    A framework for assessing the quality of green infrastructure in the built environment in the UK

    The advocacy argument for green infrastructure has largely been won. Policy and statutory guidance for green infrastructure planning and development exists at international, national and regional/local levels and the functions and benefits of green infrastructure interventions are well evidenced in academic literature. To support this, professional bodies and experts from the built and natural environment have produced a multitude of practice guidance on the delivery of individual green infrastructure features. And yet, examples of high quality green infrastructure in new development schemes remain difficult to find, and the success of statutory guidance to accelerate this phenomenon surprisingly ineffective to manage change within the development sector. This paper presents a new framework for the delivery of high quality green infrastructure. Consultation with stakeholders in the UK suggested that a key factor affecting the translation of green infrastructure evidence, and policy and practice guidance, into delivery through planning and development is a lack of confidence amongst practitioners regarding the qualities and characteristics of high quality green infrastructure in the built environment. The key characteristics of high quality green infrastructure, based on a review of both academic and grey literature, and extensive work with stakeholders are grouped into principles that underpin high quality green infrastructure (including the presence of a multifunctional network, and provision for long-term management), and principles related to health and wellbeing, water management and nature conservation. The resulting framework is presented as twenty three principles for delivering green infrastructure. This can be used internationally as a set of standards to assess the quality of green infrastructure to ensure that it contributes to quality of life, health and wellbeing of individuals and communities, flood resilient towns and cities, and places where nature can flourish and be more viable as a result of development

    © 2018 WIT Press Green infrastructure (GI) is globally recognised as an essential component of liveable and sustainable places. It is valued for its multifunctionality and the connectedness of the individual features to each other, the surrounding countryside and urban populations. It brings together many land uses (e.g. parks, gardens, cemeteries, allotments, nature reserves, surface water), urban design (e.g. street trees, landscaping) and functional features (e.g. sustainable urban drainage systems, green roofs) operating at differing spatial scales. It is widely acknowledged that GI is the primary mechanism for delivering ecosystem services in towns and cities, and there is a substantial body of research demonstrating the multiple benefits of GI to urban populations. Despite this evidence base, there is still considerable uncertainty about the best way to design, deliver and maintain GI. This paper presents an emerging benchmark that has been developed through a combination of literature review and engagement with key stakeholders. It provides a suite of standards that are flexible enough to be used across different spatial scales depending on the specific needs of the location, covering the form and function of GI including nature conservation, water management, health and well-being, environmental and design quality. It allows an assessment of GI policy, and the planning, design, delivery and long-term management of GI in new and existing places, ensuring that current good practice is adopted at all stages. The development of the benchmark to date is summarised along with the outcome of preliminary testing using the outline planning applications for two contrasting mixed-use developments. This found that the benchmark performed well, with standards set at a level to ensure that high-quality GI is rewarded but without requiring a level of GI provision and quality that would only be expected on truly exemplary developments.     Governance, democracy and accountability from a network governance perspective within the city of Ekurhuleni

    Abstract: Background: Local government as the sphere of government that directly impacts the lives of communities must create a milieu of interacting with communities in terms of planning, execution and the monitoring and evaluation of the success or failure of its programmes. Local government has a moral and a legislative duty to involve communities thereby extending and deepening democratic and accountable government processes. Aim: This study aimed to analyse how democratic and accountable governance can assist in creating sustainable processes of involvement of its communities. Local government is government’s main contributor to the socio-economic growth and development, not only of a country but more so, its people. The article emphasises improvement in the current processes of community involvement in strategic planning and the execution of such plans in general and at the city of Ekurhuleni (COE) in particular, situated in the Gauteng province of South Africa. It further aims to indicate to the city that its processes are not taking into account communities’ feelings of self-worth, humanity and identity. Methods: The study followed a qualitative approach in terms of the interpretative phenomenological analysis (IPA) and the network governance theory served as the theoretical framework. Results: The findings indicate that communities and community bodies who are to serve as actors in collaboration with the city in their planning and execution processes experience alienation from council, councillors and even ward committees and their members. Community actors are of the opinion that councillors only know them during their election campaigns, and when elections have come and gone, councillors are gone as well. Conclusion: Recommendations include that when dealing with community affairs there should be no political party affiliation and that councillors must know that communication is a two-way tool that should benefit all parties. Councillors are to commit themselves to the improvement of community members’ lives and socio-economic upliftment

The quality of green infrastructure often represents a missed opportunity to achieve this. This paper presents a review examining how built environment assessment systems evaluate the quality of green infrastructure. This was used to develop proposals for a new benchmark, which were examined by experts in terms of the demand, scope and operation. The findings suggest that current systems are not providing a robust assessment of green infrastructure and that a benchmark for green infrastructure would overcome some of the challenges associated with its planning, design and delivery

    Existence of long-lasting experience-dependent plasticity in endocrine cell networks

    Experience-dependent plasticity of cell and tissue function is critical for survival by allowing organisms to dynamically adjust physiological processes in response to changing or harsh environmental conditions. Despite the conferred evolutionary advantage, it remains unknown whether emergent experience-dependent properties are present in cell populations organized as networks within endocrine tissues involved in regulating body-wide homeostasis. Here we show, using lactation to repeatedly activate a specific endocrine cell network in situ in the mammalian pituitary, that templates of prior demand are permanently stored through stimulus-evoked alterations to the extent and strength of cell–cell connectivity. Strikingly, following repeat stimulation, evolved population behaviour leads to improved tissue output. As such, long-lasting experience-dependent plasticity is an important feature of endocrine cell networks and underlies functional adaptation of hormone release

    New genetic loci implicated in fasting glucose homeostasis and their impact on type 2 diabetes risk.

