    Causes and consequences of variability in marine benthos

    The last two decades have seen a shift of emphasis in ecology; from a focus on the drivers of biodiversity change toward a consideration for its effects on ecosystem functioning. Ecosystem functioning is affected by individual species (i.e. species with functionally dominant biological traits), but can also be influenced by other factors, such as interspecific interactions. Current evidence suggests that biotic influence over marine ecosystem functioning is largely underpinned by the effects of individual species. However, there are indications that this might not constitute a complete understanding of the link between marine biodiversity and ecosystem function (BEF). For this thesis, I applied our current understanding of marine BEF relationships (i.e. the causal link between particular biological traits and particular ecological functions) to long-term benthic community time series and investigated the causes of ecological change and its consequences for ecosystem functioning. A shift in the taxonomic composition of the species assemblage was explained by underlying variation in extrinsic drivers. However, the long-term conservation of trait composition suggests that functioning can be sustained in the face of environmental and ecological change. Experiments conducted to test BEF relationships in intertidal marine benthos reaffirmed the functional importance of the biological traits of species, but also showed that interactions among species can influence the delivery of ecological functions in various ways, including facilitation (i.e. function delivery is enhanced) and antagonism (i.e. function delivery is reduced). The results suggest that biotic influence over marine ecosystem functioning is more complex than previously suggested, and that the impacts of biodiversity change (e.g. species extinctions or shifts in species densities) could be either exacerbated or mitigated depending on the composition of the affected assemblage and the ecological function considered. To produce more realistic results, future indirect assessments of ecosystem functioning would benefit from incorporating interactions among species as well as their biological traits

    An approach for assessing and ranking fisheries management scenarios in spatially delimited marine areas

    Spatial restrictions to human activities such as bottom trawling are increasingly used to improve the ecological condition of disturbed habitats. Such management interventions typically have socio-economic consequences, which creates a challenge for those making decisions about which activities should be restricted and where restrictions should apply. We present an approach for predicting the effects of fisheries management scenarios in spatially delimited marine areas and ranking them—using a loss function—according to how well they achieve desired outcomes across a set of ecological and socio-economic indicators. This approach is demonstrated by simulating alternative fishing gear restrictions and zoning options within a hypothetical marine protected area (MPA). Relative benthic status (RBS; an indicator of ecological condition) and relative catch value (RCV; an indicator of potential economic cost) were estimated for the baseline environment and 21 potential management scenarios. The rank order depended on which indicator was prioritized (i.e. whether RBS or RCV was given greater weighting in the loss function), with the top-ranked scenarios in each case involving considerably different management measures. The methods presented can be applied anywhere using locally or strategically relevant indicators to help identify spatial fisheries management measures that minimize ecological and socio-economic trade-offs

    Change in glomerular filtration rate over time in the Oxford Renal Cohort Study:observational study

    Background: Decline in kidney function can result in adverse health outcomes. The Oxford Renal Cohort Study has detailed baseline assessments from 884 participants ≄60 years of age. Aim: To determine the proportion of participants with a decline in estimated glomerular filtration rate (eGFR), identify determinants of decline, and determine proportions with chronic kidney disease (CKD) remission. Design and setting: Observational cohort study in UK primary care. Method: Data were used from baseline and annual follow-up assessments to monitor change in kidney function. Rapid eGFR decline was defined as eGFR decrease &gt;5 ml/min/1.73 m2/year, improvement as eGFR increase &gt;5 ml/min/1.73 m2/year, and remission in those with CKD at baseline and eGFR &gt;60 ml/min/1.73 m2 during follow-up. Cox proportional hazard models were used to identify factors associated with eGFR decline. Results: There was a net decline in eGFR in the 884 participants over 5 years of follow-up. In 686 participants with &gt;2 eGFR tests with a median follow-up of 2.1 years, 164 (24%) evidenced rapid GFR decline, 185 (27%) experienced eGFR improvement, and 82 of 394 (21%) meeting CKD stage 1-4 at baseline experienced remission. In the multivariable analysis, smoking status, higher systolic blood pressure, and being known to have CKD at cohort entry were associated with rapid GFR decline. Those with CKD stage 3 at baseline were less likely to exhibit GFR decline compared with normal kidney function. Conclusion: This study established that 24% of people evidenced rapid GFR decline whereas 21% evidenced remission of CKD. People at risk of rapid GFR decline may benefit from closer monitoring and appropriate treatment to minimise risks of adverse outcomes, although only a small proportion meet the National Institute for Health and Care Excellence criteria for referral to secondary care.</p

