    MPA_Pathway_Tool: User-Friendly, Automatic Assignment of Microbial Community Data on Metabolic Pathways

    Taxonomic and functional characterization of microbial communities from diverse environments such as the human gut or biogas plants by multi-omics methods plays an ever more important role. Researchers assign all identified genes, transcripts, or proteins to biological pathways to better understand the function of single species and microbial communities. However, due to the versality of microbial metabolism and a still-increasing number of newly biological pathways, linkage to standard pathway maps such as the KEGG central carbon metabolism is often problematic. We successfully implemented and validated a new user-friendly, stand-alone web application, the MPA_Pathway_Tool. It consists of two parts, called ‘Pathway-Creator’ and ‘Pathway-Calculator’. The ‘Pathway-Creator’ enables an easy set-up of user-defined pathways with specific taxonomic constraints. The ‘Pathway-Calculator’ automatically maps microbial community data from multiple measurements on selected pathways and visualizes the results. The MPA_Pathway_Tool is implemented in Java and ReactJS

    Effects of telmisartan and ramipril on adiponectin and blood pressure in patients with type 2 diabetes

    <b>Background:</b> Adiponectin is secreted by adipose tissue and may play a role in cardiovascular disease. We examined adiponectin levels in patients with type 2 diabetes who participated in the Telmisartan vs. Ramipril in Renal Endothelial Dysfunction (TRENDY) study. <b>Methods</b> A total of 87 patients were assessed at baseline and following 9 weeks treatment with the angiotensin-receptor blocker telmisartan (final dose, 80 mg; n = 45) or the angiotensin-converting enzyme inhibitor ramipril (final dose, 10 mg; n = 42). Adiponectin levels were measured in plasma by radioimmunoassay. <b>Results:</b> Adiponectin levels were inversely correlated with systolic (SBP; r = -0.240, P < 0.05) and diastolic (DBP; r = -0.227, P < 0.05) blood pressure at baseline and following treatment with telmisartan or ramipril (SBP: r = -0.228, P < 0.05; DBP: r = -0.286, P < 0.05). Changes in adiponectin levels were related to changes in SBP (r = -0.357, P < 0.01) and DBP (r = -0.286, P < 0.01). There was a significant increase in adiponectin levels in the telmisartan (0.68 (95% confidence interval (CI), 0.27 to 1.10) <sup>µ</sup>g/ml, P < 0.01) but not in the ramipril group (0.17 (95% CI, -0.56 to 0.90) <sup>µ</sup>g/ml, P = 0.67). Blood pressure reduction in the telmisartan group (DeltaSBP: -13.5 (95% CI, -17.0 to -10.0) mm Hg; ΔDBP: -7.6 (95% CI, -9.8 to -5.3) mm Hg, each P < 0.001) was significantly (P less than or equal to 0.01 for SBP and P < 0.01 for DBP) greater than in the ramipril group (ΔSBP: -6.1 (95% CI, -6.2 to -2.0) mm Hg; ΔDBP: -2.7 (95% CI, -5.0 to -0.5) mm Hg; P < 0.01 and P < 0.05, respectively). <b>Conclusion:</b> Adiponectin is correlated with blood pressure in patients with type 2 diabetes. Whether increased adiponectin contributes to the blood pressure–lowering effect of telmisartan needs further study

    Degradation Kinetics of Lignocellulolytic Enzymes in a Biogas Reactor Using Quantitative Mass Spectrometry

    The supplementation of lignocellulose-degrading enzymes can be used to enhance the performance of biogas production in industrial biogas plants. Since the structural stability of these enzyme preparations is essential for efficient application, reliable methods for the assessment of enzyme stability are crucial. Here, a mass-spectrometric-based assay was established to monitor the structural stability of enzymes, i.e., the structural integrity of these proteins, in anaerobic digestion (AD). The analysis of extracts of Lentinula edodes revealed the rapid degradation of lignocellulose-degrading enzymes, with an approximate half-life of 1.5 h. The observed low structural stability of lignocellulose-degrading enzymes in AD corresponded with previous results obtained for biogas content. The established workflow can be easily adapted for the monitoring of other enzyme formulations and provides a platform for evaluating the effects of enzyme additions in AD, together with a characterization of the biochemical methane potential used in order to determine the biodegradability of organic substrates

    Fecal Metaproteomics Reveals Reduced Gut Inflammation and Changed Microbial Metabolism Following Lifestyle-Induced Weight Loss

