MHC-I promotes apoptosis of GABAergic interneurons in the spinal dorsal horn and contributes to cancer induced bone pain
Cancer induced bone pain (CIBP) remains one of the most intractable clinical problems due to poor understanding of its underlying mechanisms. Recent studies demonstrate the decline of inhibitory interneurons, especially GABAergic interneurons in the spinal cord, can evoke generation of chronic pain. It has also been reported that neuronal MHC-I expression renders neurons vulnerable to cytotoxic CD8+ T cells and finally lead to neurons apoptosis in a variety neurological disorders. However, whether MHC-I could induce the apoptosis of GABAergic interneurons in spinal cord and contribute to the development of CIBP remains unknown. In this study, we investigated roles of MHC-I and underlying mechanisms in CIBP on a rat model. Our results showed that increased MHC-I expression on GABAergic interneurons could deplete GABAergic interneurons by inducing their apoptosis in the spinal dorsal horn of tumor-bearing rats. Pretreatment of MHC-I RNAi-lentivirus could prevent the apoptosis of GABAergic interneurons and therefore alleviated mechanical allodynia induced by tumor cells intratibial injection. Additionally, we also found that CD8+ T cells were colocalized with MHC-I and GABAergic neurons and presented a significant and persistent increase in the spinal cord of tumor-bearing rats. Taken together, these findings indicated that MHC-I could evoke CIBP by promoting apoptosis of GABAergic interneurons in the dorsal horn, and this apoptosis was closely related to local CD8+ T cells
Visual Tracking via Feature Tensor Multimanifold Discriminate Analysis
In the visual tracking scenarios, if there are multiple objects, due to the interference of similar objects, tracking may fail in the progress of occlusion to separation. To address this problem, this paper proposed a visual tracking algorithm with discrimination through multimanifold learning. Color-gradient-based feature tensor was used to describe object appearance for accommodation of partial occlusion. A prior multimanifold tensor dataset is established through the template matching tracking algorithm. For the purpose of discrimination, tensor distance was defined to determine the intramanifold and intermanifold neighborhood relationship in multimanifold space. Then multimanifold discriminate analysis was employed to construct multilinear projection matrices of submanifolds. Finally, object states were obtained by combining with sequence inference. Meanwhile, the multimanifold dataset and manifold learning embedded projection should be updated online. Experiments were conducted on two real visual surveillance sequences to evaluate the proposed algorithm with three state-of-the-art tracking methods qualitatively and quantitatively. Experimental results show that the proposed algorithm can achieve effective and robust effect in multi-similar-object mutual occlusion scenarios
PI3K/Akt pathway: a potential therapeutic target for chronic pain
Chronic pain is among the most disabling and costly disorders, with prevalence ranging from 10% to 55%. However, current therapeutic strategies for chronic pain are unsatisfactory due to our poor understanding of its mechanisms. Thus, novel therapeutic targets need to be found in order to improve these patients' quality of life. PI3K and its downstream Akt are widely expressed in the spinal cord, particularly in the laminae I-IV of the dorsal horn, where nociceptive C and Aδ fibers of primary afferents principally terminate. The role of spinal GABAB receptors in cancer-induced bone pain in rats
Cancer-induced bone pain (CIBP) remains a major challenge in advanced cancer patients due to our lack of understanding of its mechanisms. Previous studies have demonstrated the vital role of GABAB receptors (GABABRs) in regulating nociception and various neuropathic pain models have shown diminished activity of GABABRs. However, the role of spinal GABABRs in CIBP remains largely unknown. In this study, we investigated the specific cellular mechanisms of GABABRs in the development and maintenance of CIBP in rats. Our behavioral results show that both acute and chronic intrathecal treatment with baclofen, a GABABR agonist, significantly attenuated CIBP-induced mechanical allodynia and ambulatory pain. The expression levels of GABABRs were significantly decreased in a time-dependent manner and colocalized mostly with neuron and a minority with astrocyte and microglia. Chronic treatment with baclofen restored the expression of GABABRs and markedly inhibited the activation of cAMP-dependent protein kinase (PKA) and the cAMP-response element-binding protein (CREB) signaling pathway
Operational test of bonded magnetocaloric plates
Bonded plates made by hot pressing La0.85Ce0.15Fe11.25Mn0.25Si1.5Hy particles and resin have been tested as active magnetic regenerators in a small scale magnetocaloric device. Firstly the plates were carefully characterised magnetically and thermally. The plates were prepared with 5 wt% resin, and from density measurements it was found that the volume ratio of the magnetocaloric material in the plates was 0.53, due to the resin and porosity. The best operating conditions for the plate regenerator were determined at which a temperature span of 6.4 K was measured along the plates
A Combined SERS and MCBJ Study on Molecular Junctions on Silicon Chips
We have developed a combined Surface-enhanced raman spectroscopy (SERS) and mechanically controllable break junction (MCBJ) method to detect and characterize molecular junctions formed by two electrochemically nanofabricated electrodes on silicon chips. The method allows us to obtain vibrational spectra of the molecular junction and perform electron transport measurement on the molecules simultaneously. The preliminary IN characterization and SERS measurement on an asymmetric molecule, OPE-NO(2), and a symmetric molecule, OPE, were conducted. This approach may provide new insights into not only electron transport in molecules, but also the enhancement mechanism in single-molecule SERS

Physiological and anthocyanin biosynthesis genes response induced by vanadium stress in mustard genotypes with distinct photosynthetic activity
The present study aimed to elucidate the photosynthetic performance, antioxidant enzyme activities, anthocyanin contents, anthocyanin biosynthetic gene expression, and vanadium uptake in mustard genotypes (purple and green) that differ in photosynthetic capacity under vanadium stress. The results indicated that vanadium significantly reduced photosynthetic activity in both genotypes. The activities of the antioxidant enzymes were increased significantly in response to vanadium in both genotypes, although the purple exhibited higher. The anthocyanin contents were also reduced under vanadium stress. The anthocyanin biosynthetic genes were highly expressed in the purple genotype, notably the genes TT8, F3H, and MYBL2 under vanadium stress. The results indicate that induction of TT8, F3H, and MYBL2 genes was associated with upregulation of the biosynthetic genes required for higher anthocyanin biosynthesis in purple compared with the green mustard. The roots accumulated higher vanadium than shoots in both mustard genotypes. The results indicate that the purple mustard had higher vanadium tolerance
Observation of a near-threshold enhancement in th p pbar mass spectrum from radiative J/psi-->gamma p pbar decays
We observe a narrow enhancement near 2mp in the invariant mass spectrum of
ppbar pairs from radiative J/psi-->gamma ppbar decays. The enhancement can be
fit with either an S- or P-wave Breit Wigner fuction. In the case of the S-wave
fit, the peak mass is below the 2mp threshold and the full width is less than
30 MeV. These mass and width values are not consistent with the properties of
