    A Survey on the characteristics of sausage made from Sicilian and Sardinian native pigs

    Four different mixtures were prepared using meat from native pigs. Mixtures 1 and 2 were made in Sardinia using meat from pigs of Sarda breed, mixtures 3 and 4 in Sicily, using meat from the Nera Siciliana breed. The manufacturing were different not only for the typology of the meat but also for the meat cuts used and the ingredients. Batches 1 and 3 were made in non conditioned natural environments, while batches 2 and 4 in conditioned environments. Chemical-physical and microbiological analyses were carried out on the fresh mixture, after 7 days of ripening (end of drying in conditioned environment and at the end of seasoning (28 days). The thermoigrometric environmental parameters and the variation in the weight of the production were measured during ripening. Regarding the Lactic Acid Bacteria and the Micrococcaceae, microorganisms of normal maturation, a rapid development, except for batch 1, was observed for all batches at 7 days of ripening. In batches 2 and 4 an increase of Enterobacteriaceae in the drying phase and a their decrease in the seasoning phase were noticed. In batch 3 their inactivation resulted slow. Such occurrences were probably due to the hygienic conditions and non inhibiting technological parameters. The initial humidity was less than 60% with the exception of batch 3; the quantity of NaCl ranged between 2 and 2.8%. The amount of fat varied between 14 and 24% at the beginning of the ripening, then reached values ranging between of 20 and 38% at the end of the seasoning phase. All the batches showed a decrease of aw included between 0.97 (mixture) and 0.87 (final product); the highest value of aw recorded in test 1 was attributable to the lower T recorded both during the drying and seasoning phase. The pH reached the lowest values at the end of the 7 days and then increased, with the exception of batch 3, which decreased during the whole seasoning phase

    Magnetotelluric (MT) soundings performed in the past in the volcanic area of Mt. Vesuvius by two independent research groups showed in the same places MT apparent resistivity curves with very similar shape, but statically shifted by one order of magnitude, at least. To try to resolve this ambiguity new controlled source audio-magnetotelluric (CSAMT) measurements have been carried out in the same MT sites. The interpretation of the CSAMT dataset, combined with that of two shallow dipole-dipole geoelectrical resistivity tomographies previously carried out in the area have allowed a reliable electrical structure to be recovered down to a few km of depth, which will next be used for a best constrained re-interpretation of the deep MT soundings

    Cx36 makes channels coupling human pancreatic β-cells, and correlates with insulin expression

    Previous studies have documented that the insulin-producing β-cells of laboratory rodents are coupled by gap junction channels made solely of the connexin36 (Cx36) protein, and have shown that loss of this protein desynchronizes β-cells, leading to secretory defects reminiscent of those observed in type 2 diabetes. Since human islets differ in several respects from those of laboratory rodents, we have now screened human pancreas, and islets isolated thereof, for expression of a variety of connexin genes, tested whether the cognate proteins form functional channels for islet cell exchanges, and assessed whether this expression changes with β-cell function in islets of control and type 2 diabetics. Here, we show that (i) different connexin isoforms are differentially distributed in the exocrine and endocrine parts of the human pancreas; (ii) human islets express at the transcript level different connexin isoforms; (iii) the membrane of β-cells harbors detectable levels of gap junctions made of Cx36; (iv) this protein is concentrated in lipid raft domains of the β-cell membrane where it forms gap junctions; (v) Cx36 channels allow for the preferential exchange of cationic molecules between human β-cells; (vi) the levels of Cx36 mRNA correlated with the expression of the insulin gene in the islets of both control and type 2 diabetics.     The landscape of cancer-rewired GPCR signaling axes.

