    Origin of symbol-using systems: speech, but not sign, without the semantic urge

    Natural language—spoken and signed—is a multichannel phenomenon, involving facial and body expression, and voice and visual intonation that is often used in the service of a social urge to communicate meaning. Given that iconicity seems easier and less abstract than making arbitrary connections between sound and meaning, iconicity and gesture have often been invoked in the origin of language alongside the urge to convey meaning. To get a fresh perspective, we critically distinguish the origin of a system capable of evolution from the subsequent evolution that system becomes capable of. Human language arose on a substrate of a system already capable of Darwinian evolution; the genetically supported uniquely human ability to learn a language reflects a key contact point between Darwinian evolution and language. Though implemented in brains generated by DNA symbols coding for protein meaning, the second higher-level symbol-using system of language now operates in a world mostly decoupled from Darwinian evolutionary constraints. Examination of Darwinian evolution of vocal learning in other animals suggests that the initial fixation of a key prerequisite to language into the human genome may actually have required initially side-stepping not only iconicity, but the urge to mean itself. If sign languages came later, they would not have faced this constraint

    Seed Bank Changes with Time-Since-Fire In Florida Rosemary Scrub

    The soil seed bank plays a central role in the regeneration of obligate seeding species in fire-prone habitats. We evaluated how seed density and species composition changed with time-since-fire in the Florida, USA, rosemary scrub community. Because fire affects habitat availability and plant demographic variation, we predicted that soil seed density would be low in recently burned and long-unburned stands and high at intermediate time-since-fire. Seed bank soil samples were collected from a chronosequence of time-since-fire composited from two trials conducted in 1992-1993 and 2008-2009: two sites each of 3, 6, 10, and 24 years post-fire, and 3 long-unburned (similar to 67 years) sites. The seedling emergence method was used to determine species composition of the seed bank. Across all time-since-fire age classes, herbaceous species dominated the seed bank, while long-lived subshrubs and shrubs were present in low densities. Seed banks from sites three years post-fire were distinct from the other sites. When species were sorted as functional groups, seed density was related with time-since-fire for subshrubs and ruderal herbs, but not for scrub herbs and graminoids. Subshrubs and ruderal herbs had highest seed densities in recently burned stands. Seed densities of Florida rosemary (Ceratiola ericoides Michx.) (the major obligate seeding shrub in this community) were associated with time-since-fire and showed highest densities at recently burned and long-unburned sites. The seed banks of two scrub herb species, paper nailwort (Paronychia chartacea Fernald) and nodding pinweed (Lechea cernua Small), were associated with time-since-fire. They reached peaks in density in the first ten years post-fire, corresponding with similar changes in their aboveground abundances with time-since-fire. Soil seed densities of several species and functional groups were associated with time-since-fire, but timing of peak seed density varied depending on species\u27 life span and age of reproductive maturity

    Non-invasive measurements of exhaled NO and CO associated with methacholine responses in mice

    <p>Abstract</p> <p>Background</p> <p>Nitric oxide (NO) and carbon monoxide (CO) in exhaled breath are considered obtainable biomarkers of physiologic mechanisms. Therefore, obtaining their measures simply, non-invasively, and repeatedly, is of interest, and was the purpose of the current study.</p> <p>Methods</p> <p>Expired NO (E<sub>NO</sub>) and CO (E<sub>CO</sub>) were measured non-invasively using a gas micro-analyzer on several strains of mice (C57Bl6, IL-10<sup>-/-</sup>, A/J, MKK3<sup>-/-</sup>, JNK1<sup>-/-</sup>, NOS-2<sup>-/- </sup>and NOS-3<sup>-/-</sup>) with and without allergic airway inflammation (AI) induced by ovalbumin systemic sensitization and aerosol challenge, compared using independent-sample t-tests between groups, and repeated measures analysis of variance (ANOVA) within groups over time of inflammation induction. E<sub>NO </sub>and E<sub>CO </sub>were also measured in C57Bl6 and IL-10-/- mice, ages 8–58 weeks old, the relationship of which was determined by regression analysis. S-methionyl-L-thiocitrulline (SMTC), and tin protoporphyrin (SnPP) were used to inhibit neuronal/constitutive NOS-1 and heme-oxygenase, respectively, and alter NO and CO production, respectively, as assessed by paired t-tests. Methacholine-associated airway responses (AR) were measured by the enhanced pause method, with comparisons by repeated measures ANOVA and post-hoc testing.</p> <p>Results</p> <p>E<sub>NO </sub>was significantly elevated in naïve IL-10<sup>-/- </sup>(9–14 ppb) and NOS-2<sup>-/- </sup>(16 ppb) mice as compared to others (average: 5–8 ppb), whereas E<sub>CO </sub>was significantly higher in naïve A/J, NOS-3<sup>-/- </sup>(3–4 ppm), and MKK3<sup>-/- </sup>(4–5 ppm) mice, as compared to others (average: 2.5 ppm). As compared to C57Bl6 mice, AR of IL-10<sup>-/-</sup>, JNK1<sup>-/-</sup>, NOS-2<sup>-/-</sup>, and NOS-3<sup>-/- </sup>mice were decreased, whereas they were greater for A/J and MKK3<sup>-/- </sup>mice. SMTC significantly decreased E<sub>NO </sub>by ~30%, but did not change AR in NOS-2<sup>-/- </sup>mice. SnPP reduced E<sub>CO </sub>in C57Bl6 and IL-10<sup>-/- </sup>mice, and increased AR in NOS-2<sup>-/- </sup>mice. E<sub>NO </sub>decreased as a function of age in IL-10<sup>-/- </sup>mice, remaining unchanged in C57Bl6 mice.</p> <p>Conclusion</p> <p>These results are consistent with the ideas that: 1) E<sub>NO </sub>is associated with mouse strain and knockout differences in NO production and AR, 2) alterations of E<sub>NO </sub>and E<sub>CO </sub>can be measured non-invasively with induction of allergic AI or inhibition of key gas-producing enzymes, and 3) alterations in AR may be dependent on the relative balance of NO and CO in the airway.</p

