Effect of dithiocarbamate thiram on Wistar rat growth plate and articular cartilage

Avaliou-se o efeito do tirame, ditiocarbamato largamente utilizado na agricultura como antifúngico e repelente de roedores, na ossificação endocondral de mamíferos, usando, como modelo, ratos Wistar. Não foram observadas lesões na cartilagem articular, nem nas placas de crescimento, o que pode ser atribuído à dose utilizada e à duração do ensaio. A diminuição da altura da placa de crescimento nos animais aos quais foi administrado o tirame parece traduzir o atraso verificado no crescimento em geral, e não um efeito específico na cartilagem, uma vez que as diferentes zonas da placa epifisária mantiveram as proporções dos animais do grupo-controle. Embora não tenham sido verificados, no presente trabalho, os efeitos registrados para outras espécies nos tecidos cartilaginosos, sugere-se a avaliação dos efeitos crónicos do tirame no crescimento e no desenvolvimento dos ossos longos em mamíferos

EDPR asset rotation strategy : crystalizing value while accelerating company´s growth

Mestrado Bolonha em FinançasOver the last decades the concern for the environment and the development of sustainable initiatives has soared, with renewable energy playing a vital role in attaining the green goals set by international organizations and national governments for the present and future economy. With these measures, there is a big incentive to develop renewable energy projects, such as wind and solar, which has increased the predominance of these energies exponentially. Which in turn increases the necessity of investment and financing to build wind and solar projects. As such, these renewable energy companies have been undertaking a strategy called Asset Rotation (AR), which allows them to sell portions or all assets at early stages of a project to crystallize upfront value and recycle capital to invest in new valuable projects, with lower external debt burden, with this self-funding model bringing many other accounting and market rewards. In this business case will be studied the use of AR strategy at EDPR, one of the major multinational renewable energy production, development, and operation companies. To do so, the current context of the renewable energy industry will be initially addressed, as well as all the theoretical aspects of AR that make it suitable for this capital-intensive industry. Then we will focus on EDPR and its business, to understand the fundamentals of this strategy in the company, its theoretical basis, the decision process, and its advantages. The goal of this case is firstly to understand what AR means and how it works, and if this strategy is an added value to EDPR's business strategy. After a theoretical analysis of the implementation of AR in the company, and its benefits for EDPR's business, accounts and credit reputation, the conclusion was that AR is an enhancement for the company and fits with its present and future goals. Moreover, through a practical example from EDPR, it was possible to compare the outputs of a wind project embedded in the AR strategy compared to a project held till the very end of its useful life, with AR once again proving its advantage in financial terms.Nas últimas décadas, a preocupação com o ambiente e o desenvolvimento de iniciativas sustentáveis aumentou, tendo as energias renováveis desempenhado um papel vital na consecução dos objetivos verdes estabelecidos por organizações internacionais e governos nacionais para a economia presente e futura. Com estas medidas, existe um grande incentivo para desenvolver projetos de energias renováveis, tais como eólica e solar, o que aumentou exponencialmente a predominância destas energias. O que, por sua vez, aumenta a necessidade de investimento e financiamento para a construção de projetos eólicos e solares. Como tal, estas empresas de energias renováveis têm vindo a empreender uma estratégia chamada Rotatividade de Ativos (AR), que lhes permite vender partes ou todos os ativos nas fases iniciais de um projeto para cristalizar o valor inicial e reciclar capital para investir em novos projetos de valor, com menor peso da dívida externa, sendo que este modelo de autofinanciamento traz muitas outras recompensas contabilísticas e de mercado. Neste caso empresarial será estudada a utilização da estratégia AR na EDPR, uma das maiores empresas multinacionais de produção, desenvolvimento e operação de energias renováveis. Para tal, será inicialmente abordado o contexto atual da indústria das energias renováveis, bem como todos os aspetos teóricos da AR que a tornam adequada para esta indústria de capital intensivo. Depois centrar-nos-emos na EDPR e nos seus negócios, para compreender os fundamentos desta estratégia na empresa, a sua base teórica, o processo de decisão, e as suas vantagens. O objetivo deste caso é, em primeiro lugar, compreender o que significa AR e como funciona, e se esta estratégia é uma mais-valia para a estratégia empresarial da EDPR. Após uma análise teórica da implementação da AR na empresa, e dos seus benefícios para o negócio, contas e reputação creditícia da EDPR, a conclusão foi que a AR é uma melhoria para a empresa e se enquadra nos seus objetivos presentes e futuros. Com fundamento em textos do Conselheiro Cunha Rodrigues, o Autor encontra, na base da atual prevalência da jurisprudência jusfundamental do Tribunal de Justiça, o primordial estabelecimento dos princípios do primado e do efeito direto. Chama a atenção para as consequências de tipo funcionalista da jurisprudência do Tribunal de Justiça na relação entre a União e os Estados-Membros e no nascimento do conceito de cidadania da União, contribuindo para ultrapassar o que outrora ambicionou apenas ser a construção de um mercado comum. Efeitos da semicarbazida no esqueleto de ratos Wistar em crescimento.

