    The star-switched MMC (SSMMC): a hybrid VSC for HVDC applications

    This paper presents a new hybrid VSC topology (the Star-Switched MMC) – suitable for HVDC applications. The basic structure and operating principles of the topology are described. Control strategies that regulate the power exchanged between the VSC, the AC network and the DC network are presented. A modulation strategy ensuring appropriate switching of the individual chain-link sub-modules and a capacitor voltage balancing algorithm that ensures the capacitor voltages are maintained within the required tolerance are discussed. Results from a simulation model are presented to validate the expected performance of the converter and the proposed control schemes

    Spatial analysis of malaria incidence at the village level in areas with unstable transmission in Ethiopia

    <p>Abstract</p> <p>Background</p> <p>Malaria is the leading cause of morbidity and mortality in Ethiopia, accounting for over five million cases and thousands of deaths annually. The risks of morbidity and mortality associated with malaria are characterized by spatial and temporal variation across the country. This study examines the spatial and temporal patterns of malaria transmission at the local level and implements a risk mapping tool to aid in monitoring and disease control activities.</p> <p>Methods</p> <p>In this study, we examine the global and local patterns of malaria distribution in 543 villages in East Shoa, central Ethiopia using individual-level morbidity data collected from six laboratory and treatment centers between September 2002 and August 2006.</p> <p>Results</p> <p>Statistical analysis of malaria incidence by sex, age, and village through time reveal the presence of significant spatio-temporal variations. Poisson regression analysis shows a decrease in malaria incidence with increasing age. A significant difference in the malaria incidence density ratio (IDRs) is detected in males but not in females. A significant decrease in the malaria IDRs with increasing age is captured by a quadratic model. Local spatial statistics reveals clustering or hot spots within a 5 and 10 km distance of most villages in the study area. In addition, there are temporal variations in malaria incidence.</p> <p>Conclusion</p> <p>Malaria incidence varies according to gender and age, with males age 5 and above showing a statistically higher incidence. Significant local clustering of malaria incidence occurs between pairs of villages within 1–10 km distance lags. Malaria incidence was higher in 2002–2003 than in other periods of observation. Malaria hot spots are displayed as risk maps that are useful for monitoring and spatial targeting of prevention and control measures against the disease.</p

    Nuclear Localized LSR: A Novel Regulator of Breast Cancer Behavior and Tumorigenesis

    Lipolysis Stimulated Lipoprotein Receptor (LSR) has been found in the plasma membrane and is believed to function in lipoprotein endocytosis and tight junctions. Given the impact of cellular metabolism and junction signaling pathways on tumor phenotypes and patient outcome, it is important to understand how LSR cellular localization mediates its functions. We conducted localization studies, evaluated DNA binding, and examined the effects of nuclear LSR in cells, xenografts, and clinical specimens. We found LSR within the membrane, cytoplasm, and the nucleus of breast cancer cells representing multiple intrinsic subtypes. Chromatin immunoprecipitation (ChIP) showed direct binding of LSR to DNA, and sequence analysis identified putative functional motifs and post-translational modifications of the LSR protein. While neither overexpression of transcript variants, nor pharmacological manipulation of post-translational modification significantly altered localization, inhibition of nuclear export enhanced nuclear localization, suggesting a mechanism for nuclear retention. Co-immunoprecipitation and proximal ligation assays indicated LSR-pericentrin interactions, presenting potential mechanisms for nuclear-localized LSR. The clinical significance of LSR was evaluated using data from over 1,100 primary breast tumors, which showed high LSR levels in basal-like tumors and tumors from African-Americans. In tumors histosections, nuclear localization was significantly associated with poor outcomes. Finally, in vivo xenograft studies revealed that basal-like breast cancer cells that over-express LSR exhibited both membrane and nuclear localization, and developed tumors with 100% penetrance, while control cells lacking LSR developed no tumors. These results show that nuclear LSR alters gene expression and may promote aggressive cancer phenotypes

    An epigenome-wide association study of child appetitive traits and DNA methylation

