    This conference presents the culmination of the PGR Secure project (www.pgrsecure.org) – a collaborative project involving eleven partners funded under the EU Seventh Framework Programme, THEME KBBE.2010.1.1-03, 'Characterization of biodiversity resources for wild crop relatives to improve crops by breeding', Grant agreement no. 266394. It is jointly organized with the section on genetic resources of the European Association for Research on Plant Breeding (EUCARPIA). This international conference showcases innovative and potential novel characterization techniques and conservation strategies to identify and safeguard crop wild relative (CWR) and landrace (LR) genetic diversity to increase potential options for crop improvement as a means of underpinning food security in the face of climate change. The conference brings together a wide range of biodiversity expertise from the international community to debate current and future enhanced conservation and utilization of CWR and LR diversity for improving agricultural production, increasing food security and sustaining the environment for better livelihoods. The conference represents a landmark in the plant genetic resources science arena, highlighting exotic plant germplasm as a potentially critical but neglected resource for crop improvement. Part 1 of the book of abstracts contains the abstracts of the oral presentations and Part 2, those of the posters. They are organized under the four conference themes, viz. characterization techniques, conservation strategies, facilitating CWR and LR use and informatics development. The oral presentations will be the subject of a book entitled “Enhancing Crop Genepool Use: Capturing wild relative and landrace diversity for crop improvement” that will be published by CABI as the conference proceedings. All duly registered participants will be receiving a copy of the book when it is published

    Light Ions Response of Silicon Carbide Detectors

    Silicon carbide (SiC) Schottky diodes 21 mum thick with small surfaces and high N-dopant concentration have been used to detect alpha particles and low energy light ions. In particular 12C and 16O beams at incident energies between 5 and 18 MeV were used. The diode active-region depletion-thickness, the linearity of the response, energy resolution and signal rise-time were measured for different values of the applied reverse bias. Moreover the radiation damage on SiC diodes irradiated with 53 MeV 16O beam has been explored. The data show that SiC material is radiation harder than silicon but at least one order of magnitude less hard than epitaxial silicon diodes. An inversion in the signal was found at a fluence of 10^15 ions/cm^2.Comment: 20 pages, 16 figures, submitted for publication to Nuclear Instruments and Methods in Physics Research

    Metabolic responses to the acute ingestion of two commercially available carbonated beverages: A pilot study

    <p>Abstract</p> <p>Background</p> <p>The purpose of this placebo-controlled, double-blind cross-over study was to compare the effects of two commercially available soft drinks on metabolic rate.</p> <p>Methods</p> <p>After giving informed consent, twenty healthy men and women were randomly assigned to ingest 12 ounces of Celsius™ and, on a separate day, 12 ounces of Diet Coke®. All subjects completed both trials using a randomized, counterbalanced design. Metabolic rate (via indirect calorimetry) and substrate oxidation (via respiratory exchange ratio) were measured at baseline (pre-ingestion) and at the end of each hour for 3 hours post-ingestion.</p> <p>Results</p> <p>Two-way ANOVA revealed a significant interaction (p < 0.001) between trials in metabolic rate. Scheffe post-hoc testing indicated that metabolic rate increased by 13.8% (+ 0.6 L/min, p < 0.001) 1 hr post, 14.4% (+0.63 L/min, p < 0.001) 2 hr post, and 8.5% (+0.37 L/min, p < 0.004) 3 hr post Celsius™ ingestion. In contrast, small (~4–6%) but statistically insignificant increases in metabolic rate were noted following Diet Coke<sup>® </sup>ingestion. No differences in respiratory exchange ratio were noted between trials.</p> <p>Conclusion</p> <p>These preliminary findings indicate Celsius™ has thermogenic properties when ingested acutely. The effects of repeated, chronic ingestion of Celsius™ on body composition are unknown at this time.</p

    Energy expenditure during overfeeding

    The large inter-individual variation in weight gain during standardized overfeeding together with a weight gain that is often less than theoretically calculated from the energy excess suggest that there are differences between persons in the capacity to regulate energy expenditure and hence metabolic efficiency. Adaptive thermogenesis is defined as the regulated production of heat in response to environmental changes in temperature and diet, resulting in metabolic inefficiency. The question is whether adaptive thermogenesis can be identified in overfeeding experiments. From the numerous human overfeeding experiments we selected those studies that applied suitable protocols and measurement techniques. Five studies claimed to have found evidence for adaptive thermogenesis based on weight gains smaller than expected or unaccounted increases in thermogenesis above obligatory costs. Results from the other 11 studies suggest there is no adaptive thermogenesis as weight gains were proportional to the amount of overfeeding and the increased thermogenesis was associated with theoretical costs of an increased body size and a larger food intake. These results show that in humans, evidence for adaptive thermogenesis is still inconsistent. However, they do not rule out the existence, but emphasize that if present, adaptive changes in energy expenditure may be too small to measure considering measurement errors, errors in assumptions made and small (day-to-day) differences in physical activity. In addition, it is not clear in which component or components of total energy expenditure adaptive changes can occur and whether components can overlap due to measurement limitations

    Plant Genetic Resources and Germplasm Use in India

    Plant genetic resource (PGR) scientists now recognize the importance of shifting from a singular focus on conservation to a focus on both conservation and utilization of germplasm in order to meet future challenges. This paper analyzes the patterns of distribution of pearl millet, six small millets, chickpea and pigeonpea germplasm over the last 10 years at the two major genebanks functioning in India: the National Genebank at the National Bureau of Plant Genetic Resources (NBPGR) and a Consultative Group for International Agricultural Research (CGIAR) Genebank at the International Crops Research Institute for the Semi-Arid Tropics (ICRISAT), as well as the patterns of use of germplasm by millet breeders in India. Between the years of 1999-2009, ICRISAT distributed approximately 48 per cent of all its collections to breeders in India whereas NBPGR distributed 36 per cent of their collection. A total of 20 responses (30 per cent of surveys sent) were collected through this study. Sixty-five percent of respondents said that they rarely (<50 per cent of the time) use germplasm from genebanks in their breeding programs. It is important that both genebanks look into several issues in order to improve levels of distribution and utilization, collection, duplication, engagement of the private sector, access to information, and pre-breeding

