    Synthesis and enzymatic evaluation of the guanosine analogue 2-amino-6-mercapto-7-methylpurine ribonucleoside (MESG): insights into the phosphorolysis reaction mechanism based on the blueprint transition state: SN1 or SN2?

    A modified experimental procedure for the synthesis of MESG (2-amino-6-mercapto-7-methylpurine ribonucleoside) 1 has been successfully performed and its full characterization is presented. High resolution ESI(+)-MSMS indicates both the nucleoside bond cleavage as the main fragmentation in the gas phase and a possible SN1 mechanism. Ab initio transition state calculations based on the blue print transition state support this mechanistic rationale and discard an alternative SN2 mechanism. Assays using purine nucleoside phosphorylase (PNP) enzyme (human and M. tuberculosis sources) indicate its efficiency in the phosphorolysis of MESG and allow the quantitative determination of inorganic phosphate in real time assay

    Perfil da proteína de reserva e conteúdo de aminoácidos no arroz silvestre Oryza glumaepatula

    The objective of this work was to determine the total protein profile and the contents of the four major protein fractions (albumin, globulin, prolamin and glutelin) and of the amino acids in the endosperm of the rice wild species Oryza glumaepatula. The experiment was performed with 29 accessions of this species, collected from 13 Brazilian locations, and two commercial cultivars. Protein samples were prepared using dried, polished, and ground grains to obtain homogeneous, dry flour used in the preparation of extracts. Oryza glumaepatula accessions were identified with the highest levels of total protein, albumin and glutelin protein fractions, and amino acids (with the exception of tryptophan) in comparison to the two analized rice cultivars. The albumin and glutelin profiles in SDS-Page were distinct between rice cultivars and O. glumaepatula. This wild species has the potential to increase the nutritional quality of rice storage protein through interspecific crosses.O objetivo deste trabalho foi determinar os perfis de proteína total e o conteúdo das quatro principais frações proteicas (albumina, globulina, prolamina e glutelina) e de aminoácidos no endosperma da espécie de arroz silvestre Oryza glumaepatula. O experimento foi realizado com 29 acessos dessa espécie, coletados em 13 locais no Brasil, e duas cultivares comerciais. Amostras de proteínas foram preparadas com grãos secos, polidos e moídos, para obtenção de uma farinha seca e homogênea, usada no preparo dos estratos. Acessos de O. glumaepatula foram identificados com os maiores níveis de proteína total, frações proteicas de albumina e glutelina, e aminoácidos (com exceção do triptofano), em comparação às duas cultivares de arroz avaliadas. Os perfis de albumina e glutelina em SDS-PAGE foram distintos entre as cultivares de arroz e O. glumaepatula. Essa espécie silvestre tem o potencial de aumentar a qualidade nutricional da proteína de reserva do arroz por meio de cruzamentos interespecíficos