    The etiology of childhood appetitive traits is poorly understood. Early-life epigenetic processes may be involved in the developmental programming of appetite regulation in childhood. One such process is DNA methylation (DNAm), whereby a methyl group is added to a specific part of DNA, where a cytosine base is next to a guanine base, a CpG site. We meta-analyzed epigenome-wide association studies (EWASs) of cord blood DNAm and early-childhood appetitive traits. Data were from two independent cohorts: the Generation R Study (n = 1,086, Rotterdam, the Netherlands) and the Healthy Start study (n = 236, Colorado, USA). DNAm at autosomal methylation sites in cord blood was measured using the Illumina Infinium HumanMethylation450 BeadChip. Parents reported on their child's food responsiveness, emotional undereating, satiety responsiveness and food fussiness using the Children's Eating Behaviour Questionnaire at age 4–5 years. Multiple regression models were used to examine the association of DNAm (predictor) at the individual site- and regional-level (using DMRff) with each appetitive trait (outcome), adjusting for covariates. Bonferroni-correction was applied to adjust for multiple testing. There were no associations of DNAm and any appetitive trait when examining individual CpG-sites. However, when examining multiple CpGs jointly in so-called differentially methylated regions, we identified 45 associations of DNAm with food responsiveness, 7 associations of DNAm with emotional undereating, 13 associations of DNAm with satiety responsiveness, and 9 associations of DNAm with food fussiness. This study shows that DNAm in the newborn may partially explain variation in appetitive traits expressed in early childhood and provides preliminary support for early programming of child appetitive traits through DNAm. Investigating differential DNAm associated with appetitive traits could be an important first step in identifying biological pathways underlying the development of these behaviors.</p

    Validating and Linking the GIMMS Leaf Area Index (LAI3g) with Environmental Controls in Tropical Africa

    The recent Global Inventory Modeling and Mapping Studies (GIMMS) LAI3g product provides a 30-year global times-series of remotely sensed leaf area index (LAI), an essential variable in models of ecosystem process and productivity. In this study, we use a new dataset of field-based LAITrue to indirectly validate the GIMMS LAI3g product, LAIavhrr, in East Africa, comparing the distribution properties of LAIavhrr across biomes and environmental gradients with those properties derived for LAITrue. We show that the increase in LAI with vegetation height in natural biomes is captured by both LAIavhrr and LAITrue, but that LAIavhrr overestimates LAI for all biomes except shrubland and cropland. Non-linear responses of LAI to precipitation and moisture indices, whereby leaf area peaks at intermediate values and declines thereafter, are apparent in both LAITrue and LAIavhrr, although LAITrue reaches its maximum at lower values of the respective environmental driver. Socio-economic variables such as governance (protected areas) and population affect both LAI responses, although cause and effect are not always obvious: a positive relationship with human population pressure was detected, but shown to be an artefact of both LAI and human settlement covarying with precipitation. Despite these complexities, targeted field measurements, stratified according to both environmental and socio-economic gradients, could provide crucial data for improving satellite-derived LAI estimates, especially in the human-modified landscapes of tropical Africa.Peer reviewe

    Sustainable management practices for durum wheat production: Analyzing specific agronomic interventions on productivity, grain micronutrient content, and quality

    As compared with single agronomic crop management practices during grain formation, knowledge about integrated agronomic management practices on grain mineral composition and grain technological properties in durum wheat is limited. This knowledge is important for determining management strategies aimed at increasing grain yield without affecting grain nutritional quality. Integrated agronomic practices such as foliar nutrient application × seeding rate × varieties combined with growing locations were investigated to evaluate the dynamics of yield and grain quality traits. Two durum wheat varieties, three-level of micronutrients (i.e. control, FeSO4, and ZnSO4), and four levels of seeding rate (i.e. 100, 125, 150, and 175 kg ha−1) were arranged in split-split plot design under two different growing locations (environments). The main plots were assigned to the varieties, subplots to micronutrients, and sub-sub plots to the seeding rate treatments. Zinc and iron were applied in a form of ZnSO4 and FeSO4 at the early flowering stage, both at a rate of 25 kg ha−1. Results showed a linear increment in biomass (21.5%) and grain yield (23.5%) under a high seeding rate, even though the 1000-grain weight, the number of grains spike−1, spike length, and the number of grains spike−1 were decreased. Higher varietal and environmental response of seeding rate was observed between varieties. The grain protein content, gluten, and zeleyn index decreased as the seeding rate increased. Grain micronutrient content was significantly influenced by seeding rate and varietal difference. The grain protein content was higher in a dryland environment than in a wet environment. A combined use of density-tolerant varieties, high seeding rate, and foliar-based iron application can improve the grain yield from 2.01 to 3.20 t ha−1 under a potential environment. Hence, all stakeholders should consider the genotype (G), environment (E), management (M), and their synergies, as far as grain yield and quality are considered simultaneously

    The variable prevalence of bovine tuberculosis among dairy herds in Central Ethiopia provides opportunities for targeted intervention.