The effects of semicarbazide hydrochloride on skeletal tissues were evaluated using Wistar rats. In the animals submitted to the administration of semicarbazide, radiological examination showed bone and articular alterations with growth plates enlargement. Histological exams showed irregular patterns and proliferation of chondrocytes and excessive accumulation of extracellular matrix. Immunohistochemical exams showed PCNA positive reaction in pre- and hypertrofic chondrocytes. These results confirm that semicarbazide induces important changes in skeletal tissues and that has potential utility in experimental osteolathryrism

Discondroplasia da tíbia como modelo em estudos de mecanobiologia experimental

A discondroplasia da tíbia, consiste numa anomalia espontânea, ou induzida, nas placas de crescimento epifisárias dos ossos longos, das estirpes de rápido crescimento das espécies avícolas, caracterizada pelo aparecimento de uma massa cartilagínea avascular opaca, não calcificada, que se estende até à metáfise, numa zona em que, normalmente, existe osso trabecular. Do ponto de vista citológico, a discondroplasia reflecte alterações na homeostase celular no decurso normal da ossificação endocondral, ou seja, a cartilagem não calcifica e não é substituída por osso endocondral. Deste modo, os processos de proliferação dos condrócitos (que é normal) e de degradação da cartilagem (que não se verifica), não estão em equilíbrio, resultando numa acumulação de matriz extracelular. Com a presente linha de investigação pretende-se, com base em resultados obtidos por um dos autores [Capela e Silva, 2004], designadamente no que diz respeito à expressão das caderinas, e com base noutros trabalhos relativos à expressão de proteínas de stress [Ribeiro e tal., 2004], avaliar da possibilidade de utilização da discondroplasia em estudos de mecanobiologia experimental

Effect of hydroxyethyl starch on acute renal injury in a model of hepatic ischemia-reperfusion

BACKGROUND: Hepatic vascular control techniques employed during liver surgery are usually associated with ischemia-reperfusion injury, which could cause acute renal dysfunction. The murine model has been used in the study of this injury. Hydroxyethyl starch has recognized anti-inflammatory properties and improves microcirculation. Third generation hydroxyethyl starches, namely 130/0.4, show a better safety profile than previous molecules. OBJECTIVES: Evaluation of renal injury in a murine model of partial normothermic hepatic ischemia-reperfusion injury and assessment of hydroxyethyl starch 130/0.4 effect on this injury. METHODS: Seventy-two male Wistar rats were randomized into six groups with identical characteristics (n = 12 x 6). In three of them, the ischemia-reperfusion injury groups, we placed a clamp in the vascular pedicle of the median and left liver lobes, inducing hepatic ischemia (70%), and removed the clamp 60 minutes later (IRI + HES and IRI + HS groups, with HES or hypertonic saline (7.5%) administration during reperfusion, respectively, and IRI group, without fluid therapy). The control groups were sham-operated without hepatic ischemia and treated likewise (sham + HES, sham + HS and sham groups). After 120 minutes of reperfusion in the ischemia-reperfusion injury groups and 180 minutes in the controls we drew blood from the aorta artery for creatinine, urea and alanine aminotransferase quantification and removed kidney and liver samples for histopathological analysis. RESULTS: As already published by our group, the partial hepatic ischemia-reperfusion injury model showed liver injury. In the present work, the IRI group had higher creatinine, urea and histopathological score than sham (p < 0.05). Creatinine and urea mean concentrations were significantly lower both in IRI+HES (23.08 µmol/L and 8.38 mmol/L, respectively) and IRI + HS (26.59 µmol/L and 7.82 mmol/L) when compared to IRI (40.101 µmol/L and 11.25 mmol/L). There was no significant difference between IRI + HES and IRI + HS groups (serum markers and histopathology).Conclusion: The hepatic ischemia-reperfusion injury murine model was effective in producing kidney injury. Both the hydroxyethyl starch 130/0.4 and the hypertonic saline protected the kidney in this context and were not harmful for this organ in the controls. Further studies are necessary to assess clinical implications of hydroxyethyl starch 130/0.4 administration in liver surgery
