    Lack of the serum and glucocorticoid-inducible kinase SGK1 attenuates the volume retention after treatment with the PPARγ agonist pioglitazone

    PPARgamma-agonists enhance insulin sensitivity and improve glucose utilization in diabetic patients. Adverse effects of PPARgamma-agonists include volume retention and edema formation. Recent observations pointed to the ability of PPARgamma agonists to enhance transcription of the serum and glucocorticoid-inducible kinase SGK1, a kinase that is genomically upregulated by mineralocorticoids and stimulates various renal channels and transporters including the renal epithelial Na+ channel ENaC. SGK1 has been proposed to mediate the volume retention after treatment with PPARgamma agonists. To test this hypothesis, food containing the PPARgamma agonist pioglitazone (0.02%, i.e., approximately 25 mg/kg bw/day) was administered to gene-targeted mice lacking SGK1 (sgk1-/-, n=12) and their wild-type littermates (sgk1+/+), n=12). According to in situ hybridization, quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) and immunofluorescence, treatment with pioglitazone significantly increased renal SGK1 mRNA and protein expression in sgk1+/+ mice. The treatment increased body weight significantly in both, sgk1+/+ mice (+2.2+/-0.3 g) and sgk-/- mice (+1.3+/-0.2 g), and decreased hematocrit significantly in sgk1+/+ mice (-6.5+/-1.0%) and sgk1-/- mice (-3.1+/-0.6%). Both effects were significantly (p<0.05) more pronounced in sgk1+/+ mice. According to Evans Blue distribution, pioglitazone increased plasma volume only in sgk1+/+ mice (from 50.9+/-3.9 to 63.7+/-2.5 microl/g bw) but not in sgk-/- mice (from 46.8+/-3.8 to 48.3+/-5.2 microl/g bw). Pioglitazone decreased aldosterone plasma levels and blood pressure and increased leptin plasma levels in both genotypes. We conclude that SGK1 contributes to but does not fully account for the volume retention during treatment with the PPARgamma agonist pioglitazone

    A broadly cross-reactive monoclonal antibody against hepatitis E virus capsid antigen

    To generate a hepatitis E virus (HEV) genotype 3 (HEV-3)–specific monoclonal antibody (mAb), the Escherichia coli–expressed carboxy-terminal part of its capsid protein was used to immunise BALB/c mice. The immunisation resulted in the induction of HEV-specific antibodies of high titre. The mAb G117-AA4 of IgG1 isotype was obtained showing a strong reactivity with the homologous E. coli, but also yeast-expressed capsid protein of HEV-3. The mAb strongly cross-reacted with ratHEV capsid protein derivatives produced in both expression systems and weaker with an E. coli–expressed batHEV capsid protein fragment. In addition, the mAb reacted with capsid protein derivatives of genotypes HEV-2 and HEV-4 and common vole hepatitis E virus (cvHEV), produced by the cell-free synthesis in Chinese hamster ovary (CHO) and Spodoptera frugiperda (Sf21) cell lysates. Western blot and line blot reactivity of the mAb with capsid protein derivatives of HEV-1 to HEV-4, cvHEV, ratHEV and batHEV suggested a linear epitope. Use of truncated derivatives of ratHEV capsid protein in ELISA, Western blot, and a Pepscan analysis allowed to map the epitope within a partially surface-exposed region with the amino acid sequence LYTSV. The mAb was also shown to bind to human patient–derived HEV-3 from infected cell culture and to hare HEV-3 and camel HEV-7 capsid proteins from transfected cells by immunofluorescence assay. The novel mAb may serve as a useful tool for further investigations on the pathogenesis of HEV infections and might be used for diagnostic purposes.     Die vegetationskundliche und strukturelle Zuordnung der Lebensraumtypen erfolgt nach der vorrangig von Braun-Blanquet entwickelten Vegetationsklassifizierung, einer hierarchischen Gliederung der Vegetationstypen (Syntaxonomie), die die Ebenen der Assoziation, des Verbandes, der Ordnung und der Klasse umfasst. Hierbei ist die Assoziation die grundlegende Einheit, in der die Pflanzengesellschaften zusammengefasst werden, die sich durch gleiche charakteristische Arten(gruppen)kombinationen auszeichnen. Der Verband vereinigt ähnliche Assoziationen. Das sind bereits umfassendere, jedoch standörtlich noch recht einheitliche Vegetationseinheiten. In Ordnungen werden ähnliche Verbände zusammengefasst.     Assessing land use and flood management impacts on ecosystem services in a river landscape (Upper Danube, Germany)