    Bacteria isolated from lung modulate asthma susceptibility in mice

    Asthma is a chronic, non-curable, multifactorial disease with increasing incidence in industrial countries. This study evaluates the direct contribution of lung microbial components in allergic asthma in mice. Germ-Free and Specific-Pathogen-Free mice display similar susceptibilities to House Dust Mice-induced allergic asthma, indicating that the absence of bacteria confers no protection or increased risk to aeroallergens. In early life, allergic asthma changes the pattern of lung microbiota, and lung bacteria reciprocally modulate aeroallergen responsiveness. Primo-colonizing cultivable strains were screened for their immunoregulatory properties following their isolation from neonatal lungs. Intranasal inoculation of lung bacteria influenced the outcome of allergic asthma development: the strain CNCM I 4970 exacerbated some asthma features whereas the pro-Th1 strain CNCM I 4969 had protective effects. Thus, we confirm that appropriate bacterial lung stimuli during early life are critical for susceptibility to allergic asthma in young adults

    ITPase deficiency causes a Martsolf-like syndrome with a lethal infantile dilated cardiomyopathy

    Typical Martsolf syndrome is characterized by congenital cataracts, postnatal microcephaly, developmental delay, hypotonia, short stature and biallelic hypomorphic mutations in either RAB3GAP1 or RAB3GAP2. Genetic analysis of 85 unrelated “mutation negative” probands with Martsolf or Martsolf-like syndromes identified two individuals with different homozygous null mutations in ITPA, the gene encoding inosine triphosphate pyrophosphatase (ITPase). Both probands were from multiplex families with a consistent, lethal and highly distinctive disorder; a Martsolf-like syndrome with infantile-onset dilated cardiomyopathy. Severe ITPase-deficiency has been previously reported with infantile epileptic encephalopathy (MIM 616647). ITPase acts to prevent incorporation of inosine bases (rI/dI) into RNA and DNA. In Itpa-null cells dI was undetectable in genomic DNA. dI could be identified at a low level in mtDNA without detectable mitochondrial genome instability, mtDNA depletion or biochemical dysfunction of the mitochondria. rI accumulation was detectable in proband-derived lymphoblastoid RNA. In Itpa-null mouse embryos rI was detectable in the brain and kidney with the highest level seen in the embryonic heart (rI at 1 in 385 bases). Transcriptome and proteome analysis in mutant cells revealed no major differences with controls. The rate of transcription and the total amount of cellular RNA also appeared normal. rI accumulation in RNA–and by implication rI production—correlates with the severity of organ dysfunction in ITPase deficiency but the basis of the cellulopathy remains cryptic. While we cannot exclude cumulative minor effects, there are no major anomalies in the production, processing, stability and/or translation of mRNA

    Collective Impact Partnership and Backbone Organizations as Enablers of Children’s Well-Being

    In this article, the question of partnership is approached from a perspective centred around the creation of a common agenda based on trust and from the children´s point of view. Partnership and collaboration have traditionally been viewed as mechanisms to create bridges between organisations and institutions from the private, public and non-governmental sectors in order to enhance funder collaboratives, public-private partnerships, multi-stakeholder initiatives, social sector networks and collective impact initiatives. It was not however until Kania and Kramer´s (2011) seminal work on collective impact when this subject came to be viewed as a developmental process aiming at the creation of a common agenda and mutually agreed activities and consisting of five integral parts: a common agenda, shared measurement systems, mutually reinforcing activities, continuous communication and backbone support organisations. This article, based on a systematic review of the topic, maintains that partnership – approached from the point of view of children and through the lens of collective impact – constitutes a crucial mechanism in the creation of safe and comprehensive wellbeing for children. Thus, this article – using Kania and Kramer´s (2011) definition of collective impact and focusing on the structure of partnerships and the nature of trust in organisations as the prerequisite for partnership – advocates the importance of the UN SDG17 principle as the bringer of inclusive society built upon principles and values, a shared vision, and shared goals that place people at the centre of human endeavour.© Springer Nature Switzerland AG 2020.     A Novel Human Cytomegalovirus Locus Modulates Cell Type-Specific Outcomes of Infection

