    Infrared spectroscopy of solid CO-CO2 mixtures and layers

    The spectra of pure, mixed and layered CO and CO2 ices have been studied systematically under laboratory conditions using infrared spectroscopy. This work provides improved resolution spectra (0.5 cm-1) of the CO2 bending and asymmetric stretching mode, as well as the CO stretching mode, extending the existing Leiden database of laboratory spectra to match the spectral resolution reached by modern telescopes and to support the interpretation of the most recent data from Spitzer. It is shown that mixed and layered CO and CO2 ices exhibit very different spectral characteristics, which depend critically on thermal annealing and can be used to distinguish between mixed, layered and thermally annealed CO-CO2 ices. CO only affects the CO2 bending mode spectra in mixed ices below 50K under the current experimental conditions, where it exhibits a single asymmetric band profile in intimate mixtures. In all other ice morphologies the CO2 bending mode shows a double peaked profile, similar to that observed for pure solid CO2. Conversely, CO2 induces a blue-shift in the peak-position of the CO stretching vibration, to a maximum of 2142 cm-1 in mixed ices, and 2140-2146 cm-1 in layered ices. As such, the CO2 bending mode puts clear constraints on the ice morphology below 50K, whereas beyond this temperature the CO2 stretching vibration can distinguish between initially mixed and layered ices. This is illustrated for the low-mass YSO HH46, where the laboratory spectra are used to analyse the observed CO and CO2 band profiles and try to constrain the formation scenarios of CO2.Comment: Accepted in A&

    Influence of UV radiation from a massive YSO on the chemistry of its envelope

    We have studied the influence of far ultraviolet (UV) radiation from a massive young stellar object (YSO) on the chemistry of its own envelope by extending the models of Doty et al. (2002) to include a central source of UV radiation. The models are applied to the massive star-forming region AFGL 2591 for different inner UV field strengths. Depth-dependent abundance profiles for several molecules are presented and discussed. We predict enhanced column densities for more than 30 species, especially radicals and ions. Comparison between observations and models is improved with a moderate UV field incident on the inner envelope, corresponding to an enhancement factor G0~10-100 at 200 AU from the star with an optical depth tau~15-17. Subtle differences are found compared with traditional models of Photon Dominated Regions (PDRs) because of the higher temperatures and higher gas-phase H2O abundance caused by evaporation of ices in the inner region. In particular, the CN/HCN ratio is not a sensitive tracer of the inner UV field, in contrast with the situation for normal PDRs: for low UV fields, the extra CN reacts with H2 in the inner dense and warm region and produces more HCN. It is found that the CH+ abundance is strongly enhanced and grows steadily with increasing UV field. High-J lines of molecules like CN and HCN are most sensitive to the inner dense region where UV radiation plays a role. Thus, even though the total column density affected by UV photons is small, comparison of high-J and low-J lines can selectively trace and distinguish the inner UV field from the outer one.     A review of research on the effects of Early Childhood Education and Care (ECEC) upon child development. CARE project; Curriculum Quality Analysis and Impact Review of European Early Childhood Education and Care (ECEC)

    This report considers the Early Childhood Education and Care (ECEC) curriculum throughout Europe. It explores the official curriculum, specified by national or regional governments, along with the implemented curriculum that is provided ‘on the ground’ by staff to enhance children’s development. The official curriculum documents at national or regional level are often called ‘steering documents’. Moreover, the implemented curriculum is sometimes called the ‘experienced’ or the ‘realised’ curriculum, i.e., what the staff realise in their daily practice and what the children experience day by day. The CARE project has studied European curriculum in three ways: (1) by developing a template according to which the 11 partners in the CARE Consortium described the curriculum in their own countries; (2) by analysing the responses of our partners across 11 countries to the CARE curriculum template, with the aim of identifying commonalities and differences in the broadly representative sample that comprises the CARE consortium; (3) by considering information from the templates in light of selected research literature on effectiveness - NOT through a formal literature review which is the task of another Work Package in the CARE project (Melhuish et al., forthcoming) - but by comparing the template findings with widely cited, key studies. The analytic template originated as a series of questions at a curriculum conference held in Oxford (March 2014). This template was further refined as members of the CARE consortium provided information about ECEC in their home countries. The conclusions and recommendations presented in this report are based on analysis of the completed country templates (i.e. the survey of countries represented in the CARE Consortium), but also on recent EU reports and selected international literature