    Bovine tuberculosis (bTB) is an important disease for dairy productivity, as well as having the potential for zoonotic transmission. Previous prevalence studies of bTB in the dairy sector in central Ethiopia have suggested high prevalence, however, they have been limited to relatively small scale surveys, raising concerns about their representativeness. Here we carried out a cross sectional one-stage cluster sampling survey taking the dairy herd as a cluster to estimate the prevalence of bTB in dairy farms in six areas of central Ethiopia. The survey, which to date is by far the largest in the area in terms of the number of dairy farms, study areas and risk factors explored, took place from March 2016 to May 2017. This study combined tuberculin skin testing and the collection of additional herd and animal level data by questionnaire to identify potential risk factors contributing to bTB transmission. We applied the single intradermal cervical comparative tuberculin (SICCT) test using >4mm cut-off for considering an individual animal as positive for bTB; at least one reactor animal was required for a herd to be considered bTB positive. Two hundred ninety-nine dairy herds in the six study areas were randomly selected, from which 5,675 cattle were tested. The overall prevalence of bTB after standardisation for herd-size in the population was 54.4% (95% CI 48.7-60%) at the herd level, and it was 24.5% (95% CI 23.3-25.8) at the individual animal level. A Generalized Linear Mixed Model (GLMM) with herd and area as random effect was used to explore risk factors association with bTB status. We found that herd size, age, bTB history at farm, and breed were significant risk factors for animals to be SICCT positive. Animals from large herds had 8.3 times the odds of being tuberculin reactor (OR: 8.3, p-value:0.008) as compared to animals from small herds. The effect of age was strongest for animals 8-10 years of age (the oldest category) having 8.9 times the odds of being tuberculin reactors (OR: 8.9, p-value:<0.001) compared to the youngest category. The other identified significant risk factors were bTB history at farm (OR: 5.2, p-value:0.003) and cattle breed (OR: 2.5, p-value: 0.032). Our study demonstrates a high prevalence of bTB in central Ethiopia but with a large variation in within-herd prevalence between herds, findings that lays an important foundation for the future development of control strategies

    In silico analyses of diversity and dissemination of antimicrobial resistance genes and mobile genetics elements, for plasmids of enteric pathogens

    IntroductionThe antimicrobial resistance (AMR) mobilome plays a key role in the dissemination of resistance genes encoded by mobile genetics elements (MGEs) including plasmids, transposons (Tns), and insertion sequences (ISs). These MGEs contribute to the dissemination of multidrug resistance (MDR) in enteric bacterial pathogens which have been considered as a global public health risk.MethodsTo further understand the diversity and distribution of AMR genes and MGEs across different plasmid types, we utilized multiple sequence-based computational approaches to evaluate AMR-associated plasmid genetics. A collection of 1,309 complete plasmid sequences from Gammaproteobacterial species, including 100 plasmids from each of the following 14 incompatibility (Inc) types: A/C, BO, FIA, FIB, FIC, FIIA, HI1, HI2, I1, K, M, N, P except W, where only 9 sequences were available, was extracted from the National Center for Biotechnology Information (NCBI) GenBank database using BLAST tools. The extracted FASTA files were analyzed using the AMRFinderPlus web-based tools to detect antimicrobial, disinfectant, biocide, and heavy metal resistance genes and ISFinder to identify IS/Tn MGEs within the plasmid sequences.Results and DiscussionIn silico prediction based on plasmid replicon types showed that the resistance genes were diverse among plasmids, yet multiple genes were widely distributed across the plasmids from enteric bacterial species. These findings provide insights into the diversity of resistance genes and that MGEs mediate potential transmission of these genes across multiple plasmid replicon types. This notion was supported by the observation that many IS/Tn MGEs and resistance genes known to be associated with them were common across multiple different plasmid types. Our results provide critical insights about how the diverse population of resistance genes that are carried by the different plasmid types can allow for the dissemination of AMR across enteric bacteria. The results also highlight the value of computational-based approaches and in silico analyses for the assessment of AMR and MGEs, which are important elements of molecular epidemiology and public health outcomes
