Developing a biodiversity-based indicator for large-scale environmental assessment: a case study of proposed shale gas extraction sites in Britain
1. Environmental impact assessments are important tools for predicting the consequences of development and changes in land use. These assessments generally use a small subset of total biodiversity – typically rare and threatened species and habitats – as indicators of ecological status. However, these indicators do not necessarily reflect changes in the many more widespread (but increasingly threatened) species, which are important for ecosystem functions. In addition, assessment of threatened species through field surveys is time-consuming and expensive and, therefore, only possible at small spatial scales. In contrast, planning changes in land use over large spatial scales (e.g. national infrastructure projects) require assessment and prioritization of biodiversity over large spatial extents.
2. Here, we provide a method for the assessment of biodiversity, which takes account of species diversity across larger spatial scales, based on occurrence records from 5553 species across 11 taxonomic groups. We compare the efficacy of the biodiversity-based indicator we developed against one based on threatened species only and then use it to consider spatial and temporal patterns in ecological status across Great Britain. Finally, we develop a case study to investigate biodiversity status in regions proposed for shale gas extraction in Great Britain.
3. Our results show a strong relationship between the ecological status of areas defined by all biodiversity versus only threatened species, although they also demonstrate that significant exceptions do exist where threatened species do not always accurately indicate the ecological status of wider biodiversity.
4. Spatial and temporal analyses show large variation in ecological status across Great Britain both within the area made available for shale gas licensing and within individual environmental zones. In total, however, 63% of hectads across Britain have suffered a net reduction in our biodiversity-based indicator since 1970.
5. Synthesis and applications. We provide a method and develop a biodiversity-based indicator for the assessment and prioritization of biodiversity at large spatial scales. We highlight the potential applications of this approach for the prioritization of areas that would benefit from conservation and restoration. We also emphasize the danger of insufficient consideration of more widespread species and not just rare and threatened species and habitats as indicators of ecological status when prioritizing large-scale national infrastructure projects. Our method should be a useful tool to complement existing environmental impact assessment methods
Measurement of gut permeability using fluorescent tracer agent technology
Abstract The healthy gut restricts macromolecular and bacterial movement across tight junctions, while increased intestinal permeability accompanies many intestinal disorders. Dual sugar absorption tests, which measure intestinal permeability in humans, present challenges. Therefore, we asked if enterally administered fluorescent tracers could ascertain mucosal integrity, because transcutaneous measurement of differentially absorbed molecules could enable specimen-free evaluation of permeability. We induced small bowel injury in rats using high- (15 mg/kg), intermediate- (10 mg/kg), and low- (5 mg/kg) dose indomethacin. Then, we compared urinary ratios of enterally administered fluorescent tracers MB-402 and MB-301 to urinary ratios of sugar tracers lactulose and rhamnose. We also tested the ability of transcutaneous sensors to measure the ratios of absorbed fluorophores. Urinary fluorophore and sugar ratios reflect gut injury in an indomethacin dose dependent manner. The fluorophores generated smooth curvilinear ratio trajectories with wide dynamic ranges. The more chaotic sugar ratios had narrower dynamic ranges. Fluorophore ratios measured through the skin distinguished indomethacin-challenged from same day control rats. Enterally administered fluorophores can identify intestinal injury in a rat model. Fluorophore ratios are measureable through the skin, obviating drawbacks of dual sugar absorption tests. Pending validation, this technology should be considered for human use
Role for SUR2A ED Domain in Allosteric Coupling within the KATP Channel Complex
Allosteric regulation of heteromultimeric ATP-sensitive potassium (KATP) channels is unique among protein systems as it implies transmission of ligand-induced structural adaptation at the regulatory SUR subunit, a member of ATP-binding cassette ABCC family, to the distinct pore-forming K+ (Kir6.x) channel module. Cooperative interaction between nucleotide binding domains (NBDs) of SUR is a prerequisite for KATP channel gating, yet pathways of allosteric intersubunit communication remain uncertain. Here, we analyzed the role of the ED domain, a stretch of 15 negatively charged aspartate/glutamate amino acid residues (948–962) of the SUR2A isoform, in the regulation of cardiac KATP channels. Disruption of the ED domain impeded cooperative NBDs interaction and interrupted the regulation of KATP channel complexes by MgADP, potassium channel openers, and sulfonylurea drugs. Thus, the ED domain is a structural component of the allosteric pathway within the KATP channel complex integrating transduction of diverse nucleotide-dependent states in the regulatory SUR subunit to the open/closed states of the K+-conducting channel pore
Rhamnose is superior to mannitol as a monosaccharide in the dual sugar absorption test: A prospective randomized study in children with treatment-naïve celiac disease
BACKGROUND AND AIM: We sought to correlate two different measures of gut permeability [lactulose:mannitol (L:M) and lactulose:rhamnose (L:R)] to the severity of duodenal histopathology in children with and without elevated antibodies to tissue transglutaminase (tTG). A secondary objective was to correlate gut permeability with celiac disease (CD) serology and indices of inflammation and bacterial product translocation.

