    Pharmacological Blockade of the Calcium Plateau Provides Neuroprotection Following Organophosphate Paraoxon Induced Status Epilepticus in Rats

    Organophosphate (OP) compounds which include nerve agents and pesticides are considered chemical threat agents. Currently approved antidotes are crucial in limiting OP mediated acute mortality. However, survivors of lethal OP exposure exhibit delayed neuronal injury and chronic behavioral morbidities. In this study, we investigated neuroprotective capabilities of dantrolene and carisbamate in a rat survival model of paraoxon (POX) induced status epilepticus (SE). Significant elevations in hippocampal calcium levels were observed 48-h post POX SE survival, and treatment with dantrolene (10 mg/kg, i.m.) and carisbamate (90 mg/kg, i.m.) lowered these protracted calcium elevations. POX SE induced delayed neuronal injury as characterized by Fluoro Jade C labeling was observed in critical brain areas including the dentate gyrus, parietal cortex, amygdala, and thalamus. Dantrolene and carisbamate treatment provided significant neuroprotection against delayed neuronal damage in these brain regions when administered one-hour after POX-SE. These results indicate that dantrolene or carisbamate could be effective adjuvant therapies to the existing countermeasures to reduce neuronal injury and behavioral morbidities post OP SE survival

    Developing inclusive digital health diagnostic for schistosomiasis: a need for guidance via target product profiles

    IntroductionThe INSPIRED project aims to develop inclusive Digital Optical Diagnostic Devices (DODDs) for schistosomiasis, to support disease management by enabling rapid diagnostic results, to improve efficient data management to guide decision-making and to provide healthcare workers with critical health information to facilitate follow-up action. Due to the non-availability of Target Product Profiles (TPPs) for guiding the development of digital diagnostics for schistosomiasis, we explored existing diagnostic TPPs.MethodsUsing a curated open access database (Notion database), we studied a selection of TPPs for diagnosing infectious diseases, focusing on specifications related to digital health products for Neglected Tropical Diseases (NTDs).ResultsEighteen TPPs originating from 12 documents, covering 13 specific diseases, were selected and their characteristics were labeled and entered into the database. Further exploration of the database revealed several gaps, including a lack of stakeholder input, sustainability, and TPP availability. Other significant gaps related to digital health platform interconnectivity and data stewardship specifically in relation to digital diagnostics, including DODDs.DiscussionThese findings reflect two possible scenarios: (1) there is currently no need for digital diagnostic devices for schistosomiasis and, by extension for other NTDs; or (2) those needs are not yet covered by TPPs. Therefore, we recommend that digital health diagnostics are included in the use cases for schistosomiasis control and elimination, at least in the ideal/desirable scenario, as this will guide research and incentivize investment in digital health diagnostics for schistosomiasis

The sequence of the P gene showed 97% identity with the Dutch virus isolated in 1988. There was 100% identity with the Dutch isolate from 2002 and a single nucleotide mismatch with the Danish isolate

    Longevity and relationships with children: the importance of the parental role

    <p>Abstract</p> <p>Background</p> <p>Social networks predict longevity across societies but specific mechanisms are largely unknown. The aim of this work was to examine the role of children in the longevity of elderly men and women in a cohort of community dwelling elderly people in Spain.</p> <p>Methods</p> <p>The data were taken from the "Aging in Leganes" cohort study with 15 years of follow-up. The baseline population was an age- and sex-stratified random sample of community dwelling people over 65 living in Leganés (Madrid) in 1993. Poor relationship with at least one child, emotional support and the perceived roles elders play in the lives of their children, extended family, spouse and friends were assessed at baseline. Cox proportional hazards models were fit to investigate the effects of social roles variables on longevity, adjusting for a wide range of socioeconomic, behavioural and health covariates.</p> <p>Results</p> <p>In the fully adjusted model, having a poor relationship with at least one child increased mortality by 30%. Elderly persons who felt their role in their children's lives was important (HR = 0.70; 95% CI 0.54; 0.91) had a lower mortality risk than those who felt they played a small role. Feeling loved and listened to by one's children did not have an effect on survival. Maintaining an important role in the extended family was also significantly associated with survival.</p> <p>Conclusion</p> <p>In this Mediterranean population, maintaining an important role in the lives of one's children is associated with survival. Functions of social networks related to meaning of life and different forms of social support may have important effects on mortality, and these functions may vary across cultures according to family norms and values.</p

