Gut microbiota imbalance and colorectal cancer.
International audienceThe gut microbiota acts as a real organ. The symbiotic interactions between resident micro-organisms and the digestive tract highly contribute to maintain the gut homeostasis. However, alterations to the microbiome caused by environmental changes (e.g., infection, diet and/or lifestyle) can disturb this symbiotic relationship and promote disease, such as inflammatory bowel diseases and cancer. Colorectal cancer is a complex association of tumoral cells, non-neoplastic cells and a large amount of micro-organisms, and the involvement of the microbiota in colorectal carcinogenesis is becoming increasingly clear. Indeed, many changes in the bacterial composition of the gut microbiota have been reported in colorectal cancer, suggesting a major role of dysbiosis in colorectal carcinogenesis. Some bacterial species have been identified and suspected to play a role in colorectal carcinogenesis, such as Streptococcus bovis, Helicobacter pylori, Bacteroides fragilis, Enterococcus faecalis, Clostridium septicum, Fusobacterium spp. and Escherichia coli. The potential pro-carcinogenic effects of these bacteria are now better understood. In this review, we discuss the possible links between the bacterial microbiota and colorectal carcinogenesis, focusing on dysbiosis and the potential pro-carcinogenic properties of bacteria, such as genotoxicity and other virulence factors, inflammation, host defenses modulation, bacterial-derived metabolism, oxidative stress and anti-oxidative defenses modulation. We lastly describe how bacterial microbiota modifications could represent novel prognosis markers and/or targets for innovative therapeutic strategies
Central nesfatin-1-expressing neurons are sensitive to peripheral inflammatory stimulus
Recently, a novel factor with anorexigenic properties was identified and called nesfatin-1. This protein (82 aac) is not only expressed in peripheral organs but it is also found in neurons located in specific structures including the hypothalamus and the brainstem, two sites strongly involved in food intake regulation. Here, we studied whether some of the neurons that become activated following an injection of an anorectic dose of lipopolysaccharides (LPS) exhibit a nesfatin-1 phenotype. To this end, we used double immunohistochemistry to target the expression of the immediate-early gene c-fos and of nesfatin-1 on coronal frozen sections of the rat brain. The number of c-Fos+/nesfatin-1+ neurons was evaluated in the immunosensitive structures reported to contain nesfatin-1 neurons; i.e. paraventricular hypothalamic nucleus (PVN), supraoptic nucleus (SON), arcuate nucleus (ARC) and nucleus of the solitary tract (NTS). LPS strongly increased the number of c-Fos+/nesfatin-1+ neurons in the PVN, SON and NTS, and to a lesser extent in the ARC. Triple labeling showed that a portion of the nesfatin-1 neurons activated in response to LPS within the NTS are catecholaminergic since they co-express tyrosine hydroxylase (TH). Our data therefore indicate that a portion of nesfatin-1 neurons of both the hypothalamus and brainstem are sensitive to peripheral inflammatory signals, and provide the first clues suggesting that centrally released nesfatin-1 may contribute to the neural mechanisms leading to endotoxaemic anorexia
A discrete element study of settlement in vibrated granular layers: role of contact loss and acceleration
This paper deals with the vibration of granular materials due to cyclic
external excitation. It highlights the effect of the acceleration on the
settlement speed and proves the existence of a relationship between settlement
and loss of contacts in partially confined granular materials under vibration.
The numerical simulations are carried out using the Molecular Dynamics method,
where the discrete elements consist of polygonal grains. The data analyses are
conducted based on multivariate autoregressive models to describe the
settlement and permanent contacts number with respect to the number of loading
cycles
Digital phantoms for the evaluation of a software used for an automatic analysis of the Winston-Lutz test in image guided radiation therapy
11 pagesInternational audienceAccurate isocentre positioning of the treatment machine is essential for the radiation therapy process, especially in stereotactic radio surgery and in image guided radiation therapy. We present in this paper a new method to evaluate a software which is used to perform an automatic analysis of the Winston-Lutz1, 2 test used in order to determine position and size of the isocentre. The method consists of developing digital phantoms that simulate mechanical distortions of the treatment machine as well as misalignments of the positioning laser targeting the isocentre. These Digital Test Objects (DTOs) offer a detailed and profound evaluation of the software and allow determining necessary adjustments which lead to high precision and therefore contributes to a better treatment targeting
Histology of two rice bodies isolated from the stifle of an adult draught horse stallion
In the human and equine species, different kinds of free floating intra-articular particles are related to certain disorders. Osteochondral fragments formed during osteochondrosis dissecans are the most common finding in the equine species, whereas in humans rice bodies due to rheumatoid arthritis are more frequent. Herein we report a third type of floating body inside the stifle of an adult draught horse stallion, in macroscopic appearance similar to articular rice bodies known in humans. As revealed by histologic examination, the two particles consist of polypoid degenerated structures derived from synovial villi. Their formation was probably induced by ischemia
Two Cases of Pulmonary Tuberculosis Caused by Mycobacterium tuberculosis subsp. canetti
We identified an unusual strain of mycobacteria from two patients with pulmonary tuberculosis by its smooth, glossy morphotype and, primarily, its genotypic characteristics. Spoligotyping and restriction fragment length polymorphism typing were carried out with the insertion sequence IS6110 patterns. All known cases of tuberculosis caused by Mycobacterium canetti have been contracted in the Horn of Africa