CONCLUSIONS: L:R, but not L:M, is associated with modified Marsh scores in children undergoing small bowel biopsy for suspected CD. Despite increased intestinal permeability, we see scant evidence of systemic exposure to gut microbes in these children. Gut permeability testing with L:R may predict which patients with abnormal celiac serology will have biopsy evidence for celiac disease and reduce the proportion of such patients undergoing endoscopy whose Marsh scores are ≤1. M should not be used as a monosaccharide for permeability testing in children
A pragmatic approach to resolving technological unfairness: The case of Nike’s Vaporfly & Alphafly running footwear
Background
Technology is often introduced into sport to facilitate it or to improve human performance within it. On occasion, some forms of novel technology require regulation or prevention entirely to ensure that a sport remains fair and accessible. Recently, the Nike Vaporfly and Alphafly shoes have received some concerns over their appropriateness for use in competitive distance running.
Methods
This paper evaluates the use of these shoes against an existing framework for sports technology discourse and adopts a pragmatic approach to attempt to resolve them.
Results
It is proposed that the three concerns regarding cost, access and coercion cannot be ruled out but likely remain short term issues. As a result, it is proposed that these running shoes are acceptable forms of technology but that ongoing vigilance will be required as such technologies develop further in the future.
Conclusions
The Nike Vaporfly/Alphafly shoes do push the perceived acceptability of running shoes to the limits of the current sports regulations. However, the alleged gains have not manifested themselves to a level that could be considered excessive when reviewing historical performances or when evaluated against a set of well-cited criteria. The sport will need to adopt a stance of ongoing vigilance as such technologies continue to develop or be optimised in the future

Interlaboratory Reproducibility of Contour Method Data in a High Strength Aluminum Alloy
Background
The contour method for residual stress measurement has seen significant development, but an experimental reproducibility study utilizing physical samples has not been published.
Objective
A double-blind reproducibly study is reported, having scope beginning with EDM cutting and ending with residual stress calculation.
Methods
A reinforced I-beam sample geometry is identified for its unique residual stress profile when extracted from residual stress bearing quenched aluminum bar (7050-T74). Contour measurements are prescribed on a midplane of symmetry with dimensions 24.0 mm by 50.0 mm. Fourteen identically prepared samples are fabricated from a single long bar with well characterized and uniform residual stress. Five samples throughout the bar are identified for planning measurements to validate sample uniformity and overall suitability of the residual stress field. The planning measurements employ a range of techniques: contour method, neutron diffraction, and hole-drilling. Eight samples are distributed to an international group of participants to execute their standard measurement practice. A double-blind process is followed to provide anonymity.
Results
Results are provided by eight participants: six being self-similar and two being quite different, the latter set aside as outliers. An average residual stress field is established from non-outlying results and the spatial distribution of reproducibility standard deviation is determined. The average stress field ranges from -60 to 70 MPa and the reproducibility standard deviation averages 8.1 MPa on the measurement plane. The average reproducibility standard deviation is about 3 × larger for points within 1.0 mm of plane boundaries (17.6 MPa) than for the remaining points (6.1 MPa).
