    Investigation on the influence of nematophagous fungi as feed additive on nematode infection risk of sheep and goats on pasture

    Gastrointestinal nematodes in small ruminants cause high economic losses. Thus on most farms anthelmintic treatment is required. In response to increasing problems with anthelmintic resistance, biological control, for example the use of nematophagous fungi, has received significant attention. The aim of this study was to investigate the effect of Duddingtonia flagrans orally applied to small ruminants on natural infection with gastrointestinal nematodes in a field study in Northern Germany. 20 goats and 20 sheep were fed daily for 3 months with 5x105 spores of D. flagrans per kg bodyweight. Differences in body weight, faecal egg count and larval development in faeces and on pasture in comparison with same-sized control groups were analysed. After 3 months the control goats showed significantly higher mean faecal egg count than the fungus-fed group. No significant difference was found between the two sheep groups. The maximum in larval reduction in faeces was 81.3 % in the sheep groups and 67.9 % in the goat groups (not significant). At the end of the study the body weight gain in the fungus-treated groups was 1.7 kg higher in goats and 0.7 kg higher in sheep than in the control groups (not significant). Regarding the first-year-grazing goats only, the bodyweights revealed significant differences (p<0.05). No statistically significant differences were observed in pasture larval counts. In the study presented here, no clear effect of fungus could be observed. A modified feeding regimen, perhaps with permanent release boluses or feed blocks, may improve the efficacy. Furthermore, it seems that climatic conditions during the study period could have influenced the results and displayed how sensitive the fungus application may be on such parameters

    DNA Oxidation Photoinduced by Norharmane Rhenium(I) Polypyridyl Complexes: Effect of the Bidentate N,N′-Ligands on the Damage Profile

    ReI-polypyridyl complexes have interesting and distinctive photochemical and photosensitizing properties. This work describes the capability to induce (or photoinduce) DNA damage of three ReI-complexes with a naturally occurring alkaloid called norharmane (nHo) as ligand: [Re(CO)3(nHo)(L)]CF3SO3 where L=2,2′-bipyridine (ReBpy), phenanthroline (RePhen) or dipyrido[3,2-a:2′,3′-c]phenazine (ReDppz). The interaction of the complexes with DNA was investigated by steady-state and time-resolved spectroscopy. Data show that the mode and strength of interaction depend on the chemical structure of the bidentate ligand. The complexes show a major static contribution to the overall interaction, giving rise to the formation of noncovalent adducts with DNA, and the particular trend observed was RePhen>ReDppz>ReBpy. Photo-oxidation at the purine bases represents the major DNA damaging mechanism. RePhen also induces single-strand breaks in a yield similar to that of base damage, suggesting an additional photosensitizing pathway. We also performed the Ames test to evaluate the cytotoxic and mutagenic properties of both non-irradiated and photoexcited complexes.     Urgent bedreigde typische soorten en vegetatietypen van Natura 2000-habitattypen

    Dit rapport is de weerslag van een onderzoek naar de urgent bedreigde en potentieel urgent bedreigde typische soorten en de urgent bedreigde en potentieel urgent bedreigde vegetatietypen van de habitattypen van de Natura 2000-gebieden. In totaal zijn 67 typische soorten als Urgent Bedreigd gecategoriseerd; nog eens 26 soorten zijn gecategoriseerd als Potentieel Urgent Bedreigd. Voor de vegetatietypen geldt dat er 15 (sub)associaties Urgent bedreigd zijn. Op een bijbehorende cdrom wordt voor al deze soorten en vegetaties de huidige verspreiding gegeven, of herstel binnen of buiten Natura 2000 zou moeten plaatsvinden, wat de knelpunten hierbij zijn en waar de beste herstellocaties gelegen zijn

    In photodynamic therapy (PDT), photosensitizers and light are used to cause photochemically induced cell death. The selectivity and the effectiveness of the phototoxicity in cancer can be increased by a specific uptake of the photosensitizer into tumor cells. A promising target for this goal is the folic acid receptor α (FRα), which is overexpressed on the surface of many tumor cells and mediates an endocytotic uptake. Here, we describe a polysaccharide-based nanoparticle system suitable for targeted uptake and its photochemical and photobiological characterization. The photosensitizer 5, 10, 15, 20-tetraphenyl-21H, 23H-porphyrine (TPP) was encapsulated in spermine- and acetal-modified dextran (SpAcDex) nanoparticles and conjugated with folic acid (FA) on the surface [SpAcDex(TPP)-FA]. The particles are successfully taken up by human HeLa-KB cells, and a light-induced cytotoxicity is observable. An excess of free folate as the competitor for the FRα-mediated uptake inhibits the phototoxicity. In conclusion, folate-modified SpAcDex particles are a promising drug delivery system for a tumor cell targeted photodynamic therapy

    A defective response to DNA damage is observed in several human autosomal recessive ataxias with oculomotor apraxia, including ataxia-telangiectasia. We report that senataxin, defective in ataxia oculomotor apraxia (AOA) type 2, is a nuclear protein involved in the DNA damage response. AOA2 cells are sensitive to H2O2, camptothecin, and mitomycin C, but not to ionizing radiation, and sensitivity was rescued with full-length SETX cDNA. AOA2 cells exhibited constitutive oxidative DNA damage and enhanced chromosomal instability in response to H2O2. Rejoining of H2O2-induced DNA double-strand breaks (DSBs) was significantly reduced in AOA2 cells compared to controls, and there was no evidence for a defect in DNA single-strand break repair. This defect in DSB repair was corrected by full-length SETX cDNA. These results provide evidence that an additional member of the autosomal recessive AOA is also characterized by a defective response to DNA damage, which may contribute to the neurodegeneration seen in this syndrome