    Concurrency Models with Causality and Events as Psi-calculi

    Psi-calculi are a parametric framework for nominal calculi, where standard calculi are found as instances, like the pi-calculus, or the cryptographic spi-calculus and applied-pi. Psi-calculi have an interleaving operational semantics, with a strong foundation on the theory of nominal sets and process algebras. Much of the expressive power of psi-calculi comes from their logical part, i.e., assertions, conditions, and entailment, which are left quite open thus accommodating a wide range of logics. We are interested in how this expressiveness can deal with event-based models of concurrency. We thus take the popular prime event structures model and give an encoding into an instance of psi-calculi. We also take the recent and expressive model of Dynamic Condition Response Graphs (in which event structures are strictly included) and give an encoding into another corresponding instance of psi-calculi. The encodings that we achieve look rather natural and intuitive. Additional results about these encodings give us more confidence in their correctness

    The impact of family structure and disruption on intergenerational emotional exchange in Eastern Europe

    Demographic trends across Europe involve a decrease in fertility and mortality rates, and an increase in divorce and stepfamily formation. Life courses and living arrangements have become less standardized and the structure of families has changed. In this article, we examine to what extent contemporary family structure and composition resulting from demographic changes affect emotional exchange between children and their parents, both from adult child to parent and from parent to child. Because the general level of well-being has been shown to be lower in Eastern Europe, thereby potentially affecting emotional exchange within families, we focus our research on Eastern Europe. We use the “conservation of resources theory” to derive hypotheses on how family structure may affect intergenerational emotional exchange. Family ties are assumed to be important resources of affection that people want to obtain and retain throughout their lives. Data from the Generations and Gender Survey (GGS) are used to test our hypotheses. In general, our data offer more support for the idea that families are resilient than for the often heard assumption that families are in decline as a consequence of the changed family structure and composition

    Declarative event based models of concurrency and refinement in psi-calculi

    AbstractPsi-calculi constitute a parametric framework for nominal process calculi, where constraint based process calculi and process calculi for mobility can be defined as instances. We apply here the framework of psi-calculi to provide a foundation for the exploration of declarative event-based process calculi with support for run-time refinement. We first provide a representation of the model of finite prime event structures as an instance of psi-calculi and prove that the representation respects the semantics up to concurrency diamonds and action refinement. We then proceed to give a psi-calculi representation of Dynamic Condition Response Graphs, which conservatively extends prime event structures to allow finite representations of (omega) regular finite (and infinite) behaviours and have been shown to support run-time adaptation and refinement. We end by outlining the final aim of this research, which is to explore nominal calculi for declarative, run-time adaptable mobile processes with shared resources

    Tratamento do câncer de próstata hormônio refratário atual e suas inovações: Revisão de literatura / Treatment of current prostate hormone refractory cancer and its innovations: Literature review

    Objetivo: Propôs-se realizar com esse estudo uma revisão de literatura a respeito do avanço do tratamento do câncer de próstata hormônio refratário, analisando artigos de 2015-2020. Método: todos os dados contidos no trabalho, foram selecionados mediante prévia revisão da literatura, utilizando os seguintes descritores: câncer de prostáta; tratamento; terapia de privação hormonal; câncer de próstata avançado; abiraterona; docetaxel; prostate cancer; treatment; advanced prostate cancer; androgen deprivation therapy; abiraterone , tomando como base artigos levantados nos últimos anos publicados por meio do Pubmed e da Scielo. Como critério de inclusão, foram estabelecidos artigos publicados na íntegra, nos idiomas português e inglês, publicados no período de 2015 a 2020, utilizando os descritores citados anteriormente. Como critério de exclusão, foram retirados artigos que estavam fora do período estudado e os que não tratavam de humanos. Resultados: para realização dessa revisão bibliográfica, foi encontrado um total de 231 artigos científicos, dos quais apenas 5 foram selecionados de acordo com os critérios de inclusão. Conclusão: foi observado que os homens com câncer de próstata localizado ou avançado se beneficiam do uso da abiraterona adjunto com a terapia de privação hormonal quando comparado com ela isolada ou junta ao docetaxel.