    Papel da ferritina na tolerância de arroz ao excesso de ferro

    Deficiência de ferro (Fe) ocorre freqüentemente em plantas, uma vez que este mineral é pouco disponível em condições aeróbicas. Plantas de arroz cultivadas sob alagamento, no entanto, estão sujeitas ao excesso de Fe, que pode ser extremamente tóxico. Alguns cultivares de arroz são resistentes a altas concentrações de ferro, mas os mecanismos fisiológicos responsáveis por essa resistência são pouco conhecidos. A ferritina é uma proteína de ampla distribuição e capaz de armazenar ferro, sendo considerada importante para a homeostase deste metal. Acúmulo de ferritina em condições de alta disponibilidade de ferro já foi descrito em algumas espécies vegetais. Entretanto, o papel da ferritina no mecanismo de tolerância de plantas de arroz ao excesso de ferro não é conhecido. Neste trabalho, expressamos ferritina de arroz em E. coli, produzimos um anticorpo policlonal anti-ferritina de arroz e este foi utilizado para avaliar o acúmulo de ferritina em dois cultivares de arroz (Oryza sativa) considerados suscetível (BR-IRGA 409) e tolerante (EPAGRI 108) ao excesso de ferro. O anticorpo foi capaz de reconhecer ferritina purificada de sementes de ervilha, assim como ferritina de folhas de arroz. Aumentos nos níveis de mRNA e proteína foram observados nos dois cultivares sob excesso de ferro, com maior acúmulo da proteína no cultivar EPAGRI 108. Quando submetidas a excesso do elemento, plantas deste mesmo cultivar atingiram concentrações de Fe mais baixas do que plantas do cultivar BR-IRGA409, principalmente nas partes aéreas. Sugere-se que o mecanismo de tolerância ao excesso de ferro no cultivar EPAGRI 108 inclui limitação da translocação de Fe e aumento do acúmulo de ferritina. Este é o primeiro trabalho que mostra maior acúmulo da proteína ferritina em um cultivar de Oryza sativa tolerante ao excesso de Fe, fornecendo evidência de um possível papel desta proteína nos mecanismos de tolerância a este metal.Plants ordinarily face iron (Fe) deficiency, since this mineral is poorly available in soils under aerobic conditions. Nonetheless, wetland and irrigated rice plants can be exposed to excess, highly toxic Fe. Ferritin is a ubiquitous Fe-storage protein, important for iron homeostasis. Increased ferritin accumulation resulting from higher Fe availability was shown in some plant species. However, the role of ferritin in tolerance mechanisms to Fe overload in rice is yet to be established. In this study, recombinant rice ferritin was expressed in Escherichia coli, producing an anti-rice ferritin polyclonal antibody which was used to evaluate ferritin accumulation in two rice (Oryza sativa L.) cultivars, either susceptible (BR-IRGA 409) or tolerant (EPAGRI 108) to Fe toxicity. Increased ferritin mRNA and protein levels resulting from excess Fe treatment were detected in both cultivars, with higher ferritin protein accumulation in EPAGRI 108 plants, which also reached lower shoot Fe concentrations when submitted to iron overload. The tolerance mechanism to excess Fe in EPAGRI 108 seems to include both restricted Fe translocation and increased ferritin accumulation. This is the first work that shows higher accumulation of the ferritin protein in an iron-excess tolerant Oryza sativa cultivar, providing evidence of a possible role of this protein in iron tolerance mechanisms

    Proteínas de reserva de acessos de coleção nuclear de arroz

    O objetivo deste trabalho foi quantificar o conteúdo de proteína total de reserva dos 550 acessos da Coleção Nuclear de Arroz da Embrapa (CNAE), e avaliar o perfil proteico dos 20 acessos com maior teor de proteína em SDS-PAGE. Foi encontrado alto teor de proteína total de reserva (≥ 12,0%) em 103 acessos da CNAE, teor médio (11,9 a 9,0%) em 309 acessos e teor baixo (≤ 8,9%) em 138 acessos. Seis dos 20 acessos com maior teor de proteína de reserva apresentaram um padrão qualitativo diferencial de glutelina, que é a fração proteica de reserva mais abundante do grão de arroz. Há ampla variabilidade para o teor de proteína total de reserva do grão de arroz nos acessos da CNAE, a qual pode ser explorada por programas de melhoramento genético para aumentar o valor nutricional do arroz consumido no Brasil.The objective of this work was to quantify the total storage protein content of 550 accessions from Coleção Nuclear de Arroz da Embrapa (Embrapa Rice Core Collection – CNAE), and to evaluate the SDS-PAGE profile of the 20 accessions that showed the highest storage protein content. High storage protein content was found in 103 CNAE accessions (≥12.0%), intermediate content (11.9 to 9.0%) in 309, and low protein content (≤ 8.9%) in 138. Six out of the 20 accessions with the highest storage protein content showed a differential profile for glutelin, which is the most abundant rice storage protein fraction. There is a large variability of rice storage protein in CNAE accessions, which can be explored in breeding programs to increase the nutritional value of the rice consumed in Brazil

    Cytotoxicity effect of alkaloidal extract from Prosopis juliflora Sw. D.C. (Algaroba) pods on glial cells