    Rivers and floodplains provide many regulating, provisioning and cultural ecosystem services (ES) such as flood risk regulation, crop production or recreation. Intensive use of resources such as hydropower production, construction of detention basins and intensive agriculture substantially change ecosystems and may affect their capacity to provide ES. Legal frameworks such as the European Water Framework Directive, Bird and Habitats Directive and Floods Directive already address various uses and interests. However, management is still sectoral and often potential synergies or trade‐offs between sectors are not considered. The ES concept could support a joint and holistic evaluation of impacts and proactively suggest advantageous options. The river ecosystem service index (RESI) method evaluates the capacity of floodplains to provide ES by using a standardized five‐point scale for 1 km‐floodplain segments based on available spatial data. This scaling allows consistent scoring of all ES and their integration into a single index. The aim of this article is to assess ES impacts of different flood prevention scenarios on a 75 km section of the Danube river corridor in Germany. The RESI method was applied to evaluate scenario effects on 13 ES with the standardized five‐point scale. Synergies and trade‐offs were identified as well as ES bundles and dependencies on land use and connectivity. The ratio of actual and former floodplain has the strongest influence on the total ES provision: the higher the percentage and area of an active floodplain, the higher the sum of ES.     Analyse der Surfaktantprotein A-Gene bei Patienten mit Verdacht auf einen Surfaktantproteindefekt

