    Identifying Molecular Signatures of Distinct Modes of Collective Migration in Response to the Microenvironment Using Three-Dimensional Breast Cancer Models

    Collective cell migration is a key feature of transition of ductal carcinoma in situ (DCIS) to invasive ductal carcinoma (IDC) among many other cancers, yet the microenvironmental factors and underlying mechanisms that trigger collective migration remain poorly understood. Here, we investigated two microenvironmental factors, tumor-intrinsic hypoxia and tumor-secreted factors (secretome), as triggers of collective migration using three-dimensional (3D) discrete-sized microtumor models that recapitulate hallmarks of DCIS-IDC transition. Interestingly, the two factors induced two distinct modes of collective migration: directional and radial migration in the 3D microtumors generated from the same breast cancer cell line model, T47D. Without external stimulus, large (600 µm) T47D microtumors exhibited tumor-intrinsic hypoxia and directional migration, while small (150 µm), non-hypoxic microtumors exhibited radial migration only when exposed to the secretome of large microtumors. To investigate the mechanisms underlying hypoxia- and secretome-induced directional vs. radial migration modes, we performed differential gene expression analysis of hypoxia- and secretome-induced migratory microtumors compared with non-hypoxic, non-migratory small microtumors as controls. We propose unique gene signature sets related to tumor-intrinsic hypoxia, hypoxia-induced epithelial-mesenchymal transition (EMT), as well as hypoxia-induced directional migration and secretome-induced radial migration. Gene Set Enrichment Analysis (GSEA) and protein-protein interaction (PPI) network analysis revealed enrichment and potential interaction between hypoxia, EMT, and migration gene signatures for the hypoxia-induced directional migration. In contrast, hypoxia and EMT were not enriched in the secretome-induced radial migration, suggesting that complete EMT may not be required for radial migration. Survival analysis identified unique genes associated with low survival rate and poor prognosis in TCGA-breast invasive carcinoma dataset from our tumor-intrinsic hypoxia gene signature (CXCR4, FOXO3, LDH, NDRG1), hypoxia-induced EMT gene signature (EFEMP2, MGP), and directional migration gene signature (MAP3K3, PI3K3R3). NOS3 was common between hypoxia and migration gene signature. Survival analysis from secretome-induced radial migration identified ATM, KCNMA1 (hypoxia gene signature), and KLF4, IFITM1, EFNA1, TGFBR1 (migration gene signature) to be associated with poor survival rate. In conclusion, our unique 3D cultures with controlled microenvironments respond to different microenvironmental factors, tumor-intrinsic hypoxia, and secretome by adopting distinct collective migration modes and their gene expression analysis highlights the phenotypic heterogeneity and plasticity of epithelial cancer cells

    Experiencias de voluntariado como estrategia para fomentar habilidades emocionales en jóvenes universitarios

    El objetivo del ejercicio de investigación fue desarrollar estrategias para el fomento de las habilidades emocionales en jóvenes universitarios, por medio de actividades de voluntariado. Para ello, se generaron espacios de estudio y aplicación de las habilidades emocionales y se realizó un seguimiento individual del proceso del estudiante. Se presentan los datos de 14 jóvenes miembros del voluntariado universitario de la Universidad Católica de Colombia, estudiantes de Derecho, Ingeniería, Psicología y Economía, con edades comprendidas entre los 17 y 22 años. De acuerdo con la información obtenida, se observa adecuada identificación de emociones básicas como alegría y tristeza, y algunas complejas como gratitud. La emoción que más se asocia a la actividad del voluntariado es la alegría, ya que manifiestan ser portadores y receptores de alegría durante las proyecciones sociales. A partir de esta perspectiva, consideramos que el voluntariado es un espacio que permite desarrollar habilidades emocionales, así como favorecer el desarrollo armónico de las facultades humanas, pues permite ser felices y hacer felices a los demás.1a edició

    En la historia de la humanidad son muchos los ejemplos de individuos que, poniendo en riesgo su propio bienestar, actúan en beneficio de otros. Estas conductas en las que las personas actúan para satisfacer las necesidades de los demás, muchas veces en perjuicio de sí mismas, generan un gran interés en la comunidad científica. Diversos estudios describen estas conductas, denominadas prosociales, como un fenómeno complejo que involucra acciones individuales basadas en creencias y sentimientos(Auné, Blum, Lozzia y Horacio, 2014), en el que una persona beneficia a otras de forma voluntaria (Jirsaraie, Ranby & Albeck, 2019). Esto no solo favorece la relación interpersonal y el bienestar del otro, sino que genera afecto positivo en la persona que ejecuta la acción (Richaud y Mesurado 2016).     Towards The Development of a Biologically Functional Tissue Engineered Vascular Graft