    Attenuated AMPA Receptor Expression Allows Glioblastoma Cell Survival in Glutamate-Rich Environment

    Background: Glioblastoma multiforme (GBM) cells secrete large amounts of glutamate that can trigger AMPA-type glutamate receptors (AMPARs). This commonly results in Na+ and Ca2+-permeability and thereby in excitotoxic cell death of the surrounding neurons. Here we investigated how the GBM cells themselves survive in a glutamate-rich environment. Methods and Findings: In silico analysis of published reports shows down-regulation of all ionotropic glutamate receptors in GBM as compared to normal brain. In vitro, in all GBM samples tested, mRNA expression of AMPAR subunit GluR1, 2 and 4 was relatively low compared to adult and fetal total brain mRNA and adult cerebellum mRNA. These findings were in line with primary GBM samples, in which protein expression patterns were down-regulated as compared to the normal tissue. Furthermore, mislocalized expression of these receptors was found. Sequence analysis of GluR2 RNA in primary and established GBM cell lines showed that the GluR2 subunit was found to be partly unedited. Conclusions: Together with the lack of functional effect of AMPAR inhibition by NBQX our results suggest that down-regulation and afunctionality of AMPARs, enable GBM cells to survive in a high glutamate environment without going into excitotoxic cell death themselves. It can be speculated that specific AMPA receptor inhibitors may protect normal neurons against the high glutamate microenvironment of GBM tumor

    A 3-5μ\mum VLT spectroscopic survey of embedded young low mass stars II; OCN^-

    The 4.62μ\mum (2164.5 cm1^{-1}) `XCN' band has been detected in the MM-band spectra of 34 deeply embedded young stellar objects (YSO's), observed with high signal-to-noise and high spectral resolution with the VLT-ISAAC spectrometer, providing the first opportunity to study the solid OCN^- abundance toward a large number of low-mass YSO's. It is shown unequivocally that at least two components, centred at 2165.7 cm1^{-1} (FWHM = 26 cm1^{-1}) and 2175.4 cm1^{-1} (FWHM = 15 cm1^{-1}), underlie the XCN band. Only the 2165.7-component can be ascribed to OCN^-, embedded in a strongly hydrogen-bonding, and possibly thermally annealed, ice environment based on laboratory OCN^- spectra. In order to correct for the contribution of the 2175.4-component to the XCN band, a phenomenological decomposition into the 2165.7- and the 2175.4-components is used to fit the full band profile and derive the OCN^- abundance for each line-of-sight. The same analysis is performed for 5 high-mass YSO's taken from the ISO-SWS data archive. Inferred OCN^- abundances are \leq 0.85 % toward low-mass YSO's and \leq 1 % toward high-mass YSO's, except for W33 A. Abundances are found to vary by at least a factor of 10--20 and large source-to-source abundance variations are observed within the same star-forming cloud complex on scales down to 400 AU, indicating that the OCN^- formation mechanism is sensitive to local conditions. The inferred abundances allow quantitatively for photochemical formation of OCN^-, but the large abundance variations are not easily explained in this scenario unless local radiation sources or special geometries are invoked. Surface chemistry should therefore be considered as an alternative formation mechanism

    IL6 and CRP haplotypes are associated with COPD risk and systemic inflammation: a case-control study

    <p>Abstract</p> <p>Background</p> <p>Elevated circulating levels of C-reactive protein (CRP), interleukin (IL)-6 and fibrinogen (FG) have been repeatedly associated with many adverse outcomes in patients with chronic obstructive pulmonary disease (COPD). To date, it remains unclear whether and to what extent systemic inflammation is primary or secondary in the pathogenesis of COPD.</p> <p>The aim of this study was to examine the association between haplotypes of <it>CRP</it>, <it>IL6 </it>and <it>FGB </it>genes, systemic inflammation, COPD risk and COPD-related phenotypes (respiratory impairment, exercise capacity and body composition).</p> <p>Methods</p> <p>Eighteen SNPs in three genes, representing optimal haplotype-tagging sets, were genotyped in 355 COPD patients and 195 healthy smokers. Plasma levels of CRP, IL-6 and FG were measured in the total study group. Differences in haplotype distributions were tested using the global and haplotype-specific statistics.</p> <p>Results</p> <p>Raised plasma levels of CRP, IL-6 and fibrinogen were demonstrated in COPD patients. However, COPD population was very heterogeneous: about 40% of patients had no evidence of systemic inflammation (CRP < 3 mg/uL or no inflammatory markers in their top quartile). Global test for haplotype effect indicated association of <it>CRP </it>gene and CRP plasma levels (P = 0.0004) and <it>IL6 </it>gene and COPD (P = 0.003). Subsequent analysis has shown that <it>IL6 </it>haplotype H2, associated with an increased COPD risk (p = 0.004, OR = 4.82; 1.64 to 4.18), was also associated with very low CRP levels (p = 0.0005). None of the genes were associated with COPD-related phenotypes.</p> <p>Conclusion</p> <p>Our findings suggest that common genetic variation in <it>CRP </it>and <it>IL6 </it>genes may contribute to heterogeneity of COPD population associated with systemic inflammation.</p