    A Prosopis juliflora é amplamente utilizada na alimentação humana e de várias espécies animais, especialmente bovinos. Quadros de intoxicação por esta planta, nesta espécie, têm sido relatados, principalmente quando a mesma é oferecida como única fonte alimentar, desencadeando uma doença de sintomatologia nervosa. Neste estudo, objetivou-se avaliar os efeitos in vitro da fração de alcalóides totais (FA) extraída das vagens da Prosopis juliflora utilizando cultura primária de astrócitos obtidos do córtex cerebral de ratos como modelo de estudo. A avaliação da atividade mitocondrial pelo teste do MTT demonstrou a citotoxicidade em 30 µg/ml da FA após 24 h. As concentrações de 0,3 e 3 µg/ml da FA induziram um aumento da atividade mitocondrial, indicando reatividade celular. Testes imunocitoquímicos para a GFAP, principal proteína de citoesqueleto de astrócitos, revelaram alterações morfológicas nas células após tratamento com 0,3 e 3 µg/ml da FA por 72 h. Tais resultados são consoantes à análise desta proteína por westernblot, quando as culturas foram tratadas com 0,3 e 3 µg/ml da FA por 72 h, demonstrando interferências na regulação da expressão da GFAP. A expressão de vimentina não foi significativamente alterada em nenhuma das concentrações testadas. Estes resultados sugerem que os alcalóides da P. juliflora induzem a reatividade astrocitária, o que pode estar envolvido nos efeitos neurotóxicos providos pelo consumo desta planta.Prosopis juliflora is largely used for feeding cattle and humans. Neurological signals have been reported in cattle due to intoxication with this plant. In this study, an alkaloidal fraction (AF) obtained from P. juliflora pods was tested on astrocyte primary cultures. Astrocytes display physiological functions essential to development, homeostasis and detoxification in the central nervous system (CNS). These cells are known for their role on energetic support and immune response in the CNS. Concentrations between 0.03 to 30 µg/ml AF were assayed for 24 - 72 h. The mitochondrial activity, assayed by MTT test, showed cytotoxicity at 30 µg/ml AF after 24 h. At concentrations ranging between 0.3 - 3 µg/ml, the AF induced an increase on mitochondrial activity, indicating cell reactivity. Immunocytochemistry assay for GFAP cytoskeletal protein, revealed alterations on cell morphology after treatment with 0.3 - 3 µg/ml AF for 72 h. This result corroborates with western blot analysis when cells treated with 0.3 - 3 µg/ml AF for 72 h showed GFAP upregulation. The vimentin expression was not significantly altered in all tested concentrations. These results suggest that alkaloids induce astrocyte reactivity and might be involved in the neurotoxic effects induced by P. juliflora consumption

    Mortality inequalities measured by socioeconomic indicators in Brazil: a scoping review

    OBJECTIVE Summarize the literature on the relationship between composite socioeconomic indicators and mortality in different geographical areas of Brazil. METHODS This scoping review included articles published between January 1, 2000, and August 31, 2020, retrieved by means of a bibliographic search carried out in the Medline, Scopus, Web of Science, and Lilacs databases. Studies reporting on the association between composite socioeconomic indicators and all-cause, or specific cause of death in any age group in different geographical areas were selected. The review summarized the measures constructed, their associations with the outcomes, and potential study limitations. RESULTS Of the 77 full texts that met the inclusion criteria, the study reviewed 24. The area level of composite socioeconomic indicators analyzed comprised municipalities (n = 6), districts (n = 5), census tracts (n = 4), state (n = 2), country (n = 2), and other areas (n = 5). Six studies used composite socioeconomic indicators such as the Human Development Index, Gross Domestic Product, and the Gini Index; the remaining 18 papers created their own socioeconomic measures based on sociodemographic and health indicators. Socioeconomic status was inversely associated with higher rates of all-cause mortality, external cause mortality, suicide, homicide, fetal and infant mortality, respiratory and circulatory diseases, stroke, infectious and parasitic diseases, malnutrition, gastroenteritis, and oropharyngeal cancer. Higher mortality rates due to colorectal cancer, leukemia, a general group of neoplasms, traffic accident, and suicide, in turn, were observed in less deprived areas and/or those with more significant socioeconomic development. Underreporting of death and differences in mortality coverage in Brazilian areas were cited as the main limitation. CONCLUSIONS Studies analyzed mortality inequalities in different geographical areas by means of composite socioeconomic indicators, showing that the association directions vary according to the mortality outcome. But studies on all-cause mortality and at the census tract level remain scarce. The results may guide the development of new composite socioeconomic indicators for use in mortality inequality analysis

