    An investigation of the demand for organically grown produce

    Four issues were investigated in this thesis. First, the factors which motivate organic food buying behaviour^; secondly, the reasons which cause people to avoid organics; thirdly, the cost and availability of organics in different retail outlets, and finally the opinions of retailers as to the current and future state of the organic food market. In addition, the proportion of organic buyers to non-buyers which existed among the public was estimated.Primary research was employed to survey three types of respondent. First, a national postal survey of supermarkets, wholefood shops, farm shops and greengrocers that stocked organic food was undertaken. To obtain more detailed information on the public’s attitude towards organic produce, a telephone survey of the Edinburgh and Lothian population was conducted. Finally, the particular preferences of organic food buyers were recorded by personal interviews of customers in wholefood shops around Edinburgh. Non-parametric tests, in particular chi- squared tests, were used to measure the differences between the responses of organic retailers and those between organic buyers and non-buyers.Results showed that the proportion of organic food buyers among the public in 1992 (29%) was no greater than those found by studies undertaken in 1987 and 1988. Concern for health was perceived most commonly to be the most important buying motivation for organic food by retailers, buyers and non-buyers alike, while expense was the most commonly cited non-buying reason among respondents. In general, supermarkets were found to incur greater cost when purchasing organic foods and they suffered significantly higher levels of wastage compared with wholefood shops. Yet supermarkets did enjoy a more extensive and consistent range of fresh organic produce in comparison with smaller retailers. Most retailers were optimistic about the future of the organic market and believed the high price of organic food to be the single most important barrier to market expansion.It appears that many organic purchases, ostensibly altruistic in motivation, are in fact motivated by fear or by fashion. The major non-buying reasons given by respondents fall into two types: lack of knowledge and lack of value. To encourage organic food purchases and to combat non-buying reasons, retailers must create awareness and knowledge about organics, and achieve lower prices. However, long-term expansion of the market may only be assured by legislation which is more favourable towards organic growers

    Electron Tomography of Cryofixed, Isometrically Contracting Insect Flight Muscle Reveals Novel Actin-Myosin Interactions

    BACKGROUND: Isometric muscle contraction, where force is generated without muscle shortening, is a molecular traffic jam in which the number of actin-attached motors is maximized and all states of motor action are trapped with consequently high heterogeneity. This heterogeneity is a major limitation to deciphering myosin conformational changes in situ. METHODOLOGY: We used multivariate data analysis to group repeat segments in electron tomograms of isometrically contracting insect flight muscle, mechanically monitored, rapidly frozen, freeze substituted, and thin sectioned. Improved resolution reveals the helical arrangement of F-actin subunits in the thin filament enabling an atomic model to be built into the thin filament density independent of the myosin. Actin-myosin attachments can now be assigned as weak or strong by their motor domain orientation relative to actin. Myosin attachments were quantified everywhere along the thin filament including troponin. Strong binding myosin attachments are found on only four F-actin subunits, the "target zone", situated exactly midway between successive troponin complexes. They show an axial lever arm range of 77°/12.9 nm. The lever arm azimuthal range of strong binding attachments has a highly skewed, 127° range compared with X-ray crystallographic structures. Two types of weak actin attachments are described. One type, found exclusively in the target zone, appears to represent pre-working-stroke intermediates. The other, which contacts tropomyosin rather than actin, is positioned M-ward of the target zone, i.e. the position toward which thin filaments slide during shortening. CONCLUSION: We present a model for the weak to strong transition in the myosin ATPase cycle that incorporates azimuthal movements of the motor domain on actin. Stress/strain in the S2 domain may explain azimuthal lever arm changes in the strong binding attachments. The results support previous conclusions that the weak attachments preceding force generation are very different from strong binding attachments

    Structural Changes in Isometrically Contracting Insect Flight Muscle Trapped following a Mechanical Perturbation

    The application of rapidly applied length steps to actively contracting muscle is a classic method for synchronizing the response of myosin cross-bridges so that the average response of the ensemble can be measured. Alternatively, electron tomography (ET) is a technique that can report the structure of the individual members of the ensemble. We probed the structure of active myosin motors (cross-bridges) by applying 0.5% changes in length (either a stretch or a release) within 2 ms to isometrically contracting insect flight muscle (IFM) fibers followed after 5–6 ms by rapid freezing against a liquid helium cooled copper mirror. ET of freeze-substituted fibers, embedded and thin-sectioned, provides 3-D cross-bridge images, sorted by multivariate data analysis into ∼40 classes, distinct in average structure, population size and lattice distribution. Individual actin subunits are resolved facilitating quasi-atomic modeling of each class average to determine its binding strength (weak or strong) to actin. ∼98% of strong-binding acto-myosin attachments present after a length perturbation are confined to “target zones” of only two actin subunits located exactly midway between successive troponin complexes along each long-pitch helical repeat of actin. Significant changes in the types, distribution and structure of actin-myosin attachments occurred in a manner consistent with the mechanical transients. Most dramatic is near disappearance, after either length perturbation, of a class of weak-binding cross-bridges, attached within the target zone, that are highly likely to be precursors of strong-binding cross-bridges. These weak-binding cross-bridges were originally observed in isometrically contracting IFM. Their disappearance following a quick stretch or release can be explained by a recent kinetic model for muscle contraction, as behaviour consistent with their identification as precursors of strong-binding cross-bridges. The results provide a detailed model for contraction in IFM that may be applicable to contraction in other types of muscle