    Clinical strains of HCMV encode 20 putative ORFs within a region of the genome termed ULb′ that are postulated to encode functions related to persistence or immune evasion. We have previously identified ULb′-encoded pUL138 as necessary, but not sufficient, for HCMV latency in CD34+ hematopoietic progenitor cells (HPCs) infected in vitro. pUL138 is encoded on polycistronic transcripts that also encode 3 additional proteins, pUL133, pUL135, and pUL136, collectively comprising the UL133-UL138 locus. This work represents the first characterization of these proteins and identifies a role for this locus in infection. Similar to pUL138, pUL133, pUL135, and pUL136 are integral membrane proteins that partially co-localized with pUL138 in the Golgi during productive infection in fibroblasts. As expected of ULb′ sequences, the UL133-UL138 locus was dispensable for replication in cultured fibroblasts. In CD34+ HPCs, this locus suppressed viral replication in HPCs, an activity attributable to both pUL133 and pUL138. Strikingly, the UL133-UL138 locus was required for efficient replication in endothelial cells. The association of this locus with three context-dependent phenotypes suggests an exciting role for the UL133-UL138 locus in modulating the outcome of viral infection in different contexts of infection. Differential profiles of protein expression from the UL133-UL138 locus correlated with the cell-type dependent phenotypes associated with this locus. We extended our in vitro findings to analyze viral replication and dissemination in a NOD-scid IL2Rγcnull-humanized mouse model. The UL133-UL138NULL virus exhibited an increased capacity for replication and/or dissemination following stem cell mobilization relative to the wild-type virus, suggesting an important role in viral persistence and spread in the host. As pUL133, pUL135, pUL136, and pUL138 are conserved in virus strains infecting higher order primates, but not lower order mammals, the functions encoded likely represent host-specific viral adaptations

    Collaborating with consumer and community representatives in health and medical research in Australia: results from an evaluation

    <p>Abstract</p> <p>Objective</p> <p>To collaborate with consumer and community representatives in the <it>Alcohol and Pregnancy Project </it>from 2006-2008 <url>http://www.ichr.uwa.edu.au/alcoholandpregnancy</url> and evaluate researchers' and consumer and community representatives' perceptions of the process, context and impact of consumer and community participation in the project.</p> <p>Methods</p> <p>We formed two reference groups and sought consumer and community representatives' perspectives on all aspects of the project over a three year period. We developed an evaluation framework and asked consumer and community representatives and researchers to complete a self-administered questionnaire at the end of the project.</p> <p>Results</p> <p>Fifteen researchers (93.8%) and seven (53.8%) consumer and community representatives completed a questionnaire. Most consumer and community representatives agreed that the process and context measures of their participation had been achieved. Both researchers and consumer and community representatives identified areas for improvement and offered suggestions how these could be improved for future research. Researchers thought consumer and community participation contributed to project outputs and outcomes by enhancing scientific and ethical standards, providing legitimacy and authority, and increasing the project's credibility and participation. They saw it was fundamental to the research process and acknowledged consumer and community representatives for their excellent contribution. Consumer and community representatives were able to directly influence decisions about the research. They thought that consumer and community participation had significant influence on the success of project outputs and outcomes.</p> <p>Conclusions</p> <p>Consumer and community participation is an essential component of good research practice and contributed to the <it>Alcohol and Pregnancy Project </it>by enhancing research processes, outputs and outcomes, and this participation was valued by community and consumer representatives and researchers. The National Health and Medical Research Council in Australia expects researchers to work in partnership and involve consumer and community representatives in health and medical research, and to evaluate community and consumer participation. It is important to demonstrate whether consumer and community participation makes a difference to health and medical research.</p

    Childbirth and consequent atopic disease: emerging evidence on epigenetic effects based on the hygiene and EPIIC hypotheses

    Background: In most high and middle income countries across the world, at least 1:4 women give birth by cesarean section. Rates of labour induction and augmentation are rising steeply; and in some countries up to 50 % of laboring women and newborns are given antibiotics. Governments and international agencies are increasingly concerned about the clinical, economic and psychosocial effects of these interventions. Discussion: There is emerging evidence that certain intrapartum and early neonatal interventions might affect the neonatal immune response in the longer term, and perhaps trans-generationally. Two theories lead the debate in this area. Those aligned with the hygiene (or ‘Old Friends’) hypothesis have examined the effect of gut microbiome colonization secondary to mode of birth and intrapartum/neonatal pharmacological interventions on immune response and epigenetic phenomena. Those working with the EPIIC (Epigenetic Impact of Childbirth) hypothesis are concerned with the effects of eustress and dys-stress on the epigenome, secondary to mode of birth and labour interventions. Summary: This paper examines the current and emerging findings relating to childbirth and atopic/autoimmune disease from the perspective of both theories, and proposes an alliance of research effort. This is likely to accelerate the discovery of important findings arising from both approaches, and to maximize the timely understanding of the longer-term consequences of childbirth practices
