The sparse component analysis framework is a powerful method for achieving this. Sparse orthogonal transforms, in which only few transform coefficients differ significantly from zero, are developed; once the signal has been transformed, energy is apportioned from each transform coefficient to each estimated source, and, finally, the signal is reconstructed using the inverse transform. The overriding aim of this chapter is to demonstrate how this framework, as exemplified here by two different decomposition methods which adapt to the signal to represent it sparsely, can be used to solve different problems in different mixing scenarios. To address the instantaneous (neither delays nor echoes) and underdetermined (more sources than mixtures) mixing model, a lapped orthogonal transform is adapted to the signal by selecting a basis from a library of predetermined bases. This method is highly related to the windowing methods used in the MPEG audio coding framework. In considering the anechoic (delays but no echoes) and determined (equal number of sources and mixtures) mixing case, a greedy adaptive transform is used based on orthogonal basis functions that are learned from the observed data, instead of being selected from a predetermined library of bases. This is found to encode the signal characteristics, by introducing a feedback system between the bases and the observed data. Experiments on mixtures of speech and music signals demonstrate that these methods give good signal approximations and separation performance, and indicate promising directions for future research
Survey on the use of CAD-CAM technology by UK and Irish dental technicians
Statement of the problem Digital workflows (CAD/CAM) have been introduced in dentistry during recent years. No published information exists on dental technicians' use and reporting of this technology.Purpose The aim of this cross sectional survey was to identify the extent digital technology has infiltrated the workplace and to investigate the factors affecting the use of CAD-CAM technology by dental laboratory technicians within Ireland and the UK.Materials and methods A web-based questionnaire was composed (Opinio, Object Planet Inc. Oslo, Norway) and distributed to UK and Irish dental technicians. Answers to all questions were anonymous and grouped such that general information was gathered initially, followed by branching of the survey into two sections depending on whether or not the respondent worked with CAD-CAM technology. Results were compiled and statistical analysis (Fisher's Exact test, SPSS, IBM, Armonk, New York, USA) was performed in order to investigate any correlation between various demographic variables and the answers provided.Results The survey was distributed to 760 UK technicians and 77 Irish technicians. The total number of completed surveys was 105, which yielded a total response rate of 14%. Most technicians reported using some form of CAD/CAM aspect in the workflow, and this was more significant for technicians working in large laboratories. Most training received was company-led. Large laboratories were also significantly correlated with less outsourcing of CAD/CAM work and a change in dental material use leading to the increase of zirconia and the decrease of noble alloys. Dental technicians did not report any significant change in working relationships and staffing as a result of CAD/CAM incorporation. High initial investment cost was the most common reason quoted from non-users, along with the lack of such technology in their working environment

Current resource management mechanisms and policies are inadequate for future multicore systems. Instead, a hardware/software interface based on the virtual private machine abstraction would allow software policies to explicitly manage microarchitecture resources. VPM policies, implemented primarily in software, translate application and system objectives into VPM resource assignments. Spatiotemporal variation in house spider phenology at a national scale using citizen science
The seasonal appearance of Tegenaria and Eratigena (the best known of the UK genera termed house spiders) results in considerable public and media interest. Here, we present the largest dataset ever gathered on the occurrence of house spiders anywhere in the world. We collected almost 10,000 records from different locations within the UK (amounting to âŒ250Ă more locations and 25Ă more records than any previous study) over a six-month period. Using this dataset, which contained details of sighting dates, times, location within UK, location within the home, location within rooms, and sex, we were able to investigate a number of aspects of house spider ecology. Eightytwo percent of records were males, supporting previous studies that showed house spider surges in autumn are predominantly males seeking mates. Sightings peaked in mid-September with a significant northwest progression across the UK as autumn progressed. Daily activity peaked at 19.35 hrs and spiders were seen more or less uniformly throughout different rooms; we discuss why this is more likely to be due to spider ecology than human behaviour. Within rooms, there was a sex-based difference in ecology with females more common on ceilings and doors/windows and males more common on walls, possibly because of sex-specific differences in mobility
CRISPR/Cas9 DNA cleavage at SNP-derived PAM enables both in vitro and in vivo KRT12 mutation-specific targeting
CRISPR/Cas9-based therapeutics hold the possibility for permanent treatment of genetic disease. The potency and specificity of this system has been used to target dominantly inherited conditions caused by heterozygous missense mutations through inclusion of the mutated base in the short-guide RNA (sgRNA) sequence. This research evaluates a novel approach for targeting heterozygous single-nucleotide polymorphisms (SNPs) using CRISPR/Cas9. We determined that a mutation within KRT12, which causes Meesmann's epithelial corneal dystrophy (MECD), leads to the occurrence of a novel protospacer adjacent motif (PAM). We designed an sgRNA complementary to the sequence adjacent to this SNP-derived PAM and evaluated its potency and allele specificity both in vitro and in vivo. This sgRNA was found to be highly effective at reducing the expression of mutant KRT12 mRNA and protein in vitro. To assess its activity in vivo we injected a combined Cas9/sgRNA expression construct into the corneal stroma of a humanized MECD mouse model. Sequence analysis of corneal genomic DNA revealed non-homologous end-joining repair resulting in frame-shifting deletions within the mutant KRT12 allele. This study is the first to demonstrate in vivo gene editing of a heterozygous disease-causing SNP that results in a novel PAM, further highlighting the potential for CRISPR/Cas9-based therapeutics
Childrenâs play and independent mobility in 2020: results from the British Childrenâs Play Survey
The British Childrenâs Play Survey was conducted in April 2020 with a nationally representative sample of 1919 parents/caregivers with a child aged 5â11 years. Respondents completed a range of measures focused on childrenâs play, independent mobility and adult tolerance of and attitudes towards risk in play. The results show that, averaged across the year, children play for around 3 h per day, with around half of childrenâs play happening outdoors. Away from home, the most common places for children to play are playgrounds and green spaces. The most adventurous places for play were green spaces and indoor play centres. A significant difference was found between the age that children were reported to be allowed out alone (10.74 years; SD = 2.20 years) and the age that their parents/caregivers reported they had been allowed out alone (8.91 years; SD = 2.31 years). A range of socio-demographic factors were associated with childrenâs play. There was little evidence that geographical location predicted childrenâs play, but it was more important for independent mobility. Further, when parents/caregivers had more positive attitudes around childrenâs risk-taking in play, children spent more time playing and were allowed to be out of the house independently at a younger age

