    Store-operated Ca2+ entry in primary murine lung fibroblasts is independent of classical transient receptor potential (TRPC) channels and contributes to cell migration

    Stromal interaction molecules (STIM1, 2) are acting as sensors for Ca2+ in intracellular stores and activate Orai channels at the plasma membrane for store-operated Ca2+ entry (SOCE), while classical transient receptor potential (TRPC) channel mediate receptor-operated Ca2+ entry (ROCE). Several reports, however, indicate a role for TRPC in SOCE in certain cell types. Here, we analyzed Ca2+ influx and cell function in TRPC1/6-deficient (TRPC1/6(-/-)) and STIM1/2- deficient (STIM1/2(Delta pmLF)) primary murine lung fibroblasts (pmLF). As expected, SOCE was decreased in STIM1/2- deficient pmLF and ROCE was decreased in TRPC1/6(-/-) pmLF compared to control cells. By contrast, SOCE was not significantly different in TRPC1/6(-/-) pmLF and ROCE was similar in STIM1/2-deficient pmLF compared to Wt cells. Most interestingly, cell proliferation, migration and nuclear localization of nuclear factor of activated T-cells (NFATc1 and c3) were decreased after ablation of STIM1/2 proteins in pmLF. In conclusion, TRPC1/6 channels are not involved in SOCE and STIM1/2 deficiency resulted in decreased cell proliferation and migration in pmLF

    Calcium transfer and mass balance associated with soil carbonate in a semi‐arid silicate watershed (North Cameroon): an overlooked geochemical cascade?

    International audienceCalcium is a key element of the Earth system and closely coupled to the carbon cycle. Weathering of silicate releases Ca, which is exported and sequestered in oceans. However, pedogenic calcium carbonate constitutes a second Ca-trapping pathway that has received less attention. Large accumulations of pedogenic calcium carbonate nodules, associated with palaeo-Vertisols, are widespread in North Cameroon, despite a carbonate-free watershed. A previous study suggested that a significant proportion of Ca released during weathering was trapped in palaeo-Vertisols but the pathways involved in the transfer of Ca from sources (the granite and the Saharan dust) to a temporary sink (the carbonate nodules) remain unclear. This study aims to compare the distribution of elements in carbonate nodules and their associated past and present compartments for Ca in the landscape. These compartments are all characterised by a distinctive geochemical composition, resulting from specific processes. Three end members have been defined based on geochemical data: (a) the granite and its residual products, dominated by K2O and Na2O, Ti and Zr, HREE, and a positive Ce anomaly; (b) the soil parental material and the Saharan dust, dominated by Al2O3, Fe2O3 and MgO, V, HREE, and a positive Ce anomaly; and finally (c) the carbonate nodules, which are dominated by CaO, a depletion in V, Ti and Zr, and an enrichment in REE with a negative Ce anomaly. Mass balance calculations in soil profiles demonstrated that the accumulation of Ca in carbonate nodules exceeds the Ca released by chemical weathering of the parental material, because of a continuous accumulation and contribution from lateral transfers. Consequently, at the landscape scale, carbonate nodules associated with palaeo-Vertisols constitute a temporary sink for Ca. Such a spatial relationship between sources and transient compartments opens an avenue to the new concept of 'geochemical cascade', similar in terms of geochemistry, to the concept of 'sediment cascade' developed by continental sedimentologists

    Attitude indicator format : How difficult is the transition between different reference systems?

    Abstract. A simulator study investigated the consequences of a transition between two alternative formats of the attitude indictor in aircraft cockpits, the moving-horizon and moving-aircraft format. Two groups of novices practiced performing two flight tasks (flight-path tracking and recovery from unusual attitudes) with one attitude-indicator format for six practice sessions, before transitioning to the other format. The results show that, after practice, participants were able to perform both tasks equally well with both attitude-indicator formats. However, the number of reversal errors in the recovery task increased considerably when transitioning from the moving-aircraft to moving-horizon format. No such effect emerged for the other direction. This suggests that the former transition is more difficult and represents a possible risk for flight safety

    Plasmid- and strain-specific factors drive variation in ESBL-plasmid spread in vitro and in vivo