    In Vitro Oxidative Crosslinking of Recombinant Barnacle Cyprid Cement Gland Proteins

    Barnacle adhesion is a focus for fouling-control technologies as well as the development of bioinspired adhesives, although the mechanisms remain very poorly understood. The barnacle cypris larva is responsible for surface colonisation. Cyprids release cement from paired glands that contain proteins, carbohydrates and lipids, although further compositional details are scant. Several genes coding for cement gland-specific proteins were identified, but only one of these showed database homology. This was a lysyl oxidase-like protein (lcp_LOX). LOX-like enzymes have been previously identified in the proteome of adult barnacle cement secretory tissue. We attempted to produce recombinant LOX in E. coli, in order to identify its role in cyprid cement polymerisation. We also produced two other cement gland proteins (lcp3_36k_3B8 and lcp2_57k_2F5). lcp2_57k_2F5 contained 56 lysine residues and constituted a plausible substrate for LOX. While significant quantities of soluble lcp3_36k_3B8 and lcp2_57k_2F5 were produced in E. coli, production of stably soluble lcp_LOX failed. A commercially sourced human LOX catalysed the crosslinking of lcp2_57k_2F5 into putative dimers and trimers, and this reaction was inhibited by lcp3_36k_3B8. Inhibition of the lcp_LOX:lcp2_57k_2F5 reaction by lcp3_36k_3B8 appeared to be substrate specific, with no inhibitory effect on the oxidation of cadaverine by LOX. The results demonstrate a possible curing mechanism for barnacle cyprid cement and, thus, provide a basis for a more complete understanding of larval adhesion for targeted control of marine biofouling and adhesives for niche applications

    Prediction of 7-year psychopathology from mother-infant joint attention behaviours: a nested case–control study

    &lt;br&gt;Background: To investigate whether later diagnosis of psychiatric disorder can be predicted from analysis of mother-infant joint attention (JA) behaviours in social-communicative interaction at 12 months.&lt;/br&gt; &lt;br&gt;Method: Using data from a large contemporary birth cohort, we examined 159 videos of a mother-infant interaction for joint attention behaviour when children were aged one year, sampled from within the Avon Longitudinal Study of Parents and Children (ALSPAC) cohort. Fifty-three of the videos involved infants who were later considered to have a psychiatric disorder at seven years and 106 were same aged controls. Psychopathologies included in the case group were disruptive behaviour disorders, oppositional-conduct disorder, attention-deficit/hyperactivity disorder, pervasive development disorder, anxiety and depressive disorders. Psychiatric diagnoses were obtained using the Development and Wellbeing Assessment when the children were seven years old.&lt;/br&gt; &lt;br&gt;Results: None of the three JA behaviours (shared look rate, shared attention rate and shared attention intensity) showed a significant association with the primary outcome of case–control status. Only shared look rate predicted any of the exploratory sub-diagnosis outcomes and was found to be positively associated with later oppositional-conduct disorders (OR [95% CI]: 1.5 [1.0, 2.3]; p = 0.041).&lt;/br&gt;&lt;br&gt;Conclusions: JA behaviours did not, in general, predict later psychopathology. However, shared look was positively associated with later oppositional-conduct disorders. This suggests that some features of JA may be early markers of later psychopathology. Further investigation will be required to determine whether any JA behaviours can be used to screen for families in need of intervention.&lt;/br&gt

    Vascular responses of the extremities to transdermal application of vasoactive agents in Caucasian and African descent individuals

