    Electrophoretic Analysis of Total and Membrane Proteins of Xanthomonas campestris Pathovars, the Causal Agents of the Leaf Streak of Cereals and Grasses in Iran

    Forty‐five Iranian isolates of Xanthomonas campestris obtained from wheat, barley and grasses were compared with reference strains using polyacrylamide gel electrophoresis (PAGE) of the whole‐cell and membrane proteins. The PAGE profiles of the whole‐cell and membrane proteins of the Iranian isolates obtained from barley, with the exception of IBLS 11 and IBLS 12, were identical and clearly distinguishable from those of the other isolates. The barley group isolates, which were pathogenic only to barley, were similar to UPB 458 (NCPPB 2389), the reference strain of pathovar hordei. The isolates obtained from wheat and grasses, as well as IBLS 11 and IBLS 12, which can infect wheat, barley and some wild grasses, had similar banding patterns: only IBLS 40 isolated from Hordeum sp. displayed the same profile as the barley group. Reference strains UPB 443 (NCPPB 2821) and UPB 513, which correspond to pathovars undulosa and translucens , respectively, were related to the wheat group. IBLS45, isolated from Bromus sp., had a banding pattern that differed from those observed for strains of the barley and wheat groups. The results suggest that this method can be useful for discriminating different pathovars of X. campestris attacking cereals and grasses, and sodium dodecyl sulphate (SDS)‐PAGE of membrane proteins was not more sensitive than SDS‐PAGE of total proteins for differentiating the isolates

    A cDNA microarray approach to decipher sunflower (Helianthus annuus) responses to the necrotrophic fungus Phoma macdonaldii

    To identify the genes involved in the partial resistance of sunflower (Helianthus annuus) to the necrotrophic fungus Phoma macdonaldii, we developed a 1000‐element cDNA microarray containing carefully chosen genes putatively involved in primary metabolic pathways, signal transduction and biotic stress responses. A two‐pass general linear model was used to normalize the data and then to detect differentially expressed genes. This method allowed us to identify 38 genes differentially expressed among genotypes, treatments and times, mainly belonging to plant defense, signaling pathways and amino acid metabolism. Based on a set of genes whose differential expression was highly significant, we propose a model in which negative regulation of a dual‐specificity MAPK phosphatase could be implicated in sunflower defense mechanisms against the pathogen. The resulting activation of the MAP kinase cascade could subsequently trigger defense responses (e.g. thaumatin biosynthesis and phenylalanine ammonia lyase activation), under the control of transcription factors belonging to MYB and WRKY families. Concurrently, the activation of protein phosphatase 2A (PP2A), which is implicated in cell death inhibition, could limit pathogen development. The results reported here provide a valuable first step towards the understanding and analysis of the P. macdonaldii–sunflower interaction

    Genetic analysis of partial resistance to bacterial leaf streak ( Xanthomonas campestris pv. cerealis ) in wheat

    Genetic variability of partial resistance to bacterial leaf streak was investigated in hexaploid winter wheat (Triticum aestivum. ), using 16 parental genotypes and 48 pure lines (F10) derived from a composite cross programme. Two experiments were undertaken in a controlled growth chamber. Seeds of all genotypes were grown under controlled conditions using a randomized block design with three replications. Each replication consisted of a row of 20 seedlings of each parent and pure line. An Iranian strain of bacterial leaf streak was used for the inoculation of 12‐day‐old seedlings. In a third experiment, eight genotypes from parents and F10 pure lines representing a large variability for partial resistance were inoculated with four other Iranian strains of bacterial leaf streak. A large genetic variability was observed amongst the 64 genotypes for partial resistance to the disease. Partial resistance heritability estimates were rather high (70%), indicating that the resistance factors may be transmitted by crossing. Amongst all genotypes investigated, ‘DC2‐30‐N2’ and ‘IBPT‐66’ displayed the highest partial resistance to the disease. Significant correlations between strains in the third experiment show that a genotype resistant or susceptible to one strain will have similar reactions with other strains. No significant genetic gain was observed for partial resistance in the best pure line of the 48 lines studied, when compared with the best parental line. Increasing the number of pure lines is likely to result in the identification of genotypes that might prove to be more resistant