    Horizontal gene transfer, mediated by conjugative plasmids, is a major driver of the global rise of antibiotic resistance. However, the relative contributions of factors that underlie the spread of plasmids and their roles in conjugation in vivo are unclear. To address this, we investigated the spread of clinical Extended Spectrum Beta-Lactamase (ESBL)-producing plasmids in the absence of antibiotics in vitro and in the mouse intestine. We hypothesised that plasmid properties would be the primary determinants of plasmid spread and that bacterial strain identity would also contribute. We found clinical Escherichia coli strains natively associated with ESBL-plasmids conjugated to three distinct E. coli strains and one Salmonella enterica serovar Typhimurium strain. Final transconjugant frequencies varied across plasmid, donor, and recipient combinations, with qualitative consistency when comparing transfer in vitro and in vivo in mice. In both environments, transconjugant frequencies for these natural strains and plasmids covaried with the presence/absence of transfer genes on ESBL-plasmids and were affected by plasmid incompatibility. By moving ESBL-plasmids out of their native hosts, we showed that donor and recipient strains also modulated transconjugant frequencies. This suggests that plasmid spread in the complex gut environment of animals and humans can be predicted based on in vitro testing and genetic data

    Origin of calcium in pedogenic carbonate nodules from silicate watersheds in the Far North Region of Cameroon: Respective contribution of in situ weathering source and dust input

    Significant amounts of pedogenic Ca-carbonate nodules have been observed in the Far North Region of Cameroon in the carbonate-free watershed of the Mayo Tsanaga, thus a priori not favourable for carbonate nodules accumulations. These nodules are associated with a Clay-Rich Parent Material (CRPM), covering either a granitic (upstream) or a greenstone bedrock (downstream). In this peculiar context, the amount of pedogenic carbonate nodules represents large quantities of Ca and C. Therefore, determining the Ca sources for pedogenic carbonate nodules is a key point regarding the mechanisms leading to carbonate nodule precipitation and their role in the biogeochemical cycle of Ca. Three sites, two on granite and one on greenstone, were studied by combining Sr et Nd isotope systematics in order to assess the Ca sources of carbonate nodules and the origin of the CRPM associated with nodules. Carbonate nodules have a distinct Sr isotopic composition in each profile, pointing to the contribution of a local source for Ca. Sr isotopic compositions of plagioclases and carbonate nodules display a concomitant variation in each profile, indicating that plagioclases acted as a Sr, and thus Ca, source for the carbonate nodules. Nevertheless, carbonate nodules have a higher Sr isotopic signature than plagioclases, implying the contribution of another more radiogenic Sr source. Sr and Nd isotope data from the CRPM show that it is a mixture of weathered bedrock compounds and Saharan dust. This result highlights two other potential Sr and Ca sources: biotite and Saharan dust. Calculations of their respective contributions demonstrate that in situ weathering significantly adds to the Ca source of nodules. This result contrasts with many other studies from similar settings that highlight the importance of allochthonous - marine, aeolian - inputs. This conclusion points out that a substantial part of Ca is transferred from the local bedrock to the carbonate nodules. Detailed knowledge of cell-to-cell variations with respect to cellular growth and product formation is limited, even though phenotypic heterogeneity may distinctly hamper overall production yields, especially for toxic or difficult-to-express proteins. Unraveling phenotypic heterogeneity is thus a key aspect in understanding and optimizing recombinant protein production in biotechnology and synthetic biology. Here, microfluidic single-cell analysis serves as the method of choice to investigate and unmask population heterogeneities in a dynamic and spatiotemporal fashion. In this study, we report on comparative microfluidic single-cell analyses of commonly used E. coli expression systems to uncover system-inherent specifications in the synthetic M9CA growth medium. To this end, the PT7lac/LacI, the PBAD/AraC and the Pm/XylS system were systematically analyzed in order to gain detailed insights into variations of growth behavior and expression phenotypes and thus to uncover individual strengths and deficiencies at the single-cell level. Specifically, we evaluated the impact of different system-specific inducers, inducer concentrations as well as genetic modifications that affect inducer-uptake and regulation of target gene expression on responsiveness and phenotypic heterogeneity. Interestingly, the most frequently applied expression system based on E. coli strain BL21(DE3) clearly fell behind with respect to expression homogeneity and robustness of growth. Moreover, both the choice of inducer and the presence of inducer uptake systems proved crucial for phenotypic heterogeneity. Conclusively, microfluidic evaluation of different inducible E. coli expression systems and setups identified the modified lacY-deficient PT7lac/LacI as well as the Pm/XylS system with conventional m-toluic acid induction as key players for precise and robust triggering of bacterial gene expression in E. coli in a homogeneous fashion