    We explored the dysregulation of G-protein-coupled receptor (GPCR) ligand systems in cancer transcriptomics datasets to uncover new therapeutics opportunities in oncology. We derived an interaction network of receptors with ligands and their biosynthetic enzymes. Multiple GPCRs are differentially regulated together with their upstream partners across cancer subtypes and are associated to specific transcriptional programs and to patient survival patterns. The expression of both receptor-ligand (or enzymes) partners improved patient stratification, suggesting a synergistic role for the activation of GPCR networks in modulating cancer phenotypes. Remarkably, we identified many such axes across several cancer molecular subtypes, including many involving receptor-biosynthetic enzymes for neurotransmitters. We found that GPCRs from these actionable axes, including, e.g., muscarinic, adenosine, 5-hydroxytryptamine, and chemokine receptors, are the targets of multiple drugs displaying anti-growth effects in large-scale, cancer cell drug screens, which we further validated.     Prevalence and risk factors associated with cat parasites in Italy: a multicenter study

    Background: Parasites that infect cats include protozoa, helminths and arthropods, many of which are transmissible to humans. Effective control relies on a good knowledge of parasite distribution and the risk factors for infection. The present study was aimed at evaluating the prevalence of major feline parasites in Italy and the risk factors associated with their occurrence. Methods: Over a 12-month study period, feces, hair and ectoparasites from naturally infected cats from feral colonies, shelters and private households were analyzed at 13 study centers across Italy. Samples from these cats (n = 987) were analyzed at all centers using the same diagnostic methods. Prevalence values and risk factors were evaluated statistically for the identification of predictors of risk. Results: The overall prevalence of gastro-intestinal and broncho-pulmonary (BP) nematodes was 35.9% (354/987). Toxocara cati was the most prevalent species (253/987; 25.6%), followed by Ancylostomatidae (98/987; 9.9%). Among BP nematodes, Aelurostrongylus abstrusus was the most common (76/987; 7.7%). Approximately 35.7% (352/987) of the study population was infested by ectoparasites, of which the most common were fleas (29.4%, 290/987), followed by ear mites Otodectes cynotis (9.8%, 97/987). Predictors of risk for parasite infection included age, a predominantly or exclusively outdoor lifestyle, geographic area and lack of antiparasitic treatment. Conclusions: Both ecto- and endoparasites are still common in cats throughout Italy, many of them being of zoonotic concern and vectors of pathogens to humans. Given the presence of parasites throughout the entire study period, year-round treatment should be considered. Furthermore, data confirm the need to protect the human–animal bond using proper endo- and ectoparasiticides to reduce the risk of human infection, in application of the One-Health concept

    Endothelium-Derived Netrin-4 Supports Pancreatic Epithelial Cell Adhesion and Differentiation through Integrins α2β1 and α3β1

    BACKGROUND: Netrins have been extensively studied in the developing central nervous system as pathfinding guidance cues, and more recently in non-neural tissues where they mediate cell adhesion, migration and differentiation. Netrin-4, a distant relative of Netrins 1-3, has been proposed to affect cell fate determination in developing epithelia, though receptors mediating these functions have yet to be identified. METHODOLOGY/PRINCIPAL FINDINGS: Using human embryonic pancreatic cells as a model of developing epithelium, here we report that Netrin-4 is abundantly expressed in vascular endothelial cells and pancreatic ductal cells, and supports epithelial cell adhesion through integrins α2β1 and α3β1. Interestingly, we find that Netrin-4 recognition by embryonic pancreatic cells through integrins α2β1 and α3β1 promotes insulin and glucagon gene expression. In addition, full genome microarray analysis revealed that fetal pancreatic cell adhesion to Netrin-4 causes a prominent down-regulation of cyclins and up-regulation of negative regulators of the cell cycle. Consistent with these results, a number of other genes whose activities have been linked to developmental decisions and/or cellular differentiation are up-regulated. CONCLUSIONS/SIGNIFICANCE: Given the recognized function of blood vessels in epithelial tissue morphogenesis, our results provide a mechanism by which endothelial-derived Netrin-4 may function as a pro-differentiation cue for adjacent developing pancreatic cell populations expressing adhesion receptors α2β1 and α3β1 integrins
