    Haemophilus parasuis Subunit Vaccines Based on Native Proteins with Affinity to Porcine Transferrin Prevent the Expression of Proinflammatory Chemokines and Cytokines in Pigs

    8 p.The expression of chemokines (CCL-2 and CXCL-8) and cytokines (IL-1, IL-1, IL-6, TNF-, and IL-10) was evaluated by RTqPCR in colostrum-deprived pigs vaccinated and challenged with Haemophilus parasuis serovar 5. Two vaccines containing native proteins with affinity to porcine transferrin (NPAPTim and NPAPTit) were tested, along with two control groups: one inoculated with PBS instead of antigen (challenge group (CHG)), and another one nonimmunized and noninfected (blank group). The use of NPAPTim and NPAPTit resulted in complete protection against H. parasuis (no clinical signs and/or lesions), and both vaccines were capable of avoiding the expression of the proinflammatory molecules to levels similar to physiological values in blank group. However, overexpression of all proinflammatory molecules was observed in CHG group, mainly in the target infection tissues (brain, lungs, and spleen). High expression of CCL-2, CXCL-8, IL-1, IL-1, and IL-6 can be considered one of the characteristics of H. parasuis infection by serovar 5S

    Species Diversity and Phylogeographical Affinities of the Branchiopoda (Crustacea) of Churchill, Manitoba, Canada

    The region of Churchill, Manitoba, contains a wide variety of habitats representative of both the boreal forest and arctic tundra and has been used as a model site for biodiversity studies for nearly seven decades within Canada. Much previous work has been done in Churchill to study the Daphnia pulex species complex in particular, but no study has completed a wide-scale survey on the crustacean species that inhabit Churchill's aquatic ecosystems using molecular markers. We have employed DNA barcoding to study the diversity of the Branchiopoda (Crustacea) in a wide variety of freshwater habitats and to determine the likely origins of the Churchill fauna following the last glaciation. The standard animal barcode marker (COI) was sequenced for 327 specimens, and a 3% divergence threshold was used to delineate potential species. We found 42 provisional and valid branchiopod species from this survey alone, including several cryptic lineages, in comparison with the 25 previously recorded from previous ecological works. Using published sequence data, we explored the phylogeographic affinities of Churchill's branchiopods, finding that the Churchill fauna apparently originated from all directions from multiple glacial refugia (including southern, Beringian, and high arctic regions). Overall, these microcrustaceans are very diverse in Churchill and contain multiple species complexes. The present study introduces among the first sequences for some understudied genera, for which further work is required to delineate species boundaries and develop a more complete understanding of branchiopod diversity over a larger spatial scale

    La etapa Lactante es una alternativa de abordaje en la lucha contra la obesidad. Se ha demostrado que existe una relación entre el exceso de peso en ésta etapa, con las principales problemas de salud pública; con base en lo anterior, es necesario buscar factores modificables en el comportamiento de los encargados del cuidado del infante y del equipo de salud para crear conductas que propicien el adecuado desarrollo del individuo. El objetivo de éste estudio es determinar la prevalencia del exceso de peso en lactantes del área metropolitana de Monterrey, a partir de usuarios del programa de atención del niño sano del IMSS, provenientes de seis unidades de medicina familiar. Se estudiaron 400 lactantes a través de la medición de peso y talla, para clasificarlos mediante las tablas diagnósticas nutricionales de la NCHS-CDC/OMS. Se obtuvo una prevalencia del exceso de peso de los lactantes estudiados equivalente al 43.23%. Mostrando que el exceso de peso representa un problema de salud pública creciente en México, el cual requiere el replanteamiento de los programas preventivos en éste sector demográfico, buscando aquellos niños que están adquiriendo patrones patológicos de comportamiento en su ingesta alimentaria. AbstractThe infant stage is an alternative in boarding the struggle against obesity. There has been demonstrated that a relation exists between overweight in this period with the current issues of public health. Due to this, it is necessary to search for adjustable factors in the carers and health team, in order to create appropriate behaviour that propitiates suiable development of the individual. The objective in this study was to determine the prevalence of overweight in Monterrey’s metropolitan area infants who assist the healthy child program from six primary health care units of the Mexican Institute of Social Security. There were studied 400 children under a year of age, to whom they there was realized measurement of weight and height, qualifying by means of the diagnostic nutritional tables of the NCHS-CDC/OMS. The results of this study show that the prevalence rates of excess body weight in infants under a year old in Monterrey’s Metropolitan area are 43.23%.     Genetic Predisposition to Chronic Lymphocytic Leukemia Is Mediated by a BMF Super-Enhancer Polymorphism

    SummaryChronic lymphocytic leukemia (CLL) is an adult B cell malignancy. Genome-wide association studies show that variation at 15q15.1 influences CLL risk. We deciphered the causal variant at 15q15.1 and the mechanism by which it influences tumorigenesis. We imputed all possible genotypes across the locus and then mapped highly associated SNPs to areas of chromatin accessibility, evolutionary conservation, and transcription factor binding. SNP rs539846 C>A, the most highly associated variant (p = 1.42 × 10−13, odds ratio = 1.35), localizes to a super-enhancer defined by extensive histone H3 lysine 27 acetylation in intron 3 of B cell lymphoma 2 (BCL2)-modifying factor (BMF). The rs539846-A risk allele alters a conserved RELA-binding motif, disrupts RELA binding, and is associated with decreased BMF expression in CLL. These findings are consistent with rs539846 influencing CLL susceptibility through differential RELA binding, with direct modulation of BMF expression impacting on anti-apoptotic BCL2, a hallmark of oncogenic dependency in CLL

