostras de granulometria fina apresentaram aumento das massas específica unitária e aparente e porosidade.