    Coronary artery disease (CAD) is the leading cause of death worldwide [1], and it is predicted that 23 million deaths will be attributed to CAD by 2030. Treatment of CAD has required over 400,000 coronary artery bypass graft (CABG) surgeries every year [2], representing a cost to the country of nearly $439 billion in direct and indirect expenses annually [3]. As autologous veins are often unavailable for in CABG surgeries [4, 5], and commercially available synthetic grafts have shown limited efficacy when used in small diameter vessels [5-7], a readily available tissue engineered vascular graft (TEVG) for use in CABG surgeries would provide drastic improvements in patient care. Despite significant recent progress [7, 8], the development of a biologically functional TEVG that is biocompatible, biofunctional, and anti-thrombogenic has remained elusive [9, 10]. The primary goal of this dissertation is to fabricate a TEVG that supports and modulates the growth and collagen production of vascular smooth muscle cells (VSMCs) and promotes the formation of a functional endothelium. For this purpose, an initial assessment of biocompatibility was performed by culturing SMCs in gelatin/fibrinogen electrospun scaffolds, using exogenous TGFβ2 to modulate cell response. It was demonstrated that that TGFβ2 had a differential effect on cell proliferation, migration, and collagen deposition of SMCs growing in our biopolymer materials. In order to provide additional biofunctionality to the TEVG, a tubular scaffold able to release TGFβ2 was fabricated. The combination of gelatin and PCL at different ratios allowed elution tunability. The released TGFβ2 was bioactive and was able to modulate SMCs growth in vitro and in 3D culture. To evaluate anti-thrombogenicity and perform an additional assessment of biocompatibility, human derived endothelial cells (hCB-ECs) were cultured in surface modified gelatin/fibrinogen/PCL electrospun scaffolds. hCB-ECs growing in the scaffolds showed similar or superior behavior to human umbilical vein ECs (HUVEC) in terms of platelet deposition and activation, producing eNOS, and responding to a proinflammatory stimulus.     Obtaining cellulases against oil palm empty fruit bunch from a metagenomic library

    Palm growing is one of the most promising and important agricultural sectors in Colombia. However the increment in the palm oil production has resulted in the generation of millions of tons of wastes as empty fruit bunch. Given this, it is important to generate new uses for this subproduct. One of the alternatives is the simplification of cellulose content of the empty fruit bunch in fermentable sugars as cellobiose and glucose, for a subsequent carburant alcohol production. The degradation of cellulose requires enzymes named cellulases that have the capability to hydrolase glycosidic bonds from cellulose. This study focuses on identify and characterize cellulases active against oil palm empty fruit bunch from a metagenomic library. A functional screening of the library, alloed to select four clones with the ability to degrade cellulose.     A Hybrid Model of a Genetic Regulatory Network in Mammalian Sclera

    Myopia in human and animals is caused by the axial elongation of the eye and is closely linked to the thinning of the sclera which supports the eye tissue. This thinning has been correlated with the overproduction of matrix metalloproteinase (MMP-2), an enzyme that degrades the collagen structure of the sclera. In this short paper, we propose a descriptive model of a regulatory network with hysteresis, which seems necessary for creating oscillatory behavior in the hybrid model between MMP-2, MT1-MMP and TIMP-2.     In vitro and in silico characterization of metagenomic soil-derived cellulases capable of hydrolyzing oil palm empty fruit bunch

    Diversification of raw material for biofuel production is of interest to both academia and industry. One attractive substrate is a renewable lignocellulosic material such as oil palm (Elaeis guineensis Jacq.) empty fruit bunch (OPEFB), which is a byproduct of the palm oil industry. This study aimed to characterize cellulases active against this substrate. Cellulases with activity against OPEFB were identified from a metagenomic library obtained from DNA extracted from a high-Andean forest ecosystem. Our findings show that the highest cellulolytic activities were obtained at pH and temperature ranges of 4–10 and 30 °C–60 °C, respectively. Due to the heterogeneous character of the system, degradation profiles were fitted to a fractal-like kinetic model, evidencing transport mass transfer limitations. The sequence analysis of the metagenomic library inserts revealed three glycosyl hydrolase families. Finally, molecular docking simulations of the cellulases were carried out corroborating possible exoglucanase and β-glucosidase activity