    Exploring the Association between Gambling-Related Offenses, Substance Use, Psychiatric Comorbidities, and Treatment Outcome

    Several studies have explored the association between gambling disorder (GD) and gambling-related crimes. However, it is still unclear how the commission of these offenses influences treatment outcomes. In this longitudinal study we sought: (1) to explore sociodemographic and clinical differences (e.g., psychiatric comorbidities) between individuals with GD who had committed gambling-related illegal acts (differentiating into those who had had legal consequences (n = 31) and those who had not (n = 55)), and patients with GD who had not committed crimes (n = 85); and (2) to compare the treatment outcome of these three groups, considering dropouts and relapses. Several sociodemographic and clinical variables were assessed, including the presence of substance use, and comorbid mental disorders. Patients received 16 sessions of cognitive-behavioral therapy. Patients who reported an absence of gambling-related illegal behavior were older, and showed the lowest GD severity, the most functional psychopathological state, the lowest impulsivity levels, and a more adaptive personality profile. Patients who had committed offenses with legal consequences presented the highest risk of dropout and relapses, higher number of psychological symptoms, higher likelihood of any other mental disorders, and greater prevalence of tobacco and illegal drugs use. Our findings uphold that patients who have committed gambling-related offenses show a more complex clinical profile that may interfere with their adherence to treatment

    El objetivo de este documento es actualizar las recomendaciones sobre la evaluación y el manejo de la afectación renal en pacientes con infección por el VIH del año 2010. La función renal debe monitorizarse en todos los pacientes e incluir la medida de la concentración sérica de creatinina, la estimación del filtrado glomerular (ecuación CKD-EPI), la medida del cociente proteína/creatinina en orina y un sedimento urinario. El estudio básico de la función tubular ha de incluir la concentración sérica de fosfato y la tira reactiva de orina (glucosuria). En ausencia de alteraciones, el cribado será anual. En pacientes tratados con tenofovir o con factores de riesgo para el desarrollo de enfermedad renal crónica (ERC), se recomienda una evaluación más frecuente. Se debe evitar el uso de antirretrovirales potencialmente nefrotóxicos en pacientes con ERC o factores de riesgo para evitar su progresión. También se revisan las indicaciones de la biopsia renal, cuándo enviar el paciente al nefrólogo y las indicaciones, evaluación y manejo del paciente en diálisis o del trasplante renal

    Las encuestas nacionales sobre salud bucodental tienen como función básica proporcionar una idea de conjunto sobre salud y necesidades de tratamiento poblacional con el fin de vigilar la evolución de las tasas de morbilidad. Nos permiten conocer: - La medida en que los servicios odontológicos existentes responden a las necesidades de la población. - La naturaleza y cuantía de los servicios de prevención y restauración necesarios. - Los recursos necesarios para implantar, mantener, aumentar o reducir los programas de salud bucodental, estimando las necesidades cuantitativas y el tipo de personal requerido. En 1993, el Consejo General de Colegios de Odontólogos y Estomatólogos de España (actualmente denominado Consejo General de Colegios de Dentistas de España) encargó la realización de una encuesta epidemiológica bucodental, siguiendo los criterios establecidos por la Organización Mundial de la Salud (OMS) para la ejecución de estudios transversales tipo Pathfinder. Ese estudio, publicado en 19951, se realizó una década después del anterior, desarrollado en 1984 bajo supervisión de la OMS. En los años 2000, 2005 y 20105 se realizaron las correspondientes encuestas, siempre financiadas el Consejo General de Dentistas. Transcurridos 5 años desde entonces, la necesidad de monitorizar la situación oral de nuestra población es la principal justificación de este proyecto

    Monitoring an Alien Invasion: DNA Barcoding and the Identification of Lionfish and Their Prey on Coral Reefs of the Mexican Caribbean

    BACKGROUND: In the Mexican Caribbean, the exotic lionfish Pterois volitans has become a species of great concern because of their predatory habits and rapid expansion onto the Mesoamerican coral reef, the second largest continuous reef system in the world. This is the first report of DNA identification of stomach contents of lionfish using the barcode of life reference database (BOLD). METHODOLOGY/PRINCIPAL FINDINGS: We confirm with barcoding that only Pterois volitans is apparently present in the Mexican Caribbean. We analyzed the stomach contents of 157 specimens of P. volitans from various locations in the region. Based on DNA matches in the Barcode of Life Database (BOLD) and GenBank, we identified fishes from five orders, 14 families, 22 genera and 34 species in the stomach contents. The families with the most species represented were Gobiidae and Apogonidae. Some prey taxa are commercially important species. Seven species were new records for the Mexican Caribbean: Apogon mosavi, Coryphopterus venezuelae, C. thrix, C. tortugae, Lythrypnus minimus, Starksia langi and S. ocellata. DNA matches, as well as the presence of intact lionfish in the stomach contents, indicate some degree of cannibalism, a behavior confirmed in this species by the first time. We obtained 45 distinct crustacean prey sequences, from which only 20 taxa could be identified from the BOLD and GenBank databases. The matches were primarily to Decapoda but only a single taxon could be identified to the species level, Euphausia americana. CONCLUSIONS/SIGNIFICANCE: This technique proved to be an efficient and useful method, especially since prey species could be identified from partially-digested remains. The primary limitation is the lack of comprehensive coverage of potential prey species in the region in the BOLD and GenBank databases, especially among invertebrates