    Levels of circulating glucose are tightly regulated. To identify new loci influencing glycemic traits, we performed meta-analyses of 21 genome-wide association studies informative for fasting glucose, fasting insulin and indices of beta-cell function (HOMA-B) and insulin resistance (HOMA-IR) in up to 46,186 nondiabetic participants. Follow-up of 25 loci in up to 76,558 additional subjects identified 16 loci associated with fasting glucose and HOMA-B and two loci associated with fasting insulin and HOMA-IR. These include nine loci newly associated with fasting glucose (in or near ADCY5, MADD, ADRA2A, CRY2, FADS1, GLIS3, SLC2A2, PROX1 and C2CD4B) and one influencing fasting insulin and HOMA-IR (near IGF1). We also demonstrated association of ADCY5, PROX1, GCK, GCKR and DGKB-TMEM195 with type 2 diabetes. Within these loci, likely biological candidate genes influence signal transduction, cell proliferation, development, glucose-sensing and circadian regulation. Our results demonstrate that genetic studies of glycemic traits can identify type 2 diabetes risk loci, as well as loci containing gene variants that are associated with a modest elevation in glucose levels but are not associated with overt diabetes

    Identification and validation of N-acetyltransferase 2 as an insulin sensitivity gene

    Journal ArticleDecreased insulin sensitivity, also referred to as insulin resistance (IR), is a fundamental abnormality in patients with type 2 diabetes and a risk factor for cardiovascular disease. While IR predisposition is heritable, the genetic basis remains largely unknown. The GENEticS of Insulin Sensitivity consortium conducted a genome-wide association study (GWAS) for direct measures of insulin sensitivity, such as euglycemic clamp or insulin suppression test, in 2,764 European individuals, with replication in an additional 2,860 individuals. The presence of a nonsynonymous variant of N-acetyltransferase 2 (NAT2) [rs1208 (803A>G, K268R)] was strongly associated with decreased insulin sensitivity that was independent of BMI. The rs1208 "A" allele was nominally associated with IR-related traits, including increased fasting glucose, hemoglobin A1C, total and LDL cholesterol, triglycerides, and coronary artery disease. NAT2 acetylates arylamine and hydrazine drugs and carcinogens, but predicted acetylator NAT2 phenotypes were not associated with insulin sensitivity. In a murine adipocyte cell line, silencing of NAT2 ortholog Nat1 decreased insulin-mediated glucose uptake, increased basal and isoproterenol- stimulated lipolysis, and decreased adipocyte differentiation, while Nat1 overexpression produced opposite effects. Nat1-deficient mice had elevations in fasting blood glucose, insulin, and triglycerides and decreased insulin sensitivity, as measured by glucose and insulin tolerance tests, with intermediate effects in Nat1 heterozygote mice. Our results support a role for NAT2 in insulin sensitivity

    Capture of MicroRNA–Bound mRNAs Identifies the Tumor Suppressor miR-34a as a Regulator of Growth Factor Signaling

    A simple biochemical method to isolate mRNAs pulled down with a transfected, biotinylated microRNA was used to identify direct target genes of miR-34a, a tumor suppressor gene. The method reidentified most of the known miR-34a regulated genes expressed in K562 and HCT116 cancer cell lines. Transcripts for 982 genes were enriched in the pull-down with miR-34a in both cell lines. Despite this large number, validation experiments suggested that ∼90% of the genes identified in both cell lines can be directly regulated by miR-34a. Thus miR-34a is capable of regulating hundreds of genes. The transcripts pulled down with miR-34a were highly enriched for their roles in growth factor signaling and cell cycle progression. These genes form a dense network of interacting gene products that regulate multiple signal transduction pathways that orchestrate the proliferative response to external growth stimuli. Multiple candidate miR-34a–regulated genes participate in RAS-RAF-MAPK signaling. Ectopic miR-34a expression reduced basal ERK and AKT phosphorylation and enhanced sensitivity to serum growth factor withdrawal, while cells genetically deficient in miR-34a were less sensitive. Fourteen new direct targets of miR-34a were experimentally validated, including genes that participate in growth factor signaling (ARAF and PIK3R2) as well as genes that regulate cell cycle progression at various phases of the cell cycle (cyclins D3 and G2, MCM2 and MCM5, PLK1 and SMAD4). Thus miR-34a tempers the proliferative and pro-survival effect of growth factor stimulation by interfering with growth factor signal transduction and downstream pathways required for cell division
