    A New Approach for Accurate Identification of Allele State Based on Real-Time PCR for Biomedical Tasks

    Genotyping of single nucleotide polymorphisms (SNPs) is an important task in medicine, veterinary medicine and biology. Precise differentiation of SNPs can be challenging. Methods based onTaqman can lead to false positive results due to nonspecific annealing of the probe. The aim of this research was to develop a new approach for the accurate differentiation of SNPs based on real-time PCR with Taqmanprobes and their rivals.The rivals competed with the Taqmanprobes for annealing to the site. The rivals blocked the nonspecific allele so that the Taqmanprobe could not anneal to it. Thus,the Taqmanprobe only detected specific alleles.This approach madeit possible to fine-tune the diagnostic system by selecting the ratio of Taqmanprobes and rivals (in non-equimolar amounts too).The new approach was tested on several diagonally significant SNPs in veterinary medicine.Using Taqman probes and rival probes showed a significantly greater specificity and efficiency in the determination of both homozygotes and heterozygotes than when conventional systems based only on Taqmanwere used.     The dissolution test was conducted to determine the effect of the formulation andfabrication processes on the dissolution profile in estimating the bioavailability andbioequivalence between the test and comparison products. The comparable dissolutiontest used 3 different dissolution mediums were hydrochloric acid pH 1,2 or simulatedgastric fluid without enzymes, citrate buffer medium pH 4.5 and phosphate buffer medium pH 6.8 or simulated intestinal fluid without enzymes. The first study used 12 tablets of Panadol which were randomly selected. Then, the results were comparedusing two independent model methods were the difference factor (f1) and the similarity factor (f2). Based on this, the similarity factor in the three media is in the acceptablerange of 50-100. Another study conducted testing using acetaminophen samples purchased fromRochem, Turkey. The results obtained were in accordance with USP guidelines namelyconventional acetaminophen tablets should be released at at least 80% of the labeledamount in 30 minutes. In another study also compared the dissolution profile of 4 coatedand noncoated tablets. All formulations within the first 45 minutes had a drug release of more than 85%

    Enabling IoT ecosystems through platform interoperability

    Today, the Internet of Things (IoT) comprises vertically oriented platforms for things. Developers who want to use them need to negotiate access individually and adapt to the platform-specific API and information models. Having to perform these actions for each platform often outweighs the possible gains from adapting applications to multiple platforms. This fragmentation of the IoT and the missing interoperability result in high entry barriers for developers and prevent the emergence of broadly accepted IoT ecosystems. The BIG IoT (Bridging the Interoperability Gap of the IoT) project aims to ignite an IoT ecosystem as part of the European Platforms Initiative. As part of the project, researchers have devised an IoT ecosystem architecture.     Spatiotemporal spatfall dynamics and prevailing estuarine conditions for optimal oyster (Crassostrea tulipa) spat availability in selected Gulf of Guinea brackish systems

    Uncertainties associated with wild harvests of seed and adult oysters due to unknown oceanographic oscillations are a major challenge in oyster fisheries and aquaculture development. In contribution to addressing this challenge, we proffer clarity on the spatiotemporal variations in spatfall (number of spat/m2) of the mangrove oyster Crassostrea tulipa (Lamarck, 1819) in four estuaries along the Gulf of Guinea coast. By monthly deployment of artificial substrates affixed to bamboo racks over 12 months, we find significant differences in spatfall among and within the brackish systems, and across months and seasons. Spatfall regimes were unique in each ecosystem albeit with an overall preponderance of dry season availability of spat. Locations with reef oysters had superior spatfall to mangrove root-adapted-oyster areas. Narkwa, a relatively small lagoon with reef oysters had the highest annual mean spatfall, which was 1.3, 2.5, and 9.8 folds the spatfall in Densu Delta, Benya Lagoon and Whin Estuary, respectively. Spatfall varied significantly by depth as the more frequently exposed top collectors harvested much less spat than submerged collectors. There was a year-round availability of spat, confirming continuous spawning in C. tulipa. Spatfall variability was significantly driven by fluctuations in prevailing dissolved oxygen and salinity. Prevailing dissolved oxygen and salinity levels in the estuaries for optimal spat settlement were 1.68 – 3.40 mg L-1 and 11.00 – 29.33 ppt (parts per thousand), respectively. The findings of this study are recommended as empirical reference points for sustainable seed procurement for aquaculture production and management of C. tulipa fishery in the region of the study