    Tea and coffee consumption in relation to vitamin D and calcium levels in Saudi adolescents

    Background Coffee and tea consumption was hypothesized to interact with variants of vitamin D-receptor polymorphisms, but limited evidence exists. Here we determine for the first time whether increased coffee and tea consumption affects circulating levels of 25-hydroxyvitamin D in a cohort of Saudi adolescents. Methods A total of 330 randomly selected Saudi adolescents were included. Anthropometrics were recorded and fasting blood samples were analyzed for routine analysis of fasting glucose, lipid levels, calcium, albumin and phosphorous. Frequency of coffee and tea intake was noted. 25-hydroxyvitamin D levels were measured using enzyme-linked immunosorbent assays. Results Improved lipid profiles were observed in both boys and girls, as demonstrated by increased levels of HDL-cholesterol, even after controlling for age and BMI, among those consuming 9–12 cups of coffee/week. Vitamin D levels were significantly highest among those consuming 9–12 cups of tea/week in all subjects (p-value 0.009) independent of age, gender, BMI, physical activity and sun exposure. Conclusion This study suggests a link between tea consumption and vitamin D levels in a cohort of Saudi adolescents, independent of age, BMI, gender, physical activity and sun exposure. These findings should be confirmed prospectively

    Modelling the associations between fat-free mass, resting metabolic rate and energy intake in the context of total energy balance

    © 2016 Macmillan Publishers Limited.The relationship between body composition, energy expenditure and ad libitum energy intake (EI) has rarely been examined under conditions that allow any interplay between these variables to be disclosed.Objective:The present study examined the relationships between body composition, energy expenditure and EI under controlled laboratory conditions in which the energy density and macronutrient content of the diet varied freely as a function of food choice.Methods:Fifty-nine subjects (30 men: mean body mass index=26.7±4.0 kg m-2; 29 women: mean body mass index=25.4±3.5 kg m-2) completed a 14-day stay in a residential feeding behaviour suite. During days 1 and 2, subjects consumed a fixed diet designed to maintain energy balance. On days 3-14, food intake was covertly measured in subjects who had ad libitum access to a wide variety of foods typical of their normal diets. Resting metabolic rate (RMR; respiratory exchange), total daily energy expenditure (doubly labelled water) and body composition (total body water estimated from deuterium dilution) were measured on days 3-14.Results:Hierarchical multiple regression indicated that after controlling for age and sex, both fat-free mass (FFM; P<0.001) and RMR (P<0.001) predicted daily EI. However, a mediation model using path analysis indicated that the effect of FFM (and fat mass) on EI was fully mediated by RMR (P<0.001).Conclusions:These data indicate that RMR is a strong determinant of EI under controlled laboratory conditions where food choice is allowed to freely vary and subjects are close to energy balance. Therefore, the conventional adipocentric model of appetite control should be revised to reflect the influence of RMR

    The contribution of Swiss scientists to the assessment of energy metabolism

    Although Switzerland is considered a small country, it has its share in discoveries, inventions and developments for the assessment of energy metabolism. This includes seminal contributions to respiratory and metabolic physiology and to devices for measuring energy expenditure by direct and indirect calorimetry in vivo in humans and small animals (as well as in vitro in organs/tissues), for the purpose of evaluating the basic nutritional requirements. A strong momentum came during World War II when it was necessary to evaluate the energy requirements of soldiers protecting the country by assessing their energy expenditure, as well as to determine the nutritional needs of the Swiss civil population in time of war when food rationing was necessary to ensure national neutrality and independence. A further impetus came in the 1970s at the start of the obesity epidemics, toward a better understanding of the metabolic basis of obesity, ranging from the development of whole-body concepts to molecular mechanisms. In a trip down memory lane, this review focuses on some of the earlier leading Swiss scientists who have contributed to a better understanding of the field

    Knowledge Sharing on Best Practices for Managing Crop Genebanks

    The Crop Genebank Knowledge Base (CGKB) is an initiative of the Consultative Group of International Agriculture Research (CGIAR) System-wide Genetic Resources Programme (SGRP). The CGKB was created for sharing knowledge about best practices for managing plant genetic resources (PGR), and making effective decisions about genebank management. Genebank practices from CGIAR Centers and national genebanks were gathered for nine crops (banana, barley, cassava, chickpea, forage grasses and legumes, maize, rice and wheat). This information will help PGR professionals to participate in a global crop conservation effort. An interactive approach with many partners and stakeholders was used to gather published and unpublished information about genebank management. Information on crop-specific best practices was initially collected from crop experts using pre-defined forms. In parallel, a web portal was developed using the open-source content management system (CMS) Joomla!. The CMS allows several editors to maintain pages and update them. Other participatory tools such as wiki pages, a blog, a discussion forum and online forms have been set up to gather future contributions, including information on other crops. The site provides a one-stop platform for information on conservation, characterization, regeneration and safety duplication of each of the nine crops. It also provides information on general (non-crop-specific) genebank management procedures, as well as a comprehensive bibliography of online publications, a glossary, links to relevant external websites, video and photo materials, and training modules. This paper discusses a process of collective action to develop a multi-institutional web platform, highlights important criteria for success, challenges and major lessons learned, and proposes options for the way forward