Purpose: Individuals of African descent (AFD) are more susceptible to non-freezing cold injury than Caucasians (CAU) which may be due, in part, to differences in the control of skin blood flow. We investigated the skin blood flow responses to transdermal application of vasoactive agents. Methods: Twenty-four young males (12 CAU and 12 AFD) undertook three tests in which iontophoresis was used to apply acetylcholine (ACh 1 w/v %), sodium nitroprusside (SNP 0.01 w/v %) and noradrenaline (NA 0.5 mM) to the skin. The skin sites tested were: volar forearm, non-glabrous finger and toe, and glabrous finger (pad) and toe (pad). Results: In response to SNP on the forearm, AFD had less vasodilatation for a given current application than CAU (P = 0.027–0.004). ACh evoked less vasodilatation in AFD for a given application current in the non-glabrous finger and toe compared with CAU (P = 0.043–0.014) with a lower maximum vasodilatation in the non-glabrous finger (median [interquartile], AFD n = 11, 41[234] %, CAU n = 12, 351[451] %, P = 0.011) and non-glabrous toe (median [interquartile], AFD n = 9, 116[318] %, CAU n = 12, 484[720] %, P = 0.018). ACh and SNP did not elicit vasodilatation in the glabrous skin sites of either group. There were no ethnic differences in response to NA. Conclusion: AFD have an attenuated endothelium-dependent vasodilatation in non-glabrous sites of the fingers and toes compared with CAU. This may contribute to lower skin temperature following cold exposure and the increased risk of cold injuries experienced by AFD.Published versio

    Climate forcing of an emerging pathogenic fungus across a montane multi-host community

    Changes in the timings of seasonality as a result of anthropogenic climate change are predicted to occur over the coming decades. While this is expected to have widespread impacts on the dynamics of infectious disease through environmental forcing, empirical data are lacking. Here, we investigated whether seasonality, specifically the timing of spring ice-thaw, affected susceptibility to infection by the emerging pathogenic fungus Batrachochytrium dendrobatidis (Bd) across a montane community of amphibians that are suffering declines and extirpations as a consequence of this infection. We found a robust temporal association between the timing of the spring thaw and Bd infection in two host species, where we show that an early onset of spring forced high prevalences of infection. A third highly susceptible species (the midwife toad, Alytes obstetricans) maintained a high prevalence of infection independent of time of spring thaw. Our data show that perennially overwintering midwife toad larvae may act as a year-round reservoir of infection with variation in time of spring thaw determining the extent to which infection spills over into sympatric species. We used future temperature projections based on global climate models to demonstrate that the timing of spring thaw in this region will advance markedly by the 2050s, indicating that climate change will further force the severity of infection. Our findings on the effect of annual variability on multi-host infection dynamics show that the community-level impact of fungal infectious disease on biodiversity will need to be re-evaluated in the face of climate change

    Molecular diagnosis of bird-mediated pest consumption in tropical farmland

    Biodiversity loss will likely have surprising and dramatic consequences for human wellbeing. Identifying species that benefit society represents a critical first step towards predicting the consequences of biodiversity loss. Though natural predators prevent billions of dollars in agricultural pest damage annually, characterizing which predators consume pests has proven challenging. Emerging molecular techniques may illuminate these interactions. In the countryside of Costa Rica, we identified avian predators of coffee’s most damaging insect pest, the coffee berry borer beetle (Coleoptera:Scolytidae Hypothenemus hampeii), by assaying 1430 fecal samples of 108 bird species for borer DNA. While feeding trials confirmed the efficacy of our approach, detection rates were low. Nevertheless, we identified six species that consume the borer. These species had narrow diet breadths, thin bills, and short wings; traits shared with borer predators in other systems. Borer predators were not threatened; therefore, safeguarding pest control necessitates managing species beyond those at risk of regional extinction by maintaining populations in farmland habitats. Generally, our results demonstrate potential for pairing molecular methods with ecological analyses to yield novel insights into species interactions.     Wettability decay in an oil-contaminated waste-mineral mixture with dry-wet cycles

    The dependency of soil particle wettability on soil water content implies that soils subjected to drying-wetting cycles become wettable with wetting and water repellent with drying. While this has been demonstrated widely, the results are contradictory when water repellent soils are subjected to a sequence of cycles. Added to this, past wettability measurements were seldom done in batches of samples collected from the field at natural or dry water contents, with little appreciation that slight particle size variations, different drying-wetting histories and fabric (as required by different wettability measurement methods) may alter the results. This note presents soil particle wettability—soil water content relations by means of an index test following staged drying and wetting paths over a period of 8 months for an untreated, oil-contaminated anthropogenic soil (a mixture of slag, coal particles, fly ash and mineral particles) from Barry Docks (UK), a site formally used for oil storage, which is to be remediated and redeveloped for housing. The results revealed a decrease in the water repellency and increasing mineralization and bacterial activity with the wetting and drying cycles.postprin