    Prednisolone or tetracosactide depot for infantile epileptic spasms syndrome? A prospective analysis of data embedded within two randomised controlled trials

    OBJECTIVE: To report a prospectively planned analysis of two randomised controlled trials with embedded comparisons of prednisolone versus tetracosactide depot for the treatment of infantile epileptic spasms syndrome (IESS). METHODS: Individual patient data from patients randomly allocated to prednisolone or tetracosactide depot were analysed from two trials (UKISS, ICISS). The comparison was embedded within trials in which some patients also received vigabatrin but only patients receiving monotherapy with randomly allocated hormonal treatments are included in this analysis. The main outcome was cessation of spasms (Days 13-14 after randomisation). Lead time to treatment and underlying aetiology were taken into account. Cessation of spasms on Days 14-42 inclusive, electroclinical response (EEG Day 14), plus developmental and epilepsy outcomes (at 14 months in UKISS and 18 months in ICISS) are also reported. Minimum treatment was prednisolone 40 mg per day for two weeks or tetracosactide depot 0·5 mg IM on alternate days for two weeks, all followed by a reducing dose of prednisolone over two weeks. RESULTS: 126 infants were included in this study. On tetracosactide depot, 47 of 62 (76%) were free of spasms on Days 13-14 compared to 43 of 64 (67%) on prednisolone (difference 9%, 95% CI -7·2% to +25·2%, chi square 1·15, p = 0·28). For Day 14-42 cessation of spasms, on tetracosactide depot, 41 of 61 (67%) were free of spasms compared to 35 of 62 (56%) on prednisolone (difference 11%, 95% CI -6·4% to +28·4%, chi square 1·51, p = 0·22). There was no significant difference in mean VABS score between infants who received prednisolone compared with those who received tetracosactide depot (74·8 (SD 18·3) versus 78·0 (SD 20·2) t = -0·91 p = 0·36). The proportion with ongoing epilepsy at the time of developmental assessment was 20 of 61 (33%) in the tetracosactide group compared with 26 out of 63 (41%) in the prednisolone group (difference 8%, 95% CI -9·2% to +25·2%, Chi [2] 0·95, p = 0·33). SIGNIFICANCE: With hormone monotherapy, either prednisolone or tetracosactide depot may be recommended for infantile epileptic spasms syndrome

    BACKGROUND AND AIMS In adults with congenital heart disease and systemic right ventricles, progressive right ventricular systolic dysfunction is common and is associated with adverse outcomes. Our aim was to assess the impact of the phosphodiesterase-5-inhibitor tadalafil on right ventricular systolic function. METHODS AND RESULTS This was a double-blind, randomized, placebo-controlled, multi-center superiority trial (NCT03049540) involving 100 adults with systemic right ventricles (33 women, mean age: 40.7 years, SD 10.7), comparing tadalafil 20mg once daily versus placebo (1:1-ratio). Primary endpoint was the change in right ventricular endsystolic volume after three years of therapy. Secondary endpoints were changes in right ventricular ejection fraction, exercise capacity and NT-proBNP-concentration. Primary endpoint assessment by intention to treat analysis at three years of follow up was possible in 83 patients (42 patients in the tadalafil group and 41 patients in the placebo group). No significant changes over time in right ventricular endsystolic volumes were observed in the tadalafil and the placebo-group, and no significant differences between treatment groups (3.4ml, 95% CI, -4.3 to 11.0, p=0.39). No significant changes over time were observed for the pre-specified secondary endpoints for the entire study population, without differences between the tadalafil and the placebo-group. CONLCUSIONS In this trial in adults with systemic right ventricles, right ventricular systolic function, exercise capacity and neuro-hormonal activation remained stable over a three-year follow-up period. No significant treatment effect of tadalafil was observed. Further research is needed to find effective treatment for improvement of ventricular function in adults with systemic right ventricles.