    Gut microbiota-mediated inflammation promotes obesity-associated low-grade inflammation, which represents a hallmark of metabolic syndrome. To investigate if lifestyle-induced weight loss (WL) may modulate the gut microbiome composition and its interaction with the host on a functional level, we analyzed the fecal metaproteome of 33 individuals with metabolic syndrome in a longitudinal study before and after lifestyle-induced WL in a well-defined cohort. The 6-month WL intervention resulted in reduced BMI (−13.7%), improved insulin sensitivity (HOMA-IR, −46.1%), and reduced levels of circulating hsCRP (−39.9%), indicating metabolic syndrome reversal. The metaprotein spectra revealed a decrease of human proteins associated with gut inflammation. Taxonomic analysis revealed only minor changes in the bacterial composition with an increase of the families Desulfovibrionaceae, Leptospiraceae, Syntrophomonadaceae, Thermotogaceae and Verrucomicrobiaceae. Yet we detected an increased abundance of microbial metaprotein spectra that suggest an enhanced hydrolysis of complex carbohydrates. Hence, lifestyle-induced WL was associated with reduced gut inflammation and functional changes of human and microbial enzymes for carbohydrate hydrolysis while the taxonomic composition of the gut microbiome remained almost stable. The metaproteomics workflow has proven to be a suitable method for monitoring inflammatory changes in the fecal metaproteome

    Trollthrottle -- Raising the Cost of Astroturfing

    Astroturfing, i.e., the fabrication of public discourse by private or state-controlled sponsors via the creation of fake online accounts, has become incredibly widespread in recent years. It gives a disproportionally strong voice to wealthy and technology-savvy actors, permits targeted attacks on public forums and could in the long run harm the trust users have in the internet as a communication platform. Countering these efforts without deanonymising the participants has not yet proven effective; however, we can raise the cost of astroturfing. Following the principle `one person, one voice', we introduce Trollthrottle, a protocol that limits the number of comments a single person can post on participating websites. Using direct anonymous attestation and a public ledger, the user is free to choose any nickname, but the number of comments is aggregated over all posts on all websites, no matter which nickname was used. We demonstrate the deployability of Trollthrottle by retrofitting it to the popular news aggregator website Reddit and by evaluating the cost of deployment for the scenario of a national newspaper (168k comments per day), an international newspaper (268k c/d) and Reddit itself (4.9M c/d)

    Purification and characterization of hydroquinone dioxygenase from Sphingomonas sp. strain TTNP3

    Hydroquinone-1,2-dioxygenase, an enzyme involved in the degradation of alkylphenols in Sphingomonas sp. strain TTNP3 was purified to apparent homogeneity. The extradiol dioxygenase catalyzed the ring fission of hydroquinone to 4-hydroxymuconic semialdehyde and the degradation of chlorinated and several alkylated hydroquinones. The activity of 1 mg of the purified enzyme with unsubstituted hydroquinone was 6.1 ÎŒmol per minute, the apparent Km 2.2 ÎŒM. ICP-MS analysis revealed an iron content of 1.4 moles per mole enzyme. The enzyme lost activity upon exposure to oxygen, but could be reactivated by Fe(II) in presence of ascorbate. SDS-PAGE analysis of the purified enzyme yielded two bands of an apparent size of 38 kDa and 19 kDa, respectively. Data from MALDI-TOF analyses of peptides of the respective bands matched with the deduced amino acid sequences of two neighboring open reading frames found in genomic DNA of Sphingomonas sp strain TTNP3. The deduced amino acid sequences showed 62% and 47% identity to the large and small subunit of hydroquinone dioxygenase from Pseudomonas fluorescens strain ACB, respectively.     Perilaku deteksi dini kanker leher rahim dengan metode IVA adalah bentuk tindakan pemeriksaan deteksi dini kanker leher rahim dengan metode IVA. Kesadaran perempuan Indonesia untuk melakukan deteksi kanker leher rahim secara teratur masih rendah. Hal ini terlihat dari rendahnya cakupan deteksi dini kanker leher rahim dengan metode Inspeksi Visual Asam Asetat. Untuk tingkat Jawa Tengan hanya 1,08% dari jumlah wanita usia 30-50 tahun, Puskesmas Candiroto sekitar 5%. Tujuan penelitian ini adalah menganalisis faktor yang berhubungan dengan perilaku deteksi dini kanker leher rahim metode Inspeksi Visual Asam Asetat di Puskesmas Candiroto Kabupaten Temanggung. Penelitian ini merupakan penelitian dengan pendekatan cross sectional dengan analisis data uji chi-square dengan taraf signifikansi 95%. Subjek penelitian ini adalah wanita usia subur di wilayakh kerja Puskesmas Candiroto sejumlah 100 orang dengan menggunakan simple random sampling. Hasil uji chi-square ada hubungan antara keterpaparan informasi(p=0,1), dukungan suami (p=0,026), dan dukungan teman (p<0,0001) dengan perilaku deteksi dini kanker leher rahim metode IVA, serta tidak ada hubungan antara umur (p=0,07), pendidikan (p=0,17), pekerjaan (p=0,51), pengetahuan ((p=0,36), sikap (p=0,097), jarak (p=0,1), dukungan tenaga kesehatan (p=0,1) dengan perilaku deteksi dini kanker leher rahim metode IVA. dapat disimpulkan faktor yang berhubungan dengan perilaku deteksi dini kanker leher rahim metode Inspeksi Visual Asam Asetat di Puskesmas Candiroto Kabupaten Temanggung yaitu keterpaparan informasi, dukungan suami, dan dukungan teman.
