    The radial arrangement of the human chromosome 7 in the lymphocyte cell nucleus is associated with chromosomal band gene density

Copyright @ Springer-Verlag 2008.In the nuclei of human lymphocytes, chromosome territories are distributed according to the average gene density of each chromosome. However, chromosomes are very heterogeneous in size and base composition, and can contain both very gene-dense and very gene-poor regions. Thus, a precise analysis of chromosome organisation in the nuclei should consider also the distribution of DNA belonging to the chromosomal bands in each chromosome. To improve our understanding of the chromatin organisation, we localised chromosome 7 DNA regions, endowed with different gene densities, in the nuclei of human lymphocytes. Our results showed that this chromosome in cell nuclei is arranged radially with the gene-dense/GC-richest regions exposed towards the nuclear interior and the gene-poorest/GC-poorest ones located at the nuclear periphery. Moreover, we found that chromatin fibres from the 7p22.3 and the 7q22.1 bands are not confined to the territory of the bulk of this chromosome, protruding towards the inner part of the nucleus. Overall, our work demonstrates the radial arrangement of the territory of chromosome 7 in the lymphocyte nucleus and confirms that human genes occupy specific radial positions, presumably to enhance intra- and inter-chromosomal interaction among loci displaying a similar expression pattern, and/or similar replication timing

    Genome-Wide Transcriptome Analyses of Silicon Metabolism in Phaeodactylum tricornutum Reveal the Multilevel Regulation of Silicic Acid Transporters

    BACKGROUND:Diatoms are largely responsible for production of biogenic silica in the global ocean. However, in surface seawater, Si(OH)(4) can be a major limiting factor for diatom productivity. Analyzing at the global scale the genes networks involved in Si transport and metabolism is critical in order to elucidate Si biomineralization, and to understand diatoms contribution to biogeochemical cycles. METHODOLOGY/PRINCIPAL FINDINGS:Using whole genome expression analyses we evaluated the transcriptional response to Si availability for the model species Phaeodactylum tricornutum. Among the differentially regulated genes we found genes involved in glutamine-nitrogen pathways, encoding putative extracellular matrix components, or involved in iron regulation. Some of these compounds may be good candidates for intracellular intermediates involved in silicic acid storage and/or intracellular transport, which are very important processes that remain mysterious in diatoms. Expression analyses and localization studies gave the first picture of the spatial distribution of a silicic acid transporter in a diatom model species, and support the existence of transcriptional and post-transcriptional regulations. CONCLUSIONS/SIGNIFICANCE:Our global analyses revealed that about one fourth of the differentially expressed genes are organized in clusters, underlying a possible evolution of P. tricornutum genome, and perhaps other pennate diatoms, toward a better optimization of its response to variable environmental stimuli. High fitness and adaptation of diatoms to various Si levels in marine environments might arise in part by global regulations from gene (expression level) to genomic (organization in clusters, dosage compensation by gene duplication), and by post-transcriptional regulation and spatial distribution of SIT proteins

Mostraremos que existem funções inteiras e transcendentes que levam um subconjunto dos números de Liouville nele mesmo e daremos uma resposta positiva ao Problema B de Mahler: Problema B: Existe uma função inteira e transcendente f(z) = Σn =0 ∞ a nz n com coeficientes racionais tal que f( ´Q ) ⊆ ´Q e f−1( ´Q ) ⊆ ´Q ? .Let f be an entire transcendental function.     Effect of Bi substitution on the cationic vacancy ordering in SrMoO<inf>4</inf>-based complex oxides: Structure and properties

    The electronic and crystal structures of the Sr1−3xBi2xФxMoO4 series (0.025 ≤ x ≤ 0.30, where Ф represents cation vacancies) were studied. The 0.025 ≤ x ≤ 0.15 compositions showed a defect scheelite structure. Powder X-ray and neutron diffraction patterns for compositions with 0.15 < x ≤ 0.225 exhibited a tetragonal supercell with asup ≈ √5a, csup ≈ c where a and c are the tetragonal scheelite cell parameters. Strong distortion of MoO4 polyhedra was shown by Raman spectroscopy and described by the calculated phonon spectra of “virtual crystals” of Bi2+[MoO4] and Ф2+[MoO4]. The electronic structures were calculated for both types of structure. The decrease of the calculated energy gap is consistent with experimental data from Kubelka-Munk measurements. The electroconductive properties were measured by A.C. impedance spectroscopy. For Sr1−3xBi2xФxMoO4 compositions conductivity increases with increasing x-value, with maximum values of conductivity at 948 K of 2.82 × 10−5S cm−1 for the x = 0.20 composition