    Flavonoids from the Brazilian plant Croton betulaster inhibit the growth of human glioblastoma cells and induce apoptosis

    This study investigated the effects of the flavonoids 5-hydroxy-7,4′-dimethoxyflavone, casticin, and penduletin, isolated from Croton betulaster Müll Arg., Euphorbiaceae, a plant utilized in popular medicine in Brazil, on the growth and viability of the human glioblastoma cell line GL-15. We observed that 5-hydroxy-7,4′-dimethoxyflavone and casticin were not toxic to GL-15 cells after 24 h of exposure. However, casticin and penduletin inhibited the metabolic activity of glioblastoma cells significantly at a concentration of 10 μM (p ≤ 0.05). Flavonoids casticin and penduletin also induced a significant and dose-dependent growth inhibition beginning at 24 h of exposure, and the most potent flavonoid was penduletin. It was also observed that penduletin and casticin induced an enlargement of the cell body and a reduction of cellular processes, accompanied by changes in the pattern of expression of the cytoskeletal protein vimentin. Signs of apoptosis, such as the externalization of membrane phosphatidyl serine residues, nuclear condensation, and fragmentation, were also detected in cells treated with 50–100 μM flavonoids. Our results indicate that flavonoids extracted from C. betulaster present antitumoral activity to glioblastoma cells, with penduletin proving to be the most potent of the tested flavonoids. Our results also suggest that these molecules may be promising supplementary drugs for glioblastoma treatment

    Chemical Composition and Validation of the Ethnopharmacological Reported Antimicrobial Activity of the Body Fat of Phrynops geoffroanus

    Background. Phrynops geoffroanus is a small turtle that inhabits lakes, rivers, and streams throughout South America. The body fat of this animal is used as a folk medicine in Brazil for treating illnesses such as sore throats, ear aches, mumps, rheumatism, and arthritis. The present study evaluated the antimicrobial activity of oil extracted from Phrynops geoffroanus (OPG), determined its chemical composition, and discussed the implications of its use in traditional medicine. The OPG was obtained from the ventral region of this turtle using hexane as a solvent. The antimicrobial activity of OPG was tested against standard and multiresistance strains of bacteria and fungi and its composition was determined indirectly by analyzing the methyl esters of the component fatty acids. The OPG presented a clinically relevant antifungal activity against Candida krusei ATCC 6258 (MIC 128 µg/mL). When the OPG was associated with the antibacterial and antifungal drugs, was observed a synergistic effect when associated the OPG with the gentamicin against the strain Pseudomonas aeruginosa 22. Our results indicated that OPG has clinically relevant antifungal activity against C. krusei, and demonstrated synergetic antibacterial activity in combination with commercial antibiotics against Pseudomonas aeruginosa

    Transtornos psiquiátricos na infância: diagnóstico precoce, intervenções eficazes e considerações éticas

    Reconhecer e abordar precocemente transtornos psiquiátricos na infância é vital, pois cerca de 20% das crianças enfrentam desafios que impactam emocionalmente. Com raízes genéticas evidentes desde o segundo trimestre fetal, a intervenção eficaz pode não apenas melhorar a qualidade de vida imediata, mas também reduzir o risco de complicações psiquiátricas ao longo da vida, exigindo uma gestão completa que leve em conta sintomas, contexto de desenvolvimento, ambiente familiar e estressores externos. Este estudo teve como objetivo investigar estratégias para o diagnóstico precoce, implementação de intervenções eficazes e considerações éticas no manejo de transtornos psiquiátricos na infância. Para o alcance desse propósito, realizou-se uma revisão sistemática da literatura, utilizando as base de dados Scielo, Lilacs e Medline. Com a análise dos resultados, concluiu-se que são indispensáveis avaliações clínicas extensas, aplicação de instrumentos psicométricos validados e investigação de fatores desencadeantes para garantir uma avaliação precisa. A participação ativa dos pais, especialmente na terapia cognitivo-comportamental (TCC), é destacada, assim como a necessidade de estratégias personalizadas de manejo. Além disso, a reflexão ética sobre fatores socioculturais e a consideração da criança como um sujeito de direitos são elementos cruciais para uma abordagem integral e contextualizada no cuidado de transtornos psiquiátricos na infância, visando promover o bem-estar duradouro