    Cost-effectiveness of healthy eating and/or physical activity promotion in pregnant women at increased risk of gestational diabetes mellitus

    __Background:__ Gestational diabetes mellitus (GDM) is associated with perinatal health risks to both mother and offspring, and represents a large economic burden. The DALI study is a multicenter randomized controlled trial, undertaken to add to the knowledge base on the effectiveness of interventions for pregnant women at increased risk for GDM. The purpose of this study was to evaluate the cost-effectiveness of the healthy eating and/or physical activity promotion intervention compared to usual care among pregnant women at increased risk of GDM from a societal perspective. __Methods:__ An economic evaluation was performed alongside a European multicenter-randomized controlled trial. A total of 435 pregnant women at increased risk of GDM in primary and secondary care settings in nine European countries, were recruited and randomly allocated to a healthy eating and physical activity promotion intervention (HE + PA intervention), a healthy eating promotion intervention (HE intervention), or a physical activity promotion intervention (PA intervention). Main outcome measures were gestational weight gain, fasting glucose, insulin resistance (HOMA-IR), quality adjusted life years (QALYs), and societal costs. __Results:__ Between-group total cost and effect differences were not significant, besides significantly less gestational weight gain in the HE + PA group compared with the usual care group at 35-37 weeks ( 2.3;95%CI:-3.7;-0.9). Cost-effectiveness acceptability curves indicated that the HE + PA intervention was the preferred intervention strategy. At 35-37 weeks, it depends on the decision-makers' willingness to pay per kilogram reduction in gestational weight gain whether the HE + PA intervention is cost-effective for gestational weight gain, whereas it was not cost-effective for fasting glucose and HOMA-IR.     Biomarkers of systemic inflammation and depression and fatigue in moderate clinically stable COPD

    <p>Abstract</p> <p>Introduction</p> <p>COPD is an inflammatory disease with major co-morbidities. It has recently been suggested that depression may be the result of systemic inflammation. We aimed to explore the association between systemic inflammation and symptoms of depression and fatigue in patients with mainly moderate and clinically stable COPD using a range of inflammatory biomarkers, 2 depression and 2 fatigue scales.</p> <p>Method</p> <p>We assessed 120 patients with moderate COPD (FEV<sub>1</sub>% 52, men 62%, age 66). Depression was assessed using the BASDEC and CES-D scales. Fatigue was assessed using the Manchester COPD-fatigue scale (MCFS) and the Borg scale before and after 6MWT. We measured systemic TNF-α, CRP, TNF-α-R1, TNF-α-R2 and IL-6.</p> <p>Results</p> <p>A multivariate linear model of all biomarkers showed that TNF-α only had a positive correlation with BASDEC depression score (p = 0.007). TNF-α remained positively correlated with depression (p = 0.024) after further adjusting for TNF-α-R1, TNF-α-R2, 6MWD, FEV<sub>1</sub>%, and pack-years. Even after adding the MCFS score, body mass and body composition to the model TNF-α was still associated with the BASDEC score (p = 0.044). Furthermore, patients with higher TNF-α level (> 3 pg/ml, n = 7) had higher mean CES-D depression score than the rest of the sample (p = 0.03). Borg fatigue score at baseline were weakly correlated with TNF-α and CRP, and with TNF-α only after 6MWT. Patients with higher TNF-α had more fatigue after 6MWD (p = 0.054).</p> <p>Conclusion</p> <p>This study indicates a possible association between TNF-α and two frequent and major co-morbidities in COPD; i.e., depression and fatigue.</p