    Zusammenfassung Viele Untersuchungen deuten darauf hin, dass das Surfaktantprotein A (SP-A) sowohl an der Regulation des Surfaktanthaushalts als auch als unspezifisches Opsonin an der Abwehr von Pathogenen in der Lunge beteiligt ist. Zahlreiche Polymorphismen kennzeichnen die Gene der Proteinuntereinheiten SP-A1 und 2. Die häufigste Aminosäuresubstitution Val50Leu befindet sich in der kollagenartigen Domäne, die an den Kollektinrezeptor der Phagozyten bindet. Weitere existieren in der an der Bindung an Lipopolysaccharide, Surfaktantbestandteile und Rezeptoren auf Pneumozyten beteiligten Kohlehydraterkennungsregion (CRD) der globulären Domäne. Träger des schwach exprimierten Wildtypallels 1a0 des SP-A2-Gens haben ein erhöhtes Risiko, am Atemnotsyndrom des Neugeborenen (RDS) zu erkranken. Bei der Alveolarproteinose akkumulieren die hydrophilen Surfaktantproteine A und D in den Alveolen. In der vorliegenden Arbeit wurde eine nested PCR zur isolierten Amplifikation beider SP-A-Gene etabliert. 31 Patienten mit Verdacht auf einen Surfaktantproteindefekt wurden auf neue Restriktionsfragmentlängenpolymorphismen (RFLP) im SP-A1-Gen untersucht. Der in einer Familie konstante NcoI-Polymorphismus 1162C>T in Codon 39 und der NdeI-Polymorphismus 3138T>C in Codon 184 wurden mit einer Allelfrequenz von etwa 11 % detektiert. Die Sequenzen der entsprechenden Allele wurden kloniert. Bei 14 Patienten mit idiopathischer Alveolarproteinose, therapierefraktärem Surfaktantmangel oder rezidivierender Pneumonie wurden die SP-A-Gene sequenziert. Der bisher nur SP-A1 zugeschriebene Aminosäureaustausch Val50Leu wurde als Substitution 1220G>C bei zwei Patienten im SP-A2-Gen nachgewiesen. Drei Patienten mit Alveolarproteinose waren homozygot für die Substitution Gln223Lys in der CRD des SP-A2. Bei einem Patienten handelte es sich möglicherweise um eine somatische Mutation der Leukozyten-DNA im Rahmen einer Leukämie mit sekundärer Alveolarproteinose. Ein anderer war heterozygoter Träger des seltenen Allels 6a4 mit der Aminosäuresubstitution Arg219Trp in der CRD des SP-A1 und hatte die Alveolarproteinose erst im Erwachsenenalter entwickelt. Der dritte war homozygoter Träger des sehr seltenen Allels 1a3 des SP-A2 und verstarb im Alter von 6 Wochen an konnataler Alveolarproteinose (CAP), ohne dass ein bekannter Defekt des SP-B- oder des GM-CSF-Rezeptorgens vorlag. Die SSCP-Analyse konnte allelische Varianten als Einzelstrangkonformationspolymorphismen unterscheiden, war jedoch als Suchtest in heterozygoten Proben zu unspezifisch. Der hohe Gehalt an Polymorphismusinformation (PIC) macht den SP-A-Genort sftp1 zu einem nützlichen Marker bei der Untersuchung der Surfaktantproteine und anderer auf Chromosom 10 lokalisierter Gene.Abstract Many studies give evidence of the role of surfactant protein A (SP-A) in the regulation of surfactant homeostasis and the defence from pathogens in the lung by opsonisation. The genes for the two protein subunits SP-A1 and SP-A2 are characterised by numerous polymorphisms. The most frequently substituted amino acid Val50Leu is located within the collagen-like region, which is recognised by the collectin-receptor on phagocytes. Further amino acids are substituted in the globular region, which is involved into the binding to lipopolysaccharides, surfactant particles, and receptors on pneumocytes by its carbohydrate recognition domain (CRD). Individuals carrying the weakly expressed wild-type allele 1a0 of SP-A2 have an increased risk of developing the respiratory distress syndrome (RDS) of the new-born. Alveolar proteinosis is a disease with accumulation of the hydrophilic surfactant proteins SP-A and SP-D in the alveoli. In this study a nested PCR for separate amplification of the two SP-A genes has been established. 31 patients with suspected deficiency of a surfactant protein has been investigated for new restriction fragment length polymorphisms (RFLP) in the SP-A1 gene. The NcoI-polymorphism 1162C>T in codon 39, which was constantly inherited in one family, and the NdeI-polymorphism 3138T>C in codon 184 have been detected with an allele frequency of around 11 %. The DNA sequences of these alleles have been cloned. In 14 patients suffering from idiopathic alveolar proteinosis, therapy-refractory surfactant deficiency, or recurrent pneumonia the SP-A genes have been sequenced. The substituted amino acid Val50Leu, which was previously considered exclusively in SP-A1, has been detected in SP-A2 in two patients. Three patients with alveolar proteinosis proved to be homozygous for the substitution Gln223Lys within the CRD of SP-A2. One of these patients might have a somatic mutation in the DNA of his leucocytes, with alveolar proteinosis developing secondary to his leukaemia. Another one developed alveolar proteinosis as an adult and was heterozygous for the rare allele 6a4 which includes the substituted amino acid Arg219Trp in the CRD of SP-A1. The third one proved to be homozygous for the very rare allele 1a3 of SP-A2 and died at 6 weeks of age from congenital alveolar proteinosis (CAP) without having one of the known mutations responsible for this condition within the genes for surfactant protein B (SP-B) or the GM-CSF receptor protein. The allelic variants could be differentiated by single strand conformation polymorphism but the SSCP-analysis was not enough specific for the screening of heterozygous DNA. Due to its high polymorphism information content (PIC), the SP-A gene locus sftp1 is a useful genetic marker for the analysis of the surfactant proteins and other genes located on chromosome 10

    Limit distributions of V- and U-statistics in terms of multiple stochastic Wiener-type integrals

    We describe the limit distribution of V- and U-statistics in a new fashion. In the case of V-statistics the limit variable is a multiple stochastic integral with respect to an abstract Brownian bridge GQ. This extends the pioneer work of Filippova (1961) [8]. In the case of U-statistics we obtain a linear combination of GQ-integrals with coefficients stemming from Hermite Polynomials. This is an alternative representation of the limit distribution as given by Dynkin and Mandelbaum (1983) [7] or Rubin and Vitale (1980) [13]. It is in total accordance with their results for product kernels.
