    As humans began to grow in numbers, they began to create civilizations for themselves in order to better survive, and as those civilizations grew, there came to be a divergence of roles for people to perform. The most universal of all these was the Warrior Elite, a class of people who were part of the lesser nobility and would function as officers and generals in times of conflict. For Japan it was the Samurai, for Iran it was the Persian Immortals, and for Europe in the 9th to late 15th century, there were the Knights. Let us look at what knights were, how they grew and changed through history, their eventual decline, and even a few chivalric orders

    Designing Service-Learning to Enhance Social Justice Commitments: A Critical Reflection Tool

    The COVID-19 pandemic—coupled with ongoing prominent injustice related to race, poverty, healthcare, and education—has highlighted the interlocking and reinforcing nature of systemic oppression. Now more than ever, facilitators of experiential learning are galvanized to explore and deepen their understanding of systemic change and to enhance their teaching of justice concepts, perspectives, and skills. Advancing social justice was a part of the original vision for service-learning (Stanton et al., 1999). However, scholars have long identified the ways in which service-learning can perpetuate inequitable social hierarchies, be miseducative in teaching simplistic understandings of solutions to social problems, and not equip students to participate in social movements for justice (Eby, 1998; Mitchell and Latta, 2020). By incorporating critical pedagogy, action, and reflection in and beyond the classroom, “critical service-learning” seeks to increase understanding of oppression, redistribute power, and find solutions to social issues through community-campus partnerships (Mitchell, 2008; Santiago-Ortiz, 2019). However, many service-learning practitioners report uncertainty about how to take concrete steps to align their courses with justice orientated techniques and pedagogy. In order to address this need, students, staff, and faculty in Duke Service-Learning began creating a tool to support the implementation of critical pedagogy and justice in service-learning courses in 2016 (Stith et al., 2018). For the past 8 months an expanded group has worked to update that original tool. The resulting tool organizes course design choices into five themes: reckoning with systems, authentic relationships, redistribution of power, equitable classrooms & cognitive justice, and social change skills. The resulting tool generates concrete, actionable questions for service-learning and experiential learning to interrogate how their pedagogy and practices incorporate social, economic, environmental, and racial justice. Our article summarizes the advances in our understanding and of the literature on implementing justice aligned experiential education and links to the current version of our tool. Further, our article will summarize lessons learned from conversations with practitioners who use and reflect on the implications of the tool for their own pedagogy. We will also reflect on holding the tensions in balancing the various aspects of course designs and our commitments to anti-oppressive practices. Finally, we will reflect on further implications for service-learning and experiential education that the process of creating this tool has surfaced

    Coupled ecological and management connectivity across administrative boundaries in undeveloped landscapes

    Human-induced ecological boundaries, or anthropogenic ecotones, may arise where administrative boundaries meet on undeveloped lands. Landscape-level ecological processes related to factors such as fire, invasive species, grazing, resource extraction, wildlife, and water may be affected due to unique management strategies adopted by each administrative unit. Over time, different management can result in discernible ecological differences (e.g., species composition or soil characteristics). Thus, fragmentation in the management landscape can correspond to ecological fragmentation. Different ecological patterns may emerge due to an increase in the number of management units in a region, or due to an increase in the number of different types of management units in the region. Temporal effects and collaboration history can also affect the emergence of ecotones. We use conceptual models to explore the relationship between these aspects of management fragmentation and the anthropogenic ecotones between management parcels. We then use examples of different management challenges to explore how anthropogenic ecotones can disrupt ecological flows. Our models suggest that cross-boundary collaboration that enhances management connectivity is likely essential to ecological connectivity in the face of environmental and social change

    Guidelines of the American Society of Mammalogists for the use of wild mammals in research

    Guidelines for use of wild mammal species are updated from the American Society of Mammalogists (ASM) 2007 publication. These revised guidelines cover current professional techniques and regulations involving mammals used in research and teaching. They incorporate additional resources, summaries of procedures, and reporting requirements not contained in earlier publications. Included are details on marking, housing, trapping, and collecting mammals. It is recommended that institutional animal care and use committees (IACUCs), regulatory agencies, and investigators use these guidelines as a resource for protocols involving wild mammals. These guidelines were prepared and approved by the ASM, working with experienced professional veterinarians and IACUCs, whose collective expertise provides a broad and comprehensive understanding of the biology of nondomesticated mammals in their natural environments.     Pharmacogenomic Analyses Implicate B Cell Developmental Status and MKL1 as Determinants of Sensitivity toward Anti-CD20 Monoclonal Antibody Therapy

