    Aging in the Cardiovascular System: Lessons from Hutchinson-Gilford Progeria Syndrome

    Aging, the main risk factor for cardiovascular disease (CVD), is becoming progressively more prevalent in our societies. A better understanding of how aging promotes CVD is therefore urgently needed to develop new strategies to reduce disease burden. Atherosclerosis and heart failure contribute significantly to age-associated CVD-related morbimortality. CVD and aging are both accelerated in patients suffering from Hutchinson-Gilford progeria syndrome (HGPS), a rare genetic disorder caused by the prelamin A mutant progerin. Progerin causes extensive atherosclerosis and cardiac electrophysiological alterations that invariably lead to premature aging and death. This review summarizes the main structural and functional alterations to the cardiovascular system during physiological and premature aging and discusses the mechanisms underlying exaggerated CVD and aging induced by prelamin A and progerin. Because both proteins are expressed in normally aging non-HGPS individuals, and most hallmarks of normal aging occur in progeria, research on HGPS can identify mechanisms underlying physiological aging.Progeria Research Foundation (Established Investigator Award 2014-52).     Podcasting as a popular science media tool and its ability to connect with the audience

    El periodismo científico se enfrenta a la dificultad de conectar con públicos no especializados y para ello recurre al uso de diversas plataformas digitales generando narrativas transmedia. El auge del podcasting demuestra la capacidad de formato audio online para generar comunidades en torno a plataformas de divulgación científica. El presente estudio analiza los canales Ciencia en el bar y Radio Skylab encuadrados bajo el paraguas de la marca global Naukas, para contrastar la capacidad del vídeo y el audio al servicio de la divulgación científica y su capacidad para conectar con la audiencia. El análisis recoge la interacción de los usuarios en ambos canales de (plataformas YouTube e Ivoox) para comparar índices de respuesta y valor de su participación. Los resultados apuntan a que la duración de los programas no es determinante como factor de éxito entre el público y, además, en ambos casos, la participación de la audiencia genera importantes expansiones de contenido y tiene un importante compromiso con el mismo.Scientific journalism faces the difficulty of connecting with non-specialized audiences and for this it resorts to the use of diverse digital platforms generating transmedia narratives. The rise of podcasting demonstrates the ability of online audio format to generate communities around platforms for scientific dissemination. This study analyzes the Science channels in the bar and Radio Skylab framed under the umbrella of the global brand Naukas, to contrast the capacity of video and audio at the service of scientific dissemination and its ability to connect with the audience. The analysis gathers the interaction of the users in both channels (YouTube and Ivoox platforms) to compare response rates and the value of their participation. The results suggest that the duration of the programs is not a determining factor as a factor of success among the public and, in addition, in both cases, the participation of the audience generates important expansions of content and has an important commitment to the same

    Vascular Smooth Muscle Cell-Specific Progerin Expression Provokes Contractile Impairment in a Mouse Model of Hutchinson-Gilford Progeria Syndrome that Is Ameliorated by Nitrite Treatment

    Cardiovascular disease (CVD) is the main cause of death worldwide, and aging is its leading risk factor. Aging is much accelerated in Hutchinson-Gilford progeria syndrome (HGPS), an ultra-rare genetic disorder provoked by the ubiquitous expression of a mutant protein called progerin. HGPS patients die in their teens, primarily due to cardiovascular complications. The primary causes of age-associated CVD are endothelial dysfunction and dysregulated vascular tone; however, their contribution to progerin-induced CVD remains poorly characterized. In the present study, we found that progeroid LmnaG609G/G609G mice with ubiquitous progerin expression show both endothelial dysfunction and severe contractile impairment. To assess the relative contribution of specific vascular cell types to these anomalies, we examined LmnaLCS/LCSTie2Cretg/+ and LmnaLCS/LCSSm22αCretg/+ mice, which express progerin specifically in endothelial cells (ECs) and vascular smooth muscle cells (VSMCs), respectively. Whereas vessel contraction was impaired in mice with VSMC-specific progerin expression, we observed no endothelial dysfunction in mice with progerin expression restricted to VSMCs or ECs. Vascular tone regulation in progeroid mice was ameliorated by dietary sodium nitrite supplementation.     Effect of Superior Ovarian Nerve and Plexus Nerve Sympathetic Denervation on Ovarian-Derived Infertility Provoked by Estradiol Exposure to Rats

    Sympathetic innervation of the ovary in rodents occurs via two routes: the superior ovarian nerve (SON), which runs along the ovarian ligament, and the plexus nerve (PN), which is mainly associated with the vasculature. SON and ovarian norepinephrine (NE) levels play a major role in regulating ovarian cystic health. Although it was previously described that the polycystic ovarian phenotype (PCO) is causally related to hyperstimulation of the sympathetic nerves of the ovary, much less is known, however, regarding the role of PN in ovarian physiology. We studied the role of SON and PN in relation to the maintenance of the PCO phenotype induced in the rat by exposure to estradiol valerate (EV). EV exposure at 24 days old (juvenile exposure) increases NE in the ovary for up to 90 days after EV injection. SON or PN denervation (SONX and PNX) decreased NE. SONXreversed the acyclic condition from 30 days after surgery (p &lt; 0.05), but PNXdid not. SONX was more effective than PNX to downregulate the increased number of cysts induced by EV, with the presence of the corpora lutea (CL, signifying ovulation) in the SONX group. Seventy percent of SONX rats presented with pregnancy at 60 days post-EV (6 of the 7 sperm-positive rats were pregnant); however, SONX rats had a reduced number (half) of pups compared with vehicle-treated rats and 60% more pups than EV rats. These data suggest that the SON plays a predominant role in follicular development, ovulation and pregnancy during ovarian diseases