Adventurous play for a healthy childhood: facilitators and barriers identified by parents in Britain
Rationale
Adventurous play, where children take age-appropriate risks involving uncertainty, fear, and thrill, is positively associated with children's physical health, mental health, and development. There is growing concern that children's access to and engagement with adventurous play opportunities are declining in Westernised countries, which may have negative implications for children's health.
Objective
The current study aimed to ascertain the facilitators of and barriers to children's adventurous play most identified by parents in Britain and to determine whether these differ across socio-demographic and geographic groups.
Methods
This study analysed the responses of a nationally representative sample of 1919 parents who took part in the British Children's Play Survey. Two open-ended questions asked parents to identify what they perceive to be the facilitators of and barriers to their child's adventurous play. A quantitative coding scheme, developed using the qualitative framework identified by Oliver et al. (2022), was applied to parents' responses.
Results
A diversity in the most identified facilitators and barriers was found, including concerns about the risk of injury from adventurous play and the safety of society, positive attitudes about the benefits of adventurous play, as well as factors related to child attributes. In general, these were consistently identified across different socio-demographic and geographic groups, although some differences were found in barriers.
Conclusions
The findings of this research support the identification of key targets for those working with parents to improve children's adventurous play opportunities and ultimately their physical and mental health. Future research should seek to design and tailor interventions by asking parents about the support they would value

Parent perceived barriers and facilitators of childrenâs adventurous play in Britain: a framework analysis
Abstract
Background: From a public health perspective there is growing interest in childrenâs play, including play involving risk and adventure, in relation to childrenâs physical and mental health. Regarding mental health, it is theorised that adventurous play, where children experience thrilling, exciting emotions, offers important learning opportunities that prepare children for dealing with uncertainty and help prevent anxiety. Despite these benefits, adventurous play has decreased substantially within a generation. Parents have a key role in facilitating or limiting childrenâs opportunities for adventurous play, but research identifying the barriers and facilitators parents perceive in relation to adventurous play is scarce. The present study therefore examined the barriers to and facilitators of adventurous play as perceived by parents of school-aged children in Britain.
Methods: This study analysed data from a subsample of parents in Britain (n=377) who participated in the nationally representative British Childrenâs Play Survey. Parents responded to two open-ended questions pertaining to the barriers to and facilitators of childrenâs adventurous play. Responses were analysed using a qualitative Framework Analysis, an approach suitable for managing large datasets with specific research questions.
Results: Four framework categories were identified: Social Environment; Physical Environment; Risk of Injury; Child Factors. Social Environment included barriers and facilitators related to parents, family and peers, as well as community and society. Dominant themes within the Social Environment related to perceptions about the certainty of child safety, such as supervision and the safety of society. Beliefs about the benefits of adventurous play for development and well-being were also important in the Social Environment. Physical Environment factors focused on safety and practical issues. Risk of Injury captured concerns about children being injured during play. Child Factors included child attributes, such as play preference, developmental ability and trait-like characteristics.
Conclusions: Improved understanding of what influences parent perceptions of adventurous play can inform public health interventions designed to improve childrenâs opportunities for and engagement in adventurous play, with a view to promote childrenâs physical and mental health.