    Routes to sustainability in public food procurement: An investigation of different models in primary school catering

    Increasingly, policymakers are setting ambitious goals for sustainability in public procurement, integrated across different pillars. Such ambitions are apparent in public catering services, where procurement models have been shifting towards greater localisation of supply chains and purchasing of more organically grown food. To date however, few studies have examined empirically what the impacts of different procurement models are across these multiple pillars of sustainability. This research aimed to fill the gap, by measuring and comparing the environmental, economic and nutritional outcomes of different models of school meals procurement. Case studies were undertaken of ten primary school meals services in five European countries, capturing different procurement model types. Results showed carbon emissions ranged from 0.95 kgs CO2e per meal in the lowest case to 2.41 kgs CO2e in the highest case, with adoption of low carbon food waste disposal methods and reduction of the amount of ruminant meat in the menus being the most important actions for lowering emissions. In terms of economic impact, local economic multiplier ratios ranged from 1.59 to 2.46, and although the level of local food sourcing contributed to these ratios, the effect was eclipsed, in some cases, by investment in local catering staff. Meanwhile, implementation of a robust standards regime and improving canteen environment and supervision were the most important actions for nutritional quality and intake.     Improving access to large-scale digital libraries through semantic-enhanced search and disambiguation

    With 13,000,000 volumes comprising 4.5 billion pages of text, it is currently very difficult for scholars to locate relevant sets of documents that are useful in their research from the HathiTrust Digital Libary (HTDL) using traditional lexically-based retrieval techniques. Existing document search tools and document clustering approaches use purely lexical analysis, which cannot address the inherent ambiguity of natural language. A semantic search approach offers the potential to overcome the shortcoming of lexical search, but even if an appropriate network of ontologies could be decided upon it would require a full semantic markup of each document. In this paper, we present a conceptual design and report on the initial implementation of a new framework that affords the benefits of semantic search while minimizing the problems associated with applying existing semantic analysis at scale. Our approach avoids the need for complete semantic document markup using pre-existing ontologies by developing an automatically generated Concept-in-Context (CiC) network seeded by a priori analysis of Wikipedia texts and identification of semantic metadata. Our Capisco system analyzes documents by the semantics and context of their content. The disambiguation of search queries is done interactively, to fully utilize the domain knowledge of the scholar. Our method achieves a form of semantic-enhanced search that simultaneously exploits the proven scale benefits provided by lexical indexing

    The green halo: Mechanisms and limits of the eco-label effect

    Consumers believe that “eco-labeled” products taste better, which, at least in part, may be an effect of the label. The purpose of the current series of experiments was to examine some mechanisms and limits of this eco-label effect. In Experiment 1, an eco-label effect of similar magnitude was found for taste ratings of both conventional and organic bananas. Experiment 2 showed eco-label effects for a wider range of judgmental dimensions (i.e., health, calories, vitamins/minerals, mental performance, and willingness to pay) and the effect was about the same in magnitude for judgments of grapes and raisins. Experiment 3, with water as the tasted product, found no eco-label effect on judgments of taste, calories and vitamins/minerals, but an effect on willingness to pay, judgments of health benefits and judgments of mental performance benefits. Experiments 2 and 3 also included questionnaires on social desirability traits, schizotypal traits and pro-environmental consumer traits. The last was the strongest predictor of the eco-label effect amongst the three. In all, the eco-label effect is a robust phenomenon, but depends on interactions between product type and judgmental dimension. Implications for several accounts of the effect are discussed

    Small Molecule Inhibited Parathyroid Hormone Mediated cAMP Response by N–Terminal Peptide Binding

    Ligand binding to certain classes of G protein coupled receptors (GPCRs) stimulates the rapid synthesis of cAMP through G protein. Human parathyroid hormone (PTH), a member of class B GPCRs, binds to its receptor via its N–terminal domain, thereby activating the pathway to this secondary messenger inside cells. Presently, GPCRs are the target of many pharmaceuticals however, these drugs target only a small fraction of structurally known GPCRs (about 10%). Coordination complexes are gaining interest due to their wide applications in the medicinal field. In the present studies we explored the potential of a coordination complex of Zn(II) and anthracenyl–terpyridine as a modulator of the parathyroid hormone response. Preferential interactions at the N–terminal domain of the peptide hormone were manifested by suppressed cAMP generation inside the cells. These observations contribute a regulatory component to the current GPCR–cAMP paradigm, where not the receptor itself, but the activating hormone is a target. To our knowledge, this is the first report about a coordination complex modulating GPCR activity at the level of deactivating its agonist. Developing such molecules might help in the control of pathogenic PTH function such as hyperparathyroidism, where control of excess hormonal activity is essentially required