    Zimbabwean diabetics' beliefs about health and illness: an interview study

    <p>Abstract</p> <p>Background</p> <p>Diabetes mellitus (DM) is increasing globally, with the greatest increase in Africa and Asia. In Zimbabwe a threefold increase was shown in the 1990s. Health-related behaviour is important in maintaining health and is determined by individual beliefs about health and illness but has seen little study. The purpose of the study was to explore beliefs about health and illness that might affect self-care practice and health care seeking behaviour in persons diagnosed with DM, living in Zimbabwe.</p> <p>Methods</p> <p>Exploratory study. Consecutive sample from a diabetes clinic at a central hospital. Semi-structured interviews were held with 21 persons aged 19-65 years. Data were analysed using qualitative content analysis.</p> <p>Results</p> <p>Health was described as freedom from disease and well-being, and individual factors such as compliance with advice received and drugs were considered important to promote health. A mixture of causes of DM, predominantly individual factors such as heredity, overweight and wrong diet in combination with supernatural factors such as fate, punishment from God and witchcraft were mentioned. Most respondents did not recognize the symptoms of DM when falling ill but related the problems to other diseases, e.g. HIV, malaria etc. Limited knowledge about DM and the body was indicated. Poor economy was mentioned as harmful to health and a consequence of DM because the need to buy expensive drugs, food and attend check-ups. Self-care was used to a limited extent but if used, a combination of individual measures, household remedies or herbs and religious acts such as prayers and holy water were frequently used, and in some cases health care professionals were consulted.</p> <p>Conclusions</p> <p>Limited knowledge about DM, based on beliefs about health and illness including biomedical and traditional explanations related to the influence of supernatural forces, e.g. fate, God etc., were found, which affected patients' self-care and care-seeking behaviour. Strained economy was stated to be a factor of the utmost importance affecting the management of DM and thus health. To develop cost-effective and optimal diabetes care in a country with limited resources, not only educational efforts based on individual beliefs are needed but also considering systemic and structural conditions in order to promote health and to prevent costly consequences of DM.</p

    Nature and origin of the Triassic volcanism in Albania and Othrys: a key to understanding the Neotethys opening?

    International audienceTriassic volcanic rocks, stratigraphically associated with pelagic or reef limestones, are tectonically juxtaposed with Mesozoic ophiolites in the Tethyan realm. From the central (Dinarides, Hellenides) and eastern Mediterranean (Antalya, Troodos, Baër Bassit) to the Semail nappes (Oman), they occur either associated to the tectonic sole of the ophiolitic nappes or as a distinct tectonic pile intercalated between the ophiolites and other underthrust units. In the Dinaro-Hellenic belt, the Pelagonian units represent the lower plate, which is underthrust beneath the ophiolites. Middle to Late Triassic volcanic sequences are interpreted as the eastern flank of the Pelagonian platform and are therefore considered as a distal, deep-water part of the Pelagonian margin. The Triassic volcanics from Albania and Othrys are made up of basaltic pillowed and massive flows, associated locally with dolerites and trachytes. New elemental, Nd and Pb isotopic data allow to recognize four types of volcanic suites: (1) intra-oceanic alkaline and tholeiitic basalts, (2) intra-oceanic arc-tholeiites, (3) back-arc basin basalts, (4) calc-alkaline mafic to felsic rocks. Nd and Pb isotopic initial ratios suggest that the within-plate volcanic rocks were derived from an enriched oceanic island basalt type mantle source, devoid of any continental crustal component. The lower {varepsilon}Nd value of the trachyte could be due to assimilation of oceanic altered crust or sediments in a shallow magma chamber. Island arc tholeiites and back-arc basin basalts have a similar wide range of {varepsilon}Nd. The absence of Nb negative anomalies in the back-arc basin basalts suggests that the basin floored by these basalts was wide and mature. The high Th contents of the island arc tholeiites suggest that the arc volcanoes were located not far away from the continental margin. Albania and Othrys volcanics contrast with the Late Triassic volcanism from eastern Mediterranean (SW Cyprus, SW Turkey), which displays solely features of oceanic within plate suites. The presence of back-arc basin basalts associated with arc-related volcanics in Central Mediterranean indicates that they were close to a still active subduction during the Upper Triassic, while back-arc basins developed, associated with within-plate volcanism, leading to the NeoTethys opening