Conclusions
Reproducibility standard deviation (among different labs) for contour method residual stress measurement is found to be very similar to repeatability standard deviation (in a single lab) reported in prior work. The reproducibility observed here, for the entire measurement process, is also similar to that found in a prior reproducibility study limited to contour method data analysis
Monitoring international migration flows in Europe. Towards a statistical data base combining data from different sources
The paper reviews techniques developed in demography, geography and statistics that are useful for bridging the gap between available data on international migration flows and the information required for policy making and research. The basic idea of the paper is as follows: to establish a coherent and consistent data base that contains sufficiently detailed, up-to-date and accurate information, data from several sources should be combined. That raises issues of definition and measurement, and of how to combine data from different origins properly. The issues may be tackled more easily if the statistics that are being compiled are viewed as different outcomes or manifestations of underlying stochastic processes governing migration. The link between the processes and their outcomes is described by models, the parameters of which must be estimated from the available data. That may be done within the context of socio-demographic accounting. The paper discusses the experience of the U.S. Bureau of the Census in combining migration data from several sources. It also summarizes the many efforts in Europe to establish a coherent and consistent data base on international migration.

Weighed scalar averaging in LTB dust models, part I: statistical fluctuations and gravitational entropy
We introduce a weighed scalar average formalism ("q-average") for the study
of the theoretical properties and the dynamics of spherically symmetric
Lemaitre-Tolman-Bondi (LTB) dust models models. The "q-scalars" that emerge by
applying the q-averages to the density, Hubble expansion and spatial curvature
(which are common to FLRW models) are directly expressible in terms of
curvature and kinematic invariants and identically satisfy FLRW evolution laws
without the back-reaction terms that characterize Buchert's average. The local
and non-local fluctuations and perturbations with respect to the q-average
convey the effects of inhomogeneity through the ratio of curvature and
kinematic invariants and the magnitude of radial gradients. All curvature and
kinematic proper tensors that characterize the models are expressible as
irreducible algebraic expansions on the metric and 4-velocity, whose
coefficients are the q-scalars and their linear and quadratic local
fluctuations. All invariant contractions of these tensors are quadratic
fluctuations, whose q-averages are directly and exactly related to statistical
correlation moments of the density and Hubble expansion scalar. We explore the
application of this formalism to a definition of a gravitational entropy
functional proposed by Hosoya et al (2004 Phys. Rev. Lett. 92 141302). We show
that a positive entropy production follows from a negative correlation between
fluctuations of the density and Hubble scalar, providing a brief outline on its
fulfillment in various LTB models and regions. While the q-average formalism is
specially suited for LTB and Szekeres models, it may provide a valuable
theoretical insight on the properties of scalar averaging in inhomogeneous

The rank reversal problem in multi-criteria decision making : a literature review
Despite the importance of multicriteria decision-making (MCDM) techniques for constructing effective decision models, there are many criticisms due to the occurrence of a problem called rank reversal. Nevertheless, there is a lack of a systematic literature review on this important subject which involves different methods. This study reviews the pertinent literature on rank reversal, based on 130 related articles published from 1980 to 2015 in international journals, which were gathered and analyzed according to the following perspectives: multicriteria technique, year and journal in which the papers were published, co-authorship network, rank reversal types, and research goal. Thus our survey provides recommendations for future research, besides useful information and knowledge regarding rank reversal in the MCDM field
Functional KV10.1 Channels Localize to the Inner Nuclear Membrane
Ectopically expressed human KV10.1 channels are relevant players in tumor biology. However, their function as ion channels at the plasma membrane does not totally explain their crucial role in tumors. Both in native and heterologous systems, it has been observed that a majority of KV10.1 channels remain at intracellular locations. In this study we investigated the localization and possible roles of perinuclear KV10.1. We show that KV10.1 is expressed at the inner nuclear membrane in both human and rat models; it co-purifies with established inner nuclear membrane markers, shows resistance to detergent extraction and restricted mobility, all of them typical features of proteins at the inner nuclear membrane. KV10.1 channels at the inner nuclear membrane are not all transported directly from the ER but rather have been exposed to the extracellular milieu. Patch clamp experiments on nuclei devoid of external nuclear membrane reveal the existence of channel activity compatible with KV10.1. We hypothesize that KV10.1 channels at the nuclear envelope might participate in the homeostasis of nuclear K+, or indirectly interact with heterochromatin, both factors known to affect gene expression