    Monoclonal antibody (mAb) therapy directed against CD20 is an important tool in the treatment of B cell disorders. However, variable patient response and acquired resistance remain important clinical challenges. To identify genetic factors that may influence sensitivity to treatment, the cytotoxic activity of three CD20 mAbs: rituximab; ofatumumab; and obinutuzumab, were screened in high-throughput assays using 680 ethnically diverse lymphoblastoid cell lines (LCLs) followed by a pharmacogenomic assessment. GWAS analysis identified several novel gene candidates. The most significant SNP, rs58600101, in the gene MKL1 displayed ethnic stratification, with the variant being significantly more prevalent in the African cohort and resulting in reduced transcript levels as measured by qPCR. Functional validation of MKL1 by shRNA-mediated knockdown of MKL1 resulted in a more resistant phenotype. Gene expression analysis identified the developmentally associated TGFB1I1 as the most significant gene associated with sensitivity. qPCR among a panel of sensitive and resistant LCLs revealed immunoglobulin class-switching as well as differences in the expression of B cell activation markers. Flow cytometry showed heterogeneity within some cell lines relative to surface Ig isotype with a shift to more IgG+ cells among the resistant lines. Pretreatment with prednisolone could partly reverse the resistant phenotype. Results suggest that the efficacy of anti-CD20 mAb therapy may be influenced by B cell developmental status as well as polymorphism in the MKL1 gene. A clinical benefit may be achieved by pretreatment with corticosteroids such as prednisolone followed by mAb therapy

    New Insights into Fluoroquinolone Resistance in Mycobacterium tuberculosis: Functional Genetic Analysis of gyrA and gyrB Mutations

    Fluoroquinolone antibiotics are among the most potent second-line drugs used for treatment of multidrug-resistant tuberculosis (MDR TB), and resistance to this class of antibiotics is one criterion for defining extensively drug resistant tuberculosis (XDR TB). Fluoroquinolone resistance in Mycobacterium tuberculosis has been associated with modification of the quinolone resistance determining region (QRDR) of gyrA. Recent studies suggest that amino acid substitutions in gyrB may also play a crucial role in resistance, but functional genetic studies of these mutations in M. tuberculosis are lacking. In this study, we examined twenty six mutations in gyrase genes gyrA (seven) and gyrB (nineteen) to determine the clinical relevance and role of these mutations in fluoroquinolone resistance. Transductants or clinical isolates harboring T80A, T80A+A90G, A90G, G247S and A384V gyrA mutations were susceptible to all fluoroquinolones tested. The A74S mutation conferred low-level resistance to moxifloxacin but susceptibility to ciprofloxacin, levofloxacin and ofloxacin, and the A74S+D94G double mutation conferred cross resistance to all the fluoroquinolones tested. Functional genetic analysis and structural modeling of gyrB suggest that M330I, V340L, R485C, D500A, D533A, A543T, A543V and T546M mutations are not sufficient to confer resistance as determined by agar proportion. Only three mutations, N538D, E540V and R485C+T539N, conferred resistance to all four fluoroquinolones in at least one genetic background. The D500H and D500N mutations conferred resistance only to levofloxacin and ofloxacin while N538K and E540D consistently conferred resistance to moxifloxacin only. Transductants and clinical isolates harboring T539N, T539P or N538T+T546M mutations exhibited low-level resistance to moxifloxacin only but not consistently. These findings indicate that certain mutations in gyrB confer fluoroquinolone resistance, but the level and pattern of resistance varies among the different mutations. The results from this study provide support for the inclusion of the QRDR of gyrB in molecular assays used to detect fluoroquinolone resistance in M. tuberculosis

    Listening carefully: increased perceptual acuity for species discrimination in multispecies signalling assemblages

    Communication is a fundamental component of evolutionary change because of its role in mate choice and sexual selection. Acoustic signals are a vital element of animal communication and sympatric species may use private frequency bands to facilitate intraspeciïŹc communication and identiïŹcation of conspeciïŹcs (acoustic communication hypothesis, ACH). If so, animals should show increasing rates of misclassiïŹcation with increasing overlap in frequency between their own calls and those used by sympatric heterospeciïŹcs. We tested this on the echolocation of the horseshoe bat, Rhinolophus capensis, using a classical habituation-dishabituation experiment in which we exposed R. capensis from two phonetic populations to echolocation calls of sympatric and allopatric horseshoe bat species (Rhinolophus clivosus and Rhinolophus damarensis) and different phonetic populations of R. capensis. As predicted by the ACH, R. capensis from both test populations were able to discriminate between their own calls and calls of the respective sympatric horseshoe bat species. However, only bats from one test population were able to discriminate between calls of allopatric heterospeciïŹcs and their own population when both were using the same frequency. The local acoustic signalling assemblages (ensemble of signals from sympatric conspeciïŹcs and heterospeciïŹcs) of the two populations differed in complexity as a result of contact with other phonetic populations and sympatric heterospeciïŹcs. We therefore propose that a hierarchy of discrimination ability has evolved within the same species. Frequency alone may be sufïŹcient to assess species membership in relatively simple acoustic assemblages but the ability to use additional acoustic cues may have evolved in more complex acoustic assemblages to circumvent misidentiïŹcations as a result of the use of overlapping signals. When the acoustic signal design is under strong constraints as a result of dual functions and the available acoustic space is limited because of co-occurring species, species discrimination is mediated through improved sensory acuity in the receiver