    Complex regional pain syndrome type I (CRPS I) is a disorder of one or more extremities characterized by pain, ab- normal sensitivity (allodynia), swelling, limited range of motion, vasomotor instability, fatigue and emotional distress. The symptoms may be aggravated by even minor activity or weather change. It is usually provoked by injury, surgery or injection but in a small proportion of patients CRPS I develops without a clear causative event. There are several litera- ture reports on CRPS after rubella and hepatitis B vaccination. We present a case of CRPS I affecting the left arm after diphtheria and tetanus (Di-Te) vaccination in the left deltoid muscle in a young girl having experienced profound emo- tional stress before the vaccination procedure. History data on previous minor trauma at the site of vaccination or emo- tional stress may necessitate temporary vaccination delay due to their proneness to impaired local or systemic immune response and CRPS as a complication of vaccination. If a child or an adult has prominent swelling and severe pain after vaccination, the diagnosis of CRPS I should be considered and if confirmed, the multidisciplinary treatment should start as soon as possible

    Sexual Dimorphism in Hematocrit Response Following Red Blood Cell Transfusion of Critically Ill Surgical Patients

    The change in hematocrit (ΔHct) following packed red blood cell (pRBCs) transfusion is a clinically relevant measurement of transfusion efficacy that is influenced by post-transfusion hemolysis. Sexual dimorphism has been observed in critical illness and may be related to gender-specific differences in immune response. We investigated the relationship between both donor and recipient gender and ΔHct in an analysis of all pRBCs transfusions in our surgical intensive care unit (2006–2009). The relationship between both donor and recipient gender and ΔHct (% points) was assessed using both univariate and multivariable analysis. A total of 575 units of pRBCs were given to 342 patients; 289 (49.9%) donors were male. By univariate analysis, ΔHct was significantly greater for female as compared to male recipients (3.81% versus 2.82%, resp., P < 0.01). No association was observed between donor gender and ΔHct, which was 3.02% following receipt of female blood versus 3.23% following receipt of male blood (P = 0.21). By multivariable analysis, recipient gender remained associated significantly with ΔHct (P < 0.01). In conclusion, recipient gender is independently associated with ΔHct following pRBCs transfusion. This association does not appear related to either demographic or anthropomorphic factors, raising the possibility of gender-related differences in recipient immune response to transfusion

    Clinical Evolution and Risk Factors of Hospitalized Patients with COVID-19, Haiti, March-June 2020

    In December 2019, a coronavirus epidemic emerged in China. Within months, the epidemic was considered a public health emergency of international concern. In Haiti, the first laboratory-confirmed cases of COVID-19 were reported on March 19, 2020, in a context where there was some limitations of knowledge on the modes of transmission, the severity, the clinical characteristics and the risk factors of the disease. This study has made it possible to characterize the epidemic and investigate the associations between the risk factors, co morbidities, and clinical evolution of the disease. To develop the epidemiological and clinical profile of patients with COVID-19 in Haiti, data were collected from the clinical records of patients hospitalized for COVID-19 from March 16 to June 16, 2020, in 22 healthcare facilities. Univariate, bivariate, and logistic regression model analyses were performed to describe and explore the risk factors, comorbidities, and treatments associated with patients’ clinical evolution. Statistical significance was determined using a 95% confidence interval or p-value of ?0.05.  Diabetes and high blood pressure were the main comorbidities that had a statistically significant association with the severe form of the disease and the occurrence of death. The likelihood of dying increased with age, and patients in the severe form were almost four times more likely to die. The administration of ceftriaxone to patients was significantly related to recovery from the disease. Diabetes, high blood pressure, and age were the major risk factors for the severity and mortality of people infected with COVID19. Ceftriaxone administration was protective against recovery

    Physical distancing and risk of COVID-19 in small-scale fisheries: a remote sensing assessment in coastal Ghana

    The novel coronavirus is predicted to have dire implications on global food systems including fisheries value chains due to restrictions imposed on human movements in many countries. In Ghana, food production, both agriculture and fisheries, is exempted from restrictions as an essential service. The enforcement of COVID-19 prevention protocols, particularly social distancing, has been widely reported in Ghana's agricultural markets whereas casual observations and media reports on fish landing sites suggest no such enforcements are in place. This study aimed to provide sound scientific evidence as a basis for informed policy direction and intervention for the artisanal fishing sector in these challenging times. We employed an unmanned aerial vehicle in assessing the risk of artisanal fishers to the pandemic using physical distancing as a proxy. From analysis of cumulative distribution function (G-function) of the nearest-neighbour distances, this study underscored crowding at all surveyed fish landing beaches, and identified potential "hotspots" for disease transmission. Aerial measurements taken at times of peak landing beach activity indicated that the highest proportion of people, representing 56%, 48%, 39% and 78% in Elmina, Winneba, Apam and Mumford respectively, were located at distances of less than one metre from their nearest neighbour. Risk of crowding was independent of the population at the landing beaches, suggesting that all categories of fish landing sites along the coast would require equal urgency and measured attention towards preventing and mitigating the spread of the disease
